摘要
〔目的〕探讨二甲基甲酰胺 (N ,N -dimethylformamide ,DMF)诱发的人类外周血细胞DNA断裂损伤情况。〔方法〕单细胞凝胶电泳法 (SingleCellGelElectropheresis ,SCGE)。〔结果〕接触DMF的作业工人与对照组相比 (t检验 )各指标间均有显著差异 (P <0 .0 1)。接触DMF的作业工人的 2年 -3年组、4年 -5年组、6年 -7年组与对照相比 (方差分析 )各指标间均有显著性差异 (P <0 .0 1) ,其中彗星长、尾长、Olive尾矩、尾DNA 总DNA(%)四个指标的 6年 -7年组与 2年-3年组间有显著性差异 (P <0 .0 5 )。〔结论〕长期接触DMF可引起DNA断裂损伤 ,断裂损伤程度随着接触年限的延长有加重的趋势。
Objective To investigate whether N,N-dimethyformamide (DMF) can cause DNA strand breakage of human peripheral blood cells.〔Methods〕 Single cell gel electrophoresis (SCGE) was used.〔Results〕There were significant differences in the 5 main parameters in the workers who had a long-term contact with DMF as compared with the controls ( P <0.01).Marked differences were found in the comet extent,tail extent,Olive tail moment and tail DNA/total DNA (%) between the workers contacting with DMF for 6-7 years and the controls ( P <0.05).〔Conclusions〕Long-term contact with DMF can cause DNA strand breakage.The extent of the breakage increases along with time prolongation in people having contacting with DMF.
