摘要
目的:探讨在高浓度胰岛素条件下,体外培养的兔视网膜Müller细胞的血管内皮细胞生长因子(vas-cularendothelialgrowthfactor,VEGF)的变化。方法:在不同浓度胰岛素条件下(4,8,12kU/L),体外培养兔视网膜Müller细胞,采用免疫细胞化学法、原位杂交法,定性测定Müller细胞分泌VEGF的变化,采用ELISA方法,定量测定Müller细胞分泌VEGF的变化。结果:高浓度胰岛素能明显增强VEGF的表达(P< 0.05)。结论:高浓度胰岛素可能通过刺激Müller细胞编码VEGF基因的转录,进而增强VEGF蛋白的表达,而在糖尿病视网膜病变的新生血管生成中发挥重要作用。
AIM: To study the effect of high concentration insulin on the expression of vascular endothelial growth factor (VEGF) in cultured rabbit retinal Müller cells in vitro .· METHODS: Müller cells were cultured with insulin of different concentrations (4×103 , 8×103 , 12×103 U/L). Immunocytochemistry,in situ hybridization and ELISA were conducted to assay the expression of VEGF in cultured Müller cells in vitro at different insulin concentrations qualitatively and quantitatively.· RESULTS: VEGF expression was enhanced obviously by high concentration of insulin.· CONCLUSION: Insulin plays an important role in neovascularization of diabetic retinopathy by stimulating the transcription of the VEGF gene in Müller cells so as to enhance the expression of VEGF protein.·
