摘要 对影响根癌农杆菌介导的甘薯离体遗传转化的若干因素进行了研究.结果表明:甘薯茎段外植体在浓度为OD600=0 4的菌液中浸染4min、预培养3d、共培养3d并在共培养基中加入6mg·L-1硝酸银,或25mg·L-1乙酰丁香酮,并将培养基pH值从5 8下调至pH5 0等措施均能显著提高其遗传转化频率,而菌液中加入乙酰丁香酮、在预培养基中加入硝酸银等措施均不利于甘薯遗传转化. Several factors affecting transformation of sweet potato by Agrobacterium tumefaciens were studied in this paper, The results showed that the best parameters are: the value of OD_(600) was 0.4, time for infection was 4 min, time for pre-culture and co-culture were respectively 3 days, the best suitable concentration of AgNO_3 and acetosyringone in co-culture medium were 6 mg·L^(-1) and 25 mg·L^(-1) respectively when the pH in co-medium was 5.0. On this condition, the frequency of Kan^r shoots production was extremely increased. But, the frequency will be decreased when added AgNO_3 in pre-culture medium or added acetosyringone in bacterium solution. So, a simple and effective transformation system in sweetpotato had been developed.
