摘要
目的 探讨微血管内皮细胞 (MVEC)在体外培养中形成血管样结构的细胞外基质条件 ,为体外组织工程血管化提供实验基础。方法 在纤维蛋白原浓度为 2 g/L的纤维蛋白凝胶中加入不同浓度的血管内皮细胞生长因子 165 (VEGF165) ,进行MVEC的立体培养 ,观察有无血管样结构形成。结果 在VEGF165为 10 μg/L组和 2 0 μg/L组中 ,MVEC在立体培养中形成血管样结构。VEGF165为 2 0 μg/L组比VEGF165为 10 μg/L组形成血管样结构的时间更早、结构更长。VEGF165为 5 μg/L组和对照组均无血管样结构形成 ,但可见细胞团聚。 结论 VEGF165浓度为 2 0μg/L或 10 μg/L时 ,MVEC在纤维蛋白凝胶立体培养中可形成血管样结构。VEGF165诱导形成血管样结构的长度与剂量有依赖关系。
Objective To study the conditions of extracellular matrix (ECM) for the formation of tubular structure induced by VEGF 165 in the three-dimensional (3D) culture of microvessel endothelial cells (MVEC) in vitro.Methods MVEC were cultivated in 3D fibrin-gels (containing fibrinogen at final concentration of 2 g/L) with different final concentrations of VEGF 165,then tubular structures of MVEC were observed.Results VEGF 165 induced MVEC to form tubular structures when the final concentration of VEGF 165 was at 10 μg/L or 20 μg/L.The tubular structures in the 20 μg/L VEGF group appeared earlier and grew longer in the length than the 10 μg/L VEGF group.When the final concentration of VEGF 165 was at 0 μg/L (controls) or 5 μg/L,no tubular structure appeared,but the gathering together of MVEC could be seen.Conclusion At a final concentration of 10 μg/L or 20 μg/L of VEGF 165,VEGF 165 can induce the formation of tubular structures of MVEC in 3D fibrin-gels.The length of tubular structures was dependent on the dosages of VEGF 165.
