摘要
用HE染色、免疫组织化学及聚丙烯酰胺凝胶圆盘电泳(PAGE)等技术,观察了服用雷公藤多甙(GTW)的雄性大鼠睾丸的细胞形态学与生精细胞中乳酸脱氢酶同工酶C_4(LDH-C_4)的活性变化。结果表明,大鼠服药每日30mg/kg,35d时部分曲细精管上皮已出现异常,60d,睾丸生殖上皮发生退行性变,并见大量多核巨细胞;80d,除支持细胞外,生精细胞几乎全部消失,但间质细胞高度增生。PAGE结果显示服药后,睾丸LDH—C_4含量下降,免疫组化染色曲细精管中残留的生精细胞、多核巨细胞及畸形精子的LDH-C_4仍呈阳性反应。服药35d,与雌鼠合笼,未孕。结果提示GTW具有抗生精作用,但不影响生精细胞合成LDH-C_4。
The treated Wistar male rats, body weight between 180 and 220g, were given GTW with Tween 80 solvent by gavage, 30mg/kg perday, 6 days a week for 35, 60 and 80 days. The control group was given Tween 80 solvent only. The animals of each group were sacrificed respectively. Studies were undertaken to observe the changes of the morphology of testis an 1 epididymides with HE stain, and activity of the testioular LDH-C4 with immunohistoohemistry and Disco-electroresis.Male rats antifertility effect were also observed. The results suggest that the spermatogenio cells were damaged and all the treated rats became infertility 35 days after treatment. Many multinuoleated giant cells appeared at 60 days of treatment, and spermatogenio cells almost disapeared at 80 days. However the drug did not significantly affect LDH-C4 activity and not inhibit spermatogenio cells synthesizing LDH-C4.
