破骨细胞分化因子mRNA在大鼠正畸牙槽骨改建过程中的表达和分布 被引量:6

摘要 目的 :观察大鼠正畸牙槽骨改建过程中ODFmRNA表达和分布变化情况 ,探讨ODF与牙槽骨改建和破骨细胞形成的关系。方法 :建立SD大鼠磨牙移动实验模型。于牙齿移动 1、3、5、7d后取材分别进行ODFmRNA原位杂交染色。结果 :在正常牙周组织中 ,ODFmRNA呈弱阳性主要表达于牙槽骨表面的成骨细胞和少量牙周膜成纤维细胞胞浆中 ,分布比较均匀。加力 3d后 ,压力侧牙槽骨表面可见大量多核破骨细胞及骨吸收陷窝 ,此时ODFmRNA的表达显著增强 ,主要分布于牙槽骨表面的成骨细胞、靠近牙槽骨一侧的血管周围的牙周膜成纤维细胞以及牙槽骨骨髓腔内的成骨细胞。 5d后 ,ODFmRNA表达和分布情况与第 3d相似 ;7d后 ,牙槽骨表面成骨细胞仍呈ODFmRNA阳性表达 ,但血管周围阳性细胞数明显减少。结论 :ODFmRNA的表达变化与牙槽骨改建中破骨细胞的生命过程密切相关 ,是破骨细胞来源。 AIM:To observe the change of ODF mRNA expression and distribution during experimental tooth movement in rats,and to learn about the relationship between ODF and osteoclast recruitment on orthodontic alveolar bone remodeling.METHODS:Established SD rat molar movement model. In situ hybridization analyses were performed at 1、3、5、7 d after application orthodontic force.RESULTS:The expression of ODF mRNA were weak in normal periodontium and localized mainly in cytoplasm of osteoblasts and only a few periodontal ligament fibroblasts near the alveolar bone surface,and most of periodontal ligament fibroblasts were ODF mRNA- negative responses.On day 3 after tooth movement,the expression of ODF mRNA was marked increased on the pressure side,localized in osteoblasts near the alveolar surface,periodontal ligament fibroblasts of perivascular areas on the alveolar side and osteoblasts in bone marrow cavity. The reaction were more intense on the cells on the alveolar side than on the dental root side. At the same time,a great deal of multinuclear osteoclasts appearanced on the surface of the alveolar bone.CONCLUSION:It was clear that the localization of ODF mRNA expression was concordant with the appearance of osteoclasts. ODF play an important role in the formation and recruitment of osteoclasts during orthodontic tooth movement.
