摘要
以不同籼稻品种的成熟胚为研究材料,通过对比实验,结果表明,采用DNB为诱导愈伤组织培养基,DMS与DNB培养基交替继代,籼稻愈伤组织诱导率高、质量较好;采用MSRc3(MS+casaminoacids500mg·L-1+glutamine300mg·L-1+proline500mg·L-1+mannitol36 4g·L-1+maltose30g·L-1+TDZ0 2mg·L-1+phytagel5g·L-1pH5 8)为分化培养基,愈伤组织不定芽分化率在90%以上,较对照MSR1提高300%~500%。
By the comparsion test,the paper reports the effect of media on the embryogenic callus induction rate, and its quality and the adventitious buds differentiation frequency in the mature embryo of indica rice. By using DNB as the inducing medium, DMS and DBN as the alternative medium of subculture,we obtained the high quality and high inducing rate calli. The shoots differentiation rate of the callus cultured in differentiation mdeium (MSR^c_3 MS+casamino acids 500mg·L^(-1)+glutamine 300mg·L^(-1)+proline 500mg·L^(-1)+mannitol 36.4g·L^(-1)+maltose 30g·L^(-1)+TDZ 0.2mg·L^(-1)+phytagel 5g·L^(-1) pH5.8) was over 90%, it is 300%-500%as higher as contral(MSR_1).
