转化生长因子-β及其受体在人类IgA肾病的表达

摘要 目的 探讨转化生长因子 (TGF) β及其受体 (TGF βR)在人类IgA肾病及正常肾组织中表达的差异。方法 采用逆转录聚合酶链反应 (RT PCR)及表达丰度定量法检测IgA肾病组和正常对照组肾组织中TGF β1~ 3 、TGF βRⅠ~Ⅲ的mRNA表达。 结果 IgA肾病组TGF β1的表达明显高于正常对照组 (P <0 .0 5 ) ,而TGF βRⅠ的表达在IgA肾病组明显降低 (P <0 .0 5 )。两组间TGF β2 及TGF βRⅡ、Ⅲ表达的差异无显著性。结论 IgA肾病中 ,TGF β促进系膜细胞增殖及促进细胞外基质蛋白合成和沉积的作用可能是通过非经典通道实现的 ,其生长抑制作用却因TGF βRⅠ的低表达而丧失 ,同时TGF βRⅠ表达的降低又反馈性地刺激TGF β1过度表达。而TGF β2、3 及其受体的表达可能与IgA肾病无密切关系。 Objective To study the gene expression of transforming growth factor(TGF) β and TGF β receptor in human IgA nephropathy tissues. Methods TGF β and TGF βR mRNA were analyzed by RT PCR in 18 IgA nephropathy tissues and 16 normal tissuses as control which were collected during operation and through biopsy respectively. Results The TGF β 1 gene expression in IgA nephropathy was higher ( P <0.05) and the expression of the TGF βRⅠwas lower than those in the normal tissue ( P <0.05) respectively. But the gene expressions of TGF β 2 ,TGF β 3 and their receptors showed no significant difference. Conclusion TGF β 1 stimulates mesangial cell fibrogenesis through non classical channel. The decrease of gene expression of the TGF βRⅠcauses TGF β 1 to lose its function of growth inhibition in IgA nephropathy.
