摘要
目的 观察黄连解毒汤有效部位对培养神经细胞谷氨酸兴奋毒性的保护作用。方法 体外培养新生大鼠皮层神经细胞 ,加入谷氨酸观察谷氨酸对神经细胞的兴奋毒性及黄连解毒汤有效部位的保护作用。结果 黄连解毒汤有效部位 30 5mg kg能明显增高模型细胞的活性 ;6 10、30 5、15 3mg kg能显著降低模型细胞乳酸脱氢酶 (LDH)漏出率 ,降低MDA生成 ;30 5、15 3mg kg可减少NO生成 ,6 10mg kg还可提高SOD活性。结论 黄连解毒汤有效部位对培养神经细胞谷氨酸兴奋毒性具有显著的保护作用 ,其机制可能与降低NO含量、抗过氧化作用有关。
Objective To observe the protective effects of the active fraction of Huanglianjiedu Decoction (AFHD) against glutacid-induced cytotoxicity to nerve cells in culture.Method New-born rat cortical neurons were cultivated in vitro, and glutacid and AFHD were added into the culture medium during the cultivation, in order to observe glutacid-induced cytotoxicity to cortical neurons and the protective effects of AFHD against the cytotoxicity.Results It was found that AFHD could markedly increase the activity of the model cells at the dose of 305 mg·kg -1 ; markedly decrease the leakage rate of LDH and production of MDA at the doses of 610 mg·kg -1 , 305 mg·kg -1 and 153 mg·kg -1 ; reduce the production of NO at the doses of 305 mg·kg -1 and 153 mg·kg -1 ; and increase the activity of SO, at the dose of 610 mg·kg -1 . Conclusion AFHD has a remarkable protective effect against glutacid-induced cytotoxicity to cortical neurons, the mechanism of which may be related with its decreasing the level of NO and resisting oxidation.
