摘要
目的 分析阻塞性睡眠呼吸暂停低通气综合征(OSAHS)患者瘦素受体基因Gln223Arg各基因型的分布特征和等位基因频率,探讨其基因多态性与OSAHS肥胖患者的易感性之间的关系.方法 选取经PSG监测的男性OSAHS患者为病例组(183例),其中肥胖OSAHS组(A组,90例),非肥胖OSAHS组(B组,93例);同期查体的男性健康人为对照组(201例),其中肥胖对照组(C组,105例)及正常对照组(D组,96例).用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法检测瘦素受体基因Gln223Arg的基因型并计算出等位基因频率.结果 4组Gln223Arg的基因型差异无统计学意义(x2=2.462,P=0.873);4组Gln223Arg等位基因频率比较差异无统计学意义(x2=0.617,P=0.893),但Gln223Arg中GG基因型患者颈围大于AA/AG患者颈围[(41.66±2.87) cm与(39.93±3.00) cm,t=3.045,P=0.042).结论 Gln223Arg变异可能参与调节OSAHS患者颈部脂肪的分布,但未发现瘦素受体基因Gln223Arg基因多态性与OSAHS肥胖患者发病相关.
Objective To analyze genotype distribution and allele frequency of leptin receptor gene Gln223Arg in obese patients with obstructive sleep apnea hypopnea syndrome (OSAHS) and discuss the relationship between the gene polymorphism and OSAHS.Methods A total of 384 male were monitored by polysomnography graph (PSG).They were divided into OSAHS group (183 cases) and control group (201 cases).patients in OSAHS group included 90 with obese OSAHS and 93 with non-obese OSAHS.The Leptin receptor gene Gln223Arg and genotypes were determined by using Polymerize chain reaction-Restriction fragment length polymorphism(PCR-RLFP).Results Gln223Arg genotype and allele frequency comparative in four groups was no statistically significant(x2 =2.462,P =0.873;x2 =0.617,P =0.893).But Neck circumference ((41.66±2.87) cm) of GG genotype patients in Gln223Arg was higher than that of AA/AG patients((39.93±3.00) cm,t =3.045,P =0.042).Conclusion Gln223Arg variation may be involved in regulating OSAHS patients neck fat distribution.There is no relations between the gene polymorphism in leptin receptor gene Gln223Arg and the onset of OSAHS.
