The integrity of retinal ganglion cells is tightly associated with diabetic macular degeneration that leads to damage and death of retinal ganglion cells,affecting vision.The major clinical treatments for diabetic mac...The integrity of retinal ganglion cells is tightly associated with diabetic macular degeneration that leads to damage and death of retinal ganglion cells,affecting vision.The major clinical treatments for diabetic macular edema are anti-vascular endothelial growth factor drugs and laser photocoagulation.However,although the macular thickness can be normalized with each of these two therapies used alone,the vision does not improve in many patients.This might result from the incomplete recovery of retinal ganglion cell injury.Therefore,a prospective,non-randomized,controlled clinical trial was designed to investigate the effect of anti-vascular endothelial growth factor drugs combined with laser photocoagulation on the integrity of retinal ganglion cells in patients with diabetic macular edema and its relationship with vision recovery.In this trial,150 patients with diabetic macular edema will be equally divided into three groups according to therapeutic methods,followed by treatment with anti-vascular endothelial growth factor drugs,laser photocoagulation therapy,and their combination.All patients will be followed up for 12 months.The primary outcome measure is retinal ganglion cell-inner plexiform layer thickness at 12 months after treatment.The secondary outcome measures include retinal ganglion cell-inner plexiform layer thickness before and 1,3,6,and 9 months after treatment,retinal nerve fiber layer thickness,best-corrected visual acuity,macular area thickness,and choroidal thickness before and 1,3,6,9,and 12 months after treatment.Safety measure is the incidence of adverse events at 1,3,6,9,and 12 months after treatment.The study protocol hopes to validate the better efficacy and safety of the combined treatment in patients with diabetic macula compared with the other two monotherapies alone during the 12-month follow-up period.The trial is designed to focus on clarifying the time-effect relationship between imaging measures related to the integrity of retinal ganglion cells and best-corrected visual acuity.The trial protocol was approved by the Medical Ethics Committee of the Affiliated Hospital of Beihua University with approval No.(2023)(26)on April 25,2023,and was registered with the Chinese Clinical Trial Registry(registration number:ChiCTR2300072478,June 14,2023,protocol version:2.0).展开更多
Objective:To measure the self-efficacy of nursing students taught using both flipped and traditional classroom modalities.Method:A quantitative research design was employed.The control group was taught using the tradi...Objective:To measure the self-efficacy of nursing students taught using both flipped and traditional classroom modalities.Method:A quantitative research design was employed.The control group was taught using the traditional classroom model,while the experimental group utilized the flipped classroom model.The study population consisted of undergraduate nursing majors at Shandong University of Traditional Chinese Medicine.Two out of twelve classes were selected through random sampling.Results:The study presents an analysis of the t-test results using SPSS,which shows a significant difference when the calculated P-value falls between 0.00 and 0.05.The P-value obtained was 0.00,indicating a statistically significant difference.Conclusion:The teaching model,whether flipped or traditional,has an impact on nursing students’self-efficacy.Students in the flipped classroom exhibited higher self-efficacy in mastering the curriculum.展开更多
Asphalt binder is inevitably aged by ultraviolet(UV)light during its service period.UV ageing can significantly decrease the technical properties of asphalt binder.The sensitivity of asphalt to UV ageing and thermal-o...Asphalt binder is inevitably aged by ultraviolet(UV)light during its service period.UV ageing can significantly decrease the technical properties of asphalt binder.The sensitivity of asphalt to UV ageing and thermal-oxidative ageing differs,such that the UV ageing performance cannot be determined based on the thermal-oxidative ageing performance.Previous researches mainly focused on the chemical composite and technical performance changes of asphalt binder during UV ageing,and the UV light parameters effect on the ageing rate of asphalt binder.However,the theory for characterizing and explaining the development of UV ageing depth does not get too much attentions,and the UV ageing mechanism of asphalt binder is not very clear.Therefore,it cannot guide to develop or select the good methods or anti-UV ageing additives for asphalt binders.This paper focuses on the latest researches of the mechanisms and anti-ageing methods of asphalt binders.With the increase of UV ageing time,the UV ageing of asphalt binder develops gradually from the surface to inner part.There are various methods,such as low-penetration grade asphalt,less air void ratio,UV stabilizers and UV light absorbers,that can improve the UV ageing resistance of asphalt binders.A new theory of sensitive wavelengths of asphalt UV ageing is proposed,which can enrich the basic theory of asphalt UV ageing.Depending on this theory,different wavelengths of UV light have different ageing effects on asphalt binder.The composite anti-UV ageing additives with barrier and specific absorption effects on UV light is proposed,and may have better improvement effect on the anti-UV ageing performance of asphalt binder.展开更多
Microplastics(MPs)are distributed in the oceans,freshwater,and soil environment and have become major pollutants.MPs are generally referred to as plastic particles less than 5 mm in diameter.They consist of primary mi...Microplastics(MPs)are distributed in the oceans,freshwater,and soil environment and have become major pollutants.MPs are generally referred to as plastic particles less than 5 mm in diameter.They consist of primary microplastics synthesized in microscopic size manufactured production and secondary microplastics generated by physical and environmental degradation.Plastic particles are long-lived pollutants that are highly resistant to environmental degradation.In this review,the distribution and possible sources of MPs in aquatic and terrestrial environments are described.Moreover,the adverse effects of MPs on natural creatures due to ingestion have been discussed.We also have summarized identification methods based on MPs particle size and chemical bond.To control the pollution of MPs,the biodegradation of MPs under the action of different microbes has also been reviewed in this work.This review will contribute to a better understanding of MPs pollution in the environment,as well as their identification,toxicity,and biodegradation in the ocean,freshwater,and soil,and the assessment and control of microplastics exposure.展开更多
SOX4 is highly expressed in gastric cancer(GC)and is associated with tumor grade,metastasis and prognosis,however the mechanism is not clear.We report herein that SOX4 was upregulated and overexpression of SOX4 was as...SOX4 is highly expressed in gastric cancer(GC)and is associated with tumor grade,metastasis and prognosis,however the mechanism is not clear.We report herein that SOX4 was upregulated and overexpression of SOX4 was associated with increased expression of the markers of Epithelialemesenchymal transition(EMT)and stemness in clinic patient samples.In vitro,overexpression of SOX4 promoted the invasion as showed by Transwell assay and stemness of GC cells as assessed by sphere formation assay,which was suppressed by silencing SOX4 with shRNA.Further studies showed that SOX4 up-regulated the expression of EMT transcription factors Twist1,snail1 and zeb1 and stemness transcription factors SOX2 and OCT4,and promoted the nuclear translocation of β-catenin.Moreover,we revealed that TGF-β treatment significantly up-regulated the expression of SOX4 and silencing SOX4 reversed TGF-β induced invasion and sphere formation ability of GC cells.Finally,we showed that SOX4 promoted the lung metastasis and tumor formation ability of gastric cancer cells in nude mice.Our results suggest that SOX4 is a target TGF-β signaling and mediates TGF-β-induced EMT and stem cell characteristics of GC cells,revealing a novel role of TGF-β/SOX4 axis in the regulation of malignant behavior of GC.展开更多
Growing evidence suggests that somatic hypermutational status and programmed cell death-1 overexpression are potential predictive biomarkers indicating treatment benefits from immunotherapy using immune checkpoint inh...Growing evidence suggests that somatic hypermutational status and programmed cell death-1 overexpression are potential predictive biomarkers indicating treatment benefits from immunotherapy using immune checkpoint inhibitors.However,biomarker-matched trials are still limited,and many of the genomic alterations remain difficult to target.To isolate the potential somatic hypermutational tumor from microsatellite instability low/microsatellite stability(MSI-L/MSS)cases,we employed two commercial kits to determine MSI and forensic short tandem repeat(STR)alternations in 250 gastrointestinal(GI)tumors.Three types of forensic STR alternations,namely,allelic loss,Aadd,and Anew,were identified.62.4%(156/250)of the patients with GI exhibited STR alternation,including 100%(15/15)and 60%(141/235)of the microsatellite high instability and MSI-L/MSS cases,respectively.30%(75/250)of the patients exhibited STR instability with more than 26.32%(26.32%–84.21%)STR alternation.The cutoff with 26.32%of the STR alternations covered all 15 MSI cases and suggested that it might be a potential threshold.Given the similar mechanism of the mutations of MSI and forensic STR,the widely used forensic identifier STR kit might provide potential usage for identifying hypermutational status in GI cancers.展开更多

Poly(ethylene oxide)(PEO)-based solid polymer electrolytes(SPEs)are commonly used in lithium metal batteries(LMBs)for their good Li-salt solvating ability and easy processability.However,the relatively low Li-ion cond...Poly(ethylene oxide)(PEO)-based solid polymer electrolytes(SPEs)are commonly used in lithium metal batteries(LMBs)for their good Li-salt solvating ability and easy processability.However,the relatively low Li-ion conduction ability hinders their further development.In this work,a novel hyperbranched-polyether-type composite solid polymer electrolyte(CSPE)is prepared via a quick cross-linking reaction between aldehyde-terminated polyethylene glycol(PEG)and hyperbranched poly(ethylene imine)(HPEI)in the presence of lithium salt and fluorine-containing Zr-based metal–organic framework(MOF)UiO-66-(F)_(4).The hydrogen bonds between the fluorine atoms and amino groups in the electrolyte help to the better dispersion of UiO-66-(F)_(4) in the polymer matrix,which is beneficial to solving the problem of aggregation of nanofillers.Besides,the CSPEs with the functional MOF fillers show improvements in both electrochemical and mechanical properties.Notably,the Li-ion transference number(t)is considerably enhanced from 0.23 to 0.54.All-solid-state LMBs based on the CSPE also present good cycling performances.A high specific discharge capacity of 141.4 mAh·g^(−1) is remained after 200 cycles at 0.2 C.This study not only provides an effective synthesis method of the cross-linked hyperbranched polymer electrolyte,but also puts forward a new strategy for uniform dispersion of inorganic fillers in CSPEs.展开更多
Mutations might challenge the paternity index calculation in forensic identification.While many studies have focussed on the autosomal short tandem repeats(A-STR),the mutation status of sex chromosomes and single nucl...Mutations might challenge the paternity index calculation in forensic identification.While many studies have focussed on the autosomal short tandem repeats(A-STR),the mutation status of sex chromosomes and single nucleotide polymorphism(SNP)remain blank.Next generation sequencing(NGS),known as high throughput and large sequence polymorphism,is a promising tool for forensic genetics.To describe the mutation landscapes in the paternity cases with genetic inconsistencies,a total of 63 parentage confirmed paternity cases contained at least one mismatched locus have been collected.The mutations were subsequently evaluated using Verogen’s MPSForenSeqTM DNASignature Kit and a microsatellite instability(MSI)detection kit.The result showed 98.41%(62/63)of the cases had no additional autosomal mutations even when the number of A-STRs increased to 27.As for the sex chromosomes,about 11.11%(7/63)of the cases exhibited either X-STR or Y-STR mutations.D2S1338,FGAand Penta Ewere the most frequent altered STRs,which suggested they might be the mutation hotspots.In addition,a male with sex chromosome abnormality was observed accidently,whose genotype might be 47,XXY,rather than MSI.Nearly 56.90%of the STR loci possessed isoalleles,which might result in higher STR polymorphisms.No Mendelian incompatibility was detected among the SNP markers,which indicated that SNP was a more reliable genetic marker in the genetic-inconsistent paternity cases.展开更多

The custom-designed single nucleotide polymorphism(SNP)panel amplified 231 autosomal SNPs in one PCR reaction and subsequently sequenced with massively parallel sequencing(MPS)technology and Ion Torrent personal genom...The custom-designed single nucleotide polymorphism(SNP)panel amplified 231 autosomal SNPs in one PCR reaction and subsequently sequenced with massively parallel sequencing(MPS)technology and Ion Torrent personal genome machine(PGM).SNPs were chosen from SNPforID,IISNP,HapMap,dbSNP,and related published literatures.Full concordance was obtained between available MPS calling and Sanger sequencing with 9947A and 9948 controls.Ten SNPs(rs4606077,rs334355,rs430046,rs2920816,rs4530059,rs1478829,rs1498553,rs7141285,rs12714757 and rs2189011)with low coverage or heterozygote imbalance should be optimized or excluded from the panel.Sequence data had sufficiently high coverage and gave reliable SNP calling for the remaining 221 loci with the custom MPS-SNP panel.A default DNA input amount of 10 ng per reaction was recommended by Ampliseq technology but sensitivity testing revealed positive results from as little as 1 ng input DNA.Mixture testing with this panel is possible through analysis of the F MAR(frequency of major allele reads)values at most loci with enough high coverage depth and low level of sequencing noise.These results indicate the potential advantage of the custom MPS-SNP assays and Ion Torrent PGM platform for forensic study.展开更多

文摘Poly(ethylene oxide)(PEO)-based solid polymer electrolytes(SPEs)are commonly used in lithium metal batteries(LMBs)for their good Li-salt solvating ability and easy processability.However,the relatively low Li-ion conduction ability hinders their further development.In this work,a novel hyperbranched-polyether-type composite solid polymer electrolyte(CSPE)is prepared via a quick cross-linking reaction between aldehyde-terminated polyethylene glycol(PEG)and hyperbranched poly(ethylene imine)(HPEI)in the presence of lithium salt and fluorine-containing Zr-based metal–organic framework(MOF)UiO-66-(F)_(4).The hydrogen bonds between the fluorine atoms and amino groups in the electrolyte help to the better dispersion of UiO-66-(F)_(4) in the polymer matrix,which is beneficial to solving the problem of aggregation of nanofillers.Besides,the CSPEs with the functional MOF fillers show improvements in both electrochemical and mechanical properties.Notably,the Li-ion transference number(t)is considerably enhanced from 0.23 to 0.54.All-solid-state LMBs based on the CSPE also present good cycling performances.A high specific discharge capacity of 141.4 mAh·g^(−1) is remained after 200 cycles at 0.2 C.This study not only provides an effective synthesis method of the cross-linked hyperbranched polymer electrolyte,but also puts forward a new strategy for uniform dispersion of inorganic fillers in CSPEs.

文摘Mutations might challenge the paternity index calculation in forensic identification.While many studies have focussed on the autosomal short tandem repeats(A-STR),the mutation status of sex chromosomes and single nucleotide polymorphism(SNP)remain blank.Next generation sequencing(NGS),known as high throughput and large sequence polymorphism,is a promising tool for forensic genetics.To describe the mutation landscapes in the paternity cases with genetic inconsistencies,a total of 63 parentage confirmed paternity cases contained at least one mismatched locus have been collected.The mutations were subsequently evaluated using Verogen’s MPSForenSeqTM DNASignature Kit and a microsatellite instability(MSI)detection kit.The result showed 98.41%(62/63)of the cases had no additional autosomal mutations even when the number of A-STRs increased to 27.As for the sex chromosomes,about 11.11%(7/63)of the cases exhibited either X-STR or Y-STR mutations.D2S1338,FGAand Penta Ewere the most frequent altered STRs,which suggested they might be the mutation hotspots.In addition,a male with sex chromosome abnormality was observed accidently,whose genotype might be 47,XXY,rather than MSI.Nearly 56.90%of the STR loci possessed isoalleles,which might result in higher STR polymorphisms.No Mendelian incompatibility was detected among the SNP markers,which indicated that SNP was a more reliable genetic marker in the genetic-inconsistent paternity cases.

文摘This article considers the problem of selecting two-level designs under the baseline parameterisation when some two-factor interactions are important.We propose a minimum aberration criterion,which minimises the bias caused by the non-negligible effects.Using this criterion,a class of optimal designs can be further distinguished from one another,and we present an algorithm to find the minimum aberration designs among the D-optimal designs.Sixteen-run and twenty-run designs are summarised for practical use.

文摘The custom-designed single nucleotide polymorphism(SNP)panel amplified 231 autosomal SNPs in one PCR reaction and subsequently sequenced with massively parallel sequencing(MPS)technology and Ion Torrent personal genome machine(PGM).SNPs were chosen from SNPforID,IISNP,HapMap,dbSNP,and related published literatures.Full concordance was obtained between available MPS calling and Sanger sequencing with 9947A and 9948 controls.Ten SNPs(rs4606077,rs334355,rs430046,rs2920816,rs4530059,rs1478829,rs1498553,rs7141285,rs12714757 and rs2189011)with low coverage or heterozygote imbalance should be optimized or excluded from the panel.Sequence data had sufficiently high coverage and gave reliable SNP calling for the remaining 221 loci with the custom MPS-SNP panel.A default DNA input amount of 10 ng per reaction was recommended by Ampliseq technology but sensitivity testing revealed positive results from as little as 1 ng input DNA.Mixture testing with this panel is possible through analysis of the F MAR(frequency of major allele reads)values at most loci with enough high coverage depth and low level of sequencing noise.These results indicate the potential advantage of the custom MPS-SNP assays and Ion Torrent PGM platform for forensic study.
