A validated ultra-performance liquid chromatography mass spectrometric method (UPLC-MS/MS) was used for the simultaneous quantitation of candesartan (CN) and hydrochlorothiazide (HCT) in human plasma. The analysis was performed on UPLC-MS/MS system using turbo ion spray interface. Negative ions were measured in multiple reaction monitoring (MRM) mode. The analytes were extracted using a liquid-liquid extraction (LLE) method by using 0.1 mL of plasma volume. The lower limit of quantitation for CN and HCT was 1.00 ng/mL whereas the upper limit of quantitation was 499.15 ng/mL and 601.61 ng/mL for CN and HCT respectively. CN d4 and HCT-13Cd2 were used as the internal standards for CN and HCT respectively. The chromatography was achieved within 2.0 min run time using a C18 Pheno-menex, Gemini NX (100 mm ~ 4.6 mm, 5 mm) column with organic mixture:buffer solution (80:20, v/v) at a flow rate of 0.800 mL/min. The method has been successfully applied to establish the bioequivalence of candesartan cilexetil (CNC) and HCT immediate release tablets with reference product in human subjects. 展开更多

Epithelial ovarian cancer(EOC) is the leading cause of death among all gynecological malignancies. Despite the technological and medical advances over the past four decades, such as the development of several biological markers(mRNA and proteins biomarkers), the mortality rate of ovarian cancer remains a challenge because of its late diagnosis, which is specifically attributed to low specificities and sensitivities. Under this compulsive scenario, recent advances in expression biology have shifted in identifying and developing specific and sensitive biomarkers, such as micro RNAs(miRNAs) for cancer diagnosis and prognosis. MiRNAs are a novel class of small non-coding RNAs that deregulate gene expression at the posttranscriptional level, either by translational repression or by mRNA degradation. These mechanisms may be involved in a complex cascade of cellular events associated with the pathophysiology of many types of cancer. MiRNAs are easily detectable in tissue and blood samples of cancer patients. Therefore, miRNAs hold good promise as potential biomarkers in ovarian cancer. In this review, we attempted to provide a comprehensive profile of key miRNAs involved in ovarian carcinoma to establish mi RNAs as more reliable non-invasive clinical biomarkers for early detection of ovarian cancer compared with protein and DNA biomarkers. 展开更多

In recent years, conventional rice production technologies have been leading to deterioration of soil health and declining farm profitability due to high inputs of water and labor. Conservation agriculture (CA) based resource-conserving technologies i.e. zero-tillage (ZT), raised-bed planting and direct-seeded rice (DSR) have shown promise as alternatives to conventional production technologies to overcome these problems. Present study was undertaken during 2009-2012 to establish an understanding of how permanent raised bed cropping system could be practiced to save water at the field application level to improve water productivity and also have the capability to enhance productivity, profitability and soil physical quality. The results showed that among different crop establishment techniques, conventional-tilled puddle transplanted rice (CT-TPR) required 14%-25% more water than other techniques. Compared with the CT-TPR system, zero till direct-seeded rice (ZT-DSR) consumed 6%-10%less water with almost equal system productivity and demonstrated higher water productivity. Wide raised beds saved about 15%-24% water and grain yield decrease of about 8%. Direct-seeded rice after ZT or reduced tillage or on unpuddled soil provided more net income than CT-TPR. The CT-TPR system had higher bulk density and penetration resistance due to compaction caused by the repeated wet tillage in rice. The steady-state infiltration rate and soil aggregation (〉 0.25 mm) were higher under permanent beds and ZT and lower in the CT-TPR system. Under CT-TPR, soil aggregation was static across seasons, whereas it improved under no-till and permanent beds. Similarly, mean weight diameter of aggregates was higher under ZT and permanent beds and increased over time. The study reveals that to sustain the rice productivity, CA-based planting techniques can be more viable options. However, the long-term effects of these alternative technologies need to be studied under varying agro-ecologies in western Uttar Pradesh, India. 展开更多

A highly sensitive, selective, and precise ultra-performance liquid chromatography tandem mass spectrometry method was developed and validated for simultaneous quantification of itraconazole and hydroxy itraconazole in human plasma by a single liquid-liquid extraction step. The precursor to product ion transitions of m/z 705.3/392.3, m/z 721.2/408.3 and m/z 708.2/435.4 were used to detect and quantify itraconazole, hydroxy itraconazole and itraconazole-d3 respectively. The lower limit of quantitation was found to be 0.500 ng/mL for itraconazole and 1.00 ng/mL for hydroxy itraconazole. The mean recoveries for itraconazole and hydroxy itraconazole were found to be 100.045% and 100.021%, respectively. This developed method with a chromatographic run time of 2.0 min was successfully applied to a bioequivalence study of 100 mg itraconazole capsule. 展开更多
