A major impedance to neuronal regeneration after peripheral nerve injury (PNI) is the activation of various programmed cell death mechanisms in the dorsal root ganglion. Ferroptosis is a form of programmed cell death distinguished by imbalance in iron and thiol metabolism, leading to lethal lipid peroxidation. However, the molecular mechanisms of ferroptosis in the context of PNI and nerve regeneration remain unclear. Ferroportin (Fpn), the only known mammalian nonheme iron export protein, plays a pivotal part in inhibiting ferroptosis by maintaining intracellular iron homeostasis. Here, we explored in vitro and in vivo the involvement of Fpn in neuronal ferroptosis. We first delineated that reactive oxygen species at the injury site induces neuronal ferroptosis by increasing intracellular iron via accelerated UBA52-driven ubiquitination and degradation of Fpn, and stimulation of lipid peroxidation. Early administration of the potent arterial vasodilator, hydralazine (HYD), decreases the ubiquitination of Fpn after PNI by binding to UBA52, leading to suppression of neuronal cell death and significant acceleration of axon regeneration and motor function recovery. HYD targeting of ferroptosis is a promising strategy for clinical management of PNI. 展开更多

AIM:To evaluate potential risk factors in the development of ulcerative colitis(UC) in China.METHODS:A total of 1308 patients with UC and 1308 age-matched and sex-matched controls were prospectively studied in China.The UC cases were collected from 17 hospitals in China from April 2007 to April 2010.Uniform questionnaires were designed to investigate risk factors including smoking,appendectomy,stress,socio-economic conditions,nonsteroidal antiinflammatory drugs(NSAIDs),oral contraceptives,diet,breastfeeding,infections and family sanitary conditions.Group comparisons by each factor were done using simple logistic regression analysis.Conditional logistic regression was used for multivariate analysis.RESULTS:By univariate analysis,the variables predictive of UC included feeling stress,light and heavy alcoholic drinking,spicy food,sugar consumption and infectious diarrhea,while heavy tea intake and tap water consumption were protective against UC.On multivariate analysis,the protective factor for UC was tap water consumption [odds ratios(OR) = 0.424,95%CI:0.302-0.594,P < 0.001];while the potential risk factors for UC were heavy sugar consumption(OR = 1.632,95%CI:1.156-2.305,P < 0.001),spicy food(light intake:OR = 3.329,95%CI:2.282-4.857,P < 0.001;heavy intake:OR = 3.979,95%CI:2.700-5.863,P < 0.001),and often feeling stress(OR = 1.981,95%CI:1.447-2.711,P < 0.001).Other factors,such as smoking habit,appendectomy,breastfeeding,a history of measles,rural or urban residence,education,oral contraceptives,and NSAID use have not been found to have a significant association with the development of UC in the present study.CONCLUSION:Our study showed tap water consumption was a protective factor for UC,while spicy food,heavy sugar consumption and often feeling stress were risk factors for UC in this Chinese population. 展开更多

AIM:To investigate whether MYC and BCL-2 coexpression has prognostic significance in primary gastrointestinal diffuse large B-cell lymphoma(PGI-DLBCL)patients,and explore its associations with patients’clinical parameters.METHODS:Fresh and paraffin-embedded tumor tissue samples from 60 PGI-DLBCL patients who had undergone surgery at the Tianjin Medical University Cancer Institute and Hospital from January 2005 to May 2010 were obtained,and 30 lymphoid tissue samples from reactive lymph nodes of age-and sexmatched patients represented control samples.Staging and diagnostic procedures were conducted according to the Lugano staging system.All patients had been treated with three therapeutic modalities:surgery,chemotherapy,or radiotherapy.Expression of MYC and BCL-2 were detected at both protein and m RNA levels by immunohistochemistry and real-time RT-PCR.RESULTS:Positive expression levels of MYC and BCL-2proteins were detected in 35%and 45%of patients,respectively.MYC+/BCL-2+protein was present in30%of patients.MYC and BCL-2 protein levels were correlated with high MYC and BCL-2 m RNA expression,respectively(both P<0.05).We found that advancedstage disease(atⅡE-Ⅳ)was associated with MYC and BCL-2 coexpression levels(P<0.05).In addition,MYC+/BCL-2+patients had more difficulty in achieving complete remission than others(P<0.05).Presenceof MYC protein expression only affected overall survivaland progression-free survival(PFS)when BCL-2 proteinwas coexpressed.The adverse prognostic impact ofMYC+/BCL-2+protein on PFS remained significant(P<0.05)even after adjusting for age,Lugano stage,international prognostic index,and BCL-2 proteinexpression in a multivariable model.CONCLUSION:MYC+/BCL-2+patients have worsechemotherapy response and poorer prognosis thanpatients who only express one of the two proteins,suggesting that assessment of MYC and BCL-2 expressionby immunohistochemistry has clinical significance inpredicting clinical outcomes of PGI-DLBCL patients. 展开更多

AIM:To evaluate clinical response to initial corticosteroid(CS) treatment in Chinese ulcerative colitis patients(UC) and identify predictors of clinical response.METHODS:Four hundred and twenty-three UC patients who were initially treated with oral or intravenous CS from 2007 to 2011 were retrospectively reviewed at eight inflammatory bowel disease centers in China,and 101 consecutive cases with one-year follow-up were analyzed further for clinical response and predictors.Short-term outcomes within one month were classified as primary response and primary non-response.Longterm outcomes within one year were classified as prolonged CS response,CS dependence and secondary non-response.CS refractoriness included primary and secondary non-response.Multivariate analyses were performed to identify predictors associated with clinical response.RESULTS:Within one month,95.0%and 5.0%of the cases were classified into primary response andnon-response,respectively.Within one year,41.6%of cases were assessed as prolonged CS response,while49.5%as CS dependence and 4.0%as secondary nonresponse.The rate of CS refractoriness was 8.9%,while the cumulative rate of surgery was 6.9%within one year.After multivariate analysis of all the variables,tenesmus was found to be a negative predictor of CS dependence(OR=0.336;95%CI:0.147-0.768;P=0.013)and weight loss as a predictor of CS refractoriness(OR=5.662;95%CI:1.111-28.857;P=0.040).After one-month treatment,sustained high Sutherland score(≥6)also predicted CS dependence(OR=2.347;95%CI:0.935-5.890;P=0.014).CONCLUSION:Tenesmus was a negative predictor of CS dependence,while weight loss and sustained high Sutherland score were strongly associated with poor CS response. 展开更多

目的:旨在比较化疗联合粒细胞集落刺激因子(chemotherapy plus granulocyte cyte colony stimulating factor,G-CSF)与化疗联合G-CSF和粒巨噬细胞集落刺激因子(granulocyte-macrophage colony-stimulating factor,GM-CSF)方案对多发性... 目的:旨在比较化疗联合粒细胞集落刺激因子(chemotherapy plus granulocyte cyte colony stimulating factor,G-CSF)与化疗联合G-CSF和粒巨噬细胞集落刺激因子(granulocyte-macrophage colony-stimulating factor,GM-CSF)方案对多发性骨髓瘤(multiple myeloma,MM)患者外周血造血干细胞(peripheral blood stem cell,PBSC)动员、采集及造血重建的影响。方法:回顾性分析56例2008年5月至2016年7月天津医科大学肿瘤医院血液科化疗联合G-CSF或化疗联合G-CSF和GM-CSF动员采集PBSC行自体外周血造血干细胞移植(autologous peripheral blood stem cell transplantation,APBSCT)的MM患者资料。分析患者年龄、性别、疾病类型、临床分期(DS、ISS、R-ISS分期系统)、既往治疗方案和周期数等因素对动员效果及移植后造血重建、发热和抗生素应用等的影响。结果:单因素分析发现女性、ISS分期Ⅲ期、R-ISS分期Ⅱ/Ⅲ期及化疗+G-CSF动员方案的患者采集效果较差(P<0.05)。年龄≤60岁或年龄>60岁、疾病类型、DS分期(Ⅰ+Ⅱ/Ⅲ)、动员前化疗次数(≤6次或>6次)、有无烷化剂治疗史、动员前疾病缓解状态部分缓解(partial response,PR)或完全缓解(complete response,CR)及确诊到动员间隔时间(≤18个月或>18个月)与采集获得的CD34+细胞数及采集成功率无相关性(P>0.05)。进一步多因素分析结果显示,化疗联合G-CSF和GM-CSF患者较化疗联和GCSF患者采集成功率较高(OR=12.009,95%CI:1.961~73.537),不同动员方案是影响干细胞采集结果的独立预后因素(P=0.007)。所有患者移植后均顺利获得造血重建,无移植相关性死亡。两项动员方案组的造血重建时间及回输后发热、抗生素使用情况差异均无统计学意义(P>0.05)。结论:化疗联合G-CSF和GM-CSF动员方案采集的成功率较高,而不良反应未见增加,该动员方案可作为MM患者外周血干细胞动员的一项较好选择。 展开更多

背景与目的血浆循环肿瘤DNA(circulating tumor DNA,ctDNA)是检测表皮生长因子受体(epidermal growth factor receptor,EGFR)T790M突变的理想方法。T790M突变是第一代EGFR酪氨酸激酶抑制剂(tyrosine kinase inhibitor,TKI)耐药的主要机... 背景与目的血浆循环肿瘤DNA(circulating tumor DNA,ctDNA)是检测表皮生长因子受体(epidermal growth factor receptor,EGFR)T790M突变的理想方法。T790M突变是第一代EGFR酪氨酸激酶抑制剂(tyrosine kinase inhibitor,TKI)耐药的主要机制。本研究旨在探讨ctDNA来源的T790M突变与非小细胞肺癌(non-small cell lung cancer,NSCLC)患者疾病失败部位及临床预后的关系。方法第一代TKI治疗发生进展的患者被分为局限胸内进展(chest limited,CF)、局限颅内进展(brain limited,BF)和其他部位进展(other,OF)组。采用扩增阻滞突变系统(amplification refractory mutation system,ARMS)和微滴式数字PCR(droplet digital PCR,ddPCR)对ctDNA中T790M突变进行了检测。采用Kaplan-Meier法分析了预后。结果 2种方法的总体一致性为78.3%。根据ARMS和ddPCR,OF组患者的T790M突变率明显高于BF组和CF组(P <0.001),OF组的T790M突变丰度也明显高于CF组和BF组(P <0.001)。AZD9291是T790M+患者最佳治疗选择,为EGFR-TKI治疗后进展的所有组中T790M+患者提供了最长的生存期;接受其他治疗的T790M-患者亚组的预后各不相同。结论本研究表明ctDNA中T790M突变与NSCLC患者EGFR-TKI治疗失败部位相关,失败部位和T790M突变状态对治疗选择和预后都有很大影响。 展开更多

Objective: To investigate the effects of CAL-101, particularly when combined with bortezomib(BTZ) on mantle cell lymphoma(MCL) cells, and to explore its relative mechanisms.Methods: MTT assay was applied to detect the inhibitory effects of different concentrations of CAL-101. MCL cells were divided into four groups: control group, CAL-101 group, BTZ group, and CAL-101/BTZ group. The expression of PI3K-p110σ, AKT, ERK, p-AKT and p-ERK were detected by Western blot. The apoptosis rates of CAL-101 group, BTZ group, and combination group were detected by flow cytometry. The location changes of nuclear factor kappa-B(NF-κB) of 4 groups was investigated by NF-κB Kit exploring. Western blot was applied to detect the levels of caspase-3 and the phosphorylation of AKT in different groups. Results: CAL-101 dose- and time-dependently induced reduction in MCL cell viability. CAL-101 combined with BTZ enhanced the reduction in cell viability and apoptosis. Western blot analysis showed that CAL-101 significantly blocked the PI3K/AKT and ERK signaling pathway in MCL cells. The combination therapy contributed to the inactivation of NF-κB and AKT in MCL cell lines. However, cleaved caspase-3 was up-regulated after combined treatment. Conclusion: Our study showed that PI3K/p110σ is a novel therapeutic target in MCL, and the underlying mechanism could be the blocking of the PI3K/AKT and ERK signaling pathways. These findings provided a basis for clinical evaluation of CAL-101 and a rationale for its application in combination therapy, particularly with BTZ. 展开更多

The high-temperature reactor pebble-bed mod-ule(HTR-PM)is a modular high-temperature gas-cooled reactor demonstration power plant.Its first criticality experiment is scheduled for the latter half of 2021.Before performing the first criticality experiment,a prediction calculation was performed using PANGU code.This paper presents the calculation details for predicting the HTR-PM first criticality using PANGU,including the input model and parameters,numerical results,and uncertainty analysis.The accuracy of the PANGU code was demonstrated by comparing it with the high-fidelity Monte Carlo solution,using the same input configurations.It should be noted that k eff can be significantly affected by uncertainties in nuclear data and certain input parameters,making the criticality calculation challenge.Finally,the PANGU is used to pre-dict the critical loading height of the HTR-PM first criti-cality under design conditions,which will be evaluated in the upcoming experiment later this year. 展开更多

A visible rectification effect on the current-voltage curves of metal/porous silicon/p-silicon has been observed by current- sensing atomic force microscopy. The current-voltage curves of porous silicon membranes with different porosities, prepared through variation of etching current density for a constant time, indicate that a higher porosity results in a higher resistance and thus a lower rectification, until the current reaches a threshold at a porosity >55%. We propose that the conductance mode in the porous silicon membrane with porosities >55% is mainly a hopping mechanism between nano-crystallites and an inverse static electric field between the porous silicon and p-Si interface blocks the electron injection from porous silicon to p-Si, but with porosities ≥55%, electron flows through a direct continuous channel between nano-crystallites. 展开更多

We investigate the surface structure and electronic properties ofβ-Sn islands deposited on a graphitized 6 H-SiC(0001)substrate via low temperature scanning tunneling microscopy and spectroscopy.Owing to the confinement of the island geometry,quantum well states(QWSs)are formed,manifesting as equidistant peaks in the tunneling spectra.Furthermore,a distinct strip feature appears on the surfaces of odd-layer Sn islands,ranging from 15-19 layers,which is not present on the surfaces of even-layer Sn islands.The spatial distribution of strips can be modified by applying a bias pulse,using an STM tip.Furthermore,the strip-like structure shows significant impacts on the QWS.An energy splitting of the lowest unoccupied QWSs is observed in strip regions;this may be ascribed to caused the phase shift of the wave functions of the QWSs on the top surface,due to surface distortions created by the aforementioned strips. 展开更多

Bromine is an irreplaceable reagent for many important applications including necessary additive for flame retardants and rubber treatment.However,the toxicity,causticity,and vaporability of bromine make the safe usage and transportation of bromine challenging,which can be alleviated by the bromine prefixation.Herein,vacancy-ordered bromide perovskites such as Cs^(4)Sb_(2)Br_(12)and Cs_(2)PdBr_(6)are selected out for the bromine fixation through density-functional theory calculations and experiments.The redox within the bromide perovskites is designed to provide the driving force for the reversible release and fixation of bromine.Heating and cooling the title bromide perovskites in a necked ampoule make the bromine visible and separable from the solid residue.The composition engineering(i.e.,Cs_(4)Sb_(2)Br_(12)vs Rb_(4)Sb_(2)Br_(12),and Cs_(4)Sb_(2)Br_(12)vs Cs_(2)PdBr_(6))can effectively tune the stability(against water and high temperature)and the bromine storage capacity(from9.30%to 18.76%).In addition,this work can provide a one-step preparation method for the liquid bromine without further purification. 展开更多

Lead sulfide(PbS)colloidal quantum dot(CQD)photodiodes integrated with silicon-based readout integrated circuits(ROICs)offer a promising solution for the next-generation short-wave infrared(SWIR)imaging technology.Despite their potential,large-size CQD photodiodes pose a challenge due to high dark currents resulting from surface states on nonpassivated(100)facets and trap states generated by CQD fusion.In this work,we present a novel approach to address this issue by introducing double-ended ligands that supplementally passivate(100)facets of halidecapped large-size CQDs,leading to suppressed bandtail states and reduced defect concentration.Our results demonstrate that the dark current density is highly suppressed by about an order of magnitude to 9.6 nA cm^(2) at^(-1)0 mV,which is among the lowest reported for PbS CQD photodiodes.Furthermore,the performance of the photodiodes is exemplary,yielding an external quantum efficiency of 50.8%(which corresponds to a responsivity of 0.532 A W^(-1))and a specific detectivity of 2.5×10^(12) Jones at 1300 nm.By integrating CQD photodiodes with CMOS ROICs,the CQD imager provides high-resolution(640512)SWIR imaging for infrared penetration and material discrimination. 展开更多

Effects of rhamnolipids on bacterial communities in a dioxin-contaminated soil and the gut of earthworms added to the soil

The biosurfactants rhamnolipids and the “soil ecosystem engineers” earthworms are often used to remediate contaminated soils. However, the effects of rhamnolipids on earthworm intestinal flora and microbial community in soil containing earthworms are not clearly understood. In our study, a 21-d microcosm experiment was carried out to reveal the effects of rhamnolipids on microbial abundance, composition, and metabolism, as well as contaminant degradation capacity. Both rhamnolipids and earthworms had positive effects on soil bacteria. Rhamnolipid-amended soil(RT) showed higher bacterial abundance and metabolic activity than earthworm-amended soil(ET), while the improvement in bacterial composition and contaminant degradation capacity by rhamnolipids was lower than that by earthworms. Notably, these effects were further amplified by the combined treatment of rhamnolipids and earthworms(RET). Specifically, the bacterial abundance(log-transferred) increased from 9.5 copies g-1in the control with no addition to 10.3, 10.6, and 11.1 copies g-1in ET, RT, and RET, respectively. Compared to ET, the relative abundance of the dominant phylum, Proteobacteria, increased from 41.66% to 51.67% in RET,and more pollutant-degrading bacteria were also enriched in RET. Therefore, the increases in bacterial abundance and contaminant-degrading bacteria led to the following ranking of soil dioxin removal rate: RET(77.28%) > ET(59.83%) > RT(24.65%) > control(4.71%). Moreover, the addition of rhamnolipids enhanced the abundance of bacterial functional genes involved in metabolism and environmental information processing. In addition, the composition and diversity of bacteria in the gut of earthworms were conspicuously affected by rhamnolipids, and the relative abundance of Microbacterium and Shewanella increased significantly(P < 0.05). Therefore, this study revealed that rhamnolipids remarkably influenced the abundance, composition, and metabolism of the microbial community in earthworm gut, further promoting the degradation rate of dioxin, providing theoretical support for optimizing the combined application of rhamnolipids and earthworms in soil bioremediation engineering and for the assessment of the ecological impact of rhamnolipids. 展开更多

厘清大陆深部地壳形成、演化及其动力学过程,对于了解板块构造、揭示壳幔相互作用至关重要.针对华北深部地壳物质组成、结构及演化等问题,本研究在限定其地震学结构特征基础上,结合深部地壳岩石捕虏体的来源深度(定深)、化学和物理性质... 厘清大陆深部地壳形成、演化及其动力学过程,对于了解板块构造、揭示壳幔相互作用至关重要.针对华北深部地壳物质组成、结构及演化等问题,本研究在限定其地震学结构特征基础上,结合深部地壳岩石捕虏体的来源深度(定深)、化学和物理性质(定性)和形成年龄(定年)的研究成果,构建了华北深部地壳多维度物理、化学结构模型.结果表明:华北陆块地壳厚度变化于30~48 km,全地壳和地壳内部各层均呈现东薄西厚特征.东、西部地壳虽然厚度上有明显差异,但内部结构特征有一致性,即整体上较少典型的高速基性下地壳,平均P波波速(~6.3 km/s)、密度(2.72~2.78 g/cm^(3))并呈中性岩石特征.深部地壳岩石捕虏体除斜长角闪岩外主要成分包括基性和酸性的麻粒岩.中生代时期华北陆块部分地区地壳存在榴辉岩捕虏体,指示了当时局部地区曾存在厚达45 km的地壳,但现今地震学剖面并未显示;此外,有些地区(如汉诺坝)基性岩石捕虏体揭示出新生代基性岩浆底侵,难以在地震学模型中识别.因此,岩石探针与地震探测结果的结合,将更有助于全面认识深部地壳结构组成和演化. 展开更多
