Background:Head and neck squamous cell carcinoma(HNSCC)is a prevalent form of cancer globally,with chemoresistance posing a major challenge in treatment outcomes.The efficacy of the commonly used chemotherapeutic agent...Background:Head and neck squamous cell carcinoma(HNSCC)is a prevalent form of cancer globally,with chemoresistance posing a major challenge in treatment outcomes.The efficacy of the commonly used chemotherapeutic agent,cisplatin,is diminished in patients with poor prognoses.Methods:Various bioinformatics databases were utilized to examine Carboxylesterase 1(CES1)gene expression,clinicopathologic features,patient survival analysis,and gene function.An organoid model of HNSCC was established,along with the induction of drug-resistant HNSCC in the organoid model.CES1 expression was assessed using qRT-PCR and Western Blot,and differential markers were identified through transcriptome sequencing.Knockdown and overexpression models of CES1 were created in SCC-9 and patient-derived organoid(PDO)cells using shRNA and lentivirus to investigate the tumor biology and cisplatin resistance associated with CES1.Results:Research in bioinformatics has uncovered a strong correlation between the expression level of CES1 and the prognosis of HNSCC.The data suggests a significant link between CES1 expression and tobacco smoking.RNA-sequencing revealed a notable increase in CES1 expression in HNSCC-PDOcis-R cells compared to the parental PDO cells.Subsequently,we performed in vitro studies by HNSCC-PDO and SCC-9 and found that CES1-overexpressing cells exhibited reduced sensitivity to cisplatin and stronger tumor malignant biological behavior compared with CES1-knockdown cells.Conclusion:The observed association between CES1 expression and tobacco smoking implies a potential influence of smoking on the efficacy of cisplatin-based chemotherapy in HNSCC through the regulation of CES1 expression.展开更多

In the present study, gel filtration chromatography, reverse phase-high performance liquid chromatography, and liquid chromatography-electrospray ionization-tandem mass (LC-ESI-MS/MS) were employed for purifying and identifying the ACE inhibitory peptides from hazelnut. To understand the mode of action of these peptides, the interaction between the inhibitory peptides and ACE was investigated. The results identified novel ACE inhibitory peptides Asp-Asp-Glu-Leu-Arg-Gln-Ala (DDELRQA), Asp-Asp-Glu-Leu-Arg-Ala-Ala (DDELRAA), and Asp-Gly-Glu-Leu-Arg-Glu (DGELRE). The binding free energies of DDELRQA, DDELRAA, and DGELRE for ACE were -10.2, -9.0, and -8.8 kcal/mol, respectively. The change in ocean net surface heat flux plays an important role in the climate system.It is closely related to the ocean heat content change and ocean heat transport,particularly over the North Atlantic,where the oc...The change in ocean net surface heat flux plays an important role in the climate system.It is closely related to the ocean heat content change and ocean heat transport,particularly over the North Atlantic,where the ocean loses heat to the atmosphere,affecting the AMOC(Atlantic Meridional Overturning Circulation)variability and hence the global climate.However,the difference between simulated surface heat fluxes is still large due to poorly represented dynamical processes involving multiscale interactions in model simulations.In order to explain the discrepancy of the surface heat flux over the North Atlantic,datasets from nineteen AMIP6 and eight highresSST-present climate model simulations are analyzed and compared with the DEEPC(Diagnosing Earth's Energy Pathways in the Climate system)product.As an indirect check of the ocean surface heat flux,the oceanic heat transport inferred from the combination of the ocean surface heat flux,sea ice,and ocean heat content tendency is compared with the RAPID(Rapid Climate Change-Meridional Overturning Circulation and Heat flux array)observations at 26°N in the Atlantic.The AMIP6 simulations show lower inferred heat transport due to less heat loss to the atmosphere.The heat loss from the AMIP6 ensemble mean north of 26°N in the Atlantic is about10 W m–2 less than DEEPC,and the heat transport is about 0.30 PW(1 PW=1015 W)lower than RAPID and DEEPC.The model horizontal resolution effect on the discrepancy is also investigated.Results show that by increasing the resolution,both surface heat flux north of 26°N and heat transport at 26°N in the Atlantic can be improved.展开更多

Walnut mainly containing polyunsaturated fatty acids (PUFA), especially alpha linoleic acid (18:3n-3;9 g) and linoleic acid (18:3n-;38 g). Walnuts powder was fortified with commercial high gluten wheat flour (WF) into 4 composites flour from 10% to 40%. Walnut flour improves the nutritional value due to high protein and fat content, but affects the technological properties. Parameters such as functional, pasting and rheological properties demonstrated the influence of walnut reincorporation into gluten network. The interaction of walnut flour with wheat flour is a key factor which effects on the dough quality, causing thinning and weakening the gluten structure. Flour with 30% to 40% walnut substitution was more nutritious especially in relation to protein and monounsaturated fatty acid (MUFA), but responsible for more changes in technological characteristics of dough. Present study was also directed to examine the effect of moderate walnut consumption on lipids profile, enzymatic anti-oxidant (superoxide dismutase SOD and glutathione peroxidase GSH-Px) and lipid peroxidation (Melondialdehyde MDA) in rats fed with high-fat food. The Sprague Dawley rats were housed for five weeks and assembled into normal, high fat, high fat (HF) + 10% walnut, HF + 20% walnut, HF + 30% walnut and HF + 40% walnut. The lipid profile significantly improved especially by lowering the low-density lipoprotein cholesterol (LDL-c) and high-density lipoprotein cholesterol (HDL-c) level increased in the group of HF + 40% walnut. The total anti-oxidant, SOD and GSH-Px activity were increased by supplementation of walnut and significantly higher in HF + 40% compared to the normal group. The Melondialdehyde (MDA) or lipid peroxidation status was increased in HF + 20% walnut to HF + 30% walnut as compared to normal and HF-fed diet but reduced by HF + 40% walnut. These results suggest that walnut intake could be effective to protect from oxidative stress by lowering the lipid peroxidation by improving antioxidant defense system in rats fed high-fat diet.展开更多

Te...The preferential flow plays a vital role on the infiltration of irrigation or rainfall. The objective of this study was to quantify preferential flow in the processing of irrigation infiltration in the field scale. Tests of different initial soil water contents and irrigation intensities were conducted using Brilliant Blue FCF(C.I.42090) dye tracer in Luancheng County of the North China Plain. The results showed that the percentages of infiltration by the preferential flow for irrigation depth of 25, 50, and 75 mm were 16.67%, 43.67%, and 34.17%, with 19.72%, 61.42%, 66.64% of dyed areas in the soil profile, respectively, which indicated that preferential flow was enhanced with increasing irrigation intensity, but reduced when the irrigation intensity was over 50 mm. The percentages of preferential flow for 75 and 180 mm previous irrigation producing different initial soil water contents were 23.26% and 18.97%, with 53.23% and 39.94% of dyed areas in the soil profile, respectively. Compared with the 75 mm without previous irrigation, the results indicated that higher initial soil water contents restrained the preferential flow in the field. Therefore, intermittent irrigation and low irrigation intensity patterns, and larger depth of plowing would be suggested to reduce the preferential flow which would increase the soil water utilization efficiency and reduce pollution risk of pesticide and fertilizer to groundwater.展开更多
Objective Due to the complicated compounds and the synergistic effect of multi-compounds,the quality control and assessment of Chinese materia medica(CMM)encounters a great challenge about how to identify the key comp...Objective Due to the complicated compounds and the synergistic effect of multi-compounds,the quality control and assessment of Chinese materia medica(CMM)encounters a great challenge about how to identify the key compounds,which are directly correlated with its efficacy and safety.On the guidance of study on quality marker(Q-Marker),identification of Q-Markers was performed from Hedan Tablet(HDT)by the aid of the“spider-web”mode and hepatotoxicity evaluation derived from our previous researches and literatures.Methods By the established ultra performance liquid chromatography with photodiode array detector(UPLC-PDA)method,online UPLC-DPPH·and offline antioxidant assay,21 candidate compounds of HDT were systematically investigated and comprehensively evaluated by the“spider-web”mode for them properties of Q-Marker based on“content-stability-activity”.In addition,the Q-Markers related with hepatotoxicity based on our previous researches and literatures were identified.Results Salvianolic acid B(SaB),quercetin-3-O-glucuronide(Qug),isoquercitrin(IQ)and hyperoside(Hyp)were adopted as the preferable Q-Markers of HDT according to the shaded area(A)of tested compounds in“spider-web”mode.Psoralen(Ps),isopsoralen(IP),psoralenoside(PO)and isopsoralenoside(IPO)were also strongly recommended as Q-Markers closely related with safety by considering hepatotoxicity of the accumulated Ps and IP and conversion between glycoside(PO and IPO)and aglycone(Ps and IP).Conclusion This study provided scientific evidence for quality control and assessment of HDT,and also provided a meaningful reference for application of Q-Markers in CMM.展开更多
Background:Different oscillations of brain networks could carry different dimensions of brain integration.We aimed to investigate oscillation-specific nodal alterations in patients with Parkinson’s disease(PD)across ...Background:Different oscillations of brain networks could carry different dimensions of brain integration.We aimed to investigate oscillation-specific nodal alterations in patients with Parkinson’s disease(PD)across early stage to middle stage by using graph theory-based analysis.Methods:Eighty-eight PD patients including 39 PD patients in the early stage(EPD)and 49 patients in the middle stage(MPD)and 36 controls were recruited in the present study.Graph theory-based network analyses from three oscillation frequencies(slow-5:0.01–0.027 Hz;slow-4:0.027–0.073 Hz;slow-3:0.073–0.198 Hz)were analyzed.Nodal metrics(e.g.nodal degree centrality,betweenness centrality and nodal efficiency)were calculated.Results:Our results showed that(1)a divergent effect of oscillation frequencies on nodal metrics,especially on nodal degree centrality and nodal efficiency,that the anteroventral neocortex and subcortex had high nodal metrics within low oscillation frequencies while the posterolateral neocortex had high values within the relative high oscillation frequency was observed,which visually showed that network was perturbed in PD;(2)PD patients in early stage relatively preserved nodal properties while MPD patients showed widespread abnormalities,which was consistently detected within all three oscillation frequencies;(3)the involvement of basal ganglia could be specifically observed within slow-5 oscillation frequency in MPD patients;(4)logistic regression and receiver operating characteristic curve analyses demonstrated that some of those oscillation-specific nodal alterations had the ability to well discriminate PD patients from controls or MPD from EPD patients at the individual level;(5)occipital disruption within high frequency(slow-3)made a significant influence on motor impairment which was dominated by akinesia and rigidity.Conclusions:Coupling various oscillations could provide potentially useful information for large-scale network and progressive oscillation-specific nodal alterations were observed in PD patients across early to middle stages.展开更多
Forecasting the intensity of typhoons is a difficult problem in numerical weather prediction.It is subject to many factors,among which the selection of physical parameterization schemes for the model is a hot topic of...Forecasting the intensity of typhoons is a difficult problem in numerical weather prediction.It is subject to many factors,among which the selection of physical parameterization schemes for the model is a hot topic of research.In this study,the effects of horizontal mixing length(represented by h_diff)and planetary boundary layer(PBL)schemes were investigated.Six idealized and four operational sensitivity experiments were designed based on simulation of the typhoon Lingling,which occurred over the western Pacific in 2019,using the Hurricane Weather Research and Forecasting model.The results of the idealized experiments showed that,as h_diff was increased,the slope of the typhoon eye area also increased,and its intensity became stronger.On the other hand,the results of the sensitivity experiments indicated that the intensity of the simulated typhoon was sensitive to the choice of PBL scheme,with the forecast bias of the QNSE(Quasi-Normal Scale Elimination)scheme being smaller than that of the GFDL(Geophysical Fluid Dynamics Laboratory)scheme.Angular momentum budget analyses indicated that,when increasing the h_diff,the convergence of angular momentum was larger in the boundary layer,which led to a faster spin-up of the vortex,further increasing the intensity of the typhoon.From the calculated horizontal and vertical vortex spread it was found that,when the h_diff was increased,the corresponding horizontal and vertical diffusion eddies also showed an increasing trend,which was also the reason for the strengthening of the typhoon.Meanwhile,the forecast bias decreased significantly with increasing horizontal mixing length when using the same PBL scheme.展开更多

文摘The exponential growth of bioinformatics tools in recent years has posed challenges for scientists in selecting the most suitable one for their data analysis assignments.Therefore,to aid scientists in making informed choices,a community-based platform that indexes and rates bioinformatics tools is urgently needed.In this study,we introduce Bio Treasury(http://biotreasury.rjmart.cn),an integrated communitybased repository that provides an interactive platform for users and developers to share their experiences in various bioinformatics tools.Bio Treasury offers a comprehensive collection of well-indexed bioinformatics software,tools,and databases,totaling over 10,000 entries.In the past two years,we have continuously improved and maintained Bio Treasury,adding several exciting features,including creating structured homepages for every tool and user,a hierarchical category of bioinformatics tools and classifying tools using large language model(LLM).Bio Treasury streamlines the tool submission process with intelligent auto-completion.Additionally,Bio Treasury provides a wide range of social features,for example,enabling users to participate in interactive discussions,rate tools,build and share tool collections for the public.We believe Bio Treasury can be a valuable resource and knowledge-sharing platform for the biomedical community.It empowers researchers to effectively discover and evaluate bioinformatics tools,fostering collaboration and advancing bioinformatics research.

文摘The choice of cathode and anode materials for electrochromic devices plays a key role in the performance of electrochromic smart windows.In this research,WO_(3)/Ag and TiO_(2)/NiO composite thin films were separately prepared by the hydrothermal method combined with electrodeposition.The electrochromic properties of the single WO_(3) thin film were optimized,and TiO_(2)/NiO composite films showed better electrochromic performance than that of the single NiO film.WO_(3)/Ag and TiO_(2)/NiO composite films with excellent electrochromic properties were respectively chosen as the cathode and the anode to construct a WO_(3)/Ag‒TiO_(2)/NiO electrochromic device.The response time(tc=4.08 s,tb=1.08 s),optical modulation range(35.91%),and coloration efficiency(30.37 cm^(2)·C^(-1))of this electrochromic device are better than those of WO_(3)-NiO and WO_(3)/Ag-NiO electrochromic devices.This work provides a novel research idea for the performance enhancement of electrochromic smart windows.

文摘The preferential flow plays a vital role on the infiltration of irrigation or rainfall. The objective of this study was to quantify preferential flow in the processing of irrigation infiltration in the field scale. Tests of different initial soil water contents and irrigation intensities were conducted using Brilliant Blue FCF(C.I.42090) dye tracer in Luancheng County of the North China Plain. The results showed that the percentages of infiltration by the preferential flow for irrigation depth of 25, 50, and 75 mm were 16.67%, 43.67%, and 34.17%, with 19.72%, 61.42%, 66.64% of dyed areas in the soil profile, respectively, which indicated that preferential flow was enhanced with increasing irrigation intensity, but reduced when the irrigation intensity was over 50 mm. The percentages of preferential flow for 75 and 180 mm previous irrigation producing different initial soil water contents were 23.26% and 18.97%, with 53.23% and 39.94% of dyed areas in the soil profile, respectively. Compared with the 75 mm without previous irrigation, the results indicated that higher initial soil water contents restrained the preferential flow in the field. Therefore, intermittent irrigation and low irrigation intensity patterns, and larger depth of plowing would be suggested to reduce the preferential flow which would increase the soil water utilization efficiency and reduce pollution risk of pesticide and fertilizer to groundwater.

文摘Objective Due to the complicated compounds and the synergistic effect of multi-compounds,the quality control and assessment of Chinese materia medica(CMM)encounters a great challenge about how to identify the key compounds,which are directly correlated with its efficacy and safety.On the guidance of study on quality marker(Q-Marker),identification of Q-Markers was performed from Hedan Tablet(HDT)by the aid of the“spider-web”mode and hepatotoxicity evaluation derived from our previous researches and literatures.Methods By the established ultra performance liquid chromatography with photodiode array detector(UPLC-PDA)method,online UPLC-DPPH·and offline antioxidant assay,21 candidate compounds of HDT were systematically investigated and comprehensively evaluated by the“spider-web”mode for them properties of Q-Marker based on“content-stability-activity”.In addition,the Q-Markers related with hepatotoxicity based on our previous researches and literatures were identified.Results Salvianolic acid B(SaB),quercetin-3-O-glucuronide(Qug),isoquercitrin(IQ)and hyperoside(Hyp)were adopted as the preferable Q-Markers of HDT according to the shaded area(A)of tested compounds in“spider-web”mode.Psoralen(Ps),isopsoralen(IP),psoralenoside(PO)and isopsoralenoside(IPO)were also strongly recommended as Q-Markers closely related with safety by considering hepatotoxicity of the accumulated Ps and IP and conversion between glycoside(PO and IPO)and aglycone(Ps and IP).Conclusion This study provided scientific evidence for quality control and assessment of HDT,and also provided a meaningful reference for application of Q-Markers in CMM.

文摘Background:Different oscillations of brain networks could carry different dimensions of brain integration.We aimed to investigate oscillation-specific nodal alterations in patients with Parkinson’s disease(PD)across early stage to middle stage by using graph theory-based analysis.Methods:Eighty-eight PD patients including 39 PD patients in the early stage(EPD)and 49 patients in the middle stage(MPD)and 36 controls were recruited in the present study.Graph theory-based network analyses from three oscillation frequencies(slow-5:0.01–0.027 Hz;slow-4:0.027–0.073 Hz;slow-3:0.073–0.198 Hz)were analyzed.Nodal metrics(e.g.nodal degree centrality,betweenness centrality and nodal efficiency)were calculated.Results:Our results showed that(1)a divergent effect of oscillation frequencies on nodal metrics,especially on nodal degree centrality and nodal efficiency,that the anteroventral neocortex and subcortex had high nodal metrics within low oscillation frequencies while the posterolateral neocortex had high values within the relative high oscillation frequency was observed,which visually showed that network was perturbed in PD;(2)PD patients in early stage relatively preserved nodal properties while MPD patients showed widespread abnormalities,which was consistently detected within all three oscillation frequencies;(3)the involvement of basal ganglia could be specifically observed within slow-5 oscillation frequency in MPD patients;(4)logistic regression and receiver operating characteristic curve analyses demonstrated that some of those oscillation-specific nodal alterations had the ability to well discriminate PD patients from controls or MPD from EPD patients at the individual level;(5)occipital disruption within high frequency(slow-3)made a significant influence on motor impairment which was dominated by akinesia and rigidity.Conclusions:Coupling various oscillations could provide potentially useful information for large-scale network and progressive oscillation-specific nodal alterations were observed in PD patients across early to middle stages.

文摘Forecasting the intensity of typhoons is a difficult problem in numerical weather prediction.It is subject to many factors,among which the selection of physical parameterization schemes for the model is a hot topic of research.In this study,the effects of horizontal mixing length(represented by h_diff)and planetary boundary layer(PBL)schemes were investigated.Six idealized and four operational sensitivity experiments were designed based on simulation of the typhoon Lingling,which occurred over the western Pacific in 2019,using the Hurricane Weather Research and Forecasting model.The results of the idealized experiments showed that,as h_diff was increased,the slope of the typhoon eye area also increased,and its intensity became stronger.On the other hand,the results of the sensitivity experiments indicated that the intensity of the simulated typhoon was sensitive to the choice of PBL scheme,with the forecast bias of the QNSE(Quasi-Normal Scale Elimination)scheme being smaller than that of the GFDL(Geophysical Fluid Dynamics Laboratory)scheme.Angular momentum budget analyses indicated that,when increasing the h_diff,the convergence of angular momentum was larger in the boundary layer,which led to a faster spin-up of the vortex,further increasing the intensity of the typhoon.From the calculated horizontal and vertical vortex spread it was found that,when the h_diff was increased,the corresponding horizontal and vertical diffusion eddies also showed an increasing trend,which was also the reason for the strengthening of the typhoon.Meanwhile,the forecast bias decreased significantly with increasing horizontal mixing length when using the same PBL scheme.
