Ferroptosis:a potential therapeutic target for stroke

Ferroptosis is a form of regulated cell death characterized by massive iron accumulation and iron-dependent lipid peroxidation,differing from apoptosis,necroptosis,and autophagy in several aspects.Ferroptosis is regarded as a critical mechanism of a series of pathophysiological reactions after stroke because of iron overload caused by hemoglobin degradation and iron metabolism imbalance.In this review,we discuss ferroptosis-related metabolisms,important molecules directly or indirectly targeting iron metabolism and lipid peroxidation,and transcriptional regulation of ferroptosis,revealing the role of ferroptosis in the progression of stroke.We present updated progress in the intervention of ferroptosis as therapeutic strategies for stroke in vivo and in vitro and summarize the effects of ferroptosis inhibitors on stroke.Our review facilitates further understanding of ferroptosis pathogenesis in stroke,proposes new targets for the treatment of stroke,and suggests that more efforts should be made to investigate the mechanism of ferroptosis in stroke. 展开更多

Genetic modification of miR-34a enhances efficacy of transplanted human dental pulp stem cells after ischemic stroke 被引量:1

Human dental pulp stem cells(hDPSCs) promote recovery after ischemic stro ke;however,the therapeutic efficacy is limited by the poor survival of transplanted cells.For in vitro expe riments in the present study,we used oxygen-glucose deprivation/reoxygenation in hDPSCs to mimic cell damage induced by ischemia/reperfusion.We found that miRNA-34a-5p(miR-34a) was elevated under oxygen-glucose deprivation/reoxygenation conditions in hDPSCs.Inhibition of miR-34a facilitated the prolife ration and antioxidant capacity and reduced the apoptosis of hDPSCs.Moreove r,dual-luciferase reporter gene assay showed WNT1and SIRT1 as the targets of miR-34a.In miR-34a knockdown cell lines,WNT1 suppression reduced cell prolife ration,and SIRT1 suppression decreased the antioxidant capacity.Togethe r,these results indicated that miR-34a regulates cell prolife ration and antioxidant stress via targeting WNT1 and SIRT1,respectively.For in vivo expe riments,we injected genetically modified hDPSCs(anti34a-hDPSCs) into the brains of mice.We found that anti34a-hDPSCs significantly inhibited apoptosis,reduced cerebral edema and cerebral infarct volume,and improved motor function in mice.This study provides new insights into the molecular mechanism of the cell prolife ration and antioxidant capacity of hDPSCs,and suggests a potential gene that can be targeted to improve the survival rate and efficacy of transplanted hDPSCs in brain after ischemic stroke. 展开更多

目的研究亚甲蓝介导的光动力疗法对人皮肤鳞状细胞癌A431细胞株的杀伤作用以及其诱导凋亡的机制,为亚甲蓝光动力临床应用提供理论依据以及最佳治疗参数。方法采用亚甲蓝与人皮肤鳞状细胞癌A431细胞株共同孵育,经630 nm红光照射。采用CC... 目的研究亚甲蓝介导的光动力疗法对人皮肤鳞状细胞癌A431细胞株的杀伤作用以及其诱导凋亡的机制,为亚甲蓝光动力临床应用提供理论依据以及最佳治疗参数。方法采用亚甲蓝与人皮肤鳞状细胞癌A431细胞株共同孵育,经630 nm红光照射。采用CCK-8法检测细胞生长的抑制率,确定培育时间、亚甲蓝浓度对A431细胞株的抑制作用。采用免疫组化法检测A431细胞在亚甲蓝光动力作用前后细胞色素c(Cyt-c)在细胞内的分布情况。最后,采用Western-blot方法检测亚甲蓝光动力治疗后细胞凋亡过程中,线粒体凋亡途径相关蛋白Bcl-2和Bax的表达情况。结果CCK8法检测结果显示亚甲蓝光动力组对A431细胞具有杀伤作用,其杀伤作用与亚甲蓝的药物浓度以及亚甲蓝光动力作用后的孵育时间具有密切相关。Cyt-c免疫荧光结果显示亚甲蓝光光动力作用后,细胞凋亡率空白对照0%,光照对照组8%,亚甲蓝对照组12%,光照后1 h 22%,光照后2 h 26%,光照后4 h 31%,光照后6 h 54%。Western blot的检测结果显示亚甲蓝光动力作用激活A431细胞线粒体凋亡途径,启动Bcl-2家族参与凋亡过程。随着亚甲蓝光动力作用后孵育时间的延长,促凋亡蛋白Bax上调,抗凋亡蛋白Bcl-2下调。结论亚甲蓝光动力能明显抑制A431细胞的增殖,并诱导其发生凋亡。凋亡的发生与启动细胞色素c相关的线粒体凋亡途径相关。 展开更多

Based on the elastic dislocation theory, multilayered crustal model, and rupture model obtained by seismic waveform inversion, we calculated the coand post-seismic surface deformation and gravity changes caused by the Yushu M W 6.9 earthquake occurred on April 14, 2010. The observed GPS velocity field and gravity field in Yushu areas are disturbed by the coand post-seismic effects induced by Yushu earthquake, thus the theoretical coand post-seismic deformation and gravity changes will provide important modification for the background tectonic movement of Yushu and surrounding regions. The time relaxation results show that the influences of Yushu earthquake on Yushu and surrounding areas will last as long as 30 to 50 years. The maximum horizontal displacement, vertical uplift and settlement are about 1.96, 0.27 and 0.16 m, respectively, the maximal positive and negative value of gravity changes are 8.892×10-7 m·s-2 and -4.861×10-7 m·s-2 , respectively. Significant spatial variations can be found on the coand post-seismic effects: The co-seismic effect mainly concentrates in the region near the rupture fault, while viscoelastic relaxation mostly acts on the far field. Therefore, when using the geodetic data to research tectonic motion, we should not only consider the effect of co-seismic caused by earthquake, but also pay attention to the effect of viscoelastic relaxation. 展开更多

On April 20,2013,an earthquake with magnitude 7.0 occurred in the southwest of the Longmenshan fault system in and around Lushan County,Sichuan Province,China.This devastating earthquake killed hundreds of people,injured 10 thousand others,and collapsed countless buildings.In order to analyze the potential risk of this big earthquake,we calculate the co-and post-seismic surface deformation and gravity changes of this event.In this work,a multilayered crustal model is designed,and the elastic dislocation theory is utilized to calculate the co-and post-seismic deformations and gravity changes.During the process,a rupture model obtained by seismic waveform inversion(Liu et al.Sci China Earth Sci 56(7):1187–1192,2013)is applied.The time-dependent relaxation results show that the influences on Lushan and its surrounding areas caused by the MS7.0 Lushan earthquake will last as long as 10 years.The maximum horizontal displacement,vertical uplift,and settlement are about 5 cm,21.24 cm,and 0.16 m,respectively;the maximal positive and negative values of gravity changes are 45 and-0.47 lGal,respectively.These results may be applied to evaluate the long-term potential risk caused by this earthquake and to provide necessary information for post-earthquake reconstruction. 展开更多

At GMT time 13:19, August 8, 2017, an Ms7.0 earthquake struck the Jiuzhaigou region in Sichuan Province, China,causing severe damages and casualties. To investigate the source properties, seismogenic structures, and seismic hazards, we systematically analyzed the tectonic environment, crustal velocity structure in the source region, source parameters and rupture process, Coulomb failure stress changes, and 3-D features of the rupture plane of the Jiuzhaigou earthquake. Our results indicate the following:(1) The Jiuzhaigou earthquake occurred on an unmarked fault belonging to the transition zone of the east Kunlun fault system and is located northwest of the Huya fault.(2) Both the mainshock and aftershock rupture zones are located in a region where crustal seismic velocity changes dramatically. Southeast to the source region, shear wave velocity at the middle to lower crust is significantly low, but it rapidly increases northeastward and lies close to the background velocity across the rupture fault.(3) The aftershock zone is narrow and distributes along the northwest-southeast trend, and most aftershocks occur within a depth range of 5–20 km.(4) The focal mechanism of the Jiuzhaigou earthquake indicates a left-lateral strike-slip fault, with strike, dip, and rake angles of 152°, 74° and 8°, respectively. The hypocenter depth measures 20 km, whereas the centroid depth is about 6 km. The co-seismic rupture mainly concentrates at depths of 3–13 km, with a moment magnitude(M_w) of 6.5.(5) The co-seismic rupture also strengthens the Coulomb failure stress at the two ends of the rupture fault and the east segment of the Tazang fault. Aftershocks relocation results together with geological surveys indicate that the causative fault is a near vertical fault with notable spatial variations: dip angle varies within 66°–89° from northwest to southeast and the average dip angle measures ~84°. The results of this work are of fundamental importance for further studies on the source characteristics, tectonic environment, and seismic hazard evaluation of the Jiuzhaigou earthquake. 展开更多

On April 20, 2013, an M_s 7.0 earthquake struck Lushan County in Sichuan Province, China, and caused serious damage to the source region. We investigated the rupture process of the M_s7.0 Lushan earthquake by jointly inverting waveforms of teleseismic P waveforms and local strong motion records as well as static GPS observations. The inverted results indicate that the rupture of this earthquake was dominated by the failure of an asperity with a triangular shape and that the main shock was dominated by thrust slip. The earthquake released a total seismic moment of 1.01× 10^(19)Nm, with 92% of it being released during the first 11 s. The rupture had an average slip of 0.9 m and produced an average stress drop of 1.8 MPa. Compared with our previous work that was based mainly on a unique dataset, this joint inversion result is more consistent with field observations and the distribution of aftershock zones. 展开更多

Seismic and field observations indicate that the Mw7.4 Maduo earthquake ruptured the Jiangcuo fault,which is a secondary fault~85 km south of the northern boundary of the Bayan Hor block in western China.The kinematic characteristics of the Jiangcuo fault can shed lights on the seismogenic mechanism of this earthquake.Slip rate is one of the key parameters to describe the kinematic features of a fault,which can also provide quantitative evidences for regional seismic hazard assessments.However,due to lack of effective observations,the slip rate of the Jiangcuo fault has not been studied quantitatively.In this study,we consider the interaction between the Jiangcuo fault and the eastern Kunlun fault,and estimate the slip rates of the two faults using the interseismic GPS observations across the seismogenic region.The inferred results show that the slip rates of the Jiangcuo fault and the Tuosuo Lake segment of the Kunlun fault are 1.2±0.8 and 5.4±0.3 mm a^(-1),respectively.Combining the slip rate with the average slip inferred from the coseismic slip model,the earthquake recurrence interval of the Jiangcuo fault is estimated to be 1800700+3700 years(1100–5500 years).Based on the results derived from previous studies,as well as calculations in this study,we infer that the slip rate of the Kunlun fault may decrease gradually from the Tuosuo Lake segment to the eastern tip.The Jiangcuo fault and its adjacent parallel secondary faults may have absorbed the relative motion of blocks together with the Kunlun fault. 展开更多

On May 22 nd,2021,an MS7.4 earthquake occurred near the Maduo county of the Qinghai Province,China,within the Bayan Har Block.Seismic activities have been intense in this block,thus whether the Maduo Earthquake will bring subsequent seismic hazards to its surrounding regions raises wide concerns.In this paper,we first calculated the Coulomb failure stress changes caused by the Maduo Earthquake on nearby faults,and estimated how much these faults are brought closer or further from their next failures based on their stressing rates.Next,we combined the Coulomb failure stress changes with the rate-state frictional law to estimate the seismicity rate in the study region in the next decade.A declustered catalogue before the Maduo Earthquake was adopted to calculate background seismicity rate,and rate-state parameters are constrained by fault slip rates.Our results show that the Maduo Earthquake increases stress accumulations in the northwestern portion of the Qingshuihe fault(0.02 MPa at maximum),the two ends of the Kunlun Mountain Pass-Jiangcuo fault(0.01 MPa at maximum),and the northwestern portion of the Maduo-Gande fault(on average~0.09 MPa),and seismicity rates are expected to increase near these faults.What is especially worth noting is the seismic hazard in the region extending from the eastern end of the Kunlun Mountain Pass-Jiangcuo fault to the Maqin-Maqu seismic gap on the Eastern Kunlun fault,which is calculated to have experienced a maximum stress increase of 0.67 MPa after the Maduo Earthquake.On the other hand,stress accumulations are reduced in the southern end of the Elashan fault,the Eastern Kunlun fault segment to the west of Maduo,and the northwestern portion of the Dari fault.Seismic hazards are expected to be low in these regions.For the study region as a whole,the probability of an M≥6 earthquake taking place in the next decade is estimated to be 59%,about twice the value calculated for the time period before the Maduo Earthquake. 展开更多

The point source parameters of the April 12, 2012 Mw 7.0 Santa Isabel, Mexico, earthquake indicated by teleseismic P and SH waveforms obtained by a means of traditional cut and paste(CAP) method show that the best double-couple solution of this event is: 37°/127°, 90°/81° and-9°/-180° for strike, dip and rake, respectively. Its centroid depth is 13 km. Global teleseismic waveform data exhibit that the rupture of the earthquake initiated at a focal depth of 13 km and propagated southeastward with a relatively slow rupture velocity(about 1.8 km/s on average). The maximum slip occurred at 30 km southeast of the hypocenter, with the peak slip of 3.57 m and total seismic moment of whole fault up to 3.98×1019 N·m. These observations provide some insight into properties, co- or post-seismic deformation and coulomb stress changes of future earthquake in this area. 展开更多
