The autotetraploid Carassius auratus(4nRR,4n=200,RRRR)is derived from whole-genome duplication of Carassius auratus red var.(RCC,2n=100,RR).In the current study,we demonstrated that chromatophores and pigment changes directly caused the coloration and variation of 4nRR skin(red in RCC,brownish-yellow in4nRR).To further explore the molecular mechanisms underlying coloration formation and variation in 4nRR,we performed transcriptome profiling and molecular functional verification in RCC and 4nRR.Results revealed that scarb1,associated with carotenoid metabolism,underwent significant down-regulation in 4nRR.Efficient editing of this candidate pigment gene provided clear evidence of its significant role in RCC coloration.Subsequently,we identified four divergent scarb1 homeologs in 4nRR:two original scarb1 homeologs from RCC and two duplicated ones.Notably,three of these homeologs possessed two highly conserved alleles,exhibiting biased and allelespecific expression in the skin.Remarkably,after precise editing of both the original and duplicated scarb1homeologs and/or alleles,4nRR individuals,whether singly or multiply mutated,displayed a transition from brownishyellow skin to a cyan-gray phenotype.Concurrently,the proportional areas of the cyan-gray regions displayed a gene-dose correlation.These findings illustrate the subfunctionalization of duplicated scarb1,with all scarb1genes synergistically and equally contributing to the pigmentation of 4nRR.This is the first report concerning the functional differentiation of duplicated homeologs in an autopolyploidfish,substantiallyenrichingour understanding of coloration formation and change within this group of organisms. 展开更多

The flotation mechanisms of molybdenite fines by neutral oils were investigated through microflotation test, turbidity measurements, infrared spectroscopy, and interfacial interaction calculations. The results of the flotation test show that at pH 2-11, the floatability of molybdenite fines in the presence of transformer oil is markedly better than that in the presence of kerosene and diesel oil. The addition of transformer oil, which enhances the floatability of molybdenite fines, promotes the aggregation of molybdenite particles. Fourier transform infrared measurements illustrate that physical interaction dominates the adsorption mechanism of neutral oil on molybdenite. Interfacial interaction calculations indicate that hydrophobic attraction is the crucial force that acts among the oil collector, water, and molybdenite. Strong hydrophobic attraction between the oily collector and water provides the strong dispersion capability of the collector in water. Furthermore, the dispersion capability of the collector, not the interaction strength between the oily collectors and molybdenite, has a highly significant role in the flotation system of molybdenite fines. Our findings provide insights into the mechanism of molybdenite flotation. 展开更多

The total synthesis of benzyl-substituted flavone derivatives via the acidic rearrangement of benzyl groups in flavone benzyl ethers, and the complicated regioselectivity of the rearrangement were reported. The regioselectivity was proposed to be determined by the steric hindrance as well as the ease of electrophilic substitution reaction for benzyl cations at different positions of corresponding debenzylated flavone compounds. 展开更多

A series of new combretastatin-A4 analogs were synthesized,in which a six-membered ring connects the linking bridge and A ring,and their tumor cell growth and tubulin-polymerization inhibitory activity were evaluated.These compounds appear to be potential tubulin-polymerization inhibitors.Compounds lb with amino substituted on position 3 of B ring conferred optimal bioactivity,higher than that of the lead compound 22 b and equivalent to that of CA-4.The binding modes of these compounds to tubulin were obtained by molecular docking,which can explain the structure-activity relationship.The studies presented here provide a new structural type for the development of novel antitumor agents. 展开更多
