The high mortality rate associated with gastric cancer(GC)has resulted in an urgent need to identify novel therapeutic targets for GC.This study aimed to investigate whether GAIP interacting protein,C terminus 1(GIPC1)represents a therapeutic target and its regulating mechanism in GC.GIPC1 expression was elevated in GC tissues,liver metastasis tissues,and lymph node metastases.GIPC1 knockdown or GIPC1 blocking peptide blocked the platelet-derived growth factor receptor(PDGFR)/PI3K/AKT signaling pathway,and inhibited the proliferation and migration of GC cells.Conversely,GIPC1 overexpression markedly activated the PDGFR/PI3K/AKT signaling pathway,and promoted GC cell proliferation and migration.Furthermore,platelet-derived growth factor subunit BB(PDGF-BB)cytokines and the AKT inhibitor attenuated the effect of differential GIPC1 expression.Moreover,GIPC1 silencing decreased tumor growth and migration in BALB/c nude mice,while GIPC1 overexpression had contrasting effects.Taken together,our findings suggest that GIPC1 functions as an oncogene in GC and plays a central role in regulating cell proliferation and migration via the PDGFR/PI3K/AKT signaling pathway. 展开更多

BACKGROUND Transarterial chemoembolization(TACE)combined with microwave ablation(MWA)is an effective treatment strategy for patients with advanced gastric cancer and liver metastasis.However,it may cause severe postoperative pain and inflammatory responses.The paravertebral block(PVB)is a regional anesthetic technique that provides analgesia to the thoracic and abdominal regions.AIM To evaluate the effect of PVB on postoperative analgesia and inflammatory response in patients undergoing TACE combined with MWA for advanced gastric cancer and liver metastasis.METHODS Sixty patients were randomly divided into PVB and control groups.The PVB group received ultrasound-guided PVB with 0.375%ropivacaine preoperatively,whereas the control group received intravenous analgesia with sufentanil.The primary outcome was the visual analog scale(VAS)score for pain at 6 h,12 h,24 h,and 48 h after the procedure.Secondary outcomes were the dose of sufentanil used,incidence of adverse events,and levels of inflammatory markers(white blood cell count,neutrophil percentage,C-reactive protein,and procalcitonin)before and after the procedure.RESULTS The PVB group had significantly lower VAS scores at 6 h,12 h,24 h,and 48 h after the procedure compared with the control group(P<0.05).The PVB group also had a significantly lower consumption of sufentanil and a lower incidence of nausea,vomiting,and respiratory depression than did the control group(P<0.05).Compared with the control group,the PVB group had significantly lower levels of inflammatory markers 24 h and 48 h after the procedure(P<0.05).CONCLUSION PVB can effectively reduce postoperative pain and inflammatory responses and improve postoperative comfort and recovery in patients with advanced gastric cancer and liver metastasis treated with TACE combined with MWA. 展开更多

BACKGROUND Congenital adrenal hyperplasia(CAH),which is caused by a mutation of the steroidogenic acute regulatory(StAR)gene.Affected patients are usually characterized by adrenal insufficiency in the first year of life,salt loss,glucocorticoid and mineralocorticoid deficiency,and female external genitalia,regardless of chromosomal karyotype.Patients with non-classical lipoid CAH usually develop glucocorticoid deficiency and mild mineralocorticoid deficiency at 2-4 years of age.CASE SUMMARY Herein,We report the case of a woman with non-classic lipoid CAH combined with Graves’disease.Her chromosome karyotype was 46,XX,and highthroughput sequencing revealed two missense variants in the StAR gene:c.229C>T(p.Q77X)and c.814C>T(p.R272C),which were inherited from both parents(non-close relatives).The patient was treated for Graves’disease in a timely manner and the dosage of glucocorticoid was adjusted during the treatment of Graves’disease.CONCLUSION This is the first case of non-classic lipoid CAH combined with Graves’disease reported in the Chinese population.In addition to conventional glucocorticoid replacement therapy,timely adjustments were made to the dosages of thyroid hormone and glucocorticoid to avoid adrenal crisis as a consequence of the increased demand and accelerated metabolism of glucocorticoids when the patient was diagnosed with Graves’disease. 展开更多

Partially Diffusive Helium-Silica Compound under High Pressure

Helium is the second most abundant element in the universe, and together with silica, they are important components of giant planets. Exploring the reactivity and state of helium and silica under high pressure is crucial for understanding of the evolution and internal structure of giant planets. Here, using first-principles calculations and crystal structure predictions, we identify four stable phases of a helium-silica compound with seven/eight-coordinated silicon atoms at pressure of 600–4000 GPa, corresponding to the interior condition of the outer planets in the solar system. The density of He Si O2 agrees with current structure models of the planets.This helium-silica compound exhibits a superionic-like helium diffusive state under the high-pressure and hightemperature conditions along the isentropes of Saturn, a metallic fluid state in Jupiter, and a solid state in the deep interiors of Uranus and Neptune. These results show that helium may affect the erosion of the rocky core in giant planets and may help to form a diluted core region, which not only highlight the reactivity of helium under high pressure but also provide evidence helpful for building more sophisticated interior models of giant planets. 展开更多

The world’s coral reefs are threatened by the cumulative impacts of global climate change and local stressors.Driven largely by a desire to understand the interactions between corals and their symbiotic microorganisms,and to use this knowledge to eventually improve coral health,interest in coral microbiology and the coral microbiome has increased in recent years.In this review,we summarize the role of the coral microbiome in maintaining a healthy metaorganism by providing nutrients,support for growth and development,protection against pathogens,and mitigation of environmental stressors.We explore the concept of coral microbiome engineering,that is,precise and controlled manipulation of the coral microbiome to aid and enhance coral resilience and tolerance in the changing oceans.Although coral microbiome engineering is clearly in its infancy,several recent breakthroughs indicate that such engineering is an effective tool for restoration and preservation of these valuable ecosystems.To assist with identifying future research targets,we have reviewed the common principles of microbiome engineering and its applications in improving human health and agricultural productivity,drawing parallels to where coral microbiome engineering can advance in the not-too-distant future.Finally,we end by discussing the challenges faced by researchers and practitioners in the application of microbiome engineering in coral reefs and provide recommendations for future work. 展开更多

Glycogen,amino acids,fatty acids,and other nutrient components affect the flavor and nutritional quality of oysters.Methods based on near-infrared reflectance spectroscopy(NIRS)were developed to rapidly and proximately determine the nutrient content of the Pacific oyster Crassostreagigas.Samples of C.gigas from 19 costal sites were freeze-dried,ground,and scanned for spectral data collection using a Fourier transform NIR spectrometer(Thermo Fisher Scientific).NIRS models of glycogen and other nutrients were established using partial least squares,multiplication scattering correction first-order derivation,and Norris smoothing.The R_(C) values of the glycogen,fatty acids,amino acids,and taurine NIRS models were 0.9678,0.9312,0.9132,and 0.8928,respectively,and the residual prediction deviation(RPD)values of these components were 3.15,2.16,3.11,and 1.59,respectively,indicating a high correlation between the predicted and observed values,and that the models can be used in practice.The models were used to evaluate the nutrient compositions of 1278 oyster samples.Glycogen content was found to be positively correlated with fatty acids and negatively correlated with amino acids.The glycogen,amino acid,and taurine levels of C.gigas cultured in the subtidal and intertidal zones were also significantly different.This study suggests that C.gigas NIRS models can be a cost-effective alternative to traditional methods for the rapid and proximate analysis of various slaughter traits and may also contribute to future genetic and breeding-related studies in Pacific oysters. 展开更多

Using observational data from multiple satellites,we studied seasonal variations of the shape and location of the Luzon cold eddy(LCE)northwest of Luzon Island.The shape and location of the LCE have obvious seasonal variations.The LCE occurs,develops,and disappears from December to April of the next year.During this period,the shape of the LCE changed from a flat ellipse to a circular ellipse,and the change in shape can be reflected by the increase of the ellipticity of the LCE from 0.16 to 0.82.The latitude of center location of the LCE changes from 17.4°N to 19°N,and the change in latitude can reach 1.6°.Further study showed that seasonal variation of the northeast monsoon intensity leads to the change in the shape and location of the LCE.The seasonal variation of the LCE shape can significantly alter the spatial distribution of the thermal front and chlorophyll a northwest of the Luzon Island by geostrophic advection. 展开更多

Rechargeable ion batteries require anode materials with excellent performance,presenting a key challenge for researchers.This paper explores the potential of using two-dimensional dumbbell silicene as an anode material for alkali metal ion batteries through density functional theory(DFT)calculations.Our findings demonstrate that alkali metal ions have negative adsorption energies on dumbbell silicene,and the energy barriers for Li/Na/K ion diffusion are as low as0.032 e V/0.055 e V/0.21 e V,indicating that metal ions can easily diffuse across the entire surface of dumbbell silicene.Additionally,the average open circuit voltages of dumbbell silicene as anode for Li-ion,Na-ion,and K-ion batteries are 0.42 V,0.41 V,and 0.60 V,respectively,with corresponding storage capacities of 716 m Ah/g,622 m Ah/g,and 716 m Ah/g.These results suggest that dumbbell silicene is an ideal anode material for Li-ion,Na-ion,and K-ion batteries,with high capacity,low open circuit voltage,and high ion diffusion kinetics.Moreover,our calculations show that the theoretical capacities obtained using DFT-D2 are higher than those obtained using DFT-D3,providing a valuable reference for subsequent theoretical calculations. 展开更多

This study examined the effects of using bagasse ash in replacement of ordinary Portland cement(OPC)in the treatment of expansive soils.The study concentrated on the compaction characteristics,volume change,compressive strength,splitting tensile strength,microstructure,California bearing ratio(CBR)value,and shear wave velocity of expansive soils treated with cement.Different bagasse ash replacement ratios were used to create soil samples.At varying curing times of 7,14,and 28 days,standard compaction tests,unconfined compressive strength tests,CBR tests,Brazilian split tensile testing,and bender element(BE)tests were carried out.According to X-ray diffraction(XRD)investigations,quartz and crystobalite make up the majority of the minerals in bagasse ash.Bagasse ash contains a variety of grain sizes,including numerous fiber-shaped particles,according to a scanning electronic microscope(SEM)test.For all of the treated specimens with various replacement ratios,the overall additive content has not changed.The results of the Brazilian split tensile tests demonstrate improved tensile strength for all specimens with various replacement proportions.A lower maximum dry density and a greater optimal water content would result from the substitution of bagasse ash.When the replacement ratio is not more than 20%,the CBR values of the parts replaced specimens are even higher than the cement treatments.The results of BE testing on the treated soils show that there is significant stiffness anisotropy but that it steadily diminishes with curing time and replacement ratio.According to the study,bagasse ash is a useful mineral additive,and the best replacement ratio(CBA20)is 20%. 展开更多

The implementation of China’s three-child fertility policy has led to a notable increase in multiple-child families.Notably,firstborn children experience a significant transition from being an only child to a non-only child.This transition is associated with problematic behaviors,affecting their social adjustment,sibling relationships,and family harmony.Although several studies have examined the relationship between parent-child attachment and problem behaviors exhibited byfirstborn children during family transitions,thefindings have been inconsistent.Hence,a meta-analytic study was undertaken to elucidate the inconsistencies in this relationship and explore the moderating factors that may contribute to these discrepancies.Using a systematic literature retrieval and screening method,12 effect sizes were derived from the 10 eligible articles,encompassing a sample size of 5319.The meta-analysis demonstrated a low negative association between parent-child secure attachment and problem behaviors exhibited byfirstborn children during family transitions.Furthermore,the present study investigates potential moderator factors,such as children’s age and geographic region,to gain a more nuanced understanding of the relationship.Consequently,the establishment of parent-child attachment relationships has the potential to mitigate problem behaviors observed infirst-born children during family transitions.The implications of thesefindings indicate that parents can nurture secure attachment bonds with their children by demonstrating sensitive responsiveness,employing positive parenting practices,and fostering emotional availability.These efforts contribute to the cultivation of secure internal working models and positive behavioral manifestations withinfirst-born children,which in turn affect their relationships with siblings. 展开更多

Harsh working environments and wear between blades and other unit components can easily lead to cracks and damage on wind turbine blades.The cracks on the blades can endanger the shafting of the generator set,the tower and other components,and even cause the tower to collapse.To achieve high-precision wind blade crack detection,this paper proposes a crack fault-detection strategy that integratesGated ResidualNetwork(GRN),a fusionmodule and Transformer.Firstly,GRNcan reduce unnecessary noisy inputs that could negatively impact performancewhile preserving the integrity of feature information.In addition,to gain in-depth information about the characteristics of wind turbine blades,a fusionmodule is suggested to implement the information fusion of wind turbine features.Specifically,each fan feature ismapped to a one-dimensional vector with the same length,and all one-dimensional vectors are concatenated to obtain a two-dimensional vector.And then,in the fusion module,the information fusion of the same characteristic variables in the different channels is realized through the Channel-mixing MLP,and the information fusion of different characteristic variables in the same channel is realized through the ColumnmixingMLP.Finally,the fused feature vector is input into the Transformer for feature learning,which enhances the influence of important feature information and improves the model’s anti-noise ability and classification accuracy.Extensive experimentswere conducted on the wind turbine supervisory control and data acquisition(SCADA)data froma domesticwind field.The results show that compared with other state-of-the-artmodels,including XGBoost,LightGBM,TabNet,etc.,the F1-score of proposed gated fusion based Transformer model can reach 0.9907,which is 0.4%-2.09% higher than the comparedmodels.Thismethod provides amore reliable approach for the condition detection and maintenance of fan blades in wind farms. 展开更多

随着化石燃料的消耗,全球碳排放与日俱增,由此引发的温室效应对全球气候变化带来巨大的挑战,温室气体CO_(2)的减排与利用迫在眉睫.利用太阳能、风能等可再生资源生产的电能能够将CO_(2)还原为一系列有附加值的化学品如醇、酸等有机小分... 随着化石燃料的消耗,全球碳排放与日俱增,由此引发的温室效应对全球气候变化带来巨大的挑战,温室气体CO_(2)的减排与利用迫在眉睫.利用太阳能、风能等可再生资源生产的电能能够将CO_(2)还原为一系列有附加值的化学品如醇、酸等有机小分子,人为构造碳循环网络,实现碳中和.本文综述了电催化CO_(2)还原反应(CO_(2)RR)催化剂的调控策略和结构效应.从本征结构和外部结构的角度阐述了催化剂活性位点的调控策略.同时讨论了CO_(2)RR催化剂的结构效应,如串联催化、协同效应和限域催化.关于催化剂的调控策略,从催化剂的本征结构和外部结构两方面总结了近年CO_(2)RR催化剂的研究进展.特定的元素或化合物可以表现出不同的晶体结构,造成局部电子结构差异.而局部电子结构与催化剂的催化性能密切相关,调整晶体结构成为本征结构调控的核心问题,因此本文探讨了如何通过电化学方法和纳米晶体合成调节催化剂的本征结构.与本征结构不同,催化剂的外部结构与活性位点的微环境有关.催化剂的外部结构调节主要基于各种电子结构的协同作用,对活性组分的外部配位结构进行调节.多种高效电催化剂的合成方法,例如掺杂元素、有机分子改性,甚至构建单原子材料都与活性位点的外部结构调节有关.因此,从无机元素掺杂和有机分子改性两方面讨论了外部结构调节对CO_(2)RR过程的影响.与内在结构调节不同,外部结构调节更多地依赖于对CO_(2)RR过程的理解,因此本部分涉及了CO_(2)RR的反应机制.关于催化剂的结构效应,催化剂结构中的晶面、缺陷、空位等结构分因素会影响反应中间步骤进而改变催化活性,由结构变化引发一系列催化效应,如串联催化、协同效应和限域催化.因此,研究催化剂的结构效应可以从机理层面理解CO_(2)RR,以便更好地设计催化剂.本文从基本的中间体出发,明确了*COOH和*OCHO两种中间体对于反应路径和产物趋势的影响.同时讨论了多步催化反应过程中的串联催化机制,在串联过程中,不同的中间体可以在不同的活性位点之间转移.与串联催化不同,协同效应是不同活性位点之间的非线性累积效应.在协同过程中,不同的活性位点可以在一个基元步骤上协同工作.串联机制和协同效应为催化剂设计提供了多种思路.同时讨论了反应中间体限域对CO_(2)RR的调控.最后本文总结了目前CO_(2)RR研究中面临的挑战,包括催化剂的稳定性研究和单原子催化剂的开发,以及机理研究中表征手段的一些欠缺,为设计高效的CO_(2)RR电催化剂提供一定指导,助力实现碳中和. 展开更多

为研究紫花苜蓿在叶片和根系水平上响应干旱胁迫的形态和生理的品种特异性规律,在温室内分析了干旱胁迫下WL363HQ和巨能7紫花苜蓿株高、分枝数、生物量及叶片和根系中丙二醛(MDA)、脯氨酸、抗氧化酶类物质、C、N含量、C/N、稳定性C同位... 为研究紫花苜蓿在叶片和根系水平上响应干旱胁迫的形态和生理的品种特异性规律,在温室内分析了干旱胁迫下WL363HQ和巨能7紫花苜蓿株高、分枝数、生物量及叶片和根系中丙二醛(MDA)、脯氨酸、抗氧化酶类物质、C、N含量、C/N、稳定性C同位素(δ^(13)C)和稳定性N同位素(δ^(15)N)。结果表明:干旱胁迫显著降低了供试品种地上部分和根系的干重及分枝数(P<0.05)。干旱胁迫显著降低了巨能7的株高(P<0.05),但增加了巨能7的根冠比,而WL363HQ的结果与之相反,这说明干旱胁迫下供试品种的株高和根冠比具有品种特异性的规律。干旱胁迫增加了WL363HQ和巨能7叶片和根系中MDA和脯氨酸的含量及抗氧化酶物质的活性,且在器官水平也具有品种特异性规律。干旱胁迫下巨能7叶片的MDA含量显著增加(P<0.05),而在WL363HQ根系中的MDA含量也显著增加(P<0.05)。干旱胁迫下WL363HQ叶片脯氨酸含量、POD和SOD活性,及根系SOD的活性均显著增加(P<0.05),而巨能7仅叶片SOD活性,根系脯氨酸含量、POD活性显著升高(P<0.05)。尽管干旱胁迫对供试品种叶片和根系C、N含量无显著影响(P>0.05),但干旱胁迫显著提高了WL363HQ和巨能7紫花苜蓿根系的δ^(13)C(P<0.05),且WL363HQ叶片的δ^(15)N均显著高于巨能7(P<0.05)。此外,干旱胁迫均显著提高了巨能7叶片和根系的C/N(P<0.05)。干旱胁迫下供试品种C、N代谢参数并没有在叶片和根系中表现出较为明显的品种特异性规律,深层次的机制还有待进一步研究。本研究结果将为进一步掌握紫花苜蓿叶片和根系协同抗旱机制及抗旱丰产紫花苜蓿新品种的选育提供理论依据。 展开更多

目的研究表没食子儿茶素没食子酸酯(epigallocatechin gallate,EGCG)对高脂高糖饮食诱导的肥胖大鼠心肌纤维化影响,探讨EGCG能否通过TGF-β1/Smads信号通路减轻心肌纤维化的程度。方法采用长期高脂高糖饮食形成肥胖大鼠模型,给予EGCG每... 目的研究表没食子儿茶素没食子酸酯(epigallocatechin gallate,EGCG)对高脂高糖饮食诱导的肥胖大鼠心肌纤维化影响,探讨EGCG能否通过TGF-β1/Smads信号通路减轻心肌纤维化的程度。方法采用长期高脂高糖饮食形成肥胖大鼠模型,给予EGCG每天灌胃,治疗4周后,称取各组大鼠体重,Masson染色检测心肌纤维化的程度。生化检测血清中空腹血糖(FBG)、甘油三酯(TG)、总胆固醇(TC)、丙二醛(MDA)和谷胱甘肽(GSH)的含量。Western blotting检测各组大鼠心脏组织中相关蛋白(CollagenⅠ、CollagenⅢ、TGF-β1、Smad2/3、p-Smad2/3、Smad7)的表达水平。结果与正常组比较,模型组大鼠体重增加,给予EGCG治疗后减轻。与正常组比较,模型组大鼠心肌纤维化程度严重,给予EGCG治疗后减轻。与正常组比较,模型组大鼠FBG、TC、TG、MDA高于正常组,给予EGCG治疗后下降。与正常组比较,模型组大鼠GSH活性低于正常组,给予EGCG治疗后升高。与正常组比较,模型组大鼠CollagenⅠ、CollagenⅢ、TGF-β1、Smad2/3、pSmad2/3蛋白表达升高,给予EGCG治疗后下降。与正常组比较,模型组大鼠Smad7蛋白表达低于正常组,给予EGCG治疗后升高。结论EGCG能够改善肥胖大鼠血糖和脂质代谢紊乱,减轻心肌纤维化程度,其机制可能与氧化应激介导的TGF-β1/Smads信号通路有关。 展开更多

Abnormally increased levels of iron in the brain trigger cascade amplification in Alzheimer's disease patients,resulting in neuronal death.This study investigated whether components extracted from the Chinese herbs epimedium herb,milkvetch root and kudzuvine root could relieve the abnormal expression of iron metabolism-related protein in Alzheimer's disease patients.An APPswe/PS1ΔE9 double transgenic mouse model of Alzheimer's disease was used.The intragastric administration of compounds from epimedium herb,milkvetch root and kudzuvine root improved pathological alterations such as neuronal edema,increased the number of neurons,downregulated divalent metal transporter 1 expression,upregulated ferroportin 1 expression,and inhibited iron overload in the cerebral cortex of mice with Alzheimer's disease.These compounds reduced iron overload-induced impairment of the central nervous system,indicating a new strategy for developing novel drugs for the treatment of Alzheimer's disease. 展开更多

Non-alcoholic fatty liver disease(NAFLD) is one of the leading causes of chronic liver diseases around the world due to the modern sedentary and food-abundant lifestyle, which is characterized by excessive fat accumulation in the liver related with causes other than alcohol abuse. It is widely acknowledged that insulin resistance, dysfunctional lipid metabolism, endoplasmic reticulum stress, oxidative stress, inflammation, and apoptosis/necrosis may all contribute to NAFLD. Autophagy is a protective self-digestion of intracellular organelles, including lipid droplets(lipophagy), in response to stress to maintain homeostasis. Lipophagy is another pathway for lipid degradation besides lipolysis. It is reported that impaired autophagy also contributes to NAFLD. Some studies have suggested that the histological characteristics of NAFLD(steatosis, lobular inflammation, and peri-sinusoid fibrosis) might be improved by treatment with traditional Chinese herbal extracts, while autophagy may be induced. This review will provide insights into the characteristics of autophagy in NAFLD and the related role/mechanisms of autophagy induced by traditional Chinese herbal extracts such as resveratrol, Lycium barbarum polysaccharides, dioscin, bergamot polyphenol fraction, capsaicin, and garlic-derived S-allylmercaptocysteine, which may inhibit the progression of NAFLD. Regulation of autophagy/lipophagy with traditional Chinese herbal extracts may be a novel approach for treating NAFLD, and the molecular mechanisms should be elucidated further in the near future. 展开更多
