Wettability of different clay mineral surfaces in shale:Implications from molecular dynamics simulations

Clay minerals mainly exist in nano- and micro-meter sized particles, and the pore structure is complex, which leads to its extremely complex wettability. The surface wettability of clay minerals significantly affects the oil and gas-bearing capacity of shale reservoirs. Therefore, studying the wettability of common clay minerals in shale at the nanoscale is of great significance for shale hydrocarbon exploration and development. In this study, the wetting behavior of water in n-hexane and toluene on different clay mineral surfaces at the nanoscale was systematically studied using Molecular dynamics (MD) simulation. And the influencing factors of wettability were analyzed. Through the analysis of the morphological changes of water, relative concentration of water, RDF and interaction energy, it is concluded that the following order of water wettability on the surfaces of clay minerals: montmorillonite > chlorite > kaolinite > illite. Through the analysis of interaction energy, it is concluded that the hydrophilicity of four clay minerals is stronger than that of lipophilicity. And the main interactions between water and oil and the mineral surfaces were van der Waals force and electrostatic force. In addition, the temperature, liquid hydrocarbon type, and mineralization of water affected the wettability of clay minerals. The concentration of water on the surfaces of montmorillonite, kaolinite, and illite decreased with increasing temperature, and the water wettability decreased. At 298 K, the hydrophilicity of the surfaces of the clay minerals in toluene follows the order montmorillonite > chlorite > kaolinite > illite. The higher the NaHCO3 concentration in water, the weaker the wettability of the clay mineral surfaces to water. By comparing the previous experimental results with the MD simulation results, similar wetting characteristics were obtained, and the reliability of the simulation results was verified. MD simulation was used to explore the water wetting of the surfaces of four clay minerals in a shale reservoir from the micro level. This makes up for the lack of experimental means for clarifying the flow and production mechanisms of shale oil and gas and effectively improves the evaluation technology of shale. 展开更多

The majority of oil and gas resources in the world are related to saline sediments, which mainly occur in sedimentary strata in the form of cap rocks or salt-associated shales. A large number of shale oil resources have been discovered in the saline shale sediments of the Cenozoic terrestrial lake basin in China. The hydrocarbon generation ability and the reservoir capacity of shale control the oil and gas generation. The reservoir capacity is mainly characterized by pore type, structure and porosity. Most of China’s shale oil and gas resources belong to salt-bearing formations. The role of gypsum-salt rocks in the formation and evolution of organic matter (OM) in such formations has received extensive attention. However, systematic understanding is lacking. Research on the pore formation and evolution in shale under the action of gypsum-salt rock sediments is especially weak. Taking the shales in the third member of the Shahejie Formation (Es_(3)) of the Bohai Bay Basin as an example, the influence of halite on the formation and evolution process of pores was studied in this paper. The results show that halite and gypsum minerals were associated with OM, which made them more likely to develop OM pores. The samples with a high halite mineral content (HC) are more developed regarding the pore volume and specific surface area than those with a low HC. The formation of thick salt rocks is influenced by factors of deep thermal brine upwelling, sea erosion and arid environments. The frequent alternation between humid and arid environments led to the outbreak and death of organisms and the precipitation of gypsum-salt rock, which formed the simultaneous deposition of OM and halite minerals. Finally, we have established a model of shale pore evolution under the participation of the gypsum-salt rock, and halite minerals contribute to pore development in both Stage II and Stage IV. This study provides strong microscopic evidence for the pore system formation and evolution in salt-bearing reservoirs. 展开更多

Emodin is an effective component of rhubarb with positive pharmacological effects on human health.However,it is also toxic to different cells or tissues to varying degrees.The effects of emodin on glomerular endothelial cells(GECs)remain to be tested,and the documented works were always performed in vitro and hardly reflect the real physiological situation.To study the effects of emodin on GECs in a biomimetic environment,we utilized a microfluidic chip to assess the physiological reaction of human renal glomerular endothelial cells to various concentrations of emodin in this work.The results showed that emodin caused cytotoxicity,impaired glomerular filtration barrier integrity to macromolecules,and increased barrier permeability in a dose-dependent manner.With the increase in emodin concentration,the concentration of the pro-inflammatory cytokine tumor necrosis factor-α,interleukin(IL)-6,transforming growth factor-β1,and monocyte chemoattractant protein(MCP-1)increased while the production of inflammatory cytokine IL-6 first increased and then decreased with the increase in emodin concentration.Our findings shed new light on emodin-induced nephrotoxicity and provide insights for the application of microfluidic chip devices to reveal drug-cell interactions. 展开更多

Spine degeneration is an aging-related disease,but its molecular mechanisms remain unknown,although elevatedβ-catenin signaling has been reported to be involved in intervertebral disc degeneration.Here,we determined the role ofβ-catenin signaling in spinal degeneration and in the homeostasis of the functional spinal unit(FSU),which includes the intervertebral disc,vertebra and facet joint and is the smallest physiological motion unit of the spine.We showed that pain sensitivity in patients with spinal degeneration is highly correlated withβ-catenin protein levels.We then generated a mouse model of spinal degeneration by transgenic expression of constitutively activeβ-catenin in Col2^(+) cells.We found thatβ-catenin-TCF7 activated the transcription of CCL2,a known critical factor in osteoarthritic pain.Using a lumbar spine instability model,we showed that aβ-catenin inhibitor relieved low back pain.Our study indicates thatβ-catenin plays a critical role in maintaining spine tissue homeostasis,its abnormal upregulation leads to severe spinal degeneration,and its targeting could be an avenue to treat this condition. 展开更多

The etiology of OA includes joint injury, obesity, aging, and heredity. However, the detailed molecular mechanisms of OA initiation and progression remain poorly understood and, currently,there are no interventions available to restore degraded cartilage or decelerate disease progression. The diathrodial joint is a complicated organ and its function is to bear weight, perform physical activity and exhibit a joint-specific range of motion during movement. During OA development, the entire joint organ is affected, including articular cartilage, subchondral bone, synovial tissue and meniscus. A full understanding of the pathological mechanism of OA development relies on the discovery of the interplaying mechanisms among different OA symptoms, including articular cartilage degradation, osteophyte formation, subchondral sclerosis and synovial hyperplasia, and the signaling pathway(s) controlling these pathological processes. 展开更多

To further understand the role of β-catenin in osteoarthritis(OA) development in temporomandibular joint(TMJ), we have generated β-catenin conditional activation mice(β-cat(ex3)^(Agc1CreER))by breeding Agc1-CreER mice with β-catenin^(flox(ex3)/+)mice. Results of histologic analysis showed the progressive TMJ defects in 3-and 6-month-old β-cat(ex3)^(Agc1CreER)mice(tamoxifen induction was performed at 2 weeks of age), including decreased chondrocyte numbers in the superficial layer associated with less Alcian blue staining, increased numbers of hypertrophic chondrocytes in deep layers, and rough articular surface. Compared to the TMJ phenotype of β-cat(ex3)^(Col2CreER)mice, β-cat(ex3)^(Agc1CreER)mice showed much severe morphological defects in the superficial layer of TMJ. This may reflect that Agc1-CreER mice could efficiently target cells in the superficial layer of TMJ. Results of immunostaining showed significantly increased expression of MMP13, Col-X, Adamts4,and Adamts5 in TMJ of β-cat(ex3)^(Agc1CreER)mice. Results of proliferating cell nuclear antigen(PCNA), Ki67, and terminal deoxinucleotidyl transferase-mediated d UTP-fluorescein nick end labeling(TUNEL) staining further demonstrated that cell proliferation was decreased and cell apoptosis was increased in condylar cartilage of β-cat(ex3)^(Agc1CreER)mice. Our findings indicate that abnormal upregulation of β-catenin in TMJ leads to defects assembling to OA-like phenotype, further demonstrating that β-catenin plays a critical role in TMJ pathogenesis. 展开更多

Intervertebral disc(IVD) degeneration(IVDD) is the main cause of low back pain with major social and economic burdens;however, its underlying molecular mechanisms remain poorly defined. Here we show that the focal adhesion protein Kindlin-2 is highly expressed in the nucleus pulposus(NP), but not in the anulus fibrosus and the cartilaginous endplates, in the IVD tissues. Expression of Kindlin-2 is drastically decreased in NP cells in aged mice and severe IVDD patients. Inducible deletion of Kindlin-2 in NP cells in adult mice causes spontaneous and striking IVDD-like phenotypes in lumbar IVDs and largely accelerates progression of coccygeal IVDD in the presence of abnormal mechanical stress. Kindlin-2 loss activates Nlrp3 inflammasome and stimulates expression of IL-1β in NP cells, which in turn downregulates Kindlin-2. This vicious cycle promotes extracellular matrix(ECM) catabolism and NP cell apoptosis. Furthermore, abnormal mechanical stress reduces expression of Kindlin-2, which exacerbates Nlrp3 inflammasome activation, cell apoptosis, and ECM catabolism in NP cells caused by Kindlin-2 deficiency. In vivo blocking Nlrp3 inflammasome activation prevents IVDD progression induced by Kindlin-2 loss and abnormal mechanical stress. Of translational significance, adeno-associated virus-mediated overexpression of Kindlin-2 inhibits ECM catabolism and cell apoptosis in primary human NP cells in vitro and alleviates coccygeal IVDD progression caused by mechanical stress in rat. Collectively, we establish critical roles of Kindlin-2 in inhibiting Nlrp3 inflammasome activation and maintaining integrity of the IVD homeostasis and define a novel target for the prevention and treatment of IVDD. 展开更多

Osteoarthritis(OA)is the most common degenerative joint disease that causes painful swelling and permanent damage to the joints in the body.The molecular mechanisms of OA are currently unknown.OA is a heterogeneous disease that affects the entire joint,and multiple tissues are altered during OA development.To better understand the pathological mechanisms of OA,new approaches,methods,and techniques need to be used to understand OA pathogenesis.In this review,we first focus on the epigenetic regulation of OA,with a particular focus on DNA methylation,histone modification,and microRNA regulation,followed by a summary of several key mediators in OA-associated pain.We then introduce several innovative techniques that have been and will continue to be used in the fields of OA and OA-associated pain,such as CRISPR,scRNA sequencing,and lineage tracing.Next,we discuss the timely updates concerning cell death regulation in OA pathology,including pyroptosis,ferroptosis,and autophagy,as well as their individual roles in OA and potential molecular targets in treating OA.Finally,our review highlights new directions on the role of the synovial lymphatic system in OA.An improved understanding of OA pathogenesis will aid in the development of more specific and effective therapeutic interventions for OA. 展开更多

Microfluidics has been considered as a potential technology to miniaturize the conventional equipments and technologies. It offers advantages in terms of small volume, low cost, short reaction time and highthroughput. The applications in biology and medicine research and related areas are almost the most extensive and profound. With the appropriate scale that matches the scales of cells, microfluidics is well positioned to contribute significantly to cell biology. Cell culture, fusion and apoptosis were successfully performed in microfluidics. Microfluidics provides unique opportunities for rare circulating tumor cells isolation and detection from the blood of patients, which furthers the discovery of cancer stem cell biomarkers and expands the understanding of the biology of metastasis. Nucleic acid amplification in microfluidics has extended to single-molecule, high-throughput and integration treatment in one chip. DNA computer which is based on the computational model of DNA biochemical reaction will come into practice from concept in the future. In addition, microfluidics offers a versatile platform for protein-protein interactions, protein crystallization and high-throughput screening. Although microfluidics is still in its infancy, its great potential has already been demonstrated and will provide novel solutions to the high-throughput applications. 展开更多

Orofacial pain or tenderness is a primary symptom associated with temporomandibular joint(TMJ)disorders(TMDs).To understand the pathological mechanisms underlying TMDs,several mouse models have been developed,including mechanical stimulus-induced TMD and genetic mouse models.However,a lack of feasible approaches for assessing TMD-related nociceptive behaviours in the orofacial region of mice has hindered the in-depth study of TMD-associated mechanisms.This study aimed to explore modifications of three existing methods to analyse nociceptive behaviours using two TMD mouse models:(1)mechanical allodynia was tested using von Frey filaments in the mouse TMJ region by placing mice in specially designed chambers;(2)bite force was measured using the Economical Load and Force(ELF)system;and(3)spontaneous feeding behaviour tests,including eating duration and frequency,were analysed using the Laboratory Animal Behaviour Observation Registration and Analysis System(LABORAS).We successfully assessed changes in nociceptive behaviours in two TMD mouse models,a unilateral anterior crossbite(UAC)-induced TMD mouse model and aβ-catenin conditional activation mouse model.We found that the UAC model andβ-catenin conditional activation mouse model were significantly associated with signs of increased mechanical allodynia,lower bite force,and decreased spontaneous feeding behaviour,indicating manifestations of TMD.These behavioural changes were consistent with the cartilage degradation phenotype observed in these mouse models.Our studies have shown reliable methods to analyse nociceptive behaviours in mice and may indicate that these methods are valid to assess signs of TMD in mice. 展开更多

The discharge of acid mine drainage from abandoned high sulfur(S)coal mines has caused serious pollution in the Shandi River,Yangquan,Shanxi Province.To determine the impact of long-term acid mine drainage on the microorganisms in the river,we collected river sediments from a polluted tributary(Group P)and the mainstream of Shandi River(Group R)to study the bacterial diversity and community composition.The results showed that the tributary was seriously polluted by acid drainage from abandoned coal mines,with the pH value of the sediment being<2.5,resulting in the low bacterial richness and diversity of the tributary samples.Acidophillic Fe-and S-metabolizing bacteria,such as Metallibacterium,Acidiphilium,and Acidithiobacillus,were the dominant genera in Group P samples,while the Group R was dominated by the neutral anaerobic iron-reducing bacteria Geothrix and Geobacter.Results of principal co-ordinates analysis(PCoA)revealed that the bacterial communities are significantly different between groups P and R,and the significant different species were mainly attributed to phylum Proteobacteria,Actinobacteria,and Acidobacteria.The distribution of the microbial community is mainly influenced by pH,and the Fe and Cd concentrations.Metallicactrium,the dominant genus,is negatively correlated with pH(R^(2)=-0.95)and positively correlated with Fe(R^(2)=0.99),while Geothrix and Geobacter,are mainly affected by the heavy metals.This study determined the impact of river pollution caused by abandoned coal mine drainage,especially on the microbial diversity and community composition within the river sediment. 展开更多

One-dimensional(1-D)nanostructures have been attracted much attention as a result of their exceptional properties,which are different from bulk materials.Among 1-D nanostructures,1-D heterostructures with modulated compositions and interfaces have recently become of particular interest with respect to potential applications in nanoscale building blocks of future optoelectronic devices and systems.Many kinds of methods have been developed for the synthesis of 1-D nanoscale heterostructures.This article reviews the most recent development,with an emphasize on our own recent efforts,on 1-D nanoscale heterostructures,especially those synthesized from the vapor deposition methods,in which all the reactive precursors are mixed together in the reaction chamber.Three types of 1-D nanoscale heterostructures,defined from their morphologies characteristics,are discussed in detail,which include 1-D co-axial core-shell heterostructures,1-D segmented heterostructures and hierarchical heterostructures.This article begins with a brief survey of various methods that have been developed for synthesizing 1-D nanoscale heterostructures and then mainly focuses on the synthesis,structures and properties of the above three types of nanoscale heterostructures.Finally,this review concludes with personal views towards the topic of 1-D nanoscale heterostructures. 展开更多

Objective:To validate the eighth edition of the AJCC staging system in patients with pancreatic adenocarcinoma receiving only stereotactic body radiation therapy and chemotherapy,and to propose modifications to improve prognostic accuracy.Methods:Patients with pathologically confirmed pancreatic adenocarcinoma without metastasis who were undergoing only chemoradiotherapy were included and staged according to the seventh and eighth editions of the AJCC staging system.Meanwhile,another group of stage T4 patients from the above enrollment with only portal vein involvement with or without tumor thrombi(PV±PVTT)were retrieved for survival comparisons.Modifications were proposed according to the survival comparisons.A cohort from the SEER database was used for external validation of the modified staging system.Results:A total of 683 patients were included.Patients with N2 or N1 but different T stages had significantly different survival outcomes according to the eighth edition.The survival of patients with(PV±PVTT)was comparable to that of patients with T4 tumors.The concordance index of the seventh and eighth editions,and the modified staging system was 0.744(95%CI:0.718—0.769),0.750(95%CI:0.725—0.775),and 0.788(95%CI:0.762-0.813),respectively.For external validation,the concordance index was 0.744(95%CI:0.718-0.770),0.750(95%CI:0.724-0.776),and 0.788(95%CI:0.762-0.814),respectively.Conclusions:The modified staging system is suggested to have the m ost accurate prognostic value.Hence,PV土PVTT should be added to the definition of T4 tumors regardless of tumor size.Patients with N2 or N1 in different T stages could be regrouped into different substages.Additionally,stage III should be subclassified into IIIA(T3N 2 and T4N 0)and IIIB(T4N 1-2). 展开更多

Hypoxia-inducible factors(HIFs) play essential roles in regulating cellular adaptation in the IVD under physiological conditions. Disc degeneration disease(DDD) is one of the leading causes of disability, and current therapies are ineffective. This study sought to explore the role of HIFs in DDD pathogenesis in mice. The findings of this study showed that among HIF family members, Hif1α was significantly upregulated in cartilaginous endplate(EP) and annulus fibrosus(AF) tissues from human DDD patients and two mouse models of DDD compared with controls. Conditional deletion of the E3 ubiquitin ligase Vhl in EP and AF tissues of adult mice resulted in upregulated Hif1α expression and age-dependent IVD degeneration. Aberrant Hif1α activation enhanced glycolytic metabolism and suppressed mitochondrial function. On the other hand, genetic ablation of the Hif1α gene delayed DDD pathogenesis in Vhl-deficient mice. Administration of 2-methoxyestradiol(2ME2), a selective Hif1α inhibitor, attenuated experimental IVD degeneration in mice. The findings of this study show that aberrant Hif1α activation in EP and AF tissues induces pathological changes in DDD, implying that inhibition of aberrant Hif1α activity is a potential therapeutic strategy for DDD. 展开更多

In order to investigate the mechanism of the temperature oscillation in loop heat pipes,this paper investigated the movement of the phase interface as the changed input power by a mass-spring-damper model.The model was solved with MATLAB and was used to explain the high-frequency and low-amplitude temperature oscillation.Temperature variation with the input power from 20 W to 75 W was investigated based on a LHP prototype in a literature.The model agreed well with the experimental data in the literature.The simulation results suggested that the movement of the liquid column was caused by the fluctuation of pressure difference applied on the liquid column and the stiffness coefficients of the vapor springs increasing with the input power.According to parameter analyses,the temperature oscillation at the outlet of the condenser can be weakened by increasing the mass of the liquid column and keeping the temperature at the outlet of the condenser steady. 展开更多

The middle Eocene climatic optimum(MECO,ca.-42 Ma)is a key time period for understanding Cenozoic cooling of the global climate.Still,midlatitude terrestrial records of climate evolution during MEcO epoch are rare.In this study,continuous high-resolution record of shale sediments in mid-Eocene Shahejie Formation(MES shales)in the Bohai Bay Basin were performed with major-element and wavelet analysis.The midlatitude paleoweathering and paleoclimatic evolution during MEcO epoch were analyzed in this study.The MES shales experienced weak-moderate paleoweathering under a subtropical monsoon paleoclimate with mean annual temperature of 8.3-12.9℃ and mean annual precipitation of 685-1100 mm/yr.The MES shales record a mixed provenance involving intermediate igneous rocks,and low compositional maturity.The nutrient-rich environment led to enrichment in organic matter in the MES shales.Wavelet analysis revealed good periodicity about the paleoclimate and weathering during MECO epoch.In the stage I of MES shales depositional process,the paleolake was high in nutrients,and the MES shales experienced high chemical weathering due to a relatively warmer and more humid climate.In contrast,the climate in stage II was relatively cold and dry,and the maturity of the MES shales was relatively high during this stage,suggesting a relatively stable tectonic background.This work provides more terrestrial records of MEco epoch for midlatitude region,and is benefit for better understanding of the palaeoenvironment when MES shales formed.The implication of organic matters enrichment in this study is meaningful for the shale oil/gas exploration in Nanpu Sag. 展开更多

Characteristics of extratropical cyclone variability in the Northern Hemisphere and their response to rapid changes in Arctic sea ice

Extratropical cyclones are critical weather systems that affect large-scale weather and climate changes at mid-high latitudes.However,prior research shows that there are still great difficulties in predicting extratropical cyclones for occurrence,frequency,and position.In this study,mean sea level pressure(MSLP)data from the European Centre for Medium-Range Weather Forecasts(ECMWF)reanalysis(ERA5)are used to calculate the variance statistics of the MSLP to reveal extratropical cyclone activity(ECA).Based on the analysis of the change characteristics of ECA in the Northern Hemisphere,the intrinsic link between ECA in the Northern Hemisphere and Arctic sea ice is explored.The results show that the maximum ECA mainly occurs in winter over the mid-high latitudes in the Northern Hemisphere.The maximum ECA changes in the North Pacific and the North Atlantic,which are the largest variations in the Northern Hemisphere,are independent of each other,and their mechanisms may be different.Furthermore,MSLP is a significant physical variable that affects ECA.The North Atlantic Oscillation(NAO)and North Pacific Index(NPI)are significant indices that impact ECA in the North Atlantic and North Pacific,respectively.The innovation of this paper is to explore the relationship between the activity of extratropical cyclones in the Northern Hemisphere and the abnormal changes in Arctic sea ice for the first time.The mechanism is that the abnormal changes in summer-autumn and winter Arctic sea ice lead to the phase transition of the NPI and NAO,respectively,and then cause the occurrence of ECA in the North Pacific and North Atlantic,respectively.Arctic sea ice plays a crucial role in the ECA in the Northern Hemisphere by influencing the polar vortex and westerly jets.This is the first exploration of ECAs in the Northern Hemisphere using Arctic sea ice,which can provide some references for the in-depth study and prediction of ECAs in the Northern Hemisphere. 展开更多
