P-loop,Bet v 1结构域的缺失及突变对GmPR10和Gly m 4l抑制大豆疫霉菌能力的影响 被引量:2

大豆疫霉根腐病是由大豆疫霉菌(Phytophthora sojae)引起的危害大豆生长的严重病害。课题组前期研究表明具有P-loop结构域的GmPR10(Gene Bank accession no.FJ960440)和具有P-loop、Bet v1结构域的Gly m 4l(Gene Bank accession no.HQ91... 大豆疫霉根腐病是由大豆疫霉菌(Phytophthora sojae)引起的危害大豆生长的严重病害。课题组前期研究表明具有P-loop结构域的GmPR10(Gene Bank accession no.FJ960440)和具有P-loop、Bet v1结构域的Gly m 4l(Gene Bank accession no.HQ913577.1)抑制大豆疫霉菌生长,并且过表达GmPR10和Gly m 4l的转基因大豆植株可以提高对大豆疫霉根腐病的抗性。为研究GmPR10和Gly m 4l抑菌机理,本研究利用点突变技术,获得了GmPR10的P-loop结构域突变体(Gly48/Thr48和Gly^51/Arg^51)、Gly m 4l的P-loop结构域突变体(Gly^49/Ile^49和Lys^55/Pro^55)、GmPR10和Gly m 4l的P-loop结构域以及Gly m 4l的Bet v1结构域缺失突变体,并纯化回收相应突变体蛋白,进行体外抑制大豆疫霉菌试验。结果表明,突变或缺失P-loop,Bet v1结构域的GmPR10和Gly m 4l失去了抑制大豆疫霉菌(Race 1)生长的能力,说明P-loop、Bet v 1结构域对GmPR10和Gly m 4l行使抑菌功能至关重要。 展开更多

大豆异黄酮是有助于人类身体健康的一类次级代谢产物,大豆查尔酮还原酶(chalcone reductase, CHR)基因是控制大豆异黄酮主要组分大豆苷元生物合成的关键酶之一。为拓宽大豆CHR研究范围,促进异黄酮代谢机理研究,进而推进大豆品种改良,本... 大豆异黄酮是有助于人类身体健康的一类次级代谢产物,大豆查尔酮还原酶(chalcone reductase, CHR)基因是控制大豆异黄酮主要组分大豆苷元生物合成的关键酶之一。为拓宽大豆CHR研究范围,促进异黄酮代谢机理研究,进而推进大豆品种改良,本研究利用基因工程技术对大豆CHR2-1基因进行克隆及生物信息学分析,并且构建植物表达载体pCAMBIA3301-CHR2-1,通过农杆菌介导法将目的基因整合到东农50受体大豆基因组中,PCR筛选鉴定获得大豆转化植株。结果表明:GmCHR2-1基因编码的蛋白属于非分泌型蛋白,可能在细胞质中发挥主要功能,属于非跨膜类蛋白并不在细胞中发生迁移;该蛋白存在22个潜在磷酸化位点,其中苏氨酸(Thr)4个、丝氨酸(Ser)18个;该蛋白由α-螺旋(40.63%)、延伸链(14.29%)、β-转角(4.13%)和无规则卷曲(40.95%)4个部分组成。PCR鉴定表明,转化植株基因组包含草丁膦抗性基因Bar、终止子NOS和启动子35S位点,初步确定已将GmCHR2-1转入到东农50大豆品种基因组中,并获得T;阳性植株转化苗12株。 展开更多

To elucidate the differential gene expression patterns in soybeans during infection by Phytophthora sojae,a cDNA library for suppression subtractive hybridization (SSH) was constructed with cDNAs from soybean cultivar Suinong 10 treated with sterile distilled water as the driver and cDNAs from Suinong 10 inoculated with P.sojae as the tester.A total of 2 067 recombinant colonies from the SSH library were randomly picked,amplified,and sequenced.After discarding 312 poor quality expressed sequence tags (EST),1 755 high quality ESTs were assembled and edited to 1 384 tentatively unique genes (TUG),in which,586 showed significant homology to known sequences,and 798 had low homology or no match with the known sequences.A cDNA microarray containing 307 singletons from the 586 TUGs and 222 singletons from the 798 TUGs was developed to characterize differentially expressed cDNAs in the SSH library,and eight cDNAs were identified to be up-regulated after microarray analysis and then confirmed by real-time PCR.They were homologous to the protein 10,and were also related to some proteins in disease resistance response,such as pathogen-related protein,phenylalanine ammonia-lyase,isoflavone reductase,WRKY transcription factor 31,major allergen Pru ar 1,and pleiotropic drug resistance protein 12.Most of the up-regulated cDNAs encode enzymes of phytoalexin biosynthesis and pathogenesis-related proteins involved in plant disease resistance.Here,we fist reported the Pru ar 1 in soybeans.The findings of this research have contributed to better understanding of soybean resistance to P.sojae at the molecular level. 展开更多

目的评价周边虹膜前粘连(peripheral anterior synechia,PAS)范围对激光周边虹膜成形术(laser peripheral iridoplasty,LPIP)治疗原发性闭角型青光眼急性发作期效果的影响。设计前瞻性比较性病例系列。研究对象邯郸市眼科医院就诊的急... 目的评价周边虹膜前粘连(peripheral anterior synechia,PAS)范围对激光周边虹膜成形术(laser peripheral iridoplasty,LPIP)治疗原发性闭角型青光眼急性发作期效果的影响。设计前瞻性比较性病例系列。研究对象邯郸市眼科医院就诊的急性发作期原发性闭角型青光眼患者116例(116眼)。方法 84例接受LPIP治疗(LPIP组),32例接受降眼压药物治疗(药物组)。在治疗后1、2、6小时测量眼压。眼压控制后进行视力、房角镜、眼底、角膜内皮细胞计数检查。主要指标眼压、PAS范围。结果 LPIP组和药物组PAS范围与发作持续时间均具有较强相关性(r=0.579, 0.288,P<0.05)。LPIP组PAS与术后2 h、6 h眼压具有较强相关性(r=0.296,0.334,P<0.05)。PAS范围在发作持续时间≤24 h者(2.3±2.9个钟点)显著小于发作持续时间>24 h者(6.2±4.0个钟点)(P<0.001)。药物组PAS≤6个钟点者与PAS>6个钟点者治疗后各时间点眼压无显著性差异。LPIP组PAS≤6个钟点者术后1 h(28.4±15.1 mmHg)、2 h(22.0±13.1 mmHg)、6 h(17.9±8.52 mmHg)眼压均低于PAS>6个钟点者(35.5±15.5、29.6±11.4、24.9±10.4 mmHg)(P=0.066、0.020、0.003)。结论急性发作的原发性闭角型青光眼PAS范围≤6个钟点的病例采用LPIP治疗其效果优于PAS范围>6个钟点者。 展开更多

Background Many studies indicated that short-term and long-term intraocular pressure (IOP) fluctuations in primary open angle glaucoma patients might lead to glaucomatous progression. However, seldom study has evaluated the long-term fluctuation of IOP in primary chronic angle closure diseases. The objective of this study was to investigate the long-term IOP fluctuation of primary angle closure diseases and its associations following laser peripheral iridotomy (LPI) with or without laser peripheral iridoplasty.Methods A total of 158 patients with primary angle closure suspect (PACS, n=21), primary angle closure (PAC, n=81) and primary angle closure glaucoma (PACG, n=55) had been treated by LPI with or without laser peripheral iridoplasty and followed up for more than 12 months. IOP was measured with Goldman applanation tonometer. Multivariate linear regression with generalized estimating equation (GEE) regression models was used to evaluate the association of long-term IOP fluctuation (maximum IOP minus minimum IOP) with gender, age, baseline IOP, baseline peripheral anterior synechia (PAS),baseline vertical cup/disc ratio (VCDR), baseline mean deviation (MD), need for IOP-lowering medications.Results IOP fluctuation during follow-up in PACS, PAC and PACG groups were (4.83±2.90), (5.67±3.35), and (9.40±7.14)mmHg, respectively. IOP fluctuation was strongly correlated with baseline IOP (r=0.356, P 〈0.001), PAS (r=0.374, P 〈0.001).IOP fluctuation was higher in patients with higher baseline IOP (0.18 mmHg per unit increase, 95% CI: 0.05-0.31 mmHg).Conclusions Long-term IOP fluctuation in PACG group was larger than that in PACS or PAC group. Eyes with higher baseline IOP were observed to have larger long-term IOP fluctuation. 展开更多
