Introduction:Myocardial ischemia-reperfusion(IR)injury has received widespread attention due to its damaging effects.Electroacupuncture(EA)pretreatment has preventive effects on myocardial IR injury.SLC26A4 is a Na+in...Introduction:Myocardial ischemia-reperfusion(IR)injury has received widespread attention due to its damaging effects.Electroacupuncture(EA)pretreatment has preventive effects on myocardial IR injury.SLC26A4 is a Na+independent anion reverse transporter and has not been reported in myocardial IR injury.Objectives:Tofind potential genes that may be regulated by EA and explore the role of this gene in myocardial IR injury.Methods:RNA sequencing and bioinformatics analysis were performed to obtain the differentially expressed genes in the myocardial tissue of IR rats with EA pretreatment.Myocardial infarction size was detected by TTC staining.Serum CK,creatinine kinase-myocardial band,Cardiac troponin I,and lactate dehydrogenase levels were determined by ELISA.The effect of SLC26A4 on cardiomyocyte apoptosis was explored by TUNEL staining and western blotting.The effects of SLC26A4 on inflammation were determined by HE staining,ELISA,and real-time PCR.The effect of SLC26A4 on the NF-κB pathway was determined by western blotting.Results:SLC26A4 was up-regulated in IR rats but downregulated in IR rats with EA pretreatment.Compared with IR rats,those with SLC26A4 knockdown exhibited improved cardiac function according to decreased myocardial infarction size,reduced serum LDH/CK/CK-MB/cTnI levels,and elevated left ventricular ejection fraction and fractional shortening.SLC26A4 silencing inhibited myocardial inflammation,cell apoptosis,phosphorylation,and nuclear translocation of NF-κB p65.Conclusion:SLC26A4 exhibited promoting effects on myocardial IR injury,while the SLC26A4 knockdown had an inhibitory effect on the NF-κB pathway.These results further unveil the role of SLC26A4 in IR injury.展开更多
The surface trap parameter can significantly affect the development of surface flashover in vacuum,but the effective mode and mechanism are not very clear yet.The trap parameters of three polymeric materials were test...The surface trap parameter can significantly affect the development of surface flashover in vacuum,but the effective mode and mechanism are not very clear yet.The trap parameters of three polymeric materials were tested and calculated by means of isothermal surface potential decay.The flashover experiment was developed under different applied voltages.The results show a positive correlation between the withstand voltage and the deep trap,i.e.,the deeper trap energy level is,the higher flashover voltage is.The dynamics process of charge trapping and detrapping was analyzed based on the charge transport model in dielectrics with a single trap level and two discrete trap levels.The time of charge trapping was compared with that of the discharge development.The results show that the charge trapping time is longer than the flashover development time.The way to influence flashover for a trap is not to decrease the secondary electrons in single discharge development,but to change the electric field distribution on the dielectric surface by charge capture.展开更多
AIM:To explore the alteration of tyrosine phosphatase SHP-2 protein expression in gastric cancer and to assess its prognostic values.METHODS:Three hundred and five consecutive cases of gastric cancer were enrolled int...AIM:To explore the alteration of tyrosine phosphatase SHP-2 protein expression in gastric cancer and to assess its prognostic values.METHODS:Three hundred and five consecutive cases of gastric cancer were enrolled into this study.SHP-2 expression was carried out in 305 gastric cancer specimens,of which 83 were paired adjacent normal gastric mucus samples,using a tissue microarray immunohistochemical method.Correlations were analyzed between expression levels of SHP-2 protein and tumor parameters or clinical outcomes.Serum anti-Helicobacter pylori(H.pylori) immunoglobulin G was detected with enzyme-linked immunosorbent assay.Cox proportional hazards model was used to evaluate prognostic values by compassion of the expression levels of SHP-2 and disease-specific survivals in patients.RESULTS:SHP-2 staining was found diffuse mainly in the cytoplasm and the weak staining was also observed in the nucleus in gastric mucosa cells.Thirty-two point five percent of normal epithelial specimen and 62.6% of gastric cancer specimen were identified to stain with SHP-2 antibody positively(P < 0.001).Though SHP-2 staining intensities were stronger in the H.pylori(+) group than in the H.pylori(-) group,no statistically significant difference was found in the expression levels of SHP-2 between H.pylori(+) and H.pylori(-) gastric cancer(P = 0.40).The SHP-2 expression in gastric cancer was not significantly associated with cancer stages,lymph node metastases,and distant metastasis of the tumors(P = 0.34,P = 0.17,P = 0.52).Multivariate analysis demonstrated no correlation between SHP-2 expression and disease-free survival(P = 0.86).CONCLUSION:Increased expression of SHP-2 protein in gastric cancer specimen suggesting the aberrant upregulation of SHP-2 protein might play an important role in the gastric carcinogenesis.展开更多
AIM:To investigate screening makers for gastric cancer,we assessed the association between gastric cancer and serum pepsinogens(PGs).METHODS:The subjects comprised 450 patients with gastric cancer,111 individuals with...AIM:To investigate screening makers for gastric cancer,we assessed the association between gastric cancer and serum pepsinogens(PGs).METHODS:The subjects comprised 450 patients with gastric cancer,111 individuals with gastric atrophy,and 961 healthy controls.Serum anti-Helicobacter pylori(H.pylori) immunoglobulin G(IgG),PGⅠand PG Ⅱ were detected by enzyme-linked immunosorbent assay.Gastric atrophy and gastric cancer were diagnosed by endoscopy and histopathological examinations.Odds ratios and 95%CIs were calculated using multivariate logistic regression.RESULTS:Rates of H.pylori infection remained high in Northeastern China.Rates of H.pylori IgG positivity were greater in the gastric cancer and gastric atrophy groups compared to the control group(69.1% and 75.7% vs 49.7%,P < 0.001).Higher levels of PG Ⅱ(15.9 μg/L and 13.9 μg/L vs 11.5 μg/L,P < 0.001) and lower PGⅠ/PG Ⅱ ratio(5.4 and 4.6 vs 8.4,P < 0.001) were found in patients with gastric cancer or gastric atrophy compared to healthy controls,whereas no correlation was found between the plasma PGⅠconcentration and risk of gastric cancer(P = 0.537).In addition,multivariate logistic analysis indicated that H.pylori infection and atrophic gastritis were independent risk factors for gastric cancer.Lower plasma PGⅠ/PG Ⅱ ratio was associated with higher risks of atrophy and gastric cancer.Furthermore,plasma PG Ⅱ?level?significantly?correlated?with?H.pyloriinfected gastric cancer.CONCLUSION:Serum PG Ⅱ concentration and PG Ⅰ/PG Ⅱ ratio are potential biomarkers for H.pyloriinfected gastric disease.PG Ⅱ is independently associated with risk of gastric cancer.展开更多
Aiming at the problem that traditional optimal operation of hydropower reservoir pays little attention to ecology, an optimal operation model of multi-objective hydropower reservoir with ecology consideration is estab...Aiming at the problem that traditional optimal operation of hydropower reservoir pays little attention to ecology, an optimal operation model of multi-objective hydropower reservoir with ecology consideration is established which combines the ecology and power generation. The model takes the maximum annual power generation benefit, the maximum output of the minimal output stage in the year and the minimum shortage of ecological water demand as objectives, and water quantity balance of reservoir, reservoir storage, discharge flow, output and so on as constraints. Chaotic genetic arithmetic is developed to solve the optimal model. An example is studied, showing that the annual generation of the proposed model is 8 million kW?h less than that model without ecology consideration, which is about 0.28 percent. But the proposed model is in favor of river ecology protection, and can promote the sustainable utilization of water resources. So it is worthy and necessary for the optimal operation of hydropower reservoir with ecology consideration.展开更多

Operating conditions of film capacitors are complex,and the problem of film insulation failure caused by repetitively pulsed voltage is becoming ever serious.Degradation of the film under repetitively pulsed voltage c...Operating conditions of film capacitors are complex,and the problem of film insulation failure caused by repetitively pulsed voltage is becoming ever serious.Degradation of the film under repetitively pulsed voltage cannot be accurately evaluated by the average breakdown electric field.In this paper,the effects of pulsed electric field and pulse repetition frequency on the breakdown in biaxially oriented polypropylene(BOPP)films are investigated.Three phases of BOPP degradation are proposed based on the voltage amplitude,i.e.,maintenance(M),decline(D),and near-zero(N).Evolution of the BOPP film from degradation to breakdown at different frequencies is presented.Meanwhile,transition of discharge mode and elemental composition of the film are analyzed.Experimental results show continuous heat generation under repetitive microsecond pulses is the dominant factor for degradation of BOPP film.The number of applied pulses and the repetitive stressing time decrease exponentially with increase of frequency.This research can be contributed to the safe and reliable operation of capacitors.展开更多

However, the large size of sensor and the extreme measurement conditions required strongly limit their application. It remains a challenge to reconstruct the vector of a.c. field with nanoscale spatial resolution using a single spin under ambient conditions. In this work, we choose the radio-frequency(RF) field as a typical case and realize the measurement of RF field based on a nitrogen-vacancy(NV)center in diamond. We build a solid sensor through measuring the effect of RF field on NV electron spin energy levels and the transition between them. Photothermal therapy(PTT)has shown promising applications in tumor therapies.However,due to laserinduced nonspecific heating and heat diffusion,high levels of hyperthermia(>50℃)in tumor tissues often increase the ...Photothermal therapy(PTT)has shown promising applications in tumor therapies.However,due to laserinduced nonspecific heating and heat diffusion,high levels of hyperthermia(>50℃)in tumor tissues often increase the risk of cancer recurrence and metastasis,which causes the patient pain and destroys the surrounding normal cells and tissues.It is therefore important to develop photothermal strategies that have excellent therapeutic efficiencies under low-temperature conditions(≤45℃).In addition,the heterogeneity and complexity of tumors require the development of combinatorial antitumor treatments as the therapeutic efficiency of monomodal PTT is not currently sufficient.Herein,we have adopted a stepwise synthetic approach to develop a highly efficient multimodal therapeutic agent GA@PCOF@PDA by successive bonding defect functionalization(BDF),guest encapsulation,and surface modification steps.The covalently grafted porphyrinic photosensitizers(Por),encapsulated gambogic acid(GA),and surface-modified PDA film are independently responsible for photodynamic therapy(PDT),heat-shock protein 90(HSP90)down-regulation and chemotherapy(CT),and low-temperature PTT.This proof-ofconcept study illustrates an efficient,generalized approach to design high-performance covalent organic framework(COF)-based nanoagents that can be easily tailored to combine different therapeutic modalities for improved cancer theranostics at low temperatures.展开更多

Here, we isolated a rice (Oryza sativa) mutant, w59, with a white-core floury endosperm. Map-based cloning of w59 identified a mutation in OsPKpα1, which encodes a plastidic isoform of PK (PKp). OsPKpα1 localizes to the amyloplast stroma in the developing endosperm, and the mutation of OsPKpα1 in w59 decreases the plastidic PK activity, resulting in dramatic changes to the lipid biosynthesis in seeds. The w59 grains were also characterized by a marked decrease in starch content. Consistent with a decrease in number and size of the w59 amyloplasts, large empty spaces were observed in the central region of the w59 endosperm, at the early grain-filling stage. Moreover, a phylogenetic analysis revealed four potential rice isoforms of OsPKp. We validated the in vitro PK activity of these OsPKps through reconstituting active PKp complexes derived from inactive individual OsPKps, revealing the heteromeric structure of rice PKps, which was further confirmed using a protein- protein interaction analysis. These findings suggest a functional connection between lipid and starch synthesis in rice endosperm amyloplasts.展开更多
Despite the progress made to improve therapeutic outcomes for acute myeloid leukemia(AML),many unmet clinical needs remain to be resolved.Unlike existing anti-AML strategies,here we developed a biomimetic nanocomposit...Despite the progress made to improve therapeutic outcomes for acute myeloid leukemia(AML),many unmet clinical needs remain to be resolved.Unlike existing anti-AML strategies,here we developed a biomimetic nanocomposite to efficiently eliminate the leukemia cells in the bone marrow and prevent the homing of AML.To fulfill our design,the ultra-small nanozyme was conjugated onto the surface of an oxygen-carrying nanoparticle,which was further coated with bone marrow stromal cell membrane.After entering the blood,this biomimetic nanocomposite got actively internalized by the leukemia cells in the blood and released the loaded chemotherapeutics and nanozyme inside the leukemia cells to achieve a synergistic antitumor efficacy.Meanwhile,the adhesive properties of the stromal cell membrane enabled the nanocomposite to home to the bone marrow,where the nanocomposite effectively killed the retained leukemia cells.More importantly,the biomimetic cell membrane also acted as a CXCR4 antagonism to block the CXCR4/CXCL12-mediated homing of leukemia cells to the bone marrow and infiltration to other organs like the liver and spleen.In conclusion,this proof-of-concept study demonstrated that our designed platform effectively kills leukemia cells while preventing their infiltration,thus providing a promising prospect for resolving the clinical challenges in current AML treatment.展开更多

文摘Summary What is already known about this topic?Reports detailing the clinical presentation of coronavirus disease 2019(COVID-19)are extensive in China.However,data remains limited regarding the long-term effects of the 2022 outbreak on the community and healthcare workers(HCWs).What is added by this report?In the follow-up study conducted with 1,069 community members and 3,309 HCWs infected with COVID-19,we observed that five months postoutbreak,39.2%of community members and 28.7%of HCWs reported experiencing at least one symptom.The symptoms most frequently reported included fatigue or muscle weakness,insomnia,cognitive dysfunction,hair loss,joint or muscle pain,and persistent cough.HCWs tended to experience fewer long-term physical consequences and their symptoms had an expedited recovery time compared to the community members.Nevertheless,HCWs displayed a higher prevalence of moderate to severe depression and anxiety.

文摘Detection of a.c. magnetic field is consequential for many developments in physical and biological sciences,and various designs of magnetometer have been proposed recently. However, the large size of sensor and the extreme measurement conditions required strongly limit their application. It remains a challenge to reconstruct the vector of a.c. field with nanoscale spatial resolution using a single spin under ambient conditions. In this work, we choose the radio-frequency(RF) field as a typical case and realize the measurement of RF field based on a nitrogen-vacancy(NV)center in diamond. We build a solid sensor through measuring the effect of RF field on NV electron spin energy levels and the transition between them. Both of the phase and amplitude(including the transverse and longitudinal components) are measured by this new approach.

文摘Photothermal therapy(PTT)has shown promising applications in tumor therapies.However,due to laserinduced nonspecific heating and heat diffusion,high levels of hyperthermia(>50℃)in tumor tissues often increase the risk of cancer recurrence and metastasis,which causes the patient pain and destroys the surrounding normal cells and tissues.It is therefore important to develop photothermal strategies that have excellent therapeutic efficiencies under low-temperature conditions(≤45℃).In addition,the heterogeneity and complexity of tumors require the development of combinatorial antitumor treatments as the therapeutic efficiency of monomodal PTT is not currently sufficient.Herein,we have adopted a stepwise synthetic approach to develop a highly efficient multimodal therapeutic agent GA@PCOF@PDA by successive bonding defect functionalization(BDF),guest encapsulation,and surface modification steps.The covalently grafted porphyrinic photosensitizers(Por),encapsulated gambogic acid(GA),and surface-modified PDA film are independently responsible for photodynamic therapy(PDT),heat-shock protein 90(HSP90)down-regulation and chemotherapy(CT),and low-temperature PTT.This proof-ofconcept study illustrates an efficient,generalized approach to design high-performance covalent organic framework(COF)-based nanoagents that can be easily tailored to combine different therapeutic modalities for improved cancer theranostics at low temperatures.

文摘Pyruvate kinase (PK) is a key enzyme in glycolysis and carbon metabolism. Here, we isolated a rice (Oryza sativa) mutant, w59, with a white-core floury endosperm. Map-based cloning of w59 identified a mutation in OsPKpα1, which encodes a plastidic isoform of PK (PKp). OsPKpα1 localizes to the amyloplast stroma in the developing endosperm, and the mutation of OsPKpα1 in w59 decreases the plastidic PK activity, resulting in dramatic changes to the lipid biosynthesis in seeds. The w59 grains were also characterized by a marked decrease in starch content. Consistent with a decrease in number and size of the w59 amyloplasts, large empty spaces were observed in the central region of the w59 endosperm, at the early grain-filling stage. Moreover, a phylogenetic analysis revealed four potential rice isoforms of OsPKp. We validated the in vitro PK activity of these OsPKps through reconstituting active PKp complexes derived from inactive individual OsPKps, revealing the heteromeric structure of rice PKps, which was further confirmed using a protein- protein interaction analysis. These findings suggest a functional connection between lipid and starch synthesis in rice endosperm amyloplasts.

文摘Despite the progress made to improve therapeutic outcomes for acute myeloid leukemia(AML),many unmet clinical needs remain to be resolved.Unlike existing anti-AML strategies,here we developed a biomimetic nanocomposite to efficiently eliminate the leukemia cells in the bone marrow and prevent the homing of AML.To fulfill our design,the ultra-small nanozyme was conjugated onto the surface of an oxygen-carrying nanoparticle,which was further coated with bone marrow stromal cell membrane.After entering the blood,this biomimetic nanocomposite got actively internalized by the leukemia cells in the blood and released the loaded chemotherapeutics and nanozyme inside the leukemia cells to achieve a synergistic antitumor efficacy.Meanwhile,the adhesive properties of the stromal cell membrane enabled the nanocomposite to home to the bone marrow,where the nanocomposite effectively killed the retained leukemia cells.More importantly,the biomimetic cell membrane also acted as a CXCR4 antagonism to block the CXCR4/CXCL12-mediated homing of leukemia cells to the bone marrow and infiltration to other organs like the liver and spleen.In conclusion,this proof-of-concept study demonstrated that our designed platform effectively kills leukemia cells while preventing their infiltration,thus providing a promising prospect for resolving the clinical challenges in current AML treatment.
