AIM: To assess the predictive value of Operative Link on Gastritis Assessment(OLGA) and Operative Link on Gastric Intestinal Metaplasia Assessment(OLGIM) stages in gastric cancer.METHODS: A prospective study was condu...AIM: To assess the predictive value of Operative Link on Gastritis Assessment(OLGA) and Operative Link on Gastric Intestinal Metaplasia Assessment(OLGIM) stages in gastric cancer.METHODS: A prospective study was conducted with 71 patients with early gastric cancer(EGC) and 156 patients with non-EGC. All patients underwent endoscopic examination and systematic biopsy. Outcome measures were assessed and compared, including the Japanese endoscopic gastric atrophy(EGA) classification method and the modified OLGA method as well as the modified OLGIM method. Helicobacter pylori(H. pylori) status was determined for all study participants. Stepwise logistic regression modeling was performed to analyze correlations between EGC and the EGA, OLGA and OLGIM methods.RESULTS: For patients with EGC and patients with non-EGC, the proportions of moderate-to-severe EGA cases were 64.8% and 44.9%, respectively(P = 0.005), the proportions of OLGA stages Ⅲ-Ⅳ cases were 52.1% and 22.4%, respectively(P < 0.001), and the proportions of OLGIM stages Ⅲ-Ⅳ cases were42.3% and 19.9%, respectively(P < 0.001). OLGA stage and OLGIM stage were significantly related to EGA classification; specifically, logistic regression modeling showed significant correlations between EGC and moderate-to-severe EGA(OR = 1.95, 95% CI: 1.06-3.58, P = 0.031) and OLGA stages Ⅲ-Ⅳ(OR = 3.14, 95%CI: 1.71-5.81, P < 0.001), but no significant correlation between EGC and OLGIM stages Ⅲ-Ⅳ(P = 0.781). H. pylori infection rate was significantly higher in patients with moderate-to-severe EGA(75.0% vs 54.1%, P = 0.001) or OLGA/OLGIM stages Ⅲ-Ⅳ(OLGA: 83.6% vs 55.8%, P < 0.001; OLGIM: 83.6% vs 57.8%, P < 0.001).CONCLUSION: OLGA classification is optimal for EGC screening. A surveillance program including OLGA stage and H. pylori infection status may facilitate early detection of gastric cancer.展开更多

The interaction between HSP70 and AFP in HCC cells was analyzed by immunoprecipitation and Western blot.RESULTS: Immunocytochemical staining detection showed that HCC cell BEL-7402 expressed a high level of HSP70 and AFP synchronously. Both were stained in cell plasma.AFP existed in the immunoprecipitate of anti-HSP70 mAb,while there was HSP70 in the immunoprecipitate of antiAFP mAb.CONCLUSION: HSP70 chaperones AFP in human HCCcell BEL-7402. The interaction between HSP70 and AFP in human HCC cell can be a new route to study the pathogenesis and immunotherapy of HCC.展开更多

AIM:To evaluate the effectiveness of knock-down of heat shock protein 47(HSP47)on conjunctival bleb scarring in a rat model and its possible mechanism.METHODS:Male Sprague–Dawley rats were used for glaucoma filtratio...AIM:To evaluate the effectiveness of knock-down of heat shock protein 47(HSP47)on conjunctival bleb scarring in a rat model and its possible mechanism.METHODS:Male Sprague–Dawley rats were used for glaucoma filtration surgery(GFS)and were treated with either phosphate buffered solution,shControl,mitomycin C,or sh-HSP47 using a microsyringe immediately after GFS.The morphology of filtering blebs was observed postoperatively.The levels of HSP47 were analyzed at 2,5,8,and 11d after GFS via real‑time quantitative polymerase chain reaction(PCR)and Western blot.The silencing effect of HSP47,the expression of collagen I and III,and the potential signaling pathways of HSP47 during scarification were explored 11d post GFS.The protein levels of transforming growth factor-β1(TGF-β1),phospho-Smad2(pSmad2),phospho-Smad3(p-Smad3),and phospho-p38(p-p38)were also analyzed using Western blot.RESULTS:Sh-HSP47 treatment significantly prolonged the functional filtration bleb retention.The levels of HSP47 were increased significantly at 5,8,and 11d postoperatively compared to the control group(P<0.05,P<0.01,and P<0.001).The levels of HSP47 protein at day 11 postoperatively were significantly down-regulated after HSP47 silencing using sh-HSP47 adenovirus transfection(P<0.01).Expression levels of collagen I and III within the blebs were significantly reduced in the absence of HSP47(P<0.01).Moreover,the protein levels of TGF-β1,p-Smad2/3,and p-p38 were dramatically inhibited after treatment with sh-HSP47(P<0.01).CONCLUSION:The inhibitory effects of HSP47 knockdown on scarring after GFS have the potential to be an efficacious therapeutic option for the treatment of conjunctival bleb scarring.展开更多

However, the optimal surgical approach has not been determined.CASE SUMMARY Here we report three cases of IVL treated with different surgical approaches. All patients presented with circulation symptoms. Two patients had lower extremity edema and the other had cardiopalmus. The diagnosis of IVL was confirmed based on the imagining examinations and pathological findings. All patients underwent surgical treatment and were discharged without any complications.CONCLUSION Preoperative examination is crucial for surgical planning and surgical approach is dependent on the patient's condition and tumor involvement.展开更多
The purpose of this study was to investigate the effect of four fluorescent dyes, True Blue(TB), Fluoro-Gold(FG), Fluoro-Ruby(FR), and 1,1′-dioctadecyl-3,3,3′,3′-tetramethylindocarbocyanine perchlorate(Di I), in re...The purpose of this study was to investigate the effect of four fluorescent dyes, True Blue(TB), Fluoro-Gold(FG), Fluoro-Ruby(FR), and 1,1′-dioctadecyl-3,3,3′,3′-tetramethylindocarbocyanine perchlorate(Di I), in retrograde tracing of rat spinal motor neurons. We transected the muscle branch of the rat femoral nerve and applied each tracer to the proximal stump in single labeling experiments, or combinations of tracers(FG-Di I and TB-Di I) in double labeling experiments. In the single labeling experiments, significantly fewer labeled motor neurons were observed after FR labeling than after TB, FG, or Di I, 3 days after tracer application. By 1 week, there were no significant differences in the number of labeled neurons between the four groups. In the double-labeling experiment, the number of double-labeled neurons in the FG-Di I group was not significantly different from that in the TB-Di I group 1 week after tracer application. Our findings indicate that TB, FG, and Di I have similar labeling efficacies in the retrograde labeling of spinal motor neurons in the rat femoral nerve when used alone. Furthermore, combinations of Di I and TB or FG are similarly effective. Therefore, of the dyes studied, TB, FG and Di I, and combinations of Di I with TB or FG, are the most suitable for retrograde labeling studies of motor neurons in the rat femoral nerve.展开更多

METHODS: The cathartic colon model was established by feeding rats rhubarb for 3 mo, the changes of colonic electromyography were investigated by both suspension muscle strips test and serosal recordings of colonic myoelectrical activity. Immunohistochemical staining (S-P methods) and image analysis were used to determine the changes of nociceptin/orphanin FQ in the proximal colon and distal colon of rats with cathartic colon. RESULTS: Suspension muscle strips test in vitro showed OFQ (10-9-10-6 mol/L) concentration dependently caused an immediate tonic contraction in the isolated colon. But the increase of tension in cathartic colon was less than control groups (P < 0.01). Intravenous administration of OFQ (1 μg/kg) caused phasic contractions in the proximal colon, while the amplitude of phasic contractions caused by OFQ in cathartic colon was much lower than that in the control groups (2.58 ± 0.41 vs 4.16 ± 0.53, t = -2.6, P = 0.012). OFQ was highly expressed in the myenteric plexus of the rat colon but not in the muscle cells. The immunoreactivity of OFQ in the proximal colon in cathartic colon rats decreased significantly compared with the control group (P = 0.001). CONCLUSION: Colonic smooth muscle of cathartic colon showed low sensitivity to the stimulation of OFQ, suggesting that it might be caused by the abnormal distribution of OFQ or the abnormalities of receptors, leading to the disorganization of dynamic and incoordinated contractions.展开更多

The results of PeonyGlycyrrhiza decoction(PGD) as a potentially useful adjunctive treatment for hyperprolactinaemia are inconsistent.Aim This meta-analysis of randomised controlled trials(RCTs) examined the efficacy and safety of adjunctive PGD therapy for AP-induced hyperprolactinaemia.Methods English(PubMed, Embase, Cochrane Library,PsycINFO) and Chinese(Chinese National Knowledge Infrastructure, Wanfang Data) databases were systematically searched up to 10 June 2018. The inclusion criteria were based on PIC0 S一/^rticipants: adult patients with schizophrenia; Aitervention: PGD plus APs; Cbmparison: APs plus placebo or AP monotherapy;(Xjtcomes: efficacy and safety; Study design: RCTs. The weighted mean difference(WMD) and risk ratio(RR) along with their 95% CIs were calculated using Review Manager(RevMan) V.5.3 software.Results Five RCTs(n=450) were included and analysed.Two RCTs(n=140) were double-blind and four RCTs(n=409) reported ‘random' assignment with specific description. The PGD group showed a significantly lower serum prolactin level at endpoint than the control group(n=380, WMD:-32.69 ng/mL(95% Cl-41.66 to 23.72),/X0.00001, ^=97%). Similarly, the superiority of PGD over the control groups was also found in the improvement of hyperprolactinaemia-related symptoms. No difference was found in the improvement of psychiatric symptoms assessed by the Positive and Negative Syndrome Scale(n=403,WMD:-0.62(95% Cl-2.38 to 1.15), p=0.49, ^=0%). There were similar rates of all-cause discontinuation(n=330, RR0.93(95% Ci 0.63 to 1.37),/t=0.71, ^=0%) and adverse drug reactions between the two groups. According to the Grading of Recommendations Assessment, Development and Evaluation approach, the level of evidence of primary and secondary outcomes ranged from ‘very low'(14.3%),‘low'(42.8%),‘moderate'(14.3%),to ‘high,(28.6%>.Conclusions Current evidence supports the adjunctive use of PGD to suppress elevated prolactin and improve prolactininduced symptoms without significant adverse events in adult patients with AP-induced hyperprolactinaemia. Highquality RCTs with longer duration are needed to confirm these findings.Trial registration number 42016037017.展开更多

The networks of the 30 two-state proteins and 15 three-state proteins were constructed by complex networks analysis at three length scales: Protein Contact Networks, Long-range Interaction Networks and Short-range Interaction Networks. To uncover the relationship between structural properties and folding kinetics of the proteins, the correlations of protein network parameters with protein folding rate and topology parameters contact order were analyzed. The results show that Protein Contact Networks and Short-range Interaction Networks (for both two-state and three-state proteins) exhibit the “small-world” property and Long-range Interaction networks indicate “scale-free” behavior. Our results further indicate that all Protein Contact Networks and Short- range Interaction networks are assortative type. While some of Long-range Interaction Networks are of assortative type, the others are of disassortative type. For two-state proteins, the clustering coefficients of Short-range Interaction Networks show prominent correlation with folding rate and contact order. The assortativity coefficients of Short-range Interaction Networks also show remarkable correlation with folding rate and contact order. Similar correlations exist in Protein Contact Networks of three-state proteins. For two-state proteins, the correlation between contact order and folding rate is determined by the numbers of local contacts. Short- range interactions play a key role in determining the connecting trend among amino acids and they impact the folding rate of two-state proteins directly. For three-state proteins, the folding rate is determined by short-range and long-range interactions among residues together.展开更多
We investigate the structural phase transitions and electronic properties of GaAs nanowires under high pressure by using synchrotron x-ray diffraction and infrared reflectance spectroscopy methods up to 26.2 GPa at ro...We investigate the structural phase transitions and electronic properties of GaAs nanowires under high pressure by using synchrotron x-ray diffraction and infrared reflectance spectroscopy methods up to 26.2 GPa at room temperature.The zinc-blende to orthorhombic phase transition was observed at around 20.0 GPa.In the same pressure range, pressureinduced metallization of GaAs nanowires was confirmed by infrared reflectance spectra.The metallization originates from the zinc-blende to orthorhombic phase transition.Decompression results demonstrated that the phase transition from zincblende to orthorhombic and the pressure-induced metallization are reversible.Compared to bulk materials, GaAs nanowires show larger bulk modulus and enhanced transition pressure due to the size effects and high surface energy.展开更多
AIM To develop a novel hepatocyte serum-free medium based on sericin,and to explore the effect of sericin on the hepatocyte transcriptome.METHODS A controlled trial comparing novel serum-free medium and other media: C...AIM To develop a novel hepatocyte serum-free medium based on sericin,and to explore the effect of sericin on the hepatocyte transcriptome.METHODS A controlled trial comparing novel serum-free medium and other media: C3 A cells were cultured in our novel serum-free medium,Hepato ZYME,complete medium(DMEM/F12 with 100 m L/L FBS),and DMEM/F12,andthen cell attachment,proliferation,and function as well as the biocompatibility of the media were assessed.A comparative study of serum-free media with or without 2 mg/m L sericin: the effect of sericin on C3 A growth was assessed by cell viability and proliferation,the effect of sericin on C3 A cell cycle distribution was determined by flow cytometry,and the effect of sericin on the C3 A transcriptome was assessed by gene-chip array and RT-q PCR.RESULTS More C3 A cells attached to the plate containing our serum-free medium than to those containing Hepato ZYME and DMEM/F12 at 24 h post-seeding.Both the viability and proliferation rate of C3 A cells in sericin-based serum-free medium were superior to those of cells in Hepato ZYME and DMEM/F12(P < 0.001).The content of albumin and urea in our serum-free medium was significantly higher than that in Hepato ZYME and DMEM/F12 throughout the whole culture period(P < 0.001) and was similar to that in complete medium at day 3,4,and 5.In part 2,cell viability and proliferation were greater in the presence of 2 mg/m L sericin(P < 0.001),as was the proportion of cells in S phase(16.21% ± 0.98% vs 12.61% ± 0.90%,P < 0.01).Gene-chip array analysis indicated that the expression of CCR6,EGFR,and FOS were up-regulated by 2 mg/m L sericin,and RT-q PCR revealed that the expression of CCR6,EGFR,FOS,AKT1,JNK1,NFk B1,MMP-9,MEK2,ERK1/2 and MYC was upregulated by 2 mg/m L sericin(P < 0.05).CONCLUSION We developed a novel hepatocyte serum-free medium.Sericin probably enhances cell attachment through the CCR6-Akt-JNK-NF-κB pathway and promotes cell proliferation through CCR6-mediated activation of the ERK1/2-MAPK pathway.展开更多

Methods: A total of 120 patients with diabetic nephropathy admitted to People's Hospital of De Yang, Sichuan, China were randomly divided into control group and observation group, with 60 cases in each group. Patients in the control group were given routine treatment, while patients in the observation group were given atorvastatin on the basis of routine treatment. Before and after treatment, the levels of blood urea nitrogen (BUN), creatinine (Scr), cystatin C (CysC), matrix metalloproteinase inhibitor-1 (TIMP-1), transforming growth factor-β1 (TGF-β1), malondialdehyde (MDA), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), matrix metalloprotein-9 (MMP-9) and tumor necrosis factor-α (TNF-α) were detected and compared between the two groups.Results: After treatment, the levels of BUN, Scr, CysC, TIMP-1, TGF-β1, MDA, MMP-9 and TNF-α of the patients in the control group were significantly decreased (P<0.05), while the levels of SOD and GSH-Px were significantly increased (P<0.05). After treatment, the levels of BUN, Scr, CysC, TIMP-1, TGF-β1, MDA, MMP-9, and TNF-α in the serum of the patients in the observation group were significantly decreased (P<0.05), while the levels of SOD and GSH-Px were significantly increased (P<0.05). The above indexes of the patients in the observation group were better than those of the control group (P<0.05). Conclusions: Atorvastatin can effectively alleviate renal function damage, renal fibrosis and inflammatory stress in patients with diabetic nephropathy, enhance the antioxidant capacity of the body, and have a certain protective effect on the kidney.展开更多

Salt,saline-alkali conditions,and drought are major environmental factors limiting plant growth and productivity.The vacuolar H+-translocating inorganic pyrophosphatase(V-H+-PPase) is an electrogenic proton pump that ...Salt,saline-alkali conditions,and drought are major environmental factors limiting plant growth and productivity.The vacuolar H+-translocating inorganic pyrophosphatase(V-H+-PPase) is an electrogenic proton pump that translocates protons into vacuoles in plant cells.Expression of V-H+-PPase increases in plants under a number of abiotic stresses,and is thought to have an important role in adaptation to abiotic stress.In this work,we report the isolation and characterization of the gene,ScVP,encoding a vacuolar inorganic pyrophosphatase(V-H+-PPase) from the halophyte,Suaeda corniculata.Semiquantitative reverse transcription-polymerase chain reaction analysis showed that ScVP was induced in roots,stems and leaves under treatment with salt,saline-alkali and drought.Compared with wild-type(WT) Arabidopsis,transgenic plants overexpressing ScVP accumulated more Na+ in leaves and roots,and showed increased tolerance to high salinity,saline-alkali and drought stresses.The germination percentage of transgenic Arabidopsis seeds was higher than that of WT seeds under the abiotic stresses.The root length of transgenic plants under salt stress was longer than that of WT plants.Furthermore,the rate of water loss during drought stress was higher in WT than in transgenic plants.Collectively,these results indicate that ScVP plays an important role in plant tolerance to salt,saline-alkali and drought stress.展开更多

Methods Liposomes of rutin were prepared by film dispersion method and the encapsulation efficiency(EE) was determined by RP-HPLC. Human umbilical vein endothelial cells(HUVECs) were injuried by H_2O_2 and treated with either free aqueous rutin or the RL delivery systems. The viability of HUVECs was determined by MTT and ELISA. Results The drug delivery system showed uniform rutin loaded nanoparticles with average particle size of(147.20 ± 1.42) nm, polydispersity index of(0.191 ± 0.003) nm, Zeta potential of(-20.0 ± 1.0) mV, and the drug EE was closed to 90.0%. The antioxidant effect of the drug delivery system to H_2O_2-damaged HUVECs showed that RL could increase injury cells viability compared to free aqueous rutin, which was accompanied with an obvious decrease in malondialdehyde(MDA), lactate dehydrogenase(LDH) while increase the level of nitrogen oxide(NOS). This study analyzed the effects of male age and abstinence time on semen quality and explored the best abstinence time for Chinese males among different age groups.Semen parameters,including sperm kinetics,morphology,...This study analyzed the effects of male age and abstinence time on semen quality and explored the best abstinence time for Chinese males among different age groups.Semen parameters,including sperm kinetics,morphology,and DNA fragmentation index(DFI),were reviewed from 2952 men.Samples were divided into six age groups(≤25 years,26–30 years,31–35 years,36–40 years,41–45 years,and>45 years)and were divided into six groups according to different abstinence time(2 days,3 days,4 days,5 days,6 days,and 7 days).The differences in semen quality between the groups were compared,and the effect of age and abstinence time on semen quality was analyzed.Significant differences were observed in semen volume,progressive motility(PR),and DFI among the age groups(all P<0.05),and no significant differences were observed in sperm morphological parameters(all P>0.05).There were significant differences in semen volume,PR,and DFI among different abstinence time groups(all P<0.05)and no significant differences in sperm morphological parameters(all P>0.05).Pearson analysis showed that male age and abstinence time were both significantly correlated with sperm kinetics and DFI(both P<0.05),while no significant correlation was found with sperm morphological parameters(all P>0.05).The box plots and histograms of men’s age,abstinence time,and semen quality show that most semen quality parameters differ significantly between the 2 days and 7 days abstinence groups and other groups at different ages.Except for the sperm morphology parameters,sperm kinetic parameters and sperm DFI are linearly related to male age and abstinence time.展开更多

文摘Well-ordered aluminosilicates(MAs)were prepared by in-situ assembly of pre-crystallized units of zeolite Y precursors at a commercial scale,and applied in an industrial fluid catalytic cracking unit for the first time.Compared with incumbent equilibrium catalyst,the surface area of trial equilibrium catalysts(30%inventory ratio)increased from 110 m^(2)g^(-1)to 120m^(2)g^(-1).Moreover,a significant increase of the mesoporous surfaceareaof trial equlibrium catalysts(30%inventoryrati)from 33 m g/to 40magi(22%increase).Furthermore,the equilibrium catalyst that contain 80%LPC-65 yields significantly lower heavy oil(0.23%)and higher total liquids(0.53%)compared with LDO-70.The industrial results demonstrated excellent hydrothermal stability and superior catalytic cracking properties,showing the promising futurein the industrial units.
文摘AIM: To assess the predictive value of Operative Link on Gastritis Assessment(OLGA) and Operative Link on Gastric Intestinal Metaplasia Assessment(OLGIM) stages in gastric cancer.METHODS: A prospective study was conducted with 71 patients with early gastric cancer(EGC) and 156 patients with non-EGC. All patients underwent endoscopic examination and systematic biopsy. Outcome measures were assessed and compared, including the Japanese endoscopic gastric atrophy(EGA) classification method and the modified OLGA method as well as the modified OLGIM method. Helicobacter pylori(H. pylori) status was determined for all study participants. Stepwise logistic regression modeling was performed to analyze correlations between EGC and the EGA, OLGA and OLGIM methods.RESULTS: For patients with EGC and patients with non-EGC, the proportions of moderate-to-severe EGA cases were 64.8% and 44.9%, respectively(P = 0.005), the proportions of OLGA stages Ⅲ-Ⅳ cases were 52.1% and 22.4%, respectively(P < 0.001), and the proportions of OLGIM stages Ⅲ-Ⅳ cases were42.3% and 19.9%, respectively(P < 0.001). OLGA stage and OLGIM stage were significantly related to EGA classification; specifically, logistic regression modeling showed significant correlations between EGC and moderate-to-severe EGA(OR = 1.95, 95% CI: 1.06-3.58, P = 0.031) and OLGA stages Ⅲ-Ⅳ(OR = 3.14, 95%CI: 1.71-5.81, P < 0.001), but no significant correlation between EGC and OLGIM stages Ⅲ-Ⅳ(P = 0.781). H. pylori infection rate was significantly higher in patients with moderate-to-severe EGA(75.0% vs 54.1%, P = 0.001) or OLGA/OLGIM stages Ⅲ-Ⅳ(OLGA: 83.6% vs 55.8%, P < 0.001; OLGIM: 83.6% vs 57.8%, P < 0.001).CONCLUSION: OLGA classification is optimal for EGC screening. A surveillance program including OLGA stage and H. pylori infection status may facilitate early detection of gastric cancer.

文摘AIM: To investigate the interaction between heat shock protein 70 (HSP70) and α-fetoprotein (AFP) in human hepatocellular carcinoma (HCC) cell line BEL7402.METHODS: The expression and localization of HSP70 and AFP in human HCC cell line BEL-7402 were determined by immunocytochemistry and indirect immunofluorescence cytochemical staining. The interaction between HSP70 and AFP in HCC cells was analyzed by immunoprecipitation and Western blot.RESULTS: Immunocytochemical staining detection showed that HCC cell BEL-7402 expressed a high level of HSP70 and AFP synchronously. Both were stained in cell plasma.AFP existed in the immunoprecipitate of anti-HSP70 mAb,while there was HSP70 in the immunoprecipitate of antiAFP mAb.CONCLUSION: HSP70 chaperones AFP in human HCCcell BEL-7402. The interaction between HSP70 and AFP in human HCC cell can be a new route to study the pathogenesis and immunotherapy of HCC.

文摘Giant cell arteritis(GCA)is a commonly occurring large vacuities characterized by angiopathy of medium and large-sized vessels.GCA granulomatous formation plays an important role in the pathogenesis of GCA.Analysis of T cell lineages and signaling pathways in GCA have revealed the essential role of T cells in the pathology of GCA.T cells are the dominant population present in GCA lesions.CD4+T cell subtypes that are present include Th1,Th2,Th9,Th17,follicular helper T(Tfh)cells,and regulatory T(Treg)cells.CD8 T cells can primarily differentiate into cytotoxic CD8+T lymphocytes and Treg cells.The instrumental part of GCA is the interplay between dendritic cells,macrophages and endothelial cells,which can result in the vascular injury and the characteristics granulomatous infiltrates formation.During the inflammatory loop of GCA,several signaling pathways have been reported to play an essential role in recruiting,activating and differentiating T cells,including T-cell receptor(TCR)signaling,vascular endothelial growth factor(VEGF)-Jagged-Notch signaling and the Janus kinase and signal transducer and activator of transcription(STAT)pathway(JAK-STAT)pathway.In this review,we have focused on the role of T cells and their potential signaling mechanism(s)that are involved in the pathogenesis of GCA.A better understanding of the role of T cells mediated complicated orchestration during the homeostasis and the changes could possibly favor developments of novel treatment strategies against immunological disorders associated with GCA.

文摘AIM:To evaluate the effectiveness of knock-down of heat shock protein 47(HSP47)on conjunctival bleb scarring in a rat model and its possible mechanism.METHODS:Male Sprague–Dawley rats were used for glaucoma filtration surgery(GFS)and were treated with either phosphate buffered solution,shControl,mitomycin C,or sh-HSP47 using a microsyringe immediately after GFS.The morphology of filtering blebs was observed postoperatively.The levels of HSP47 were analyzed at 2,5,8,and 11d after GFS via real‑time quantitative polymerase chain reaction(PCR)and Western blot.The silencing effect of HSP47,the expression of collagen I and III,and the potential signaling pathways of HSP47 during scarification were explored 11d post GFS.The protein levels of transforming growth factor-β1(TGF-β1),phospho-Smad2(pSmad2),phospho-Smad3(p-Smad3),and phospho-p38(p-p38)were also analyzed using Western blot.RESULTS:Sh-HSP47 treatment significantly prolonged the functional filtration bleb retention.The levels of HSP47 were increased significantly at 5,8,and 11d postoperatively compared to the control group(P<0.05,P<0.01,and P<0.001).The levels of HSP47 protein at day 11 postoperatively were significantly down-regulated after HSP47 silencing using sh-HSP47 adenovirus transfection(P<0.01).Expression levels of collagen I and III within the blebs were significantly reduced in the absence of HSP47(P<0.01).Moreover,the protein levels of TGF-β1,p-Smad2/3,and p-p38 were dramatically inhibited after treatment with sh-HSP47(P<0.01).CONCLUSION:The inhibitory effects of HSP47 knockdown on scarring after GFS have the potential to be an efficacious therapeutic option for the treatment of conjunctival bleb scarring.
文摘BACKGROUND Intravenous leiomyomatosis(IVL) is a rare and complicated disease, which requires surgery by a multidisciplinary team. However, the optimal surgical approach has not been determined.CASE SUMMARY Here we report three cases of IVL treated with different surgical approaches. All patients presented with circulation symptoms. Two patients had lower extremity edema and the other had cardiopalmus. The diagnosis of IVL was confirmed based on the imagining examinations and pathological findings. All patients underwent surgical treatment and were discharged without any complications.CONCLUSION Preoperative examination is crucial for surgical planning and surgical approach is dependent on the patient's condition and tumor involvement.

文摘The purpose of this study was to investigate the effect of four fluorescent dyes, True Blue(TB), Fluoro-Gold(FG), Fluoro-Ruby(FR), and 1,1′-dioctadecyl-3,3,3′,3′-tetramethylindocarbocyanine perchlorate(Di I), in retrograde tracing of rat spinal motor neurons. We transected the muscle branch of the rat femoral nerve and applied each tracer to the proximal stump in single labeling experiments, or combinations of tracers(FG-Di I and TB-Di I) in double labeling experiments. In the single labeling experiments, significantly fewer labeled motor neurons were observed after FR labeling than after TB, FG, or Di I, 3 days after tracer application. By 1 week, there were no significant differences in the number of labeled neurons between the four groups. In the double-labeling experiment, the number of double-labeled neurons in the FG-Di I group was not significantly different from that in the TB-Di I group 1 week after tracer application. Our findings indicate that TB, FG, and Di I have similar labeling efficacies in the retrograde labeling of spinal motor neurons in the rat femoral nerve when used alone. Furthermore, combinations of Di I and TB or FG are similarly effective. Therefore, of the dyes studied, TB, FG and Di I, and combinations of Di I with TB or FG, are the most suitable for retrograde labeling studies of motor neurons in the rat femoral nerve.
文摘AIM: To demonstrate the change and effect of nociceptin/orphanin FQ in the colon of rats with cathartic colon. METHODS: The cathartic colon model was established by feeding rats rhubarb for 3 mo, the changes of colonic electromyography were investigated by both suspension muscle strips test and serosal recordings of colonic myoelectrical activity. Immunohistochemical staining (S-P methods) and image analysis were used to determine the changes of nociceptin/orphanin FQ in the proximal colon and distal colon of rats with cathartic colon. RESULTS: Suspension muscle strips test in vitro showed OFQ (10-9-10-6 mol/L) concentration dependently caused an immediate tonic contraction in the isolated colon. But the increase of tension in cathartic colon was less than control groups (P < 0.01). Intravenous administration of OFQ (1 μg/kg) caused phasic contractions in the proximal colon, while the amplitude of phasic contractions caused by OFQ in cathartic colon was much lower than that in the control groups (2.58 ± 0.41 vs 4.16 ± 0.53, t = -2.6, P = 0.012). OFQ was highly expressed in the myenteric plexus of the rat colon but not in the muscle cells. The immunoreactivity of OFQ in the proximal colon in cathartic colon rats decreased significantly compared with the control group (P = 0.001). CONCLUSION: Colonic smooth muscle of cathartic colon showed low sensitivity to the stimulation of OFQ, suggesting that it might be caused by the abnormal distribution of OFQ or the abnormalities of receptors, leading to the disorganization of dynamic and incoordinated contractions.

文摘Background Hyperprolactinaemia is a common adverse effect of antipsychotics(APs). The results of PeonyGlycyrrhiza decoction(PGD) as a potentially useful adjunctive treatment for hyperprolactinaemia are inconsistent.Aim This meta-analysis of randomised controlled trials(RCTs) examined the efficacy and safety of adjunctive PGD therapy for AP-induced hyperprolactinaemia.Methods English(PubMed, Embase, Cochrane Library,PsycINFO) and Chinese(Chinese National Knowledge Infrastructure, Wanfang Data) databases were systematically searched up to 10 June 2018. The inclusion criteria were based on PIC0 S一/^rticipants: adult patients with schizophrenia; Aitervention: PGD plus APs; Cbmparison: APs plus placebo or AP monotherapy;(Xjtcomes: efficacy and safety; Study design: RCTs. The weighted mean difference(WMD) and risk ratio(RR) along with their 95% CIs were calculated using Review Manager(RevMan) V.5.3 software.Results Five RCTs(n=450) were included and analysed.Two RCTs(n=140) were double-blind and four RCTs(n=409) reported ‘random' assignment with specific description. The PGD group showed a significantly lower serum prolactin level at endpoint than the control group(n=380, WMD:-32.69 ng/mL(95% Cl-41.66 to 23.72),/X0.00001, ^=97%). Similarly, the superiority of PGD over the control groups was also found in the improvement of hyperprolactinaemia-related symptoms. No difference was found in the improvement of psychiatric symptoms assessed by the Positive and Negative Syndrome Scale(n=403,WMD:-0.62(95% Cl-2.38 to 1.15), p=0.49, ^=0%). There were similar rates of all-cause discontinuation(n=330, RR0.93(95% Ci 0.63 to 1.37),/t=0.71, ^=0%) and adverse drug reactions between the two groups. According to the Grading of Recommendations Assessment, Development and Evaluation approach, the level of evidence of primary and secondary outcomes ranged from ‘very low'(14.3%),‘low'(42.8%),‘moderate'(14.3%),to ‘high,(28.6%>.Conclusions Current evidence supports the adjunctive use of PGD to suppress elevated prolactin and improve prolactininduced symptoms without significant adverse events in adult patients with AP-induced hyperprolactinaemia. Highquality RCTs with longer duration are needed to confirm these findings.Trial registration number 42016037017.
文摘It is a challenging task to investigate the different in- fluence of long-range and short-range interactions on two-state and three-state folding kinetics of protein. The networks of the 30 two-state proteins and 15 three-state proteins were constructed by complex networks analysis at three length scales: Protein Contact Networks, Long-range Interaction Networks and Short-range Interaction Networks. To uncover the relationship between structural properties and folding kinetics of the proteins, the correlations of protein network parameters with protein folding rate and topology parameters contact order were analyzed. The results show that Protein Contact Networks and Short-range Interaction Networks (for both two-state and three-state proteins) exhibit the “small-world” property and Long-range Interaction networks indicate “scale-free” behavior. Our results further indicate that all Protein Contact Networks and Short- range Interaction networks are assortative type. While some of Long-range Interaction Networks are of assortative type, the others are of disassortative type. For two-state proteins, the clustering coefficients of Short-range Interaction Networks show prominent correlation with folding rate and contact order. The assortativity coefficients of Short-range Interaction Networks also show remarkable correlation with folding rate and contact order. Similar correlations exist in Protein Contact Networks of three-state proteins. For two-state proteins, the correlation between contact order and folding rate is determined by the numbers of local contacts. Short- range interactions play a key role in determining the connecting trend among amino acids and they impact the folding rate of two-state proteins directly. For three-state proteins, the folding rate is determined by short-range and long-range interactions among residues together.

文摘We investigate the structural phase transitions and electronic properties of GaAs nanowires under high pressure by using synchrotron x-ray diffraction and infrared reflectance spectroscopy methods up to 26.2 GPa at room temperature.The zinc-blende to orthorhombic phase transition was observed at around 20.0 GPa.In the same pressure range, pressureinduced metallization of GaAs nanowires was confirmed by infrared reflectance spectra.The metallization originates from the zinc-blende to orthorhombic phase transition.Decompression results demonstrated that the phase transition from zincblende to orthorhombic and the pressure-induced metallization are reversible.Compared to bulk materials, GaAs nanowires show larger bulk modulus and enhanced transition pressure due to the size effects and high surface energy.

文摘AIM To develop a novel hepatocyte serum-free medium based on sericin,and to explore the effect of sericin on the hepatocyte transcriptome.METHODS A controlled trial comparing novel serum-free medium and other media: C3 A cells were cultured in our novel serum-free medium,Hepato ZYME,complete medium(DMEM/F12 with 100 m L/L FBS),and DMEM/F12,andthen cell attachment,proliferation,and function as well as the biocompatibility of the media were assessed.A comparative study of serum-free media with or without 2 mg/m L sericin: the effect of sericin on C3 A growth was assessed by cell viability and proliferation,the effect of sericin on C3 A cell cycle distribution was determined by flow cytometry,and the effect of sericin on the C3 A transcriptome was assessed by gene-chip array and RT-q PCR.RESULTS More C3 A cells attached to the plate containing our serum-free medium than to those containing Hepato ZYME and DMEM/F12 at 24 h post-seeding.Both the viability and proliferation rate of C3 A cells in sericin-based serum-free medium were superior to those of cells in Hepato ZYME and DMEM/F12(P < 0.001).The content of albumin and urea in our serum-free medium was significantly higher than that in Hepato ZYME and DMEM/F12 throughout the whole culture period(P < 0.001) and was similar to that in complete medium at day 3,4,and 5.In part 2,cell viability and proliferation were greater in the presence of 2 mg/m L sericin(P < 0.001),as was the proportion of cells in S phase(16.21% ± 0.98% vs 12.61% ± 0.90%,P < 0.01).Gene-chip array analysis indicated that the expression of CCR6,EGFR,and FOS were up-regulated by 2 mg/m L sericin,and RT-q PCR revealed that the expression of CCR6,EGFR,FOS,AKT1,JNK1,NFk B1,MMP-9,MEK2,ERK1/2 and MYC was upregulated by 2 mg/m L sericin(P < 0.05).CONCLUSION We developed a novel hepatocyte serum-free medium.Sericin probably enhances cell attachment through the CCR6-Akt-JNK-NF-κB pathway and promotes cell proliferation through CCR6-mediated activation of the ERK1/2-MAPK pathway.
文摘Objective: To investigate the effects of atorvastatin on renal function, renal fibrosis, oxidative stress and inflammatory factors in patients with diabetic nephropathy. Methods: A total of 120 patients with diabetic nephropathy admitted to People's Hospital of De Yang, Sichuan, China were randomly divided into control group and observation group, with 60 cases in each group. Patients in the control group were given routine treatment, while patients in the observation group were given atorvastatin on the basis of routine treatment. Before and after treatment, the levels of blood urea nitrogen (BUN), creatinine (Scr), cystatin C (CysC), matrix metalloproteinase inhibitor-1 (TIMP-1), transforming growth factor-β1 (TGF-β1), malondialdehyde (MDA), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), matrix metalloprotein-9 (MMP-9) and tumor necrosis factor-α (TNF-α) were detected and compared between the two groups.Results: After treatment, the levels of BUN, Scr, CysC, TIMP-1, TGF-β1, MDA, MMP-9 and TNF-α of the patients in the control group were significantly decreased (P<0.05), while the levels of SOD and GSH-Px were significantly increased (P<0.05). After treatment, the levels of BUN, Scr, CysC, TIMP-1, TGF-β1, MDA, MMP-9, and TNF-α in the serum of the patients in the observation group were significantly decreased (P<0.05), while the levels of SOD and GSH-Px were significantly increased (P<0.05). The above indexes of the patients in the observation group were better than those of the control group (P<0.05). Conclusions: Atorvastatin can effectively alleviate renal function damage, renal fibrosis and inflammatory stress in patients with diabetic nephropathy, enhance the antioxidant capacity of the body, and have a certain protective effect on the kidney.

文摘Objective:To investigate the effect of paraoxonase 1 gene polymorphism on carotid plaque stability with cerebral infarction in Hainan population.Methods:277 patients of caroticl plaque With cerebral infarction who underwent physical examination in a hospital in Hainan from 2015 to another awarding 2018 were selected as the experimental group and the 363 people who no cerebral infarction as the Analytical methods:control group.The clinical data analyzed.DNA was collected from peripheral blood of two groups of patients and genotyped by flight mass analytical methods.''AG and GG could be detected by rs3917538.The distribution frequencies of The three genotypes in The control group accorded with Hardy-Weinberg equilibrium.Results:The distribution frequencies of AA,AG and GG in the control group were 97(26.7%),175(48.2%)and 91(25.1%)respectively.In the experimental group,the distribution frequencies were 76(27.4%),136(49.1%)and 65(23.5%).There were no statistical differences among the three detection methods of co-dominant model,Dominant model and recessive model.There was no difference in the frequency of allele A and G between groups.Conclusion:Polymorphism of paraoxonase 1 gene rs3917538 has No significant effect on carotid plaque formation and cerebral infarction in Hainan population.The Supplementary sample size to add more SNP research sites for further study,It is expected to further Revral the relationship between PON1and carotial piaque complicatecl with cerebral infarction in Hainan.

文摘Salt,saline-alkali conditions,and drought are major environmental factors limiting plant growth and productivity.The vacuolar H+-translocating inorganic pyrophosphatase(V-H+-PPase) is an electrogenic proton pump that translocates protons into vacuoles in plant cells.Expression of V-H+-PPase increases in plants under a number of abiotic stresses,and is thought to have an important role in adaptation to abiotic stress.In this work,we report the isolation and characterization of the gene,ScVP,encoding a vacuolar inorganic pyrophosphatase(V-H+-PPase) from the halophyte,Suaeda corniculata.Semiquantitative reverse transcription-polymerase chain reaction analysis showed that ScVP was induced in roots,stems and leaves under treatment with salt,saline-alkali and drought.Compared with wild-type(WT) Arabidopsis,transgenic plants overexpressing ScVP accumulated more Na+ in leaves and roots,and showed increased tolerance to high salinity,saline-alkali and drought stresses.The germination percentage of transgenic Arabidopsis seeds was higher than that of WT seeds under the abiotic stresses.The root length of transgenic plants under salt stress was longer than that of WT plants.Furthermore,the rate of water loss during drought stress was higher in WT than in transgenic plants.Collectively,these results indicate that ScVP plays an important role in plant tolerance to salt,saline-alkali and drought stress.

Human umbilical vein endothelial cells(HUVECs) were injuried by H_2O_2 and treated with either free aqueous rutin or the RL delivery systems. The viability of HUVECs was determined by MTT and ELISA. Results The drug delivery system showed uniform rutin loaded nanoparticles with average particle size of(147.20 ± 1.42) nm, polydispersity index of(0.191 ± 0.003) nm, Zeta potential of(-20.0 ± 1.0) mV, and the drug EE was closed to 90.0%. The antioxidant effect of the drug delivery system to H_2O_2-damaged HUVECs showed that RL could increase injury cells viability compared to free aqueous rutin, which was accompanied with an obvious decrease in malondialdehyde(MDA), lactate dehydrogenase(LDH) while increase the level of nitrogen oxide(NOS). Conclusion The nanostructured RL is improved on the antioxidant effect and may be treating the different diseases caused by free radicals.

文摘This study analyzed the effects of male age and abstinence time on semen quality and explored the best abstinence time for Chinese males among different age groups.Semen parameters,including sperm kinetics,morphology,and DNA fragmentation index(DFI),were reviewed from 2952 men.Samples were divided into six age groups(≤25 years,26–30 years,31–35 years,36–40 years,41–45 years,and>45 years)and were divided into six groups according to different abstinence time(2 days,3 days,4 days,5 days,6 days,and 7 days).The differences in semen quality between the groups were compared,and the effect of age and abstinence time on semen quality was analyzed.Significant differences were observed in semen volume,progressive motility(PR),and DFI among the age groups(all P<0.05),and no significant differences were observed in sperm morphological parameters(all P>0.05).There were significant differences in semen volume,PR,and DFI among different abstinence time groups(all P<0.05)and no significant differences in sperm morphological parameters(all P>0.05).Pearson analysis showed that male age and abstinence time were both significantly correlated with sperm kinetics and DFI(both P<0.05),while no significant correlation was found with sperm morphological parameters(all P>0.05).The box plots and histograms of men’s age,abstinence time,and semen quality show that most semen quality parameters differ significantly between the 2 days and 7 days abstinence groups and other groups at different ages.Except for the sperm morphology parameters,sperm kinetic parameters and sperm DFI are linearly related to male age and abstinence time.
