Objective Retinoblastoma(RB)is a prevalent type of eye cancer in youngsters.Prospero homeobox 1(Prox1)is a homeobox transcriptional repressor and downstream target of the proneural gene that is relevant in lymphatic,h...Objective Retinoblastoma(RB)is a prevalent type of eye cancer in youngsters.Prospero homeobox 1(Prox1)is a homeobox transcriptional repressor and downstream target of the proneural gene that is relevant in lymphatic,hepatocyte,pancreatic,heart,lens,retinal,and cancer cells.The goal of this study was to investigate the role of Prox1 in RB cell proliferation and drug resistance,as well as to explore the underlying Notch1 mechanism.Methods Human RB cell lines(SO-RB50 and Y79)and a primary human retinal microvascular endothelial cell line(ACBRI-181)were used in this study.The expression of Prox1 and Notch1 mRNA and protein in RB cells was detected using quantitative real time-polymerase chain reaction(RT-qPCR)and Western blotting.Cell proliferation was assessed after Prox1 overexpression using the Cell Counting Kit-8 and the MTS assay.Drug-resistant cell lines(SO-RB50/vincristine)were generated and treated with Prox1 to investigate the role of Prox1 in drug resistance.We employed pcDNA-Notch1 to overexpress Notch1 to confirm the role of Notch1 in the protective function of Prox1.Finally,a xenograft model was constructed to assess the effect of Prox1 on RB in vivo.Results Prox1 was significantly downregulated in RB cells.Overexpression of Prox1 effectively decreased RB cell growth while increasing the sensitivity of drug-resistant cells to vincristine.Notch1 was involved in Prox1’s regulatory effects.Notch1 was identified as a target gene of Prox1,which was found to be upregulated in RB cells and repressed by increased Prox1 expression.When pcDNA-Notch1 was transfected,the effect of Prox1 overexpression on RB was removed.Furthermore,by downregulating Notch1,Prox1 overexpression slowed tumor development and increased vincristine sensitivity in vivo.Conclusion These data show that Prox1 decreased RB cell proliferation and drug resistance by targeting Notch1,implying that Prox1 could be a potential therapeutic target for RB.展开更多
BACKGROUND:Edaravone(3-methyl-1-penyl-2-pyrazolin-5-one) is a potent free-radical scavenger and has the antioxidant ability to inhibit lipid peroxidation.The study aimed to examine the effect of edaravone on protectin...BACKGROUND:Edaravone(3-methyl-1-penyl-2-pyrazolin-5-one) is a potent free-radical scavenger and has the antioxidant ability to inhibit lipid peroxidation.The study aimed to examine the effect of edaravone on protecting the acute injury of human type II alveolar epithelial cells(A549cells) induced by paraquat(PQ) and the change of production of reactive oxygen species(ROS),malondialdehyde(MDA),superoxide dismutase(SOD).METHODS:A549 cells were cultured and divided into PQ group(group P),edaravone-treated group(group E) and normal control group(group C).The cells in group P were exposed to paraquat(600 umol/L),and the cells in group E were treated with edaravone(100 umol/L) additionally,and no drug intervention was given to the cells in group C.Real-time monitoring by LSCM was used to detect the cell response and the intracellular dynamic change of ROS level in A549 cells after administration of PQ and edaravone.And the levels of SOD and MDA were detected respectively by biochemistry colorimetry.Data were expressed as mean ± standard error of the mean.Statistical analysis was carried out with the soft SPSS 16.0.RESULTS:The concentration of intracellular ROS significantly increased when PQ was given to A549 cells.But after administration of edaravone,the concentration of intracellular ROS was decreased.Compared to the PQ group,the levels of SOD in the edaravone group were significantly increased while the levels of MDA were markedly decreased.CONCLUSIONS:Paraquat can increase the oxidative stress,and induce the lipid peroxidation of A549 cells.Edaravone has the effect to scavenge reactive oxygen species,and to protect against the PQ-induced lung toxicity.展开更多
New samples returned by China Chang’e-5(CE-5)mission offer an opportunity for studying the lunar geologic longevity,space weathering,and regolith evolution.The age determination of the CE-5 samples was among the firs...New samples returned by China Chang’e-5(CE-5)mission offer an opportunity for studying the lunar geologic longevity,space weathering,and regolith evolution.The age determination of the CE-5 samples was among the first scientific questions to be answered.However,the precious samples,most in the micrometer size range,challenge many traditional analyses on large single crystals of zircon developed for massive bulk samples.Here,we developed a non-destructive rapid screening of individual zirconium-containing particle for isotope geochronology based on a Micro X-ray fluorescence analysis(μXRF).The selected particles were verified via scanning electron microscopy(SEM),3D X-ray microscopy(XRM),and focused ion beam scanning electron microscopy(FIB-SEM)techniques,which showed that zirconium-bearing minerals with several microns were precisely positioned and readily suitable for site-specific isotopic dating by second ion mass spectrometry(SIMS).Such protocol could be also appli-cable in non-destructively screening other types of particles for different scientific purposes.We there-fore proposed a correlative workflow for comprehensively studying the CE-5 lunar samples from single particles on nanometer to atomic scales.Linking various microscopic and spectromicroscopic instru-ments together,this workflow consists of six steps:(1)single-particle selection with non-destructive μXRF technique,(2)2D/3D morphological and structural characterization with a correlative submicron 3D XRM and nanoscale resolution FIB-SEM imaging methods,(3)SEM analysis of the surface morphology and chemistry of the selected particle,(4)a series of microscopic and microbeam analyses(e.g.,SEM,electron probe microanalysis,and SIMS)on the cross-section of the selected particle to obtain structural,mineralogical,chemical,and isotopic features from the micron to nanometer scale,(5)advanced 2D/3D characterization and site-specific sample preparation of thin foil/tip specimens on a microregion of inter-est in the selected particle with FIB-SEM technique,and(6)comprehensive analyses on the FIB-milled specimens at nanometer to atomic scale with synchrotron-based scanning transmission X-ray micro-scopy,analytic transmission electron microscopy,and atom probe tomography.Following this technical roadmap,one can integrate multiple modalities into a uniform frame of multimodal and multiscale cor-related datasets to acquire high-throughput information on the limited or precious terrestrial and extraterrestrial samples.展开更多
Supercritical flows are ubiquitous in natural environments;however,there is rare 3D anatomy of their deposits.This study uses high-quality 3D seismic datasets from the world’s largest submarine fan,Bengal Fan,to inte...Supercritical flows are ubiquitous in natural environments;however,there is rare 3D anatomy of their deposits.This study uses high-quality 3D seismic datasets from the world’s largest submarine fan,Bengal Fan,to interpret 3D architectures and flow processes of Pliocene undulating bedforms that were related to supercritical flows.Bengal undulating bedforms as documented in this study were developed in unconfined settings,and are seismically imaged as strike-elongated,crescentic bedforms in plan view and as rhythmically undulating,upstream migrating,erosive,discontinuous reflections in section view.Their lee sides are overall 3 to 4 times steeper(0.280 to 1.19°in slope)and 3 to 4 times shorter(117 to 419 m in length)than their stoss flanks and were ascribed to faster(high flow velocities of 2.70 to 3.98 m/s)supercritical flows(Froude numbers of 1.53 to 2.27).Their stoss sides,in contrast,are overall 3 to 4 times gentler(0.120 to 0.270 in slope)and 3 to 4 times longer(410 to 1139 m in length)than their lee flanks and were related to slower(low velocities of 2.35 to 3.05 m/s)subcritical flows(Froude numbers of 0.58 to 0.97).Bengal wave-like features were,thus,created by supercriticalto-subcritical flow transformations through internal hydraulic jumps(i.e.,cyclic steps).They have crests that are positive relative to the surrounding region of the seafloor,suggesting the predominant deposition of draping sediments associated with net-depositional cyclic steps.Turbidity currents forming Bengal wave-like features were,thus,dominated by deposition,resulting in net-depositional cyclic steps.Sandy deposits associated with Bengal net-depositional cyclic steps are imaged themselves as closely spaced,strike-elongated high RMS-attribute patches,thereby showing closely spaced,long and linear,strike-elongated distribution patterns.展开更多
Objective:To observe the expression of Long non-coding RNA antisense mitochondrial non-coding RNA-2 (ASncmtRNA-2) in high glucose (HG) treated human renal mesangial cells (HRMCs) and the role of ASncmtRNA-2 in oxidati...Objective:To observe the expression of Long non-coding RNA antisense mitochondrial non-coding RNA-2 (ASncmtRNA-2) in high glucose (HG) treated human renal mesangial cells (HRMCs) and the role of ASncmtRNA-2 in oxidative stress mediated diabetic nephropathy (DN) fibrosis.Methods: The expression levels of ASncmtRNA-2、transforming growth factorβ1 (TGF-β1) and fibronectin (FN) mRNA in cultured HRMCs were measured by qRT-PCR. In addition, relative reactive oxygen species (ROS) levels in HRMCs were detected with the non-fluorescent probe DCFH-DA assays.Results: Compared with 0h, the expression of ASncmtRNA-2 remained unchanged in all groups at 8 h post treatment. However, the level of ASncmtRNA-2 mRNA was increased significantly in HG and HG+NG-nitro-L-Arginine methylester (L-NAME) treated cells compared with low glucose (LG) treated cells from 16h onwards, while the level of ASncmtRNA-2 mRNA in the HG+L-NAME group was decreased compared with the HG group. Moreover, ROS fluorescence was significantly up-regulated in HG-treated cells compared with LG-treated cells, while the ROS fluorescence in HG+L-NAME group was suppressed compared with HG-treated cells. In addition, Levels of ASncmtRNA-2, TGF-β1 and FN mRNA were significantly up-regulated in HG treated cells compared with LG treated cells while Levels of ASncmtRNA-2, TGF-β1 and FN mRNA in HG+L-NAME group were down-regulated compared with HG group. Finally, the expression of ASncmtRNA-2, TGF-β1 and FN mRNA were significantly decreased in HG+ASncmtRNA-2 siRNA group compared with HG group.Conclusion: ASncmtRNA-2 was up-regulated in HG treated cells and may promote glomerular fibrosis in DN via positively regulating the expression of pro-fibrotic factors. These findings may provide novel potential therapeutic treatments for DN.展开更多
Since the consecutive discovery of several gas fields from 2004 to present,the Rakhine Basin has been an active area for petroleum exploration in the Bay of Bengal.High-resolution 3D seismic data and well data from bl...Since the consecutive discovery of several gas fields from 2004 to present,the Rakhine Basin has been an active area for petroleum exploration in the Bay of Bengal.High-resolution 3D seismic data and well data from blocks AD1,AD6 and AD8 offshore northwest Myanmar are used to study the Miocene–Pleistocene depositional architecture and sedimentary evolution in the Rakhine Basin.Analysis of seismic facies and seismic attributes indicates that deep-water architectural elements include submarine canyons,confined slope channel complex systems,aggradational channel–levee complexes,isolated channels,frontal splays and mass-transport complexes,which have variable characters(shape,dimension,sedimentary architecture)within predominantly background deep-water slope-basin floor facies.Most of the sediments are interpreted to be sourced from the Ganges–Brahmaputra fluvio-deltaic system to the north with only minor lateral input from the IndoMyanmar Ranges to the east.Investigation of the depositional evolution and architectural elements transformation during the filling history of the Rakhine Basin suggests the Rakhine Basin experienced rapid progradation during the Oligocene–Middle/Upper Miocene,gradual retrogradation during the Middle/Upper Miocene–Early Pliocene and gradual progradation during the Early Pliocene–Pleistocene.Published exploration results indicate that the main reservoirs of the discoveries in blocks A1 and A3 are Pliocene frontal splays and channel–levee fills,dominated by fine and very fine-grained sandstones,in structural and structural–stratigraphic traps.Analytic results from seismic characters and several exploration wells indicate that channel complexes and associated overbanks and frontal splays with fine-grained sandstones and siltstones trapped by the four-way closures are primary reservoir targets.展开更多
Previous studies suggest that serotonin(5-HT) might interact with brain-derived neurotrophic factor(BDNF) during the stress response.However,the relationship between 5-HT and BDNF expression under purely psychological...Previous studies suggest that serotonin(5-HT) might interact with brain-derived neurotrophic factor(BDNF) during the stress response.However,the relationship between 5-HT and BDNF expression under purely psychological stress is unclear.In this study,one hour before psychological stress exposure,the 5-HT1 A receptor agonist 8-OH-DPAT or antagonist MDL73005,or the 5-HT2 A receptor agonist DOI or antagonist ketanserin were administered to rats exposed to psychological stress.Immunohistochemistry and in situ hybridization revealed that after psychological stress,with the exception of the ventral tegmental area,BDNF protein and m RNA expression levels were higher in the 5-HT1 A and the 5-HT2 A receptor agonist groups compared with the solvent control no-stress or psychological stress group in the CA1 and CA3 of the hippocampus,prefrontal cortex,central amygdaloid nucleus,dorsomedial hypothalamic nucleus,dentate gyrus,shell of the nucleus accumbens and the midbrain periaqueductal gray.There was no significant difference between the two agonist groups.In contrast,after stress exposure,BDNF protein and m RNA expression levels were lower in the 5-HT1 A and 5-HT2 A receptor antagonist groups than in the solvent control non-stress group,with the exception of the ventral tegmental area.Our findings suggest that 5-HT regulates BDNF expression in a rat model of acute psychological stress.展开更多
Severe fever with thrombocytopenia syndrome(SFTS)is an emerging infectious disease caused by severe fever with thrombocytopenia syndrome virus(SFTSV).SFTSV has been found in humans,ticks and animals,and SFTS has high ...Severe fever with thrombocytopenia syndrome(SFTS)is an emerging infectious disease caused by severe fever with thrombocytopenia syndrome virus(SFTSV).SFTSV has been found in humans,ticks and animals,and SFTS has high mortality and increasing prevalence in East Asia.In the study,the samples(heart,liver,lung,kidney,spleen,brain tissue and serum)were collected from 374 domestic animals and 241 wild animals in Pingqiao District and Xinxian County of Xinyang in Henan Province,China.275(44.72%,275/615)animals were positive for anti-SFTSV antibodies,the anti-SFTSV antibodies positive ratios of domestic and wild animals were 43.58%(163/374)and 46.47%(112/241),respectively.There was no significant difference in domestic and wild animals,but significant differences were detected among different species of animals(x^2=112.59,P<0.0001).Among 615 animals,105(17.07%,105/615)animals were positive for SFTSV RNA,and only one SFTSV strain was isolated from heart tissue of a yellow weasel.The phylogenetic analysis shows that the sequence from animals belonged to the same group with viral sequences obtained from humans.The animals maybe play a reservoir host in maintaining the life cycle of SFTSV in nature.展开更多

However, the level of ASncmtRNA-2 mRNA was increased significantly in HG and HG+NG-nitro-L-Arginine methylester (L-NAME) treated cells compared with low glucose (LG) treated cells from 16h onwards, while the level of ASncmtRNA-2 mRNA in the HG+L-NAME group was decreased compared with the HG group. Moreover, ROS fluorescence was significantly up-regulated in HG-treated cells compared with LG-treated cells, while the ROS fluorescence in HG+L-NAME group was suppressed compared with HG-treated cells. In addition, Levels of ASncmtRNA-2, TGF-β1 and FN mRNA were significantly up-regulated in HG treated cells compared with LG treated cells while Levels of ASncmtRNA-2, TGF-β1 and FN mRNA in HG+L-NAME group were down-regulated compared with HG group. Finally, the expression of ASncmtRNA-2, TGF-β1 and FN mRNA were significantly decreased in HG+ASncmtRNA-2 siRNA group compared with HG group.Conclusion: ASncmtRNA-2 was up-regulated in HG treated cells and may promote glomerular fibrosis in DN via positively regulating the expression of pro-fibrotic factors. These findings may provide novel potential therapeutic treatments for DN.
文摘Since the consecutive discovery of several gas fields from 2004 to present,the Rakhine Basin has been an active area for petroleum exploration in the Bay of Bengal.High-resolution 3D seismic data and well data from blocks AD1,AD6 and AD8 offshore northwest Myanmar are used to study the Miocene–Pleistocene depositional architecture and sedimentary evolution in the Rakhine Basin.Analysis of seismic facies and seismic attributes indicates that deep-water architectural elements include submarine canyons,confined slope channel complex systems,aggradational channel–levee complexes,isolated channels,frontal splays and mass-transport complexes,which have variable characters(shape,dimension,sedimentary architecture)within predominantly background deep-water slope-basin floor facies.Most of the sediments are interpreted to be sourced from the Ganges–Brahmaputra fluvio-deltaic system to the north with only minor lateral input from the IndoMyanmar Ranges to the east.Investigation of the depositional evolution and architectural elements transformation during the filling history of the Rakhine Basin suggests the Rakhine Basin experienced rapid progradation during the Oligocene–Middle/Upper Miocene,gradual retrogradation during the Middle/Upper Miocene–Early Pliocene and gradual progradation during the Early Pliocene–Pleistocene.Published exploration results indicate that the main reservoirs of the discoveries in blocks A1 and A3 are Pliocene frontal splays and channel–levee fills,dominated by fine and very fine-grained sandstones,in structural and structural–stratigraphic traps.Analytic results from seismic characters and several exploration wells indicate that channel complexes and associated overbanks and frontal splays with fine-grained sandstones and siltstones trapped by the four-way closures are primary reservoir targets.

文摘Previous studies suggest that serotonin(5-HT) might interact with brain-derived neurotrophic factor(BDNF) during the stress response.However,the relationship between 5-HT and BDNF expression under purely psychological stress is unclear.In this study,one hour before psychological stress exposure,the 5-HT1 A receptor agonist 8-OH-DPAT or antagonist MDL73005,or the 5-HT2 A receptor agonist DOI or antagonist ketanserin were administered to rats exposed to psychological stress.Immunohistochemistry and in situ hybridization revealed that after psychological stress,with the exception of the ventral tegmental area,BDNF protein and m RNA expression levels were higher in the 5-HT1 A and the 5-HT2 A receptor agonist groups compared with the solvent control no-stress or psychological stress group in the CA1 and CA3 of the hippocampus,prefrontal cortex,central amygdaloid nucleus,dorsomedial hypothalamic nucleus,dentate gyrus,shell of the nucleus accumbens and the midbrain periaqueductal gray.There was no significant difference between the two agonist groups.In contrast,after stress exposure,BDNF protein and m RNA expression levels were lower in the 5-HT1 A and 5-HT2 A receptor antagonist groups than in the solvent control non-stress group,with the exception of the ventral tegmental area.Our findings suggest that 5-HT regulates BDNF expression in a rat model of acute psychological stress.

文摘Severe fever with thrombocytopenia syndrome(SFTS)is an emerging infectious disease caused by severe fever with thrombocytopenia syndrome virus(SFTSV).SFTSV has been found in humans,ticks and animals,and SFTS has high mortality and increasing prevalence in East Asia.In the study,the samples(heart,liver,lung,kidney,spleen,brain tissue and serum)were collected from 374 domestic animals and 241 wild animals in Pingqiao District and Xinxian County of Xinyang in Henan Province,China.275(44.72%,275/615)animals were positive for anti-SFTSV antibodies,the anti-SFTSV antibodies positive ratios of domestic and wild animals were 43.58%(163/374)and 46.47%(112/241),respectively.There was no significant difference in domestic and wild animals,but significant differences were detected among different species of animals(x^2=112.59,P<0.0001).Among 615 animals,105(17.07%,105/615)animals were positive for SFTSV RNA,and only one SFTSV strain was isolated from heart tissue of a yellow weasel.The phylogenetic analysis shows that the sequence from animals belonged to the same group with viral sequences obtained from humans.The animals maybe play a reservoir host in maintaining the life cycle of SFTSV in nature.
