Invasive breast carcinoma(BRCA)is associated with poor prognosis and high risk of mortality.Therefore,it is critical to identify novel biomarkers for the prognostic assessment of BRCA.Methods:The expression data of po...Invasive breast carcinoma(BRCA)is associated with poor prognosis and high risk of mortality.Therefore,it is critical to identify novel biomarkers for the prognostic assessment of BRCA.Methods:The expression data of polo-like kinase 1(PLK1)in BRCA and the corresponding clinical information were extracted from TCGA and GEO databases.PLK1 expression was validated in diverse breast cancer cell lines by quantitative real-time polymerase chain reaction(qRT-PCR)and western blotting.Single sample gene set enrichment analysis(ssGSEA)was performed to evaluate immune infiltration in the BRCA microenvironment,and the random forest(RF)and support vector machine(SVM)algorithms were used to screen for the hub infiltrating cells and calculate the immunophenoscore(IPS).The RF algorithm and COX regression model were applied to calculate survival risk scores based on the PLK1 expression and immune cell infiltration.Finally,a prognostic nomogram was constructed with the risk score and pathological stage,and its clinical potential was evaluated by plotting calibration charts and DCA curves.The application of the nomogram was further validated in an immunotherapy cohort.Results:PLK1 expression was significantly higher in the tumor samples in TCGA-BRCA cohort.Furthermore,PLK1 expression level,age and stage were identified as independent prognostic factors of BRCA.While the IPS was unaffected by PLK1 expression,the TMB and MATH scores were higher in the PLK1-high group,and the TIDE scores were higher for the PLK1-low patients.We also identified 6 immune cell types with high infiltration,along with 11 immune cell types with low infiltration in the PLK1-high tumors.A risk score was devised using PLK1 expression and hub immune cells,which predicted the prognosis of BRCA patients.In addition,a nomogram was constructed based on the risk score and pathological staging,and showed good predictive performance.Conclusions:PLK1 expression and immune cell infiltration can predict post-immunotherapy prognosis of BRCA patients.展开更多

Kansui and licorice, one of T...Although compatibility is highly advocated in traditional Chinese medicine (TCM), inappropriate combination of some herbs may reduce the therapeutic action and even produce toxic effects. Kansui and licorice, one of TCM “Eighteen Incompatible Medicaments”, are the most representative cases of improper herbal combination, which may still be applied simultaneously under given conditions. However, the potential mechanism of their compatibility and incompatibility is unclear. In the present study, two different ratios of kansui and licorice, representing their compatibility and incompatibility respectively, were designed to elucidate their interaction by comparative plasma/tissue metabolomics and a heatmap with relative fold change. As a result, glycocholic acid, prostaglandin F2a, dihydroceramide and sphinganine were screened out as the principal alternative biomarkers of compatibility group;sphinganine, dihydroceramide, arachidonic acid, leukotriene B4, acetoacetic acid and linoleic acid were those of incompatibility group. Based on the values of biomarkers in each tissue, the liver was identified as the compatible target organ, while the heart, liver, and kidney were the incompatible target organs. Furthermore, important pathways for compatibility and incompatibility were also constructed. These results help us to better understand and utilize the two herbs, and the study was the first to reveal some innate characters of herbs related to TCM “Eighteen Incompatible Medicaments”.展开更多
Segment Routing(SR)is a new routing paradigm based on source routing and provide traffic engineering(TE)capabilities in IP network.By extending interior gateway protocol(IGP),SR can be easily applied to IP network.How...Segment Routing(SR)is a new routing paradigm based on source routing and provide traffic engineering(TE)capabilities in IP network.By extending interior gateway protocol(IGP),SR can be easily applied to IP network.However,upgrading current IP network to a full SR one can be costly and difficult.Hybrid IP/SR network will last for some time.Aiming at the low flexibility problem of static TE policies in the current SR networks,this paper proposes a Deep Reinforcement Learning(DRL)based TE scheme.The proposed scheme employs multi-path transmission and use DRL to dynamically adjust the traffic splitting ratio among different paths based on the network traffic distribution.As a result,the network congestion can be mitigated and the performance of the network is improved.Simulation results show that our proposed scheme can improve the throughput of the network by up to 9%than existing schemes.展开更多
Objective To investigate the clinical value of intravenous lidocaine in reducing CRBD in male patients who need catheterization after general anesthesia.Methods A total of 210 male patients who underwent elective gene...Objective To investigate the clinical value of intravenous lidocaine in reducing CRBD in male patients who need catheterization after general anesthesia.Methods A total of 210 male patients who underwent elective general anesthesia in our hospital from May 2017 to April 2018 were selected.Random number table method was used to divide into the lidocaine group(1.5 mg?kg-1 of intravenous lidocaine infusion during anesthesia induction,and continuous intravenous infusion of lidocaine 2 mg?kg-1?h-1 after intubation),Dexmedetomidine group(0.5μg?kg-1 of dexmedetomidine intravenously during anesthesia induction,continuous intravenous infusion of dexmedetomidine 0.4μg?kg-1?h-1 after intubation).Control group(intravenous saline infusion during anesthesia induction,continuous intravenous infusion of the same amount of saline after intubation),70 cases in each group.All three groups were placed in the bladder with a 16F comfortable urethral catheter lubricated with paraffin oil.The amount of sufentanil used during the operation,the amount of sufentanil used after surgery,and the CRBD score and VAS at different time points after extubation were compared.Scoring,HR,SBP.Results T1 to T4,the CRBD scores of the lidocaine group were lower than those of the dexmedetomidine group and the control group(P<0.05).The VAS scores of the lidocaine group were lower than those of the control group(P<0.05),the CRBD score and VAS score of the dexmedetomidine group were lower than those of the control group(P<0.05);From T0 to T4,the differences in HR and SBP between the three groups were not statistically significant(P>0.05).Conclusion Lidocaine administration in male patients requiring catheterization after general anesthesia can significantly reduce the incidence of CRBD.展开更多
Objective:To investigate the effects of sublingual dust mite drops on Pulmonary function, immunoglobulin and T lymphocyte subsets in patients with allergic rhinitis.Methods:One hundred and ten patients with allergic r...Objective:To investigate the effects of sublingual dust mite drops on Pulmonary function, immunoglobulin and T lymphocyte subsets in patients with allergic rhinitis.Methods:One hundred and ten patients with allergic rhinitis treated in our hospital from June 2014 to December 2016 were retrospectively analyzed. According to the treatment methods, the patients were divided into control group and observation group, with 53 cases in control group and 57 cases in observation group. The patients were given oral kastitium and risperidone nasal spray. The observation group was given sublingual dust mite droplet on the basis of the control group. Afterwards, the Pulmonary function, immunoglobulin and T lymphocyte subsets Group level testing.Results: After treatment, the levels of FEV1, FVC and PEF increased significantly in the two groups. After treatment, the levels of FEV1, FVC and PEF in the observation group were (2.97±0.09) L, (3.96±0.07) L and (6.61±0.03) L/S, were significantly higher than the control group;After treatment, the levels of sIgE and T-IgE in both groups were significantly decreased, sIgG4 levels were significantly increased,After treatment, the levels of sIgE and T-IgE in the observation group were (12.22±1.08) kUA/L and (155.32 ± 9.51) kUA/L, respectively, which were significantly lower than those in the control group,The level of sIgG4 was (434.17±12.31) mg/L, which was significantly higher than that of the control group;After treatment, the levels of CD3+, CD4+ and CD4+/CD8+ in both groups were significantly higher than those before treatment, and the levels of CD8+ in the two groups were significantly lower than those before treatment. The levels of CD3+, CD4+ and CD4+/CD8 in the two groups were (65.90±2.11)%, (39.18±0.33)% and (1.75±0.07), respectively, were significantly higher than that of the control group after treatment, the level of CD8+ was (24.13±0.77)%, which was significantly lower than that of the control group Significance.Conclusion: Sublingual dust mite drops can effectively improve pulmonary ventilation in patients with allergic rhinitis, regulate patient immune function, a significant effect, worthy of further clinical application.展开更多

Gut microbes exhibit complex interactions with their hosts and shape an organism's immune system throughout its lifespan.As the largest secondary lymphoid organ,the spleen has a wide range of immunological functio...Gut microbes exhibit complex interactions with their hosts and shape an organism's immune system throughout its lifespan.As the largest secondary lymphoid organ,the spleen has a wide range of immunological functions.To explore the role of microbiota in regulating and shaping the spleen,we employ scRNA-seq and Stereo-seq technologies based on germ-free(GF)mice to detect differences in tissue size,anatomical structure,cell types,functions,and spatial molecular characteristics.We identify 18 cell types,9 subtypes of T cells,and 7 subtypes of B cells.Gene differential expression analysis reveals that the absence of microorganisms results in alterations in erythropoiesis within the red pulp region and congenital immune deficiency in the white pulp region.Stereo-seq results demonstrate a clear hierarchy of immune cells in the spleen,including marginal zone(MZ)macrophages,MZ B cells,follicular B cells and T cells,distributed in a well-defined pattern from outside to inside.However,this hierarchical structure is disturbed in GF mice.Ccr7 and Cxcl13 chemokines are specifically expressed in the spatial locations of T cells and B cells,respectively.We speculate that the microbiota may mediate the structural composition or partitioning of spleen immune cells by modulating the expression levels of chemokines.展开更多

Yes-associated protein 1(YAP1) interacts with transcription factor TEA domain 1(TEAD1) and plays an important role in cell proliferation and survival. However, the role of YAP1 in Dox-induced cardiomyopathy has not been reported. In this study, the expression of YAP1 was reduced in clinical human failing hearts with dilated cardiomyopathy and Dox-induced in vivo and in vitro cardiotoxic model. Ectopic expression of Yap1 significantly blocked Dox-induced cardiomyocytes apoptosis in TEAD1 dependent manner. Isorhapontigenin(Isor) is a new derivative of stilbene and responsible for a wide range of biological processes. Here, we found that Isor effectively relieved Doxinduced cardiomyocytes apoptosis in a dose-dependent manner in vitro. Administration with Isor(30 mg/kg/day, intraperitoneally, 3 weeks) significantly protected against Dox-induced cardiotoxicity in mice. Interestingly, Isor increased Dox-caused repression in YAP1 and the expression of its target genes in vivo and in vitro. Knockout or inhibition of Yap1 blocked the protective effects of Isor on Dox-induced cardiotoxicity. In conclusion, YAP1 may be a novel target for Dox-induced cardiotoxicity and Isor might be a new compound to fight against Dox-induced cardiotoxicity by increasing YAP1 expression.展开更多
Background:Developing effective spinal cord repair strategies for spinal cord injury(SCI)is of great importance.Emerging evidence suggests that microRNAs(miRNAs)are closely linked to SCI recovery.This study aimed to i...Background:Developing effective spinal cord repair strategies for spinal cord injury(SCI)is of great importance.Emerging evidence suggests that microRNAs(miRNAs)are closely linked to SCI recovery.This study aimed to investigate the function of miR-34c in the neuronal recovery in rats with SCI.Methods:A rat model with SCI was established.Differentially expressed miRNAs were identified by a microarray analysis.MiR-34c expression in rats was measured by reverse transcription quantitative polymerase chain reaction.Altered expression of miR-34c or C-X-C motif ligand 14(CXCL14)was introduced in SCI rats to measure their roles in neuronal recovery.Western blot analysis was performed to determine the phosphorylation of Janus kinase 2(JAK2)and signal transducer and activator of transcription-3(STAT3).Neuronal apoptosis in rat spinal cord tissues was detected.The concentrations of SCI recovery-related proteins thyrotropin releasing hormone(TRH),prostacyclin(PGI2),and ganglioside(GM)were evaluated by enzyme-linked immunosorbent assay.Data were analyzed using a t-test with a one-way or two-way analysis of variance.Results:Rats with SCI presented decreased grip strength(112.03±10.64 vs.17.32±1.49 g,P<0.01),decreased miR-34c expression(7 days:3.78±0.44 vs.0.95±0.10,P<0.05),and increased CXCL14 expression(7 days:0.61±0.06 vs.2.91±0.27,P<0.01).MiR-34c was found to directly bind to CXCL14.Overexpression of miR-34c increased grip strength(11.23±1.08 vs.31.26±2.99 g,P<0.01)and reduced neuronal apoptosis in spinal cord tissues(53.61%±6.07%vs.24.59%±3.32%,P<0.01),and silencing of CXCL14 also increased the grip strength(12.76±1.13 vs.29.77±2.75 g,P<0.01)and reduced apoptosis in spinal cord tissues(55.74%±6.24%vs.26.75%±2.84%,P<0.01).In addition,miR-34c upregulation or CXCL14 downregulation increased the concentrations of TRH,PGI2,and GM,and reduced phosphorylation of JAK2 and STAT3 in rats with SCI(all P<0.01).Conclusion:The study provided evidence that miR-34c could promote neuronal recovery in rats with SCI through inhibiting CXCL14 expression and inactivating the JAK2/STAT3 pathway.This study may offer new insights into SCI treatment.展开更多

The chemical st...Nanorod hydroxyapatite (NRHA)/graphene oxide (GO)composites with weight ratios of 0.4,1.5, and 5have been fabricated by a facile ultrasonic-assisted method at room temperature and atmospheric pressure. The chemical structure properties and morphology of the composites were characterized by field emission source scanning electron microscope,X-ray diffraction,transmission electron microscopy,and high-resolution transmission electron microscopy.The results indicate that the NRHA/ GO composites have an irregular surface with different degree wrinkles and are stable,and NRHA are well combined with GO.In addition,the biomimetic mineralization mechanism of hydroxyapatite on the NRHA/GO composites in simulated body fluid (SBF)is presented.The presence of a bone-like apatite layer on the composite surface indicate that the NRHA/GO composites facilitate the nucleation and growth of hydroxyapatite crystals in SBF for biomimetic mineralization.Moreover,the NRHA-1.5/GO composite and pure GO were cultured with MC3T3-E1cells to investigate the proliferation and adhesion of cells.In vitro cytocompatibility evaluation demonstrated that the NRHA/GO composite can act as a good template for the growth and adhesion of cells. Therefore,the NRHA/GO composite could be applied as a GO-based,free-template,non-toxic,and bioactive composite to substitute for a damaged or defect bone.展开更多
Composite materials have elicited much interest because of their superior performance in the removal of toxic and radioactive uranyl ions from aqueous solutions.With polyethyleneimine as a functional group,carboxylate...Composite materials have elicited much interest because of their superior performance in the removal of toxic and radioactive uranyl ions from aqueous solutions.With polyethyleneimine as a functional group,carboxylated chitosan as a matrix,and oxidizing activated carbon as a nanofiller,this study synthesized a novel environment-friendly polyethylenimine-functionalized carboxylated chitosan/oxidized activated charcoal(PCO)biocomposite with a unique three-dimensional porous structure.PCO was synthesized through an easy chemical cross-linking method.Detailed characterization certified the formation of the unique three-dimensional porous structure.The obtained PCO was used to remove uranyl ions from an aqueous solution,demonstrating the maximum adsorption capacity of 450 mg·g^(−1).The adsorption capacity of PCO decreased by less than 7.51%after five adsorption-desorption cycles.PCO exhibited good adsorption selectivity(K_(d)=3.45×10^(4) mL·g^(−1))for uranyl ions.The adsorption mechanism of PCO was also discussed.The material showed good potential for application in the treatment of wastewater containing uranyl ions.展开更多
Little is known about how to precisely promote the selective production of either colloidal semiconductor metal chalcogenide(ME),magic-size clusters(MSCs),or quantum dots(QDs).Recently,a two-pathway model has been pro...Little is known about how to precisely promote the selective production of either colloidal semiconductor metal chalcogenide(ME),magic-size clusters(MSCs),or quantum dots(QDs).Recently,a two-pathway model has been proposed to comprehend their evolution;here,we reveal for the first time that the size of precursors plays a decisive role in the selected evolution pathway of MSCs and QDs.With the reaction of cadmium myristate(Cd(MA)2)and tri-n-octylphosphine selenide(SeTOP)in 1-octadecene(ODE)as a model system,the size of Cd precursors was manipulated by the steric hindrance of carboxylic acid(RCOOH)additive.Without RCOOH,the reaction produced both CdSe MSCs and QDs(from 100 to 240℃).With RCOOH,the reaction produced MSCs or QDs when R was small(such as CH3−)or large(such as C6H5−),respectively.According to the twopathway model,the selective evolution is attributed to the promotion and suppression of the self-assembly of Cd and Se precursors,respectively.We propose that the addition of carboxylic acid may occur ligand exchange with Cd(MA)2,causing the different sizes of Cd precursor.The results suggest that the size of Cd precursors regulates the self-assemble behavior of the precursors,which dictates the directed evolution of either MSCs or QDs.The present findings bring insights into the two-pathway model,as the size of M and E precursors determine the evolution pathways of MSCs or QDs,the understanding of which is of great fundamental significance toward mechanism-enabled design and predictive synthesis of functional nanomaterials.展开更多
Three novel rare-earth,nitrogen-rich and oxygen heterocyclic supramolecular complexes,namely[Nd(BTF)_(2)(H_(2)O)_(5)]_(n),[Sm(BTF)_(2)(H_(2)O)_(5)]_(n),and[Eu(BTF)_(2)(H_(2)O)_(5)]_(n),were synthesized.A single crysta...Three novel rare-earth,nitrogen-rich and oxygen heterocyclic supramolecular complexes,namely[Nd(BTF)_(2)(H_(2)O)_(5)]_(n),[Sm(BTF)_(2)(H_(2)O)_(5)]_(n),and[Eu(BTF)_(2)(H_(2)O)_(5)]_(n),were synthesized.A single crystal was obtained by the solvent evaporation method,and the structure and coordination mode of metal complexes were determined by single crystal X-ray diffraction.Results show that the supramolecular complexes contain many hydrogen bonds and thus have good thermal stability(T_(dec)>540 K).The thermal decomposition of ammonium perchlorate(AP)catalyzed by the complexes was investigated by differential thermal analysis,which reveals a pre-eminent catalytic effect on AP.The high temperature decomposition peak of AP can be advanced by nearly 90 K at the amount of added complexes of 10 wt%,and the activation energy of AP descent range is from 70 to 150 kJ/mol.The other properties were fully characterized through elemental analysis and Fourier transform infrared spectroscopy.展开更多

文摘Objective To investigate the clinical value of intravenous lidocaine in reducing CRBD in male patients who need catheterization after general anesthesia.Methods A total of 210 male patients who underwent elective general anesthesia in our hospital from May 2017 to April 2018 were selected.Random number table method was used to divide into the lidocaine group(1.5 mg?kg-1 of intravenous lidocaine infusion during anesthesia induction,and continuous intravenous infusion of lidocaine 2 mg?kg-1?h-1 after intubation),Dexmedetomidine group(0.5μg?kg-1 of dexmedetomidine intravenously during anesthesia induction,continuous intravenous infusion of dexmedetomidine 0.4μg?kg-1?h-1 after intubation).Control group(intravenous saline infusion during anesthesia induction,continuous intravenous infusion of the same amount of saline after intubation),70 cases in each group.All three groups were placed in the bladder with a 16F comfortable urethral catheter lubricated with paraffin oil.The amount of sufentanil used during the operation,the amount of sufentanil used after surgery,and the CRBD score and VAS at different time points after extubation were compared.Scoring,HR,SBP.Results T1 to T4,the CRBD scores of the lidocaine group were lower than those of the dexmedetomidine group and the control group(P<0.05).The VAS scores of the lidocaine group were lower than those of the control group(P<0.05),the CRBD score and VAS score of the dexmedetomidine group were lower than those of the control group(P<0.05);From T0 to T4,the differences in HR and SBP between the three groups were not statistically significant(P>0.05).Conclusion Lidocaine administration in male patients requiring catheterization after general anesthesia can significantly reduce the incidence of CRBD.
文摘Objective:To investigate the effects of sublingual dust mite drops on Pulmonary function, immunoglobulin and T lymphocyte subsets in patients with allergic rhinitis.Methods:One hundred and ten patients with allergic rhinitis treated in our hospital from June 2014 to December 2016 were retrospectively analyzed. According to the treatment methods, the patients were divided into control group and observation group, with 53 cases in control group and 57 cases in observation group. The patients were given oral kastitium and risperidone nasal spray. The observation group was given sublingual dust mite droplet on the basis of the control group. Afterwards, the Pulmonary function, immunoglobulin and T lymphocyte subsets Group level testing.Results: After treatment, the levels of FEV1, FVC and PEF increased significantly in the two groups. After treatment, the levels of FEV1, FVC and PEF in the observation group were (2.97±0.09) L, (3.96±0.07) L and (6.61±0.03) L/S, were significantly higher than the control group;After treatment, the levels of sIgE and T-IgE in both groups were significantly decreased, sIgG4 levels were significantly increased,After treatment, the levels of sIgE and T-IgE in the observation group were (12.22±1.08) kUA/L and (155.32 ± 9.51) kUA/L, respectively, which were significantly lower than those in the control group,The level of sIgG4 was (434.17±12.31) mg/L, which was significantly higher than that of the control group;After treatment, the levels of CD3+, CD4+ and CD4+/CD8+ in both groups were significantly higher than those before treatment, and the levels of CD8+ in the two groups were significantly lower than those before treatment. The levels of CD3+, CD4+ and CD4+/CD8 in the two groups were (65.90±2.11)%, (39.18±0.33)% and (1.75±0.07), respectively, were significantly higher than that of the control group after treatment, the level of CD8+ was (24.13±0.77)%, which was significantly lower than that of the control group Significance.Conclusion: Sublingual dust mite drops can effectively improve pulmonary ventilation in patients with allergic rhinitis, regulate patient immune function, a significant effect, worthy of further clinical application.
文摘To the Editor:Atopic dermatitis(AD)is a chronic,recurrent,and inflammatory skin disease that can be accompanied by asthma,allergic rhinitis,conjunctivitis,and other diseases.[1]Topical corticosteroids are mostly used in patients with AD.Considering the side effects of the long-term use of corticosteroids,additional topical treatments could be useful.

文摘Gut microbes exhibit complex interactions with their hosts and shape an organism's immune system throughout its lifespan.As the largest secondary lymphoid organ,the spleen has a wide range of immunological functions.To explore the role of microbiota in regulating and shaping the spleen,we employ scRNA-seq and Stereo-seq technologies based on germ-free(GF)mice to detect differences in tissue size,anatomical structure,cell types,functions,and spatial molecular characteristics.We identify 18 cell types,9 subtypes of T cells,and 7 subtypes of B cells.Gene differential expression analysis reveals that the absence of microorganisms results in alterations in erythropoiesis within the red pulp region and congenital immune deficiency in the white pulp region.Stereo-seq results demonstrate a clear hierarchy of immune cells in the spleen,including marginal zone(MZ)macrophages,MZ B cells,follicular B cells and T cells,distributed in a well-defined pattern from outside to inside.However,this hierarchical structure is disturbed in GF mice.Ccr7 and Cxcl13 chemokines are specifically expressed in the spatial locations of T cells and B cells,respectively.We speculate that the microbiota may mediate the structural composition or partitioning of spleen immune cells by modulating the expression levels of chemokines.

文摘As an effective anticancer drug, the clinical limitation of doxorubicin(Dox) is the time-and dose-dependent cardiotoxicity. Yes-associated protein 1(YAP1) interacts with transcription factor TEA domain 1(TEAD1) and plays an important role in cell proliferation and survival. However, the role of YAP1 in Dox-induced cardiomyopathy has not been reported. In this study, the expression of YAP1 was reduced in clinical human failing hearts with dilated cardiomyopathy and Dox-induced in vivo and in vitro cardiotoxic model. Ectopic expression of Yap1 significantly blocked Dox-induced cardiomyocytes apoptosis in TEAD1 dependent manner. Isorhapontigenin(Isor) is a new derivative of stilbene and responsible for a wide range of biological processes. Here, we found that Isor effectively relieved Doxinduced cardiomyocytes apoptosis in a dose-dependent manner in vitro. Administration with Isor(30 mg/kg/day, intraperitoneally, 3 weeks) significantly protected against Dox-induced cardiotoxicity in mice. Interestingly, Isor increased Dox-caused repression in YAP1 and the expression of its target genes in vivo and in vitro. Knockout or inhibition of Yap1 blocked the protective effects of Isor on Dox-induced cardiotoxicity. In conclusion, YAP1 may be a novel target for Dox-induced cardiotoxicity and Isor might be a new compound to fight against Dox-induced cardiotoxicity by increasing YAP1 expression.

文摘Background:Developing effective spinal cord repair strategies for spinal cord injury(SCI)is of great importance.Emerging evidence suggests that microRNAs(miRNAs)are closely linked to SCI recovery.This study aimed to investigate the function of miR-34c in the neuronal recovery in rats with SCI.Methods:A rat model with SCI was established.Differentially expressed miRNAs were identified by a microarray analysis.MiR-34c expression in rats was measured by reverse transcription quantitative polymerase chain reaction.Altered expression of miR-34c or C-X-C motif ligand 14(CXCL14)was introduced in SCI rats to measure their roles in neuronal recovery.Western blot analysis was performed to determine the phosphorylation of Janus kinase 2(JAK2)and signal transducer and activator of transcription-3(STAT3).Neuronal apoptosis in rat spinal cord tissues was detected.The concentrations of SCI recovery-related proteins thyrotropin releasing hormone(TRH),prostacyclin(PGI2),and ganglioside(GM)were evaluated by enzyme-linked immunosorbent assay.Data were analyzed using a t-test with a one-way or two-way analysis of variance.Results:Rats with SCI presented decreased grip strength(112.03±10.64 vs.17.32±1.49 g,P<0.01),decreased miR-34c expression(7 days:3.78±0.44 vs.0.95±0.10,P<0.05),and increased CXCL14 expression(7 days:0.61±0.06 vs.2.91±0.27,P<0.01).MiR-34c was found to directly bind to CXCL14.Overexpression of miR-34c increased grip strength(11.23±1.08 vs.31.26±2.99 g,P<0.01)and reduced neuronal apoptosis in spinal cord tissues(53.61%±6.07%vs.24.59%±3.32%,P<0.01),and silencing of CXCL14 also increased the grip strength(12.76±1.13 vs.29.77±2.75 g,P<0.01)and reduced apoptosis in spinal cord tissues(55.74%±6.24%vs.26.75%±2.84%,P<0.01).In addition,miR-34c upregulation or CXCL14 downregulation increased the concentrations of TRH,PGI2,and GM,and reduced phosphorylation of JAK2 and STAT3 in rats with SCI(all P<0.01).Conclusion:The study provided evidence that miR-34c could promote neuronal recovery in rats with SCI through inhibiting CXCL14 expression and inactivating the JAK2/STAT3 pathway.This study may offer new insights into SCI treatment.

文摘Nanorod hydroxyapatite (NRHA)/graphene oxide (GO)composites with weight ratios of 0.4,1.5, and 5have been fabricated by a facile ultrasonic-assisted method at room temperature and atmospheric pressure. The chemical structure properties and morphology of the composites were characterized by field emission source scanning electron microscope,X-ray diffraction,transmission electron microscopy,and high-resolution transmission electron microscopy.The results indicate that the NRHA/ GO composites have an irregular surface with different degree wrinkles and are stable,and NRHA are well combined with GO.In addition,the biomimetic mineralization mechanism of hydroxyapatite on the NRHA/GO composites in simulated body fluid (SBF)is presented.The presence of a bone-like apatite layer on the composite surface indicate that the NRHA/GO composites facilitate the nucleation and growth of hydroxyapatite crystals in SBF for biomimetic mineralization.Moreover,the NRHA-1.5/GO composite and pure GO were cultured with MC3T3-E1cells to investigate the proliferation and adhesion of cells.In vitro cytocompatibility evaluation demonstrated that the NRHA/GO composite can act as a good template for the growth and adhesion of cells. Therefore,the NRHA/GO composite could be applied as a GO-based,free-template,non-toxic,and bioactive composite to substitute for a damaged or defect bone.

文摘Composite materials have elicited much interest because of their superior performance in the removal of toxic and radioactive uranyl ions from aqueous solutions.With polyethyleneimine as a functional group,carboxylated chitosan as a matrix,and oxidizing activated carbon as a nanofiller,this study synthesized a novel environment-friendly polyethylenimine-functionalized carboxylated chitosan/oxidized activated charcoal(PCO)biocomposite with a unique three-dimensional porous structure.PCO was synthesized through an easy chemical cross-linking method.Detailed characterization certified the formation of the unique three-dimensional porous structure.The obtained PCO was used to remove uranyl ions from an aqueous solution,demonstrating the maximum adsorption capacity of 450 mg·g^(−1).The adsorption capacity of PCO decreased by less than 7.51%after five adsorption-desorption cycles.PCO exhibited good adsorption selectivity(K_(d)=3.45×10^(4) mL·g^(−1))for uranyl ions.The adsorption mechanism of PCO was also discussed.The material showed good potential for application in the treatment of wastewater containing uranyl ions.

文摘Little is known about how to precisely promote the selective production of either colloidal semiconductor metal chalcogenide(ME),magic-size clusters(MSCs),or quantum dots(QDs).Recently,a two-pathway model has been proposed to comprehend their evolution;here,we reveal for the first time that the size of precursors plays a decisive role in the selected evolution pathway of MSCs and QDs.With the reaction of cadmium myristate(Cd(MA)2)and tri-n-octylphosphine selenide(SeTOP)in 1-octadecene(ODE)as a model system,the size of Cd precursors was manipulated by the steric hindrance of carboxylic acid(RCOOH)additive.Without RCOOH,the reaction produced both CdSe MSCs and QDs(from 100 to 240℃).With RCOOH,the reaction produced MSCs or QDs when R was small(such as CH3−)or large(such as C6H5−),respectively.According to the twopathway model,the selective evolution is attributed to the promotion and suppression of the self-assembly of Cd and Se precursors,respectively.We propose that the addition of carboxylic acid may occur ligand exchange with Cd(MA)2,causing the different sizes of Cd precursor.The results suggest that the size of Cd precursors regulates the self-assemble behavior of the precursors,which dictates the directed evolution of either MSCs or QDs.The present findings bring insights into the two-pathway model,as the size of M and E precursors determine the evolution pathways of MSCs or QDs,the understanding of which is of great fundamental significance toward mechanism-enabled design and predictive synthesis of functional nanomaterials.

文摘Three novel rare-earth,nitrogen-rich and oxygen heterocyclic supramolecular complexes,namely[Nd(BTF)_(2)(H_(2)O)_(5)]_(n),[Sm(BTF)_(2)(H_(2)O)_(5)]_(n),and[Eu(BTF)_(2)(H_(2)O)_(5)]_(n),were synthesized.A single crystal was obtained by the solvent evaporation method,and the structure and coordination mode of metal complexes were determined by single crystal X-ray diffraction.Results show that the supramolecular complexes contain many hydrogen bonds and thus have good thermal stability(T_(dec)>540 K).The thermal decomposition of ammonium perchlorate(AP)catalyzed by the complexes was investigated by differential thermal analysis,which reveals a pre-eminent catalytic effect on AP.The high temperature decomposition peak of AP can be advanced by nearly 90 K at the amount of added complexes of 10 wt%,and the activation energy of AP descent range is from 70 to 150 kJ/mol.The other properties were fully characterized through elemental analysis and Fourier transform infrared spectroscopy.
