Src homology 2-containing protein tyrosine phosphatase 2(SHP2), encoded by PTPN11, is widely expressed in the human body and plays a role in inflammation through various mechanisms. Therefore, SHP2 is considered a potential target for the treatment of inflammation-related diseases. However, its role in secondary inflammation after spinal cord injury remains unclear. In this study, SHP2 was found to be abundantly expressed in microglia at the site of spinal cord injury. Inhibition of SHP2 expression using siRNA and SHP2 inhibitors attenuated the microglial inflammatory response in an in vitro lipopolysaccharide-induced model of inflammation. Notably, after treatment with SHP2 inhibitors, mice with spinal cord injury exhibited significantly improved hind limb locomotor function and reduced residual urine volume in the bladder. Subsequent in vitro experiments showed that, in microglia stimulated with lipopolysaccharide, inhibiting SHP2 expression promoted M2 polarization and inhibited M1 polarization. Finally, a co-culture experiment was conducted to assess the effect of microglia treated with SHP2 inhibitors on neuronal cells. The results demonstrated that inflammatory factors produced by microglia promoted neuronal apoptosis, while inhibiting SHP2 expression mitigated these effects. Collectively, our findings suggest that SHP2 enhances secondary inflammation and neuronal damage subsequent to spinal cord injury by modulating microglial phenotype. Therefore, inhibiting SHP2 alleviates the inflammatory response in mice with spinal cord injury and promotes functional recovery postinjury.展开更多

文摘Purple-leafed plants not only have a higher resistance to biotic and abiotic stresses, but also have higher ornamental value.Anthocyanins are vital for leaf color formation, growth and development of purple leaves. However, the molecular mechanism underlying purple leaf formation in Lagerstroemia indica remains unclear.Metabolomic and transcriptomic analysis of purple-leafed cultivar‘Ebony Embers’and greenleafed cultivar‘Arapahoe’showed that the high expression of anthocyanin structure genes induced hyperaccumulation of cyanidin and pelargonidin derivatives, making the leaves purple.LfiHY5,LfiMYB75 and LfibHLH1 were identified using correlation analysis and weighted gene co-expression network analysis. In‘Arapahoe’‘Ebony Embers’population,LfiHY5 and LfiMYB75 showed significant positive correlation with leaf anthocyanin content.Transient expression of LfiMYB75 and LfiHY5 in tobacco and purple-leafed crape myrtle indicated that the two genes activated anthocyanin synthesis.Yeast two-hybrid analysis showed that LfiMYB75 and LfibHLH1 could form a complex that enhanced anthocyanin synthesis.Yeast monohybrid and dual-luciferase assays confirmed that LfiHY5 activated the expression of LfiMYB75,to activate the transcription of anthocyanin structural genes LfiCHS and LfiANS.Moreover, there were three alleles of LfiHY5 in crape myrtle, and the different sequences had different activation effects on LfiMYB75.In conclusion, the results showed that LfiHY5 led to upregulate the transcription of LfiMYB75,and LfiMYB75 formed a complex with LfibHLH1,which increased the transcription level of LfiCHS and LfiANS to affect anthocyanin synthesis in crape myrtle.

文摘Electrocatalytic water splitting has been identified as a potential candidate for producing clean hydrogen energy with zero carbon emission. However, the sluggish kinetics of oxygen evolution reaction on the anode side of the watersplitting device significantly hinders its practical applications.Generally, the efficiency of oxygen evolution processes depends greatly on the availability of cost‐effective catalysts with high activity and selectivity. In recent years, extensive theoretical and experimental studies have demonstrated that cobalt(Co)‐based nanomaterials, especially low‐dimensional Co‐based nanomaterials with a huge specific surface area and abundant unsaturated active sites, have emerged as versatile electrocatalysts for oxygen evolution reactions, and thus, great progress has been made in the rational design and synthesis of Co‐based nanomaterials for electrocatalytic oxygen evolution reactions.Considering the remarkable progress in this area,in this timely review,we highlight the most recent developments in Co‐based nanomaterials relating to their dimensional control, defect regulation(conductivity),electronic structure regulation, and so forth.Furthermore,a brief conclusion about recent progress achieved in oxygen evolution on Co‐based nanomaterials,as well as an outlook on future research challenges,is given.

文摘Along with the progression of Internet of Things(IoT)technology, network terminals are becoming continuously more intelligent.IoT has been widely applied in various scenarios, including urban infrastructure, transportation, industry, personal life, and other socio-economic fields. The introduction of deep learning has brought new security challenges, like an increment in abnormal traffic, which threatens network security.Insufficient feature extraction leads to less accurate classification results. In abnormal traffic detection, the data of network traffic is high-dimensional and complex. This data not only increases the computational burden of model training but also makes information extraction more difficult. To address these issues, this paper proposes an MD-MRD-ResNeXt model for abnormal network traffic detection. To fully utilize the multi-scale information in network traffic,a Multi-scale Dilated feature extraction(MD)block is introduced. This module can effectively understand and process information at various scales and uses dilated convolution technology to significantly broaden the model’s receptive field. The proposed Max-feature-map Residual with Dual-channel pooling(MRD)block integrates the maximum feature map with the residual block. This module ensures the model focuses on key information, thereby optimizing computational efficiency and reducing unnecessary information redundancy.Experimental results show that compared to the latest methods, the proposed abnormal traffic detection model improves accuracy by about 2%.
文摘Background:Caffeic acid(CA)is considered a promising phytochemical that has inhibited numerous cancer cell proliferation. Therefore,it is gaining increasing attention due to its safe and pharmacological applications. In this study,we investigated the role of CA in inhibiting the Interleukin-6(IL-6)/Janus kinase(JAK)/Signal transducer and activator of transcription-3(STAT-3)mediated suppression of the proliferation signaling in human prostate cancer cells.Materials and Methods:The role of CA in proliferation and colony formation abilities was studied using 3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide(MTT)assay and colony formation assays.Tumour cell death and cell cycle arrest were identified usingflow cytometry techniques.CA treatment-associated protein expression of mitogen-activated protein kinase(MAPK)families,IL-6/JAK/STAT-3,proliferation,and apoptosis protein expressions in PC-3 and LNCaP cell lines were measured using Western blot investigation. Results:We have obtained that treatment with CA inhibits prostate cancer cells(PC-3 and LNCaP)proliferation and induces reactive oxygen species(ROS),cell cycle arrest, and apoptosis cell death in a concentration-dependent manner. Moreover,CA treatment alleviates the expression phosphorylated form of MAPK families,i.e.,extracellular signal-regulated kinase 1(ERK1),c-Jun N-terminal kinase(JNK),and p38 in PC-3 cells.IL-6 mediated JAK/STAT3 expressions regulate the proliferation and antiapoptosis that leads to prostate cancer metastasis and migration. Therefore,to mitigate the expression of IL-6/JAK/STAT-3 is considered an important target for the treatment of prostate cancer. In this study,we have observed that CA inhibits the expression of IL-6,JAK1,and phosphorylated STAT-3 in both PC-3 and LNCaP cells.Due to the inhibitory effect of IL-6/JAK/STAT-3,it resulted in decreased expression of cyclin-D1,cyclin-D2,and CDK1 in both PC-3 cells. In addition,CA induces apoptosis by enhancing the expression of Bax and caspase-3;and decreased expression of Bcl-2 in prostate cancer cells. Conclusions:Thus,CA might act as a therapeutical application against prostate cancer by targeting the IL-6/JAK/STAT3 signaling axis.

文摘The haloscope based on the TM_(010)mode cavity is a well-established technique for detecting QCD axions. However, the method has limitations in detecting high-mass axion due to significant volume loss in the high-frequency cavity.Utilizing a higher-order mode cavity can effectively reduce the volume loss of the high-frequency cavity. The rotatable dielectric pieces as a tuning mechanism can compensate for the degradation of the form factor of the higher-order mode.Nevertheless, the introduction of dielectric causes additional volume loss.To address these issues, this paper proposes a novel design scheme by adding a central metal rod to the higher-order mode cavity tuned by dielectrics, which improves the performance of the haloscope due to the increased effective volume of the cavity detector. The superiority of the novel design is demonstrated by comparing its simulated performance with previous designs. Moreover, the feasibility of the scheme is verified by the full-wave simulation results of the mechanical design model.

文摘Objective:Candida albicans is a common fungal pathogen that triggers complex host defense mechanisms, including coordinated innate and adaptive immune responses,to neutralize invading fungi effectively.Exploring the immune microenvironment has the potential to inform the development of therapeutic strategies for fungal infections. Methods:The study analyzed individual immune cell profiles in peripheral blood mononuclear cells from Candida albicans-infected mice and healthy control mice using single-cell transcriptomics, fluorescence quantitative PCR,and Western blotting. We investigated intergroup differences in the dynamics of immune cell subpopulation infiltration, pathway enrichment, and differentiation during Candida albicans infection. Results:Our findings indicate that infiltration of CD4^(+)naive cells, regulatory T(Treg)cells, and Microtubules(MT)-associated cells increased after infection, along with impaired T cell activity.Notably,CD4^(+) T cells and plasma cells were enhanced after infection, suggesting that antibody production is dependent on T cells. In addition,we screened 6 hub genes, transcription factor forkhead box protein 3(Foxp3),cytotoxic T-lymphocyte associated protein 4(CTLA4),Interleukin 2 Receptor Subunit Beta(Il2rb),Cd28,C-C Motif Chemokine Ligand 5(Ccl5),and Cd27 for alterations associated with CD4^(+) T cell differentiation. Conclusions:These results provide a comprehensive immunological landscape of the mechanisms of Candida albicans infection and greatly advance our understanding of adaptive immunity in fungal infections.
