Nanometer-sized silica spheres were covalently attached onto the surfaces of micrometer-sized silica spheres. Crosslinked polymer shells on the nano-sized spheres outside the attached area were formed by "graft-from" strategy through ATRP. After removal of the silica cores, single-hole hollow crosslinked polymer nanospheres were obtained. In this strategy, most of ATRP monomers may be used and thus many functional groups can be easily incorporated into the single-hole hollow crosslinked polymer nanospheres.展开更多

文摘Background Recombinant human growth hormone(rhGH)therapy has shown to improve height and body composition in children with Prader–Willi syndrome(PWS),the evidence of early rhGH treatment on motor and mental development is still accumulating. This study explored the time effect on psychomotor development, anthropometric indexes, and safety for infants and young children with PWS.Methods A phase 3,single-arm,multicenter,self-controlled study was conducted in six sites.Patients received rhGH at 0.5 mg/m2/day for first four weeks, and 1 mg/m2/day thereafter for up to 52 weeks.Motor development was measured using Peabody Developmental Motor Scales-second edition, mental development using Griffiths Development Scales-Chinese(GDS-C).Height standard deviation score(SDS),body weight SDS,and body mass index(BMI)SDS were also assessed. Results Thirty-five patients were enrolled totally.Significant improvements were observed in height, body weight, and BMI SDS at week 52;GDS-C score showed significant improvement in general quotient(GQ)and sub-quotients.In a linear regression analysis, total motor quotient(TMQ),gross motor quotient(GMQ),and fine motor quotient were negatively correlated with age;however, treatment may attenuate deterioration of TMQ and GMQ.Changes in GQ and locomotor sub-quotient in<9-month group were significantly higher than≥9-month group.Mild to moderate severity adverse drug reactions were reported in six patients. Conclusion Fifty-two-week treatment with rhGH improved growth,BMI,mental development, and lessened the deterioration of motor function in infants and young children with PWS.Improved mental development was more pronounced when instituted in patients<9 months old.

文摘Background:Hepatocellular carcinoma(HCC)has a poor long-term prognosis. The competition of circular RNAs(circRNAs)with endogenous RNA is a novel tool for predicting HCC prognosis.Based on the alterations of circRNA regulatory networks, the analysis of gene modules related to HCC is feasible. Methods:Multiple expression datasets and RNA element targeting prediction tools were used to construct a circRNA-microRNA-mRNA network in HCC.Gene function, pathway, and protein interaction analyses were performed for the differentially expressed genes(DEGs)in this regulatory network. In the proteinprotein interaction network, hub genes were identified and subjected to regression analysis, producing an optimized four-gene signature for prognostic risk stratification in HCC patients.Anti-HCC drugs were excavated by assessing the DEGs between the low-and high-risk groups.A circRNA-microRNA-hub gene subnetwork was constructed,in which three hallmark genes,KIF4A,CCNA2,and PBK,were subjected to functional enrichment analysis. Results:A four-gene signature(KIF4A,CCNA2,PBK,and ZWINT)that effectively estimated the overall survival and aided in prognostic risk assessment in the The Cancer Genome Atlas(TCGA)cohort and International Cancer Genome Consortium(ICGC)cohort was developed.CDK inhibitors,PI3K inhibitors,HDAC inhibitors, and EGFR inhibitors were predicted as four potential mechanisms of drug action(MOA)in high-risk HCC patients.Subsequent analysis has revealed that PBK,CCNA2,and KIF4A play a crucial role in regulating the tumor microenvironment by promoting immune cell invasion, regulating microsatellite instability(MSI),and exerting an impact on HCC progression. Conclusions:The present study highlights the role of the circRNA-related regulatory network, identifies a four-gene prognostic signature and biomarkers, and further identifies novel therapy for HCC.

文摘With the changing lifestyle and the acceleration of aging in the Chinese population, the incidence and mortality of colorectal cancer(CRC)have risen in the last decades. On the contrary, the incidence and mortality of CRC have continued to decline in the USA since the 1980s,which is mainly attributed to early screening and standardized diagnosis and treatment.Rectal cancer accounts for the largest proportion of CRC in China, and its treatment regimens are complex. At present, surgical treatment is still the most important treatment for rectal cancer.Since the first Chinese guideline for diagnosis and treatment of CRC was issued in 2010,the fourth version has been revised in 2020.These guidelines have greatly promoted the standardization and internationalization of CRC diagnosis and treatment in China. And with the development of comprehensive treatment methods such as neoadjuvant chemoradiotherapy, targeted therapy, and immunotherapy, the post-operative quality of life and prognosis of patients with rectal cancer have improved. We believe that the inflection point of the rising incidence and mortality of rectal cancer will appear in the near future in China. This article reviewed the current status and research progress on surgical therapy of rectal cancer in China.

文摘There have been several investigations into the genus Heterobasidion, however, differentiation of species in these studies have depended on the gene regions analyzed.Reliable defining of species, establishing species divergence times and establishing species biogeographical distributions have been challenging. Here,we used a multilocus phylogenetic approach and maximum parsimony, maximum likehood, and Bayesian analyses to infer the phylogenetic relationships of Heterobasidion species. In addition,we focused on a fungus fossil-based approach and used the largest subunit of RNA polymerase II-the second subunit of RNA polymerase II(RPB1-RPB2)dataset to perform maximum likelihoodbased estimation and Bayesian binary analyses,to assess the divergence and biogeographical distributions of Heterobasidion species. The Heterobasidion annousum/H.insulare species complex clusters in three groups in the phylogenetic analyses.Molecular dating suggests that ancestral Heterobasidion species originated in Eurasia during the Early Miocene, followed by dispersal and speciation to other continents during the Middle Miocene and Early Pliocene. Our data are compatible with the previous viewpoint that H.irregulare and H.occidentale colonized North America via different routes, which has been interpreted as Beringian and Thulean North Atlantic vicariance. In addition,we propose that the occurrence of H.araucariae in the southern Hemisphere was probably due to recent human-mediated introductions.Plate tectonics and long-distance dispersal are the most likely factors that influenced Heterobasidion speciation and biogeography.

文摘Single-hole hollow polymer nanospheres were fabricated by raspberry-like template method using "graft-from" strategy through atom transfer radical polymerization (ATRP). In this strategy, most of ATRP monomers may be used and thus many functional groups can be easily incorporated into the single-hole hollow crosslinked polymer nanospheres.

文摘To the Editor:Adrenal and gonadal function can be evaluated by analyzing steroid hormone levels in different age groups, especially adolescent girls.Liquid chromatography-tandem mass spectrometry(LC-MS/MS or highperformance LC[HPLC]-MS/MS)offers a combination of the physical separation capabilities of HPLC and the mass analysis capabilities of tandem MS,which improves sensitivity and specificity in measuring steroid hormone levelsJ1,21 This method is more sensitive, effective, specific, and homogenous than other methods for analyzing children’s steroid hormone levels in the clinic.Studies on the reference intervals of steroid sex hormones have more commonly performed with Caucasian subjects than with Chinese subjects.^31 LC-MS/MS-based reference intervals for steroid hormones in China, especially for healthy girls, have not yet been reported. With LC-MS/MS becoming increasingly common, the establishment of reference intervals is growing more urgent. Here,we define appropriate reference ranges for pregnenolone,17ahydroxyprogesterone,corticosterone,dehydroepiandrosterone,androstenedione,and free testosterone.
