[Objectives]To establish a TLC and content determination method of Pileostegia tomentellal,with umbelliferone as the indicator component.[Methods]TLC identification was performed by silica gel G thin layer plate with n-hexane-ethyl acetate(4:3)as the developing agent,and the plate was examined by UV lamp(365 nm).The umbelliferone content was determined by HPLC:Inertsil ODS-3 C 18 column(4.60 mm×250 mm,5μm);mobile phase acetonitrile-0.2%phosphoric acid gradient elution;detection wavelength 320 nm,flow rate 1.0 mL/min,column temperature 30℃,injection volume 10μL.[Results]The chromatogram of P.tomentellal showed the same color spot in the same position as that of reference medicinal material,and the spot was clear with good specificity.Umbelliferone showed a good linear relationship when the injection volume was 2.63-131.27μg/mL(R^(2)=0.9997).The average recovery of umbelliferone in the low,middle and high adding groups of P.tomentellal was 99.57%and the RSD was 2.15%.[Conclusions]The method can effectively identify Yao medicine P.tomentellal and accurately determine the content of umbelliferone in medicinal materials,which will provide a scientific basis for the development and utilization of medicinal resources of Yao medicine P.tomentellal. 展开更多

[Objectives]To establish the quality standard of hospital preparation Jiedu Shengxue granules.[Methods]Scleromitrion diffusum and Prunella vulgaris in Jiedu Shengxue granules were qualitatively identified by thin layer chromatography(TLC).A high performance liquid chromatography(HPLC)was established to determine the content of notoginsenoside R1 in the granule.[Results]The traditional Chinese medicinal materials in Jiedu Shengxue granules could be identified by TLC,and the characteristic spots were stable and clear.Notoginsenoside R1 had a good linear relationship in the range of 10.45-104.5μg/mL,with an average recovery of 98.52%and RSD=2.36%.[Conclusions]TLC and HPLC,as the quality control methods of Jiedu Shengxue granules,have high accuracy and good repeatability,which lays a foundation for the quality control of this mixture. 展开更多

[Objectives]To establish the TLC identification methods of Llicis Rotundae Cortex and Mahoniae Caulis and the determination method of syringin content in Zhuang Medicine Xiaoyan Zhiyang Lotion.[Methods]Thin layer chromatography(TLC)was used to qualitatively identify Llicis Rotundae Cortex and Mahoniae Caulis in Zhuang Medicine Xiaoyan Zhiyang Lotion.The quantitative analysis of syringin in Zhuang Medicine Xiaoyan Zhiyang Lotion was carried out by high-performance liquid chromatography(HPLC).Chromatographic conditions are as follows:Luna 5μm C_(18)((2)100 A(250 mm×4.6 mm,5μm)column was adopted;acetonitrile(10%)-water(90%)was as the mobile phase,and the flow rate was 1.0 mL/min;the detection wavelength was 265 nm;the sample size was 5μL;the column temperature was 30℃.[Results]Syringin had a good linear relationship with its peak area in the range of 0.190-2.856μg(R^(2)=0.9995).The RSD values of precision,stability and repeatability experiments were all lower than 2.5%,and the average sampling recovery rate(n=6)was 100.37%(RSD=-0.88%).[Conclusions]The method had the advantages of simple operation,strong specificity,good repeatability and stability,and can be used for the quality control of Zhuang Medicine Xiaoyan Zhiyang Lotion. 展开更多

[Objectives]To establish the extraction process and quality standard method of Zhuang medicine fumigation lotion.[Methods]The orthogonal design method was employed to optimize the water extraction process with the amount of water added,decocting time and extraction times as factors,and syringin content and dry extract yield as indexes.The content of syringin was determined by high performance liquid chromatography.[Results]The best water extraction process was:soaking in water for 1 h,decocting twice,added 10 times the amount of water each time,decocting for 1 h.The average content of syringin in 3 batches was 0.98 mg/g,and the average dry extract yield was 26.07%.[Conclusions]The project adopts water extraction method to prepare Zhuang medicine fumigation lotion,which has the characteristics of high efficiency and suitable for large-scale production.The quality control method is reliable,rapid and accurate,and can effectively control the quality of the lotion. 展开更多

[Objectives]To observe the clinical efficacy and safety of Zhuang Medicine Xiaoyan Zhiyang Formula by retrospective analysis of clinical use.[Methods]A total of 225 cases were collected,including 117 males and 108 females,115 cases of contact dermatitis and 110 cases of acute eczema.Zhuang Medicine Xiaoyan Zhiyang Formula was administered to 225 patients with contact dermatitis and acute eczema and the clinical efficacy was evaluated.[Results]After one week of treatment,among 225 patients with contact dermatitis and acute eczema,76 cases were cured(33.78%),133 cases showed marked response(59.11%),16 cases(7.11%)were improved,0 case was ineffective,and the overall response rate was 92.89%.After one week of Zhuang Medicine Xiaoyan Zhiyang Formula treatment,the symptoms of itching,pain and swelling were significantly improved in all patients,and the rash was partially crusted without new symptom,and there were no complications and adverse reactions.[Conclusions]Zhuang Medicine Xiaoyan Zhiyang Formula therapy has a remarkable effect in the treatment of contact dermatitis and acute eczema. 展开更多

Anti-tumor Effect of Paclitaxel Enhanced by Psoralen at the Cellular Level

[Objectives]To explore the effect of psoralen combined with paclitaxel on the apoptosis of MCF-7 cells.[Methods]The effects of different concentrations of psoralen,paclitaxel,or the combination of psoralen and paclitaxel on cell viability were detected using CCK-8 assay kit.Cell cycle distribution and apoptosis after 24 h of psoralen(0.16,0.32,0.64 mmol/L),paclitaxel(0.1μmol/L),combined action of psoralen(0.32 mmol/L)and paclitaxel(0.1μmol/L)were detected using flow cytometry.[Results]Lower concentration of psoralen(0.04-0.32 mmol/L)showed no significant inhibitory effect on cells.After combined with paclitaxel,the inhibitory effect on MCF-7 cell proliferation was significantly higher than that of the group treated alone.Compared with the paclitaxel group,the cell apoptosis rate in the drug combination group was significantly increased.Different low concentrations of psoralen can block the cell cycle of MCF-7 at G 0/G 1 phase,while paclitaxel can block the cell cycle at G 2/M phase.After combined action,the number of cells blocked at G 2/M phase decreased.[Conclusions]Overall,the combined effect of psoralen and paclitaxel can enhance anti-tumor ability by inhibiting cell proliferation,inducing apoptosis,and blocking cell cycle. 展开更多

[Objectives]This study aimed to observe the effects of different processed products of Radix Codonopsis on intestinal flora in rats with spleen deficiency.[Methods]Rat models with spleen deficiency were established by bitter-cold purgation method with Radix et Rhizoma Rhei.Normal group,model group,Radix Codonopsis group,fried Radix Codonopsis group,rice-fried Radix Codonopsis group,honey-fried Radix Codonopsis group and bran-fired Radix Codonopsis group were designed.After subjecting to corresponding treatments,the changes in the quantity of intestinal microorganisms of the rats were detected.[Results]There was no significant change in the quantity of intestinal microorganisms of the rats in the normal group and model group.The rats in the rice-fried Radix Codonopsis group were administered after successful modeling,and the abundance of Bifidobacterium and Lactobacillus in the intestines of the rats increased,and the quantity of Escherichia coli and Staphylococci reduced.The rats in the Radix Codonopsis group,fried Radix Codonopsis groups,honey-fried Radix Codonopsis group and bran-fried Radix Codonopsis group were administered after successful modeling,and the abundance of Bifidobacterium and Lactobacillus in the intestines of the rats increased(the increases were smaller than those in the rice-fried Radix Codonopsis group),and the abundance of E.coli and Staphylococci reduced,close to normal levels.[Conclusions]Different processed products of Radix Codonopsis have obvious regulation effect on intestinal flora of rats with spleen deficiency,and the regulation effect of rice-fried Radix Codonopsis on rats with spleen deficiency is better than that of Radix Codonopsis,fried Radix Codonopsis,honey-fried Radix Codonopsis and bran-fried Radix Codonopsis. 展开更多

[Objectives]To observe the effect of drug-containing serum from different extracts of Phyllanthus reticulatus leaf on the proliferation of human gastric cancer SGC-7901 cells and the effect of in vivo on the life extension rate of mice with H22 ascites tumor,and to investigate the effects of chemical components such as Corilagin,ethyl gallate,ethyl brevifolincarboxylate,and gallic acid on the proliferation of human gastric cancer SGC-7901 cells.[Methods]MTT method was used to observe the inhibitory effect of drug-containing serum of different extracts of P.reticulatus leaf on human gastric cancer SGC-7901 cells in vitro to determine its active site.The active site was used as the research object to establish a Kunming mouse ascites tumor model,to investigate the effect on the life extension rate of mice with H22 ascites tumor,to further separate the monomer components from the effective fraction and investigate its effect on human gastric cancer SGC-7901 cells.[Results]Compared with the 10%blank serum control group,10%drug-containing serum of ethyl acetate and n-butanol in P.reticulatus leaf had significant inhibitory effect on the proliferation of human gastric cancer SGC-7901 cells,and the inhibition rate was 34.99%and 28.68%,respectively(P<0.05).Compared with the blank group,the survival time of the high dose group of ethyl acetate in P.reticulatus leaf was significant(P<0.05).Gallic acid,Corilagin,and Brevifolincarboxylic acid ethyl ester had a significant inhibitory effect on the proliferation of human gastric cancer SGC-7901 cells,with IC 50 of 26.52,70.45,and 158.86μg/mL,respectively.Ethyl gallate had no inhibitory effect on the proliferation of human gastric cancer SGC-7901 cells,and its IC 50 was 251.96μg/mL.[Conclusions]The drug-containing serum of ethyl acetate and n-butanol extract of P.reticulatus can inhibit the proliferation of human gastric cancer SGC-7901 cells.Ethyl acetate of P.reticulatus leaves can increase the life extension rate of mice with H22 ascites tumor.Gallic acid,Corilagin,and Brevifolincarboxylic acid ethyl ester isolated from its active site are the material basis for inhibiting the proliferation of human gastric cancer SGC-7901 cells. 展开更多

[Objectives] To study the differences in the effects of Rheum officinale and wine prepared Rheum palmatum Dachengqi Decoction(DCQ) on laxative effect of mice.[Methods]First,72 mice were randomly divided into positive control group,blank control group,6 and10 g/kg dose R.officinale DCQ groups,6 and 10 g/kg dose wine prepared R.palmatum DCQ groups.After administration at the dose of20 m L/kg,metabolic cage method and carbon powder propulsion were used to conduct Na+-K+-ATPasc activity experiment,and the comparison was made for the differences in the normal laxative effect of R.officinale and wine prepared R.palmatum on DCQ,the carbon powder propulsion rate,and Na+-K+-ATPasc activit.[Results]There are differences in the laxative effect of R.officinale and wine prepared R.palmatum on DCQ,but their differences in changes are,to some extent,connected with R.officinale and wine prepared R.palmatum.From comparison of R.officinale DCQ group,wine prepared R.palmatum DCQ group,blank control group,and positive control group,it is found that there are differences in the normal laxative effect of normal mice,carbon powder propulsion,and inhibition of Na+-K+-ATPase activity.In addition,the comparison of drug administration groups indicates that the differences in the effects of R.officinale DCQ group are most significant,and there are significant differences in the 6 and 10 g/kg groups.[Conclusions]There are certain differences in the influence of R.officinale and wine prepared R.palmatum Dachengqi Decoction on normal laxative effect of mice,which may be connected with the fact that changes in main chemical components of king drugs R.officinale and wine prepared R.palmatum lead to changes in the dissolution amount of anthraquinones in the compound Dachengqi Decoction. 展开更多

L. alata in different production areas of Guangxi contained the caffeic acid,and there was a great difference in the caffeic acid. L. alata in Baise had the highest content of caffeic acid,while that in Guilin had the lowest content of caffeic acid. [Conclusions]This method can accurately determine the content of caffeic acid and is expected provide a scientific basis for the development and utilization of herbal medicine L. alata. 展开更多

[Objectives]To optimize the extraction process of total flavonoids of Zhuang medicine Clerodendrum japonicum(Thumb.)Sweet and establish a method for its extraction and content determination.[Methods]The total flavonoids of C.japonicum were extracted by reflux extraction method.Through a single factor experiment,the effects of extraction method,extraction solvent concentration,extraction volume and extraction time on the total flavonoids content of medicinal materials were investigated to select the optimal extraction process of the total flavonoids of C.japonicum.[Results]The optimal extraction process of the total flavonoids of C.japonicum was 75%ethanol,1∶20 solid-to-liquid ratio,and 50 min reflux extraction time.[Conclusions]This method can effectively determine the content of total flavonoids in C.japonicum and provide a certain scientific basis for the study of the quality standard of this Zhuang medicine.This method has high reproducibility.It is stable and feasible in extraction of total flavonoids from Zhuang medicine C.japonicum. 展开更多

[Objectives]To determine the content of five kinds of free anthraquinones in raw rhubarb and stewed rhubarb by HPLC,and compare their content difference,so as to reveal the change rule of material basis before and after processing of traditional Chinese medicine.[Methods]The content was determined by HPLC with Thermo BDS HYPERSII C18 column(4.6 mm×250 mm,5μm)as chromatographic column and methanol-0.1%phosphoric acid(85∶15)aqueous solution as mobile phase;the detection wavelength was 256 nm,the flow rate was 1.0 mL/min,the column temperature was 30℃,and the injection volume was 10μL.[Results]Aloe-emodin,rhein,emodin,chrysophanol and emodin methyl ether showed a good linear relationship in the linear range of 0.0207-0.414(R2=0.9999),0.1721-3.442(R2=0.9996),0.0203-0.406(R2=0.9999),0.148-2.96(R2=0.9998)and 0.164-3.28(R2=0.9993),respectively;the content of aloe-emodin decreased by about 8.26%,and rhein decreased by about 9.21%;the content of emodin increased by about 16.44%,chrysophanol increased by about 16.12%,and emodin methyl ether increased by about 20.01%,indicating that the processing technology had different effects on the components of rhubarb.[Conclusions]The content of five free anthraquinones in rhubarb was affected to different degrees after processing,and this method could be used as a quality control method for rhubarb and its different processed products. 展开更多

[Objectives]To study the anti-inflammatory effects of water extract and ethanol extract from Excoecaria cochinchinensis Lour.[Methods]The acute and chronic inflammation models of mouse paw edema induced by cotton ball granuloma and carrageenan were established respectively,and the anti-inflammatory effects of water and ethanol extract from E.cochinchinensis Lour.on acute and chronic inflammation in mice were evaluated.The experimental mice were equally divided into three groups of high,medium and low dose of E.cochinchinensis Lour.(60,40 and 20 g/kg),dexamethasone(0.006 g/kg)group and blank group(distilled water).In the cotton ball granuloma test in mice,the mice were given intragastric administration continuously for 7 d after cotton ball implantation.In the experiment of inflammation induced by carrageenan,the mice in each group were given intragastric administration once a day for 7 d,and the inflammatory model was made after the last administration.[Results]The granuloma inhibition rates of mice in the high,medium and low dose groups of water extract and ethanol extract from E.cochinchinensis Lour were significantly lower than those in the blank group(P<0.05).The degree of paw edema in each dose group of water and ethanol extract from E.cochinchinensis Lour.was significantly lower than that in the blank group(P<0.05).[Conclusions]The water and ethanol extracts from E.cochinchinensis Lour.had significant anti-inflammatory effects. 展开更多

Effects of Different Processed Products of Ilex hainanensis Merr on Rats with Hyperlipidemia

[Objectives] This study aimed to investigate the hypolipidemic effects of different processed products of Ilex hainanensis Merr to provide experimental basis for the research on the hypolipidemic mechanism of I. hainanensis Merr. [Methods] SD rats were fed high-fat diet to establish hyperlipidemia rat models. The hyperlipidemia rats were administered with different processed products of I. hainanensis Merr by gavage to investigate the preventive and therapeutic effects of the medicinal material. [Results] The levels of total cholesterol(TC), triglyceride(TG), low density lipoprotein cholesterol(LDL-C) in the rats fed high-fat diet were significantly higher than those in of the control group(P<0.01), indicating successful modeling. Among different processing methods, the hypolipidemic effects were in the order as baking=traditional processing=frying=microwaving>drying in shade. Among different baking times, the hypolipidemic effects ranked as 180 ℃>200 ℃=220 ℃. Among different baking times, the hypolipidemic effects ranked as 20 min=40 min>30 min. The hypolipidemic effects of processed product of I. hainanensis Merr with different amounts of active ingredients were different. [Conclusions] The processed products of I. hainanensis Merr all had hypolipidemic effects, but the effects were different. This study will provide a scientific basis for expanding the resources of I. hainanensis Merr. 展开更多
