Daidzein has been widely used in pharmaceuticals,nutraceuticals,cosmetics,feed additives,etc.Its preparation process and related reaction mechanism need to be further investigated.A cost-effective process for synthesi...Daidzein has been widely used in pharmaceuticals,nutraceuticals,cosmetics,feed additives,etc.Its preparation process and related reaction mechanism need to be further investigated.A cost-effective process for synthesizing daidzein was developed in this work.In this article,a two-step synthesis of daidzein(Friedel–Crafts acylation and[5+1]cyclization)was developed via the employment of trifluoromethanesulfonic acid(TfOH)as an effective promoting reagent.The effect of reaction conditions such as solvent,the amount of TfOH,reaction temperature,and reactant ratio on the conversion rate and the yield of the reaction,respectively,was systematically investigated,and daidzein was obtained in 74.0%isolated yield under optimal conditions.Due to the facilitating effect of TfOH,the Friedel–Crafts acylation was completed within 10 min at 90℃ and the[5+1]cyclization was completed within 180 min at 25℃.In addition,a possible reaction mechanism for this process was proposed.The results of the study may provide useful guidance for industrial production of daidzein on a large scale.展开更多
Herein,the co-pyrolysis reaction characteristics of corn straw(CS)and bituminous coal in the presence of ilmenite oxygen carriers(OCs)are investigated via thermogravimetry coupled with mass spectrometry.The results re...Herein,the co-pyrolysis reaction characteristics of corn straw(CS)and bituminous coal in the presence of ilmenite oxygen carriers(OCs)are investigated via thermogravimetry coupled with mass spectrometry.The results reveal that the participation of OCs weakens the devolatilization intensity of co-pyrolysis.When the CS blending ratio is<50%,the mixed fuel exhibits positive synergistic effects.The fitting results according to the Coats-Redfern integral method show that the solid-solid interaction between OCs and coke changes the reaction kinetics,enhancing the co-pyrolysis reactivity at the high-temperature zone(750-950C).The synergistic effect is most prominent at a 30%CS blending ratio,with copyrolysis activation energy in the range of 26.35-40.57 kJ·mol^(-1).展开更多

This study investigated the feasibility of hyperspectral imaging techniques to estimate the vigor of heatdamaged Quercus variabilis seeds.Four thermal damage grades were classified according to heat treatment duration...This study investigated the feasibility of hyperspectral imaging techniques to estimate the vigor of heatdamaged Quercus variabilis seeds.Four thermal damage grades were classified according to heat treatment duration(0,2,5,and 10 h).After obtaining hyperspectral images with a 370–1042 nm hyperspectral imager that included visible and near infrared light,germination was tested to confirm estimates.The Savitzky–Golay(SG)second derivative was used to preprocess the spectrum to reduce any noise impact.The successive projections algorithm(SPA),principal component analysis,and local linear embedding algorithm were used to extract the characteristic spectral bands related to seed vigor.Finally,a model for seed vigor classifi-cation of Q.variabili s based on partial least squares support vector machine(LS-SVM)with different spectral data sets was developed.The results show that the spectrum after SG second derivative preprocessing was better for developing the model,and SPA performed the best among the three feature band selection methods.The combination SG second derivative-LS-SVM provided the best classification model for Q.variabilis seed vigor,with the prediction set reaching 98.81%.This study provides an important basis for rapid and nondestructive assessment of the vigor of heat-damaged seeds using hyperspectral imaging techniques.展开更多
The images of three diseases of black spot,anthracnose and leaf blight in Sichuan Ophiopogon japonicus leaves were taken the research object,and the bimodal method,Otsu threshold segmentation method and K-means cluste...The images of three diseases of black spot,anthracnose and leaf blight in Sichuan Ophiopogon japonicus leaves were taken the research object,and the bimodal method,Otsu threshold segmentation method and K-means clustering segmentation algorithm were compared and analyzed on the images of O.japonicus.The segmentation effect showed that the K-means clustering algorithm combined with the mathematical morphology processing method could meet the segmentation requirements.Then,the color,shape,and texture information of the lesion image were extracted and fused into a feature vector.Next,the variance analysis and principal component analysis method were applied to eliminate the feature parameters with poor disease characterization ability and reduce the eigenvector dimension to 10 dimensions.Finally,the classifiers for disease identification were designed by the support vector machine,and the recognition rate reached 90%after testing.The method has the advantages of low cost,simple algorithm and high efficiency,and basically meets the requirements of practical applications.展开更多

The purpose of this study is to clarify the significance of postoperative radiotherapy for N2 lung cancer.This study aimed to investigate the effect of postoperative radiotherapy on the survival and prognosis of patie...The purpose of this study is to clarify the significance of postoperative radiotherapy for N2 lung cancer.This study aimed to investigate the effect of postoperative radiotherapy on the survival and prognosis of patients with N2 lung cancer.Data from 12,000 patients with N2 lung cancer were extracted from the Surveillance,Epidemiology,and End Results database(2004-2012).Age at disease onset and 5-year survival rates were calculated.Survival curves were plotted using the Kaplan-Meier method.The univariate log-rank test was performed.Multivariate Cox regression were used to examine factors affecting survival.Patients’median age was 67 years(mean 66.46±10.03).The 5-year survival rate was 12.55%.Univariate analysis revealed age,sex,pathology,and treatment regimen as factors affecting prognosis.In multivariate analysis,when compared to postoperative chemotherapy,postoperative chemoradiotherapy was better associated with survival benefits(hazard ratio[HR]=0.85,95%confidence interval[CI]:0.813-0.898,P<0.001).Propensity score matching revealed that patients who had received postoperative chemoradiotherapy had a better prognosis than did patients who had received postoperative chemotherapy(HR=0.869,95%CI:0.817-0.925,P<0.001).Female patients and patients aged<65 years had a better prognosis than did their counterparts.Patients with adenocarcinoma had a better prognosis than did patients with squamous cell carcinoma.Moreover,prognosis worsened with increasing disease T stage.Patients who had received postoperative chemoradiotherapy had a better prognosis than did patients who had received postoperative chemotherapy.Postoperative radiotherapy was an independent prognostic factor in this patient group.展开更多

In the first phase, the fast global K-means clustering algorithm is used to obtain the cluster number and the prototypes. Then, the prototypes of all these clusters and representatives of points belonging to these clusters are regarded as the input data set of the second phase. Afterwards, all the prototypes are clustered according to a DED measure which makes data points locating in the same structure to possess high similarity with each other. In experimental studies, the authors test the proposed algorithm on seven artificial as well as seven UCI data sets. The results demonstrate that the proposed algorithm is flexible to different data distributions and has a stronger ability in clustering data sets with complex non-convex distribution when compared with the comparison algorithms.展开更多

The aim of this study was to investigate the role of HIF-1α in the regu- lation of vascular endothelial growth factor C (VEGF-C) expression and its effect on lymphangiogenesis and an- giogenesis in breast cancer. Lymphatic vessel density (LVD), microvessel density (MVD) and the expressions of HIF-1α and VEGF-C proteins were evaluated by immunohistochemistry in 75 breast cancer samples. There was a significant correlation between HIF-1α and VEGF-C (P = 0.014, r = 0.273, Spearman's coefficient of correlation). HIF-1α and VEGF-C overexpression was significantly correlated with higher LVD (P = 0.003 and P = 0.017, re- spectively), regional lymph nodal involvement (P = 0.002 and P = 0.004, respectively) and advanced tumor, node, metastasis (TNM) classification (P = 0.001 and P = 0.01, respectively). Higher MVD was observed in the group expressing higher levels of HIF-1α and VEGF-C (P = 0.033 and P = 0.037, respectively). Univariate analysis showed shorter survival time in patients expressing higher levels of HIF-1α and VEGF-C. HIF-1α was also found to be an independent prognostic factor of overall survival in multivariate analysis. The results suggest that HIF-1α may affect VEGF-C expression, thus acting as a crucial regulator of lymphangiogenesis and angiogenesis in breast cancer. This study highlights promising potential of HIF- 1α as a therapeutic target against tumor lymph node me- tastasis.展开更多

Eight stabilizers or their b...Azilsartan(AZL), a poorly soluble drug, was considered to be fit for nanocrystals to improve its solubility. Our study intended to prepare AZL nanocrystals by means of bead milling method. Eight stabilizers or their binary combination and the milling time were set to be variable factors to optimize AZL nanosuspension formulation, and six types of freezedrying supports were investigated to reduce the aggregation of particles during the solidification. AZL nanocrystals with or without sodium deoxycholate(NaDC) as combined stabilizer with Poloxamer 188(F68) were prepared owning mean particle sizes of about 300 nm and 460 nm. During the screening processes, the formulation containing NaDC showed a smaller particle size and better stability during lyophilization. The irregular shape and crystal form changing in AZL nanocrystals were discovered by various characterizations. And with physical mixture as reference, nanocrystals showed its improvement about in-vitro dissolution and in-vivo bioavailability. In conclusion, the nanocrystals of AZL could be prepared well in our study. Additionally, our results suggested that NaDC was an appreciated excipient on the nanocrystals platform, which can exhibit the abilities of size-reduction and stabilitymaintaining on freeze-drying.展开更多
In this work,a model of hydrogen production by double chemical looping is introduced.The efficiency benefit obtained was investigated.The chemical looping hydrogen generation unit is connected in series to the downstr...In this work,a model of hydrogen production by double chemical looping is introduced.The efficiency benefit obtained was investigated.The chemical looping hydrogen generation unit is connected in series to the downstream of a chemical looping gasification unit as an additional system for 100 MWh coal gasification,with the function of supplementary combustion to produce hydrogen.Using Aspen Plus software for process simulation,the production of H_(2) and N_(2) in the series system is higher than that in the independent Chemical looping gasification and Chemical looping hydrogen generation systems,and the production of hydrogen is approximately 25.63%and 12.90%higher,respectively;The study found that when the gasification temperature is 900C,steam-carbon ratio is 0.84 and oxygen-carbon ratio is 1.5,the hydrogen production rate of the system was the maximum.At the same time,through heat exchange between logistics,high-pressure steam at 8.010×10^(4) kg·h^(-1) and medium-pressure steam at 1.101×10^(4) kg·h^(-1) are generated,and utility consumption is reduced by 61.58%,with utility costs decreasing by 48.69%.An economic estimation study found that the production cost of ammonia is 108.66 USD(t NH_(3))^(-1).Finally,cost of equipment is the main factors influencing ammonia production cost were proposed by sensitivity analysis.展开更多

Epithelial ovarian cancer cell line SKOV-3 was treated with me- droxyprogestogen, phosphatidylinositol 3-kinase inhibitor LY294002 and vehicle control. Akt, phospho-Akt, Bcl-2 and phospho-Bad proteins were examined by immunoblotting assays. Medroxyprogestogen-induced apoptosis was assessed by MTT assays and Annexin V apoptosis assay. We found no significant difference in Akt and Bad expression in both the medroxyprogestogen groups and the control group. The levels of phospho-Akt, Bcl-2 and phospho-Bad were decreased in all the medroxyprogestogen groups and significantly decreased in the high dose mitogen-activated protein (MAP) group (10 gmol/L). Viability of SKOV-3 was reduced and apparent apoptosis of SKOV-3 cells was observed with increased doses of MAP. The findings suggest that medroxyprogestogen can induce SKOV-3 cell apoptosis by inhibiting Akt phosphorylation.展开更多

Melanoma is the most lethal type of skin cancer,originating from the malignant transformation of melanocyte.While the development of targeted therapy and immunotherapy has gained revolutionary advances in potentiating...Melanoma is the most lethal type of skin cancer,originating from the malignant transformation of melanocyte.While the development of targeted therapy and immunotherapy has gained revolutionary advances in potentiating the therapeutic effect,the prognosis of patients with melanoma is still suboptimal.During tumor progression,melanoma frequently encounters stress from both endogenous and exogenous sources in tumor microenvironment.SIRT7 is a nuclear-localized deacetylase of which the activity is highly dependent on intracellular nicotinamide adenine dinucleotide(NAD+),with versatile biological functions in maintaining cell homeostasis.Nevertheless,whether SIRT7 regulates tumor cell biology and tumor immunology in melanoma under stressful tumor microenvironment remains elusive.Herein,we reported that SIRT7 orchestrates melanoma progression by simultaneously promoting tumor cell survival and immune evasion via the activation of unfolded protein response.We first identified that SIRT7 expression was the most significantly increased one in sirtuins family upon stress.Then,we proved that the deficiency of SIRT7 potentiated tumor cell death under stress in vitro and suppressed melanoma growth in vivo.Mechanistically,SIRT7 selectively activated the IRE1α-XBP1 axis to potentiate the pro-survival ERK signal pathway and the secretion of tumorpromoting cytokines.SIRT7 directly de-acetylated SMAD4 to antagonize the TGF-β-SMAD4 signal,which relieved the transcriptional repression on IRE1αand induced the activation of the IRE1α-XBP1 axis.Moreover,SIRT7 up-regulation eradicated anti-tumor immunity by promoting PD-L1 expression via the IRE1α-XBP1 axis.Additionally,the synergized therapeutic effect of SIRT7 suppression and anti-PD-1 immune checkpoint blockade was also investigated.Taken together,SIRT7 can be employed as a promising target to restrain tumor growth and increase the effect of melanoma immunotherapy.展开更多
Background:Hepatocellular carcinoma(HCC)is one of the most common malignant tumors in the world.The ubiquitin-specific peptidase 25(USP25)protein has been reported to participate in the development of several cancers....Background:Hepatocellular carcinoma(HCC)is one of the most common malignant tumors in the world.The ubiquitin-specific peptidase 25(USP25)protein has been reported to participate in the development of several cancers.However,few studies have reported its association with HCC.In this study,we aimed to investigate the function and mechanism of USP25 in the progression of HCC.Methods:We analyzed USP25 protein expression in HCC based on The Cancer Genome Atlas(TCGA)and International Cancer Genome Consortium(ICGC)database cohorts.Then,we constructed USP25-overexpressing and USP25-knockdown HepG2,MHCC97H,and L-O2 cells.We detected the biological function of USP25 by performing a series of assays,such as Cell Counting Kit-8(CCK-8),colony formation,transwell,and wound healing assays.Western blotting and quantitative real-time polymerase chain reaction(qRT-PCR)analyses were performed to detect the interaction between USP25 and the Wnt/β-catenin signaling pathway.The relationship between USP25 and tripartite motif-containing 21(TRIM21)was assessed through mass spectrometry and co-immunoprecipitation(Co-IP)analysis.Finally,we constructed a mouse liver cancer model with the USP25 gene deletion to verify in vivo role of USP25.Results:USP25 was highly expressed in HCC tissue and HCC cell lines.Importantly,high expression of USP25 in tissues was closely related to a poor prognosis.USP25 knockdown markedly reduced the proliferation,migration,and invasion of HepG2 and MHCC97H cells,whereas USP25 overexpression led to the opposite effects.In addition,we demonstrated that USP25 interacts with TRIM21 to regulate the expression of proteins related to epithelial-mesenchymal transition(EMT;E-cadherin,N-cadherin,and Snail)and the Wnt/β-catenin pathway(β-catenin,Adenomatous polyposis coli,Axin2 and Glycogen synthase kinase 3 beta)and those of their downstream proteins(C-myc and Cyclin D1).Finally,we verified that knocking out USP25 inhibited tumor growth and distant metastasis in vivo.Conclusions:In summary,our data showed that USP25 was overexpressed in HCC.USP25 promoted the proliferation,migration,invasion,and EMT of HCC cells by interacting with TRIM21 to activate theβ-catenin signaling pathway.展开更多

文摘This study investigated the feasibility of hyperspectral imaging techniques to estimate the vigor of heatdamaged Quercus variabilis seeds.Four thermal damage grades were classified according to heat treatment duration(0,2,5,and 10 h).After obtaining hyperspectral images with a 370–1042 nm hyperspectral imager that included visible and near infrared light,germination was tested to confirm estimates.The Savitzky–Golay(SG)second derivative was used to preprocess the spectrum to reduce any noise impact.The successive projections algorithm(SPA),principal component analysis,and local linear embedding algorithm were used to extract the characteristic spectral bands related to seed vigor.Finally,a model for seed vigor classifi-cation of Q.variabili s based on partial least squares support vector machine(LS-SVM)with different spectral data sets was developed.The results show that the spectrum after SG second derivative preprocessing was better for developing the model,and SPA performed the best among the three feature band selection methods.The combination SG second derivative-LS-SVM provided the best classification model for Q.variabilis seed vigor,with the prediction set reaching 98.81%.This study provides an important basis for rapid and nondestructive assessment of the vigor of heat-damaged seeds using hyperspectral imaging techniques.

文摘The images of three diseases of black spot,anthracnose and leaf blight in Sichuan Ophiopogon japonicus leaves were taken the research object,and the bimodal method,Otsu threshold segmentation method and K-means clustering segmentation algorithm were compared and analyzed on the images of O.japonicus.The segmentation effect showed that the K-means clustering algorithm combined with the mathematical morphology processing method could meet the segmentation requirements.Then,the color,shape,and texture information of the lesion image were extracted and fused into a feature vector.Next,the variance analysis and principal component analysis method were applied to eliminate the feature parameters with poor disease characterization ability and reduce the eigenvector dimension to 10 dimensions.Finally,the classifiers for disease identification were designed by the support vector machine,and the recognition rate reached 90%after testing.The method has the advantages of low cost,simple algorithm and high efficiency,and basically meets the requirements of practical applications.

文摘The purpose of this study is to clarify the significance of postoperative radiotherapy for N2 lung cancer.This study aimed to investigate the effect of postoperative radiotherapy on the survival and prognosis of patients with N2 lung cancer.Data from 12,000 patients with N2 lung cancer were extracted from the Surveillance,Epidemiology,and End Results database(2004-2012).Age at disease onset and 5-year survival rates were calculated.Survival curves were plotted using the Kaplan-Meier method.The univariate log-rank test was performed.Multivariate Cox regression were used to examine factors affecting survival.Patients’median age was 67 years(mean 66.46±10.03).The 5-year survival rate was 12.55%.Univariate analysis revealed age,sex,pathology,and treatment regimen as factors affecting prognosis.In multivariate analysis,when compared to postoperative chemotherapy,postoperative chemoradiotherapy was better associated with survival benefits(hazard ratio[HR]=0.85,95%confidence interval[CI]:0.813-0.898,P<0.001).Propensity score matching revealed that patients who had received postoperative chemoradiotherapy had a better prognosis than did patients who had received postoperative chemotherapy(HR=0.869,95%CI:0.817-0.925,P<0.001).Female patients and patients aged<65 years had a better prognosis than did their counterparts.Patients with adenocarcinoma had a better prognosis than did patients with squamous cell carcinoma.Moreover,prognosis worsened with increasing disease T stage.Patients who had received postoperative chemoradiotherapy had a better prognosis than did patients who had received postoperative chemotherapy.Postoperative radiotherapy was an independent prognostic factor in this patient group.
文摘Here, the authors propose a novel two-phase clustering algorithm with a density exploring distance (DED) measure. In the first phase, the fast global K-means clustering algorithm is used to obtain the cluster number and the prototypes. Then, the prototypes of all these clusters and representatives of points belonging to these clusters are regarded as the input data set of the second phase. Afterwards, all the prototypes are clustered according to a DED measure which makes data points locating in the same structure to possess high similarity with each other. In experimental studies, the authors test the proposed algorithm on seven artificial as well as seven UCI data sets. The results demonstrate that the proposed algorithm is flexible to different data distributions and has a stronger ability in clustering data sets with complex non-convex distribution when compared with the comparison algorithms.

文摘Hypoxia-induced factor-1 alpha (HIF-1α) affects many effector molecules and regulates tumor lymphangio- genesis and angiogenesis during hypoxia. The aim of this study was to investigate the role of HIF-1α in the regu- lation of vascular endothelial growth factor C (VEGF-C) expression and its effect on lymphangiogenesis and an- giogenesis in breast cancer. Lymphatic vessel density (LVD), microvessel density (MVD) and the expressions of HIF-1α and VEGF-C proteins were evaluated by immunohistochemistry in 75 breast cancer samples. There was a significant correlation between HIF-1α and VEGF-C (P = 0.014, r = 0.273, Spearman's coefficient of correlation). HIF-1α and VEGF-C overexpression was significantly correlated with higher LVD (P = 0.003 and P = 0.017, re- spectively), regional lymph nodal involvement (P = 0.002 and P = 0.004, respectively) and advanced tumor, node, metastasis (TNM) classification (P = 0.001 and P = 0.01, respectively). Higher MVD was observed in the group expressing higher levels of HIF-1α and VEGF-C (P = 0.033 and P = 0.037, respectively). Univariate analysis showed shorter survival time in patients expressing higher levels of HIF-1α and VEGF-C. HIF-1α was also found to be an independent prognostic factor of overall survival in multivariate analysis. The results suggest that HIF-1α may affect VEGF-C expression, thus acting as a crucial regulator of lymphangiogenesis and angiogenesis in breast cancer. This study highlights promising potential of HIF- 1α as a therapeutic target against tumor lymph node me- tastasis.

文摘Azilsartan(AZL), a poorly soluble drug, was considered to be fit for nanocrystals to improve its solubility. Our study intended to prepare AZL nanocrystals by means of bead milling method. Eight stabilizers or their binary combination and the milling time were set to be variable factors to optimize AZL nanosuspension formulation, and six types of freezedrying supports were investigated to reduce the aggregation of particles during the solidification. AZL nanocrystals with or without sodium deoxycholate(NaDC) as combined stabilizer with Poloxamer 188(F68) were prepared owning mean particle sizes of about 300 nm and 460 nm. During the screening processes, the formulation containing NaDC showed a smaller particle size and better stability during lyophilization. The irregular shape and crystal form changing in AZL nanocrystals were discovered by various characterizations. And with physical mixture as reference, nanocrystals showed its improvement about in-vitro dissolution and in-vivo bioavailability. In conclusion, the nanocrystals of AZL could be prepared well in our study. Additionally, our results suggested that NaDC was an appreciated excipient on the nanocrystals platform, which can exhibit the abilities of size-reduction and stabilitymaintaining on freeze-drying.

文摘In this work,a model of hydrogen production by double chemical looping is introduced.The efficiency benefit obtained was investigated.The chemical looping hydrogen generation unit is connected in series to the downstream of a chemical looping gasification unit as an additional system for 100 MWh coal gasification,with the function of supplementary combustion to produce hydrogen.Using Aspen Plus software for process simulation,the production of H_(2) and N_(2) in the series system is higher than that in the independent Chemical looping gasification and Chemical looping hydrogen generation systems,and the production of hydrogen is approximately 25.63%and 12.90%higher,respectively;The study found that when the gasification temperature is 900C,steam-carbon ratio is 0.84 and oxygen-carbon ratio is 1.5,the hydrogen production rate of the system was the maximum.At the same time,through heat exchange between logistics,high-pressure steam at 8.010×10^(4) kg·h^(-1) and medium-pressure steam at 1.101×10^(4) kg·h^(-1) are generated,and utility consumption is reduced by 61.58%,with utility costs decreasing by 48.69%.An economic estimation study found that the production cost of ammonia is 108.66 USD(t NH_(3))^(-1).Finally,cost of equipment is the main factors influencing ammonia production cost were proposed by sensitivity analysis.

文摘We sought to assess the effect of progestin on the apoptosis of epithelial ovarian cancer cell line SKOV-3 and via regulation of phosphorylation signaling in. Epithelial ovarian cancer cell line SKOV-3 was treated with me- droxyprogestogen, phosphatidylinositol 3-kinase inhibitor LY294002 and vehicle control. Akt, phospho-Akt, Bcl-2 and phospho-Bad proteins were examined by immunoblotting assays. Medroxyprogestogen-induced apoptosis was assessed by MTT assays and Annexin V apoptosis assay. We found no significant difference in Akt and Bad expression in both the medroxyprogestogen groups and the control group. The levels of phospho-Akt, Bcl-2 and phospho-Bad were decreased in all the medroxyprogestogen groups and significantly decreased in the high dose mitogen-activated protein (MAP) group (10 gmol/L). Viability of SKOV-3 was reduced and apparent apoptosis of SKOV-3 cells was observed with increased doses of MAP. The findings suggest that medroxyprogestogen can induce SKOV-3 cell apoptosis by inhibiting Akt phosphorylation.

文摘Picea mongolica is a conifer with a limited distribution in China and its taxonomic status is controversial.In order to explore genetic diff erences between P.mongolica and other nearby Picea species and to investigate its taxonomic status,phylogenetic relationships were analyzed between P.mongolica and Picea koraiensis,Picea meyeri and Picea wilsonii by three chloroplast gene sequences matK,chlB and atpA.The length of joint chloroplast sequence is 2379 bp.The fi fteen haplotypes were identifi ed by haplotype network analysis,among which two were major haplotypes and nine were unique.In addition,the genetic diversity of the sample collection species was inferred.Based on the haplotype network and Neighbor Joining phylogenetic tree analysis,P.mongolica was located on the basal clade of the phylogenetic tree which had more primitive taxa,and the genetic diversity of P.mongolica was highest.The signifi cant diff erences between P.mongolica and these other Picea species were identifi ed by this research.

文摘Melanoma is the most lethal type of skin cancer,originating from the malignant transformation of melanocyte.While the development of targeted therapy and immunotherapy has gained revolutionary advances in potentiating the therapeutic effect,the prognosis of patients with melanoma is still suboptimal.During tumor progression,melanoma frequently encounters stress from both endogenous and exogenous sources in tumor microenvironment.SIRT7 is a nuclear-localized deacetylase of which the activity is highly dependent on intracellular nicotinamide adenine dinucleotide(NAD+),with versatile biological functions in maintaining cell homeostasis.Nevertheless,whether SIRT7 regulates tumor cell biology and tumor immunology in melanoma under stressful tumor microenvironment remains elusive.Herein,we reported that SIRT7 orchestrates melanoma progression by simultaneously promoting tumor cell survival and immune evasion via the activation of unfolded protein response.We first identified that SIRT7 expression was the most significantly increased one in sirtuins family upon stress.Then,we proved that the deficiency of SIRT7 potentiated tumor cell death under stress in vitro and suppressed melanoma growth in vivo.Mechanistically,SIRT7 selectively activated the IRE1α-XBP1 axis to potentiate the pro-survival ERK signal pathway and the secretion of tumorpromoting cytokines.SIRT7 directly de-acetylated SMAD4 to antagonize the TGF-β-SMAD4 signal,which relieved the transcriptional repression on IRE1αand induced the activation of the IRE1α-XBP1 axis.Moreover,SIRT7 up-regulation eradicated anti-tumor immunity by promoting PD-L1 expression via the IRE1α-XBP1 axis.Additionally,the synergized therapeutic effect of SIRT7 suppression and anti-PD-1 immune checkpoint blockade was also investigated.Taken together,SIRT7 can be employed as a promising target to restrain tumor growth and increase the effect of melanoma immunotherapy.

文摘Background:Hepatocellular carcinoma(HCC)is one of the most common malignant tumors in the world.The ubiquitin-specific peptidase 25(USP25)protein has been reported to participate in the development of several cancers.However,few studies have reported its association with HCC.In this study,we aimed to investigate the function and mechanism of USP25 in the progression of HCC.Methods:We analyzed USP25 protein expression in HCC based on The Cancer Genome Atlas(TCGA)and International Cancer Genome Consortium(ICGC)database cohorts.Then,we constructed USP25-overexpressing and USP25-knockdown HepG2,MHCC97H,and L-O2 cells.We detected the biological function of USP25 by performing a series of assays,such as Cell Counting Kit-8(CCK-8),colony formation,transwell,and wound healing assays.Western blotting and quantitative real-time polymerase chain reaction(qRT-PCR)analyses were performed to detect the interaction between USP25 and the Wnt/β-catenin signaling pathway.The relationship between USP25 and tripartite motif-containing 21(TRIM21)was assessed through mass spectrometry and co-immunoprecipitation(Co-IP)analysis.Finally,we constructed a mouse liver cancer model with the USP25 gene deletion to verify in vivo role of USP25.Results:USP25 was highly expressed in HCC tissue and HCC cell lines.Importantly,high expression of USP25 in tissues was closely related to a poor prognosis.USP25 knockdown markedly reduced the proliferation,migration,and invasion of HepG2 and MHCC97H cells,whereas USP25 overexpression led to the opposite effects.In addition,we demonstrated that USP25 interacts with TRIM21 to regulate the expression of proteins related to epithelial-mesenchymal transition(EMT;E-cadherin,N-cadherin,and Snail)and the Wnt/β-catenin pathway(β-catenin,Adenomatous polyposis coli,Axin2 and Glycogen synthase kinase 3 beta)and those of their downstream proteins(C-myc and Cyclin D1).Finally,we verified that knocking out USP25 inhibited tumor growth and distant metastasis in vivo.Conclusions:In summary,our data showed that USP25 was overexpressed in HCC.USP25 promoted the proliferation,migration,invasion,and EMT of HCC cells by interacting with TRIM21 to activate theβ-catenin signaling pathway.

文摘Aiming at the problems of single classification method and high classification cost of kiwifruit in China,we proposed a grading method based on kiwifruit surface defects.A set of kiwifruit image acquisition system was built.The K-means clustering segmentation algorithm was used to segment the surface defects,and then color contrast was performed to determine whether it was a piece of defective fruit.Then,the shape features of normal fruit were extracted and an SVM classifier was designed to further determine its grade.This method has the advantages of low cost,simple algorithm and high efficiency,which opens a new way for fruit classification,and is of great significance to promoting the development of fruit classification industry in China and enhancing international competitiveness.
