目的:探讨普乐沙福联合硫培非格司亭的稳态动员策略在淋巴瘤和多发性骨髓瘤(multiple myeloma,MM)患者中自体外周血造血干细胞(peripheral blood hematopoietic stem cell,PBSC)动员的有效性。方法:回顾性分析2016年4月至2023年4月于绵... 目的:探讨普乐沙福联合硫培非格司亭的稳态动员策略在淋巴瘤和多发性骨髓瘤(multiple myeloma,MM)患者中自体外周血造血干细胞(peripheral blood hematopoietic stem cell,PBSC)动员的有效性。方法:回顾性分析2016年4月至2023年4月于绵阳市中心医院接受普乐沙福联合硫培非格司亭(P+Mecapeg),普乐沙福联合重组人粒细胞集落刺激因子(P+rhG-CSF),单用硫培非格司亭(Mecapeg)稳态动员方案进行自体PBSC动员的50例淋巴瘤和MM患者。分别比较3组稳态动员方案的采集疗效、移植后造血重建,并探讨了每组接受稳态动员方案患者采集失败的危险因素。结果:50例患者中,应用P+Mecapeg为25例(50.0%),P+rhG-CSF为12例(24.0%),单用Mecapeg为13例(26.0%)。PBSC动员成功率分别为100%、83.3%和76.9%;中位CD34+细胞数分别为4.23×10^(6)/kg、3.43×10^(6)/kg和3.16×10^(6)/kg;中位采集天数分别为1.0、1.5、2.0天。P+Mecapeg组采集第1天单个核细胞显著高于P+rhG-CSF(P=0.030)及单用Mecapeg组(P=0.005),PBSC采集成功率高于单用Mecapeg动员方案(P=0.034),P+Mecapeg动员方案采集成功率较高(P=0.025)。二元Logistic回归分析显示一线自体造血干细胞移植(autologous hematopoietic stem cell transplantation,ASCT)是PBSC采集的独立保护因素(P=0.008,OR=36,95%CI:2.585~501.268)。50例患者中36例完成ASCT,其中P+Mecapeg动员组19例,对照组(P+rhG-CSF及单用Mecapeg动员组)17例,P+Mecapeg与对照组中性粒细胞中位植活时间均为10天,血小板中位植活时间均为12天,两者在造血重建方面比较差异无统计学意义。结论:普乐沙福联合硫培非格司亭是一种高效、安全、可替代的PBSC稳态动员策略,能有效提高淋巴瘤和MM患者PBSC动员达标成功率,一线ASCT前进行PBSC动员与采集可显著提高淋巴瘤和MM患者采集成功率。 展开更多

A Rapid Determination of 30 Prohibited Pesticide Residues in Lycii Fructus by UPLC-MS/MS

Prohibited pesticide residues have become one of the main factors affecting the quality and safety of Lycii Fructus,However,rarely studies focus on the rapid determination of these residues.Here,a total of 30 kinds of prohibited pesticide residues were determined by ultra-performance liquid chromatography tandem triple quadrupole mass spectrometry(UPLC-MS/MS)in five different process ways.Pretreatment methods,chromatographic separation and detection conditions in mass spectrometry were all optimized accordingly.Among the five different pretreatment methods,the first and third solid phase extraction failed to provide high recoveries of sulfosulfuron compounds(both lower than 60%).Recovery of chlorphenamidine by the Quick Easy Cheap Effective Rugged and Safe multiresidue method(QuEChERS)was lower than 60%,which did not meet the requirements of trace determination.The concentrations of 30 prohibited pesticides residues treated by straightforward and solid phase extraction showed good linearity in their corresponding ranges,with correlation coefficients over 0.99.The average recoveries of straightforward ranged from 78.13%to 110.9%,while RSD ranged from 1.3%to 16.9%,albeit poor purification was observed.The recovery yield from solid phase extraction was between 67.75%and 103.08%with RSD value from 0.8%to 14.0%,which met the requirements of trace determination,this method has good precision and stability.These results could be employed to other Traditional Chinese Medicines(TCMs)in detecting prohibited pesticide residues. 展开更多

As a natural analog of resveratrol,piceatannol(Pic)exhibits good antioxidant and anti-inflammatory activities in different disease models.However,the role of Pic in type 1 diabetes mousemodel has not been reported yet.In this study,we investigated the in vivo effect of Pic in streptozotocin(STZ)-induced type 1 diabetic mice.Mice were injected with STZ to establish the type 1 diabetesmellitus(T1DM)model.After stable hyperglycemia was achieved,mice were then orally treated with Pic(40 mg/kg b.w.,i.g.)for 30 days.The results indicated that Pic supplementation efficiently alleviated the typical symptoms associated with T1DM,including body weight loss,polydipsia,hyperglycemia,and hypoinsulinemia.Pic treatment also improved the glucose tolerance of STZ-induced diabetic mice.In addition,Pic supplementation markedly decreased the expression of pro-inflammatory molecules TNF-αand IL-6,the expression of endoplasmic reticulum(ER)stress markers GRP78 and CHOP,and the level of oxidative stress in T1DM mice.Moreover,Pic administration also partly reversed the metabolic profiles of STZ-treated mice as detected by 1H Nuclear Magnetic Resonance(NMR)-based metabolomics.Our study suggested that the therapeutic potential of Pic in type 1 diabetes and the anti-diabetic effects of Pic may be associated with its activities to suppress oxidative stress,inflammation,and ER stress. 展开更多

Wheat production requires at least-2.4%increase per year rate by 2050 globally to meet food demands.However,heat stress results in serious yield loss of wheat worldwide.Correspondingly,wheat has evolved genetic basis and molecular mechanisms to protect themselves from heat-induced damage.Thus,it is very urgent to understand the underlying genetic basis and molecular mechanisms responsive to elevated temperatures to provide important strategies for heat-tolerant varieties breeding.In this review,we focused on the impact of heat stress on morphology variation at adult stage in wheat breeding programs.We also summarize the recent studies of genetic and molecular factors regulating heat tolerance,including identification of heat stress tolerance related QTLs/genes,and the regulation pathway in response to heat stress.In addition,we discuss the potential ways to improve heat tolerance by developing new technologies such as genome editing.This review of wheat responses to heat stress may shed light on the understanding heat-responsive mechanisms,although the regu-latory network of heat tolerance is still ambiguous in wheat. 展开更多
