Inhibiting the death receptor 3(DR3)signaling pathway in group 3 innate lymphoid cells(ILC3s)presents a promising approach for promoting mucosal repair in individuals with ulcerative colitis(UC).Paeoniflorin,a promine...Inhibiting the death receptor 3(DR3)signaling pathway in group 3 innate lymphoid cells(ILC3s)presents a promising approach for promoting mucosal repair in individuals with ulcerative colitis(UC).Paeoniflorin,a prominent component of Paeonia lactiflora Pall.,has demonstrated the ability to restore barrier function in UC mice,but the precise mechanism remains unclear.In this study,we aimed to delve into whether paeoniflorin may promote intestinal mucosal repair in chronic colitis by inhibiting DR3 signaling in ILC3s.C57BL/6 mice were subjected to random allocation into 7 distinct groups,namely the control group,the 2%dextran sodium sulfate(DSS)group,the paeoniflorin groups(25,50,and 100 mg/kg),the anti-tumor necrosis factor-like ligand 1A(anti-TL1A)antibody group,and the IgG group.We detected the expression of DR3 signaling pathway proteins and the proportion of ILC3s in the mouse colon using Western blot and flow cytometry,respectively.Meanwhile,DR3-overexpressing MNK-3 cells and 2%DSS-induced Rag1^(-/-)mice were used for verification.The results showed that paeoniflorin alleviated DSS-induced chronic colitis and repaired the intestinal mucosal barrier.Simultaneously,paeoniflorin inhibited the DR3 signaling pathway in ILC3s and regulated the content of cytokines(interleukin-17A,granulocyte-macrophage colony stimulating factor,and interleukin-22).Alternatively,paeoniflorin directly inhibited the DR3 signaling pathway in ILC3s to repair mucosal damage independently of the adaptive immune system.We additionally confirmed that paeoniflorin-conditioned medium(CM)restored the expression of tight junctions in Caco-2 cells via coculture.In conclusion,paeoniflorin ameliorates chronic colitis by enhancing the intestinal barrier in an ILC3-dependent manner,and its mechanism is associated with the inhibition of the DR3 signaling pathway.展开更多
The ocean is one of the essential fields of national defense in the future,and more and more attention is paid to the lightweight research of Marine equipment and materials.This study it is to develop a Machine learni...The ocean is one of the essential fields of national defense in the future,and more and more attention is paid to the lightweight research of Marine equipment and materials.This study it is to develop a Machine learning(ML)-based prediction method to study the evolution of the mechanical properties of Al-Li alloys in the marine environment.We obtained the mechanical properties of Al-Li alloy samples under uniaxial tensile deformation at different exposure times through Marine exposure experiments.We obtained the strain evolution by digital image correlation(DIC).The strain field images are voxelized using 2D-Convolutional Neural Networks(CNN)autoencoders as input data for Long Short-Term Memory(LSTM)neural networks.Then,the output data of LSTM neural networks combined with corrosion features were input into the Back Propagation(BP)neural network to predict the mechanical properties of Al-Li alloys.The main conclusions are as follows:1.The variation law of mechanical properties of2297-T8 in the Marine atmosphere is revealed.With the increase in outdoor exposure test time,the tensile elastic model of 2297-T8 changes slowly,within 10%,and the tensile yield stress changes significantly,with a maximum attenuation of 23.6%.2.The prediction model can predict the strain evolution and mechanical response simultaneously with an error of less than 5%.3.This study shows that a CNN/LSTM system based on machine learning can be built to capture the corrosion characteristics of Marine exposure experiments.The results show that the relationship between corrosion characteristics and mechanical response can be predicted without considering the microstructure evolution of metal materials.展开更多

TLJN has been shown to protect primary cultured hippocampal neurons. How-ever, its mechanism of action remains unclear. In the present study, primary cultured hippocampal neurons treated with Aβ1-42 (10 μmol/L) signiifcantly increased the release of lactate dehydroge-nase, which was markedly reduced by TLJN (2 μL/mL), speciifcally by the component geniposide (26 μmol/L), but not ginsenoside Rg1 (2.5 μmol/L). hTe estrogen receptor inhibitor, ICI182780 (1 μmol/L), did not block TLJN-or geniposide-mediated decrease of lactate dehydrogenase under Aβ1-42-exposed conditions. In this paper, we...Lane line detection is a fundamental step in applications like autonomous driving and intelligent traffic monitoring. Emerging applications today have higher requirements for accurate lane detection. In this paper, we present a precise information extraction algorithm for lane lines. Specifically, with Gaussian Mixture Model(GMM), we solved the issue of lane line occlusion in multi-lane scenes. Then, Progressive Probabilistic Hough Transform(PPHT) was used for line segments detection. After K-Means clustering for line segments classification, we solved the problem of extracting precise information that includes left and right edges as well as endpoints of each lane line based on geometric characteristics. Finally, we fitted these solid and dashed lane lines respectively. Experimental results indicate that the proposed method performs better than the other methods in both single-lane and multi-lane scenarios.展开更多
Background and aims:Cytokeratin 19-positive(CK19t)hepatocellular carcinoma(HCC)is an aggressive subtype with poor outcomes.The initiation and development of CK19t HCC in the background of liver cirrhosis remains uncle...Background and aims:Cytokeratin 19-positive(CK19t)hepatocellular carcinoma(HCC)is an aggressive subtype with poor outcomes.The initiation and development of CK19t HCC in the background of liver cirrhosis remains unclear.This study investigated the role of the cirrhosis-related gene C-C motif chemokine ligand 16(CCL16)in the development of CK19t HCC.Methods:Datasets from Gene Expression Omnibus(GEO),The Cancer Genome Atlas(TCGA)and International Cancer Genome Consortium(ICGC)were analyzed to screen and validate the genes associated with CK19t HCC.A total of 102 HCC patients were included for tissue microarray analysis.Gain-of-function experiments were conducted to investigate the biological functions of CCL16.CIBERSORT was used to investigate the correlation of CCL16 and immune infiltration.Results:GEO dataset analysis showed that CK19t HCC had lower expression of CCL16.In both TCGA dataset and our HCC cohort,CCL16 expression was negatively correlated with CK19 expression(P<0.05)and its expression was higher in para-tumor than tumor tissues(P<0.001).Moreover,low CCL16 expression was related to advanced stage and poor overall survival(P<0.05).CCL16 overexpression downregulated CK19 expression and impacted the sphere formation ability of HCC cells.Overexpression of CCL16 inhibited the cell proliferation,migration,and invasion of HCC cell lines.Immune analysis showed HCC with high CCL16 expression had more infiltration of mast cells.HCC patients with both low CCL16 expression and low mast cells had the worst prognosis(P<0.001).Conclusion:Our data indicated that CCL16 downregulated the expression of CK19 and inhibited the malignant phenotype of HCC.展开更多
OBJECTIVE: To investigate the traditional Chinese Medicine (TCM) etiology and pathogenesis of ac- quired immune deficiency syndrome (AIDS) by 18-month observation of Chinese rhesus macaques infected with simian i...OBJECTIVE: To investigate the traditional Chinese Medicine (TCM) etiology and pathogenesis of ac- quired immune deficiency syndrome (AIDS) by 18-month observation of Chinese rhesus macaques infected with simian immunodeficiency virus (SIV) mac239. METHODS: Thirty-five healthy Chinese rhesus ma- caques were divided into a model group (n=30) and a control group (n=5). The model was estab- lished by inoculating monkeys intravenously with SIVmac239. Changes in TCM symptoms after SIV in- fection within 18 months were then observed and recorded. Routine blood tests, SIV viral load, T-lym-phocyte subsets, plasma triiodothyronine (T3), tet- raiodothyronine (14), adrenocorticotropic hormone (ACTH) and cortisol (Cor) were tested periodically during the experiment. RESULTS: During the acute infection period of SIV, model monkeys temporarily showed clinical symp- toms such as diarrhea, dysphoria and slight weight loss. Decrease percentages of CD4~ T-lymphocytes were observed but levels of T3, 14, Cot, and ACTH were relatively unchanged. Monkeys in the model group during the early and middle periods of infec- tion showed no obvious symptoms, except few monkeys exhibited transient diarrhea and reduced food intake. All variables at this stage showed nor- mal fluctuations. In the middle period model group monkeys showed chronic and persistent diarrhea, weight loss, reduced food intake and low levels of 13 and Cot. In the late period, symptoms including emaciation, weight loss, listlessness, crouching in corners and low levels of T3appeared. CONCLUSION: The results suggest that the rhesus monkey SIWSAIDS model can be applied to re- search on TCM etiology and pathogenesis of AIDS. According to this model, the etiology of disease is the SIV virus. The pathogenesis manifests as the in- vasion of SIV virus, incubation of the virus, balance between virus and healthy "Qi", damage to spleen and kidney as the disease progressed, exhaustion of vitality and finally the failure of five zang and six fu organs.展开更多
