AIM: To transfer hepatitis E virus (HEV) ORF2 partial gene to tomato plants, to investigate its expression in transformants and the immunoactivity of expression products, and to explore the feasibility of developing a...AIM: To transfer hepatitis E virus (HEV) ORF2 partial gene to tomato plants, to investigate its expression in transformants and the immunoactivity of expression products, and to explore the feasibility of developing a new type of plantderived HEV oral vaccine.METHODS: Plant binary expression vector p1301E2,carrying a fragment of HEV open reading frame-2 (named HEV-E2), was constructed by linking the fragment to a constitutive CaMV35s promoter and nos terminator, then directly introduced into Agrobacterium tumefaciens EHA105. With leaf-disc method, tomato plants medicated by EHA105 were transformed and hygromycin-resistant plantlets were obtained in selective medium containing hygromycin. The presence and integration of foreign DNA in transgenic tomato genome were confirmed by Gus gene expression, PCR amplification and Southern dot blotting.The immunoactivity of recombinant protein extracted from transformed plants was examined by enzyme-linked immunosorbant assay (ELISA) using a monoclonal antibody specifically against HEV. ELISA was also used to estimate the recombinant protein content in leaves and fruits of the transformants.RESULTS: Seven positive lines of HEV-E2-transgenic tomato plants confirmed by PCR and Southern blotting were obtained and the immunoactivity of recombinant protein could be detected in extracts of transformants. The expression levels of recombinant protein were 61.22 ng/g fresh weight in fruits and 6.37-47.9 ng/g fresh weight in leaves of the transformants.CONCLUSION: HEV-E2 gene was correctly expressed in transgenic tomatoes and the recombinant antigen derived from them has normal immunoactivity. Transgenic tomatoes may hold a good promise for producing a new type of lowcost oral vaccine for hepatitis E virus.展开更多

The expressions of COX-2 and bcl-2 in cancerous tissues and corresponding para-cancerous tissues were investigated by immunohistochemistry using COX-2 polyclonal antibody and bcl-2 monoclonal antibody. The normal gastric mucosa tissues were used as control.RESULTS: The expressions of COX-2 and bcl-2 in gastric carcinoma were significantly higher than that in the paracancerous tissues (77.8% vs 47.2%, P<0.01, 80.56% vs 58.33%, P<0.05). The expression of COX-2 in cardiac adenocarcinoma was remarkably higher than that in the distal gastric carcinoma (93.8% vs 65.0%, P<0.01). The expression of COX-2 was mainly localized in the cytoplasm of tumor cells and partly in the nucleus. There is a transition of the COX-2 cytoplasmic positivity to nucleic in tumor cells with the increase of gastric carcinoma pathological grade. Interstitial macrophages, fibroblasts and vascular endothelial cells also expressed COX-2. The tissues with higher expression of COX-2 also expressed high level of bcl-2 protein.CONCLUSION: Abnormal expression pattern of COX-2within the tissues of human gastric cancer is correlated with tumor location and lymph node metastasis. COX-2may regulate expression of apoptosis suppressor gene (bcl-2) through interaction of tumor cells and stromal cells and play an important role in the generation and development of tumors, which will be of great help in developing new methods for antitumor therapy.展开更多

Patients were divided into 2 groups; those with squamocolumnar junction (SCJ) beyond gastroesophageal junction (GEJ)≥3cm (group A), and those with SCJ beyond GE.1 <3cm (group B). Biopsy specimens were obtainedend escopically from just below the SCJ, gastric antrum along the greater and lesser curvature. Pathological changes and Hpylorr infection were determined by HE staining, Alcian blue staining and Giemsa staining.RESULTS:The prevalence of Hpyloriinfection was 46.93%.There was no difference in the prevalence between males and females.The prevalence of Hpyloriinfection decreased stepwise significantly from RE grade I to Ⅲ.There was no difference in the prevalence between the two groups, and between long-segment and short-segment BE. In distal stomach, prevalence of Hpyloriinfection was significantly higher in patients with IM than those without IM.CONCLUSION: There is a protective role of Hpyloriinfectuion to GERD. There may be no relationship between Hpylori infection of stomach and BE. Hpyloriinfection is associated with the development of IN in the distal stomach.展开更多

METHODS: This study was carded out in a large prospective series of 391 patients who had undergone upper endoscopy. The patients were divided into 3 groups according to the position of squamocolumnar junction (SC3). Reflux esophagitis (RE) and its degree were recorded. Intestinal metaplasia (IM) in biopsy specimen was typed according to histochemistry and HE and alcian blue (pH2.5) staining separately. Results correlating with clinical, endoscopic, and pathological data were analysed. RESULTS: The prevalence of IM endoscopically appearing Long-segment Barrett's Esophagus (LSBE) was 26.53%, Short-segment Barrett's Esophagus (SSBE) was 33.85% and gastroesophageal junction (GEJ) was 34.00%. IM increased with age of above 40 years old and no difference was found between male and female. Twelve were diagnosed as dysplasia (7 low -grade, 5 high-grade), 16 were diagnosed as cardiac adenocarcinoma and 1 as esophageal adenocarcinoma. The more far away the SCJ moved upward above GEJ, the higher the prevalence and the more severe the RE were. CONCLUSION: There was no difference of the prevalence of IM in different places of SCJ, and IM increased with age of above 40 years old. It is important to pay attention to dysplasia in the distal esophagus and gastro-esophageal junction, and adenocarcinoma is more common in cardia than in esophagus. BE is a consequence of gastroesophageal reflux disease.展开更多
AIM:Modified Heller's myotomy is still the first choice for achalasia and the assessment of surgical outcomes is usually made based on the subjective sensation of patients.This study was to objectively assess the ...AIM:Modified Heller's myotomy is still the first choice for achalasia and the assessment of surgical outcomes is usually made based on the subjective sensation of patients.This study was to objectively assess the long-term outcomes of esophageal myotomy for achalasia using esophageal manometry, 24-hour pH monitoring,esophageal scintigraphy and fiberoptic esophagoscopy.METHODS:From February 1979 to October 2000, 176 patients with achalasia underwent modified Heller's myotomy, including esophageal myotomy alone in 146 patients, myotomy in combination with Gallone or Dor antirefiux procedure in 22 and 8 patients, respectively. Clinical score,pressure of the lower esophageal sphincter (LES),esophageal clearance rate and gastroesophageal reflux were determined before and i to 22 years after surgery.RESULTS: After a median follow-up of 14 years, 84.5% of patients had a good or excellent relief of symptoms,and clinical scores as well as resting pressures of the esophageal body and LES were reduced compared with preoperative values (P<0.001).However,there was no significant difference in DeMeester score between pre-and postoperative patients(P=0.51).Esophageal transit was improved in postoperative patients, but still slower than that in normal controls. The incidence of gastroesophageal reflux in patients who underwent esophageal myotomy alone was 63.6% compared to 27.3% in those who underwent myotomy and antirefiux procedure (P=-0.087). Three (1.7%) patients were complicated with esophageal cancer after surgery.CONCLUSION: Esophageal myotomy for achalasia can reduce the resting pressures of the esophageal body and LES and improve esophageal transit and dysphagia. Myotomy in combination with antireflux procedure can prevent gastroesophageal reflux to a certain extent,but further randomized studies should be carried out to demonstrate its efficacy.展开更多

The mixed probe was hybridized with three pieces of BiostarH-40 s double dot human whole gene chip. The chips were scanned with a ScanArray 4000. The acquired images were analyzed using GenePix Pro 3.0 software.RESULTS: A total of 141 genes were screened out that exhibited different expression in all three chips. There were 74 upregulated and 67 downregulated genes in gene expression profiles of BE which were two times of that in CIM.CONCLUSION: There is a difference in gene expression level between BE and CIM epithelia. These 141 genes probably relate to the occurrence and development of BE and the progression to adenocarcinoma.展开更多

METHODS: By u...AIM:To explore the expression of Sp1 in gastric carcinoma as well as its association with other clinicopathologic features, and to evaluate the role of Spl as a prognostic indicator of gastric carcinoma. METHODS: By using immunohistochemistry, we examined the Spl expression patterns in 65 cases of human gastric cancer, and 40 normal gastric mucosa specimens. Simultaneously, the correlation between Spl expression and clinical outcome or clinicopathologic features was investigated. RESULTS: The percentage of Spl expression was 12.5% (5/40) in normal gastric mucosa, and the Spl protein was mainly expressed in the nuclei of cells located in the mucous neck region. In sharp contrast, strong Spl expression was detected in tumor cells, whereas no or faint Spl staining was detected in stromal cells and normal glandular cells surrounding the tumors. The expression rate of Spl in gastric cancer lesions was 53.85% (35/65). The medium survival duration in patients who had a tumor with negative, weak and strong Spl expressions was 1 700, 1 560 and 1 026 d, respectively (P<0.05). Spl protein expression was closely related to the depth of tumor infiltration (X2 = 13.223, P<0.01) and TNM stage (X2= 11.009, P<0.05), but had no relationship with the number of lymph nodes and Lauren's classification (P>0.05). Cox regression model for multivariate analysis revealed that high Spl expression (P<0.05) and advanced stage (P<0.01) were independent predictors of poor survival. CONCLUSION: Normal and malignant gastric tissues have unique Spl expression patterns. Spl might serve as an independent prognostic factor, by influencing the tumor infiltration and progression.展开更多

METHODS: The expression of somatostatin receptor 2 (SSTR2) messenger RNA (mRNA) in human HCC cell line HepG2 was examined by reverse transcriptase-polymerase chain reaction (RT-PCR). Uptake of 99mTc-HYNIC-TOC was evaluated in the human HCC implanted into BALB/c nude mice. ANMIS2000 nuclear medicine analysis system was used to calculate the ratio of 99mTc uptake between tumor tissue and vital organs. RESULTS: We demonstrated the expression of SSTR2 mRNA in human HCC cell line HepG2 by RT-PCR. The size of the RT-PCR products was 364 bp detected by sequence analysis of the human SSTR2 mRNA. Scintigraphy proved that 99mTc-HYNIC-TOC was uptaken in the tumor tissue, liver and kidney of the tumor-bearing mice. CONCLUSION: Based on expression of the SSTR2 mRNA in human HCC, 99mTc-HYNIC-TOC can markedly bind with and be uptaken by human HCC tissues as compared with normal liver tissue. The significant retention of radionudide in kidney and bladder is probably related to non-specific peptide uptake in the tubulus cells of kidney and possibly due to excretion by kidney. Our results show that localizable diagnosis and targeting radiotherapy with radionuclide-labeled somatostatin analog for HCC are of great value to be further studied.展开更多

For various aging temperatures, the time to finish precipitation of α2 phase is different. The facts that various distribution characteristics and growth speed of α2 ordered phase are caused by changed aging condition imply optimal selection and control for precipitation of α2 ordered phase reachable. Aging at a moderate temperature can promote the tendency of uniformprecipitation of alpha_2 phase in both alpha_p and beta_t. OBJECTIVE To investigate the effect of scutellarin(SCU),which is the main effective component of Erigeron breviscapus(Vant.)Hand-Mazz native to Yunnan in China,on vascular dysfunction(VD)of cardiac coronary artery(CA)...OBJECTIVE To investigate the effect of scutellarin(SCU),which is the main effective component of Erigeron breviscapus(Vant.)Hand-Mazz native to Yunnan in China,on vascular dysfunction(VD)of cardiac coronary artery(CA)and cerebral basilar artery(BA)caused by hypertension in SHR rats.METHODS 1.BA and CA vesselsrings from 40 weeks of SHR rats were isolated and equilibrated in organ bath with MOPS-PSS buffer and ring tension was recorded,comparing with a normal control of WKY rats.SCU was treated accumulatively after pre-contracted with vasoconstrictor U46619(1μmol·L-1).2.ACH and SNP were treated accumulatively after pre-incubation with SCU(300μmol·L-1,100μmol·L-1)and pre-contracted by U46619(1μmol·L-1),K+60mmol·L-1,respectively.While U46619 was added accumulatively to BA/CA rings pre-incubated with SCU(300and 100μmol·L-1).ACH-induced relaxation rate was to evaluate endothelium-dependent relaxation,and SNP-induced relaxation rate to evaluate the artery non-endothelium-dependent relaxation.RESULTS SCU significantly dilated both BA and CA rings pre-contracted by U46619 in old rats.EC50 value of SCU in WKY ratswas less than that in SHR(P<0.05),which showing VD of CA and BA in SHR rats.Compared with WKY group,ACH relaxation curve of SHR group shifted to the right,suggesting that hypertension induced VD.When SCU 300μmol·L-1 pre-treated CA groups and SCU 100μmol·L-1 pre-treated BA groups,EC50 to ACH was significantly lower(P<0.05).Likewise,the vasodilatation of CA/BA rings to SNP was also improved obviously when pre-treated with SCU,and Emax to SNP was decreased significantly(P<0.05).Moreover,EC50 to U46619was significantly lower when pre-treated by SCU.CONCLUSION In SHR rats,SCU antagonized U46619 on CA/BA rings in a noncompetitive manner.Furthermore,SCU should appear to protect VD induced by hypertension.展开更多

The distribution and size of the α2 ordered p...The precipitation characteristics of the α2 ordered phase in Ti-AI-Sn-Zr-Mo-Si-Nd alloys with various content of Al, under different aging conditions, were investigated. The distribution and size of the α2 ordered phase changed with temperature and Al content. The dislocations were the only places where the α2 ordered phase could precipitate at higher temperature near the critical transformation temperature for each alloy experimented. With the addition of Al content, the critical transformation temperature of α2 ordered phase increased. When the aging temperature was relatively low (650℃), the precedent precipitation of α2 ordered phase took place in primary a phase at the early stage of aging, in the duplex microstructure (the primary a with the transformed (3) of the alloys with lower Al content. But after certain aging time (50 h), the size of α2 particles was almost equal in both the primary a and the transformed β. And no obvious growth of α2 particles could be observed after 50 h.展开更多

These alloys were designed with varied electron concentration values and named as Sn-rich alloys and Al-rich alloys, respectively. The precipita- tion and growth of α2 ordered phase in the tested alloys under various heat treatment conditions were investigated. Some compari- sons among the experimental results were performed and discussed in detail. Stronger precipitation and growth of α2 ordered phase were caused in Al-rich alloys but relatively weak change in Sn-rich alloys with increasing the electron concentration. The precipita- tion of α2 ordered phase in Al-rich alloys is stronger than that in Sn-rich alloys when the electron concentration value is the same for the two alloys.展开更多

文摘AIM:Modified Heller's myotomy is still the first choice for achalasia and the assessment of surgical outcomes is usually made based on the subjective sensation of patients.This study was to objectively assess the long-term outcomes of esophageal myotomy for achalasia using esophageal manometry, 24-hour pH monitoring,esophageal scintigraphy and fiberoptic esophagoscopy.METHODS:From February 1979 to October 2000, 176 patients with achalasia underwent modified Heller's myotomy, including esophageal myotomy alone in 146 patients, myotomy in combination with Gallone or Dor antirefiux procedure in 22 and 8 patients, respectively. Clinical score,pressure of the lower esophageal sphincter (LES),esophageal clearance rate and gastroesophageal reflux were determined before and i to 22 years after surgery.RESULTS: After a median follow-up of 14 years, 84.5% of patients had a good or excellent relief of symptoms,and clinical scores as well as resting pressures of the esophageal body and LES were reduced compared with preoperative values (P<0.001).However,there was no significant difference in DeMeester score between pre-and postoperative patients(P=0.51).Esophageal transit was improved in postoperative patients, but still slower than that in normal controls. The incidence of gastroesophageal reflux in patients who underwent esophageal myotomy alone was 63.6% compared to 27.3% in those who underwent myotomy and antirefiux procedure (P=-0.087). Three (1.7%) patients were complicated with esophageal cancer after surgery.CONCLUSION: Esophageal myotomy for achalasia can reduce the resting pressures of the esophageal body and LES and improve esophageal transit and dysphagia. Myotomy in combination with antireflux procedure can prevent gastroesophageal reflux to a certain extent,but further randomized studies should be carried out to demonstrate its efficacy.

文摘AIM: To study the difference of gene expression profile changes in Barrett's esophagus (BE) and cardia intestinal metaplasia (CIM) and to screen the novel genes in the early stage by cDNA microarray.METHODS: cDNA retrotranscribed from an equal amount of mRNA from BE and CIM epithelial tissues was labeled with Cy3 and Cy5 fluorescence as probes. The mixed probe was hybridized with three pieces of BiostarH-40 s double dot human whole gene chip. The chips were scanned with a ScanArray 4000. The acquired images were analyzed using GenePix Pro 3.0 software.RESULTS: A total of 141 genes were screened out that exhibited different expression in all three chips. There were 74 upregulated and 67 downregulated genes in gene expression profiles of BE which were two times of that in CIM.CONCLUSION: There is a difference in gene expression level between BE and CIM epithelia. These 141 genes probably relate to the occurrence and development of BE and the progression to adenocarcinoma.

文摘AIM:To explore the expression of Sp1 in gastric carcinoma as well as its association with other clinicopathologic features, and to evaluate the role of Spl as a prognostic indicator of gastric carcinoma. METHODS: By using immunohistochemistry, we examined the Spl expression patterns in 65 cases of human gastric cancer, and 40 normal gastric mucosa specimens. Simultaneously, the correlation between Spl expression and clinical outcome or clinicopathologic features was investigated. RESULTS: The percentage of Spl expression was 12.5% (5/40) in normal gastric mucosa, and the Spl protein was mainly expressed in the nuclei of cells located in the mucous neck region. In sharp contrast, strong Spl expression was detected in tumor cells, whereas no or faint Spl staining was detected in stromal cells and normal glandular cells surrounding the tumors. The expression rate of Spl in gastric cancer lesions was 53.85% (35/65). The medium survival duration in patients who had a tumor with negative, weak and strong Spl expressions was 1 700, 1 560 and 1 026 d, respectively (P<0.05). Spl protein expression was closely related to the depth of tumor infiltration (X2 = 13.223, P<0.01) and TNM stage (X2= 11.009, P<0.05), but had no relationship with the number of lymph nodes and Lauren's classification (P>0.05). Cox regression model for multivariate analysis revealed that high Spl expression (P<0.05) and advanced stage (P<0.01) were independent predictors of poor survival. CONCLUSION: Normal and malignant gastric tissues have unique Spl expression patterns. Spl might serve as an independent prognostic factor, by influencing the tumor infiltration and progression.
文摘AIM: To investigate the uptake of 99mTc-HYNIC-Tyr3-octreotide (99mTc-HYNIC-TOC) in human hepatocellular carcinoma (HCC), which can provide the localizable diagnosis in hepatic carcinoma. METHODS: The expression of somatostatin receptor 2 (SSTR2) messenger RNA (mRNA) in human HCC cell line HepG2 was examined by reverse transcriptase-polymerase chain reaction (RT-PCR). Uptake of 99mTc-HYNIC-TOC was evaluated in the human HCC implanted into BALB/c nude mice. ANMIS2000 nuclear medicine analysis system was used to calculate the ratio of 99mTc uptake between tumor tissue and vital organs. RESULTS: We demonstrated the expression of SSTR2 mRNA in human HCC cell line HepG2 by RT-PCR. The size of the RT-PCR products was 364 bp detected by sequence analysis of the human SSTR2 mRNA. Scintigraphy proved that 99mTc-HYNIC-TOC was uptaken in the tumor tissue, liver and kidney of the tumor-bearing mice. CONCLUSION: Based on expression of the SSTR2 mRNA in human HCC, 99mTc-HYNIC-TOC can markedly bind with and be uptaken by human HCC tissues as compared with normal liver tissue. The significant retention of radionudide in kidney and bladder is probably related to non-specific peptide uptake in the tubulus cells of kidney and possibly due to excretion by kidney. Our results show that localizable diagnosis and targeting radiotherapy with radionuclide-labeled somatostatin analog for HCC are of great value to be further studied.
文摘The characteristic of the precipitation and growth of α2 ordered phase during aging treatment in near α Ti alloys have been investigated in terms of the influences of aging temperature, aging time and aging manner. The results exhibit that aging temperatures influence the distribution of α2 phase precipitated and cause the changes in growth speed of α2 phase. For various aging temperatures, the time to finish precipitation of α2 phase is different. The facts that various distribution characteristics and growth speed of α2 ordered phase are caused by changed aging condition imply optimal selection and control for precipitation of α2 ordered phase reachable. Some discussions on adoptable aging steps are presented.
文摘The precipitation of ordered alpha_2 phase in a near - alphaTi-6.3Al-4.8Sn-2.0Zr-1.0Mo-0.34Si-0.9Nd (mass fraction) alloy with duplex microstructure, duringaging at various temperatures, was investigated. It is concluded that the precipitation and growthof the alpha_2 phase can be controlled by suitable selection of the aging temperature. Aging athigher temperatures can result in the uniform precipitation and growth of alpha_2 ordered phase inalpha_p whereas the alpha_2 ordered phase precipitated only at the lamella boundaries anddislocations in beta_t. Aging at a moderate temperature can promote the tendency of uniformprecipitation of alpha_2 phase in both alpha_p and beta_t. Aging at a relatively low temperature issuitable for the uniform precipitation and growth of fine alpha_2 particles throughout both thealpha_p and beta_t matrix.

文摘OBJECTIVE To investigate the effect of scutellarin(SCU),which is the main effective component of Erigeron breviscapus(Vant.)Hand-Mazz native to Yunnan in China,on vascular dysfunction(VD)of cardiac coronary artery(CA)and cerebral basilar artery(BA)caused by hypertension in SHR rats.METHODS 1.BA and CA vesselsrings from 40 weeks of SHR rats were isolated and equilibrated in organ bath with MOPS-PSS buffer and ring tension was recorded,comparing with a normal control of WKY rats.SCU was treated accumulatively after pre-contracted with vasoconstrictor U46619(1μmol·L-1).2.ACH and SNP were treated accumulatively after pre-incubation with SCU(300μmol·L-1,100μmol·L-1)and pre-contracted by U46619(1μmol·L-1),K+60mmol·L-1,respectively.While U46619 was added accumulatively to BA/CA rings pre-incubated with SCU(300and 100μmol·L-1).ACH-induced relaxation rate was to evaluate endothelium-dependent relaxation,and SNP-induced relaxation rate to evaluate the artery non-endothelium-dependent relaxation.RESULTS SCU significantly dilated both BA and CA rings pre-contracted by U46619 in old rats.EC50 value of SCU in WKY ratswas less than that in SHR(P<0.05),which showing VD of CA and BA in SHR rats.Compared with WKY group,ACH relaxation curve of SHR group shifted to the right,suggesting that hypertension induced VD.When SCU 300μmol·L-1 pre-treated CA groups and SCU 100μmol·L-1 pre-treated BA groups,EC50 to ACH was significantly lower(P<0.05).Likewise,the vasodilatation of CA/BA rings to SNP was also improved obviously when pre-treated with SCU,and Emax to SNP was decreased significantly(P<0.05).Moreover,EC50 to U46619was significantly lower when pre-treated by SCU.CONCLUSION In SHR rats,SCU antagonized U46619 on CA/BA rings in a noncompetitive manner.Furthermore,SCU should appear to protect VD induced by hypertension.
文摘The precipitation characteristics of the α2 ordered phase in Ti-AI-Sn-Zr-Mo-Si-Nd alloys with various content of Al, under different aging conditions, were investigated. The distribution and size of the α2 ordered phase changed with temperature and Al content. The dislocations were the only places where the α2 ordered phase could precipitate at higher temperature near the critical transformation temperature for each alloy experimented. With the addition of Al content, the critical transformation temperature of α2 ordered phase increased. When the aging temperature was relatively low (650℃), the precedent precipitation of α2 ordered phase took place in primary a phase at the early stage of aging, in the duplex microstructure (the primary a with the transformed (3) of the alloys with lower Al content. But after certain aging time (50 h), the size of α2 particles was almost equal in both the primary a and the transformed β. And no obvious growth of α2 particles could be observed after 50 h.

文摘Some experimental α+α2 alloys were prepared by the addition of tin or aluminum elements into Ti-55 alloy. These alloys were designed with varied electron concentration values and named as Sn-rich alloys and Al-rich alloys, respectively. The precipita- tion and growth of α2 ordered phase in the tested alloys under various heat treatment conditions were investigated. Some compari- sons among the experimental results were performed and discussed in detail. Stronger precipitation and growth of α2 ordered phase were caused in Al-rich alloys but relatively weak change in Sn-rich alloys with increasing the electron concentration. The precipita- tion of α2 ordered phase in Al-rich alloys is stronger than that in Sn-rich alloys when the electron concentration value is the same for the two alloys.
