Three fluid-structure interaction numerical methods including Fluid FEM and Fluid BEM were used in this case. The comparison of numerical and theoretical results by the present method shows that ANSYS/Fluid80 is more credible, the NASTRAN/Virtual Mass Method is more suitable for engineering calculations and results of the same-phase vibration by the present method is more accurate.展开更多
Moderate leaf rolling is one of the most important morphological traits in rice breeding for plant ideotype.Previous studies have shown that the rl(t)gene has a high breeding potential for developing hybrid-rice varie...Moderate leaf rolling is one of the most important morphological traits in rice breeding for plant ideotype.Previous studies have shown that the rl(t)gene has a high breeding potential for developing hybrid-rice varieties with an ideal ideotype,because it leads to an appropriate leaf rolling index(LRI)of about 30%in the heterozygous state,and had a positive effect on grain yield.In this study,we isolated rl(t)and performed a preliminary investigation of its function in regulating leaf rolling in rice.DNA sequencing identified a single base change(G to T)in the finely mapped region(11 kb)containing rl(t),and this is located in 30-untranslated region(30-UTR)of the only predicted gene,Roc5(Rice outermost cell-specific).The expression level of Roc5 is significantly higher in the rl(t)mutant than in the wild-type.Using RNAi and overexpression analysis,we found that the expression level of Roc5 correlated with LRI and leaf bulliform area,and wasalso associated with leaf abaxial or adaxial rolling.These results confirmed that Roc5 controls leaf rolling in a dosage-dependent manner.Bioinformatics analysis revealed a conserved 17-nt sequence(called the GU-rich element)in the 30-UTR of HD-GL2(Homeodomain-Glabra2)family genes including Roc5.Based on the model of this element in regulating mRNA stability in mammals,we speculate that the single nucleotide change in this element accounts for the higher expression level of Roc5 in the rl(t)mutant compared to the wild-type,which ultimately leads to adaxial rolling of the leaf.This discovery further enhances our knowledge of the molecular mechanisms underlying leaf rolling in rice.展开更多
Meiotic recombination is essential for reciprocal exchange of genetic information between homologous chromosomes and their subsequent proper segregation in sexually reproducing organisms. MLH1 and MLH3 belong to meios...Meiotic recombination is essential for reciprocal exchange of genetic information between homologous chromosomes and their subsequent proper segregation in sexually reproducing organisms. MLH1 and MLH3 belong to meiosis-specific members of the Mut L-homolog family, which are required for normal level of crossovers(COs) in some eukaryotes. However, their functions in plants need to be further elucidated.Here, we report the identification of Os MLH1 and reveal its functions during meiosis in rice. Using CRISPRCas9 approach, two independent mutants, Osmlh1-1 and Osmlh1-2, are generated and exhibited significantly reduced male fertility. In Osmlh1-1, the clearance of PAIR2 is delayed and partial ZEP1 proteins are not loaded into the chromosomes, which might be due to the deficient in resolution of interlocks at late zygotene. Thus, Os MLH1 is required for the assembly of synapsis complex. In Osmlh1-1, CO number is dropped by ~53% and the distribution of residual COs is consistent with predicted Poisson distribution,indicating that Os MLH1 is essential for the formation of interference-sensitive COs(class I COs). Os MLH1 interacts with Os MLH3 through their C-terminal domains. Mutation in Os MLH3 also affects the pollen fertility. Thus, our experiments reveal that the conserved heterodimer Mut Lg(Os MLH1-Os MLH3) is essential for the formation of class I COs in rice.展开更多
Intersections appear where one street crosses another,acting as fundamental nodes embedded in the network of urban public space.Though discussed in configurationa and perceptive studies,limited attention has been paid...Intersections appear where one street crosses another,acting as fundamental nodes embedded in the network of urban public space.Though discussed in configurationa and perceptive studies,limited attention has been paid to the morphological aspects of inter.sections.This study proposes and tested a new approach to modeling the urban intersection form.First,effective cylinder was introduced to define the related space for each intersection based on its scale.Second,a set of morphological indicators were presented to measure the physical properties of the intersection form.Third,the affinity propagation algorithm was used to examine the patterns of intersection form.Using the Old City of Nanjing,China as the study area,844 intersections were analyzed to test this new method.As a result,we were able to classify the intersections into eight types.This study shows that the intersection can be modelled as a volumetric and integral spatial unit of urban form,which may demand more attention from urban designers and architects in the future for shaping the built environment.The quantitative nature of our method could also open more possibilities for intersection-based studies.展开更多
