Objective:To study the expression of TRPC6 among prostate cancer cells,establish high expression cell lines of TRPC6,and to provide potential cell mode for prostate cancer oncogenesis and development.Methods:Occurrenc...Objective:To study the expression of TRPC6 among prostate cancer cells,establish high expression cell lines of TRPC6,and to provide potential cell mode for prostate cancer oncogenesis and development.Methods:Occurrence and development of prostate cancer cells,PC3,PC—3m DU145,22 rvl,LNCaP and normal prostate epithelial cells in the PrEC TRPC6 expression level were detected by QPCR method.Calcium phosphate transfection method was used to package retrovirus pLEGFP-Nl-TRPC6 and pLEGFP-Nl-vector and infect the prostate cancer cells,a stable high expression of TRPC6 prostate cancer cells.Sable cell lines of TRPC6,matrix metalloproteinase(MMP)2,MMP9 expression was detected by QPCR and Western blot.Change of cell invasion ability was detected by Transwell.Results:The expression level of prostate cancer cells TRPC6 were higher than control group PrEC cells.Among TPRC6 the expression of cell line PC 3 transfer potential wre the lowest,and high transfer cell tone PC-3M express was the highest.Real-time fluorescent quantitative PCR and western blot results showed that after filter,the seventh generation of cell TRPC6 protein and mRNA expression levels were higher than the control group obviously.Transwell experimental results showed that the overexpression of TRPC6could promote the invasion ability of PC.3 prostate cancer cells.Conclusions:TRPC6 expressed in prostate cancer cells is in disorder,and its action may be associated with the invasion and metastasis of prostate cancer cells;successful establishment of stable high expression of TRPC6prostate cancer cells primarily confirm the invasion-trigger ability of TRPC6 on prostate cancer,and lay down the foundation for exploring the TRPC6's role in the occurrence and development of prostate cancer展开更多

Based on the theory of approximati...Polynomial-basis response surface method has some shortcomings for truss structures in structural optimization,concluding the low fitting accuracy and the great computational effort. Based on the theory of approximation, a response surface method based on Multivariate Rational Function basis(MRRSM) is proposed. In order to further reduce the computational workload of MRRSM, focusing on the law between the cross-sectional area and the nodal displacements of truss structure, a conjecture that the determinant of the stiffness matrix and the corresponding elements of adjoint matrix involved in displacement determination are polynomials with the same order as their respective matrices, each term of which is the product of cross-sectional areas, is proposed. The conjecture is proved theoretically for statically determinate truss structure, and is shown corrected by a large number of statically indeterminate truss structures. The theoretical analysis and a large number of numerical examples show that MRRSM has a high fitting accuracy and less computational effort. Efficiency of the structural optimization of truss structures would be enhanced.展开更多
Background:Whether the time course of post-stroke depression (PSD) can be used to predict the quality of life (QoL) of patients with late-stage stroke remains unclear,this study investigated whether persistent depress...Background:Whether the time course of post-stroke depression (PSD) can be used to predict the quality of life (QoL) of patients with late-stage stroke remains unclear,this study investigated whether persistent depression at 1 year after stroke predicts QoL at 5 years following stroke.Methods:We analyzed the demographic and clinical data of patients with stroke in 56 hospitals across China that participated in the Prospective Cohort Study on the Incidence and Outcome of Patients with PSD in China Study.Follow-up assessments were performed at the following time points after stroke:in person,2 weeks,3 months,6 months,and 1 year;by telephone,5 years.National Institutes of Health Stroke Scale (NIHSS) score on admission,recurrence,disability,depression,QoL,and chronic complications were recorded.Depression was diagnosed using the Diagnostic and Statistical Manual of Mental Disorders.QoL was measured using short form-12 (SF-12).Multivariable ordinal logistic regression analysis was used to identify factors that independently affected the physical component summary (PCS) and mental component summary (MCS) scores of the SF-12.Results:Of the 801 patients evaluated in this study,80 had persistent depression.The multivariable regression analysis of data obtained at 5 years showed that persistent depression at 1 year (odds ratio [OR]:0.48;95% confidence interval [CI]:0.29-0.81) and disability at 5 years (OR:0.34;95% CI:0.23-0.49) were associated with poor MCS scores at 5 years.Old age,a high NIHSS score on admission,disability at 5 years,and stroke recurrence within 5 years were associated with poor PCS scores at the 5-year follow-up.Conclusions:Persistent depression at the 1-year follow-up could predict poor MCS scores at the 5-year follow-up.The development of interventional strategies targeting post-stroke patients with persistent depression is warranted.展开更多
Objective Insomnia is a highly prevalent disorder among patients suffering from stroke.The association between insomnia and stroke mortality is less studied,particularly using the latest diagnostic criteria.The curren...Objective Insomnia is a highly prevalent disorder among patients suffering from stroke.The association between insomnia and stroke mortality is less studied,particularly using the latest diagnostic criteria.The current study examined the relationship between insomnia and mortality among patients with first-evonal hazard models were used to calculate HRs for stroke er stroke in China.Methods Patients with acute cerebrovascular diseases(stroke)were recruited from 56 hospitals in China's Mainland.Insomnia was defined as difficulty falling asleep,or difficulty staying asleep or waking up early,for at least two consecutive visits.Demographic data,medical history and clinical data were collected.Four follow-up visits occurred within the first year after stroke,and the last follow-up call was conducted 6 years later.Cox proportional hazard models were used to calculate HRs for stroke mortality.results Insomnia was reported by 38.4%(489/1273)of patients at baseline.During the 6 years of follow-up,after adjusting for all confounders,insomnia was found to be associated with increased mortality(HR=1.66,95%CI 1.10 to 2.48).Old age(HR=1.08,95%CI 1.06 to 1.10),stroke recurrence in the first year of follow-up(HR=2.53,95%CI 1.48 to 4.31)and stroke survivors with hypertension(HR=1.62,95%CI 1.04 to 2.53)had substantially higher risk of mortality.Conclusions Besides old age,stroke recurrence in the first year of follow-up and hypertension,insomnia is associated with increased risk of mortality in patients with first-ever stroke in China.More studies about prompt and efficient interventions for insomnia are expected in the future.Trial registration number rctn62169508.展开更多
Objective:To investigate the therapeutic effect of moxibustion on sarcomas from mesenchymal tissues,which have a low response rate to chemotherapy and radiotherapy.Methods:S180 sarcoma cell line was inoculated in C57 ...Objective:To investigate the therapeutic effect of moxibustion on sarcomas from mesenchymal tissues,which have a low response rate to chemotherapy and radiotherapy.Methods:S180 sarcoma cell line was inoculated in C57 BL/6 mice to form transplanted tumor.Moxibustion therapy was directly applied at the transplanted tumor sites,at a distance of 3.0 cm,10 min per session,till skin temperature reached 45°C,once a day,for 14 consecutive days of intervention.After the mice were killed,serum was collected and used to detect concentrations of interleukin-10(IL-10),transforming growth factor-b1(TGF-b1),IL-4 and interferon-c(IFN-c)by Luminex liquid suspension chip.The numbers of Treg^(^(+) )T cells and CD4^(+) CD25^(+) Forkhead Box P3(Foxp3)^(+) T cells were detected by flow cytometry.Fluorescence in situ hybridization was used to analyze the changes of CD4,CD8,Foxp3 and TGF-b1 in the tumor microenvironment(TME).Results:Weight of S180 transplanted tumor in the control group was(2.03±0.54)g,and that in the moxibustion group was(1.27±0.29)g,which was statistically different(P=0.023).The mean value of Foxp3^(+) T cells in the normal group was 2.01%,which increased to 3.63%after the formation of transplanted tumor,and decreased to 1.48%after moxibustion treatment.The moxibustion group also had reduced numbers of CD4^(+) CD25^(+) Foxp3^(+) T cells in the spleen of mice with transplanted tumor.The concentrations of IL-10,TGF-b1 and IL-4 decreased in the serum of mice with transplanted tumor,while the concentration of IFN-c increased.Moxibustion was associated with downregulation in expression of Foxp3,IL-10 and TGF-b1 genes in the transplanted tumor,and increases in the gene expression of CD4^(+) T cells and CD8^(+) T cells in the TME.Conclusion:Moxibustion may have therapeutic effects on sarcomas by reducing the number of Treg cells in the blood and controlling the infiltration of Treg cells in the TME.展开更多
Objective:We investigated the influence of β-receptor blocker metoprolol on return of spontaneous circulation(ROSC) during cardiopulmonary resuscitation(CPR) in rats with induced myocardial infarction(MI).Methods:Mal...Objective:We investigated the influence of β-receptor blocker metoprolol on return of spontaneous circulation(ROSC) during cardiopulmonary resuscitation(CPR) in rats with induced myocardial infarction(MI).Methods:Male Sprague-Dawley rats were randomly divided into three groups:the sham-operated group,the MI group without metoprolol,which was fed the vehicle,and the MI+metoprolol group receiving intragastric metoprolol.Each group was further divided randomly into three subgroups,depending on the dosage of epinephrine administered during subsequent CPR applied after the induction of asphyxial cardiac arrest.Results:The ROSC rate was significantly decreased in the low dose subgroup of MI group,unchanged in the medium dose subgroup of MI group,and significantly decreased in the high dose subgroup of MI group,compared with the same dose subgroup of sham-operated group.MI+metoprolol group had a lower ROSC rate than MI group in the medium dose subgroup,and a higher ROSC rate than MI group in the high dose subgroup.There was no difference in blood K+ values of successful rats between MI group and MI+metoprolol group.The rats with successful CPR had lower blood K+ values than rats with unsuccessful CPR in each of the three treatment groups.Conclusions:Metoprolol administered to MI rats over a long period significantly improved ROSC rates under an appropriate dose of epinephrine during CPR.An increasing high blood K+ value would attenuate the rate of a successful CPR.展开更多
