Inhibiting the death receptor 3(DR3)signaling pathway in group 3 innate lymphoid cells(ILC3s)presents a promising approach for promoting mucosal repair in individuals with ulcerative colitis(UC).Paeoniflorin,a prominent component of Paeonia lactiflora Pall.,has demonstrated the ability to restore barrier function in UC mice,but the precise mechanism remains unclear.In this study,we aimed to delve into whether paeoniflorin may promote intestinal mucosal repair in chronic colitis by inhibiting DR3 signaling in ILC3s.C57BL/6 mice were subjected to random allocation into 7 distinct groups,namely the control group,the 2%dextran sodium sulfate(DSS)group,the paeoniflorin groups(25,50,and 100 mg/kg),the anti-tumor necrosis factor-like ligand 1A(anti-TL1A)antibody group,and the IgG group.We detected the expression of DR3 signaling pathway proteins and the proportion of ILC3s in the mouse colon using Western blot and flow cytometry,respectively.Meanwhile,DR3-overexpressing MNK-3 cells and 2%DSS-induced Rag1^(-/-)mice were used for verification.The results showed that paeoniflorin alleviated DSS-induced chronic colitis and repaired the intestinal mucosal barrier.Simultaneously,paeoniflorin inhibited the DR3 signaling pathway in ILC3s and regulated the content of cytokines(interleukin-17A,granulocyte-macrophage colony stimulating factor,and interleukin-22).Alternatively,paeoniflorin directly inhibited the DR3 signaling pathway in ILC3s to repair mucosal damage independently of the adaptive immune system.We additionally confirmed that paeoniflorin-conditioned medium(CM)restored the expression of tight junctions in Caco-2 cells via coculture.In conclusion,paeoniflorin ameliorates chronic colitis by enhancing the intestinal barrier in an ILC3-dependent manner,and its mechanism is associated with the inhibition of the DR3 signaling pathway. 展开更多

Immune checkpoint inhibitors(ICIs)constitute a pivotal class of immunotherapeutic drugs in cancer treatment.However,their widespread clinical application has led to a notable surge in immune-related adverse events(irAEs),significantly affecting the efficacy and survival rates of patients undergoing ICI therapy.While conventional hematological and imaging tests are adept at detecting organ-specific toxicities,distinguishing adverse reactions from those induced by viruses,bacteria,or immune diseases remains a formidable challenge.Consequently,there exists an urgent imperative for reliable biomarkers capable of accurately predicting or diagnosing irAEs.Thus,a thorough review of existing studies on irAEs biomarkers is indispensable.Our review commences by providing a succinct over-view of major irAEs,followed by a comprehensive summary of irAEs biomarkers across various dimensions.Furthermore,we delve into innovative methodologies such as machine learning,single-cell RNA sequencing,multiomics analysis,and gut microbiota profiling to identify novel,robust biomarkers that can facilitate precise irAEs diagnosis or prediction.Lastly,this review furnishes a concise exposition of irAEs mechanisms to augment understanding of irAEs prediction,diagnosis,and treatment strategies. 展开更多

A Problem Not to Be Ignored: The Influencing Factors of Mobile Phone Addiction and Its Influence on Sleep Quality

Background and Objective: With the popularity and widespread use of mobile phones, the effects of mobile phone dependence and addiction on individuals’ physical and mental health have attracted more and more attention. The present study aims to analyze the current state of mobile phone addiction and its impact on sleep quality within the population, while also exploring the influence of related factors on sleep quality. Ultimately, this research will provide a scientific foundation for targeted intervention measures and strategies. Methods: A total of 253 permanent residents in Nanjing were randomly selected as study subjects. The Mobile Phone Addiction Index (MPAI) and Pittsburgh Sleep Quality Index (PSQI) were used to evaluate the degree of smartphone addiction and sleep quality of the study subjects. Body mass index (BMI) was measured according to standardized procedures. Independent sample t-test, Chi-square test, rank sum test and multiple linear regression were used to analyze the correlation between mobile phone addiction and sleep quality, and P Results: 117 people (46.2%) were addicted to mobile phones. Chi-square test showed that the rate of mobile phone addiction in drinking group was significantly higher than that in non-drinking group (P P P P P P P P P P Conclusion: Mobile phone addiction may lead to shorter sleep duration and reduce sleep efficiency. The withdrawal of mobile phone addiction may have a negative impact on sleep quality. According to the characteristics of the population, appropriate comprehensive intervention measures should be taken to build an effective evaluation system, so as to reduce the impact of mobile phone addiction and withdrawal problems on sleep and improve sleep quality. 展开更多

详细评估成岩作用对碳酸岩锶同位素组成以及元素含量的影响是应用其揭示古环境变化的重要前提。本文探究来自美国Monitor Range的一个晚奥陶世剖面碳酸岩^(87)Sr/^(86)的主要控制因素。结果显示,在生物带D.ornatus的中上部,^(87)Sr/^(86... 详细评估成岩作用对碳酸岩锶同位素组成以及元素含量的影响是应用其揭示古环境变化的重要前提。本文探究来自美国Monitor Range的一个晚奥陶世剖面碳酸岩^(87)Sr/^(86)的主要控制因素。结果显示,在生物带D.ornatus的中上部,^(87)Sr/^(86)从0.70794升高至0.70830,显著高于已发表的同时期样品的比值(~0.7079)。这一异常高的^(87)Sr/^(86)比值指示了后期、局部过程对研究剖面碳酸岩原始锶同位素信息的改变,因而不能代表同时期海水的锶同位素组成。另外,^(87)Sr/^(86)与成岩作用指标(Mn、Fe含量,Mn/Sr与Fe/Sr比值)呈显著的正相关关系,表明成岩蚀变是所测异常^(87)Sr/^(86)的主控因素。通过成岩流体-岩石反应的数值模型计算,再次证实所测的锶同位素以及元素数据可用成岩作用造成的碳酸岩化学组成变化进行很好的解释。本文结果表明,即使碳酸岩样品满足“[Sr]>300 ppm,[Mn]<300 ppm,[Fe]<1000 ppm,Mn/Sr<0.2,Fe/Sr<1.6”这一严苛的锶同位素保存评判标准,后期成岩作用仍可能显著改变其原始锶同位素组成,提示在应用碳酸岩^(87)Sr/^(86)示踪古环境变化时需更加注意成岩作用的影响。 展开更多

利用STMAS-WRF模式,对宁波鄞州风电场2021年12月23-26日的一次强冷空气大风过程进行数值模拟试验,并采用风机的观测资料进行误差检验,对此次大风过程的模式模拟能力进行评估。结果表明,STMAS-WRF模式对此次强冷空气大风过程具有较好的... 利用STMAS-WRF模式,对宁波鄞州风电场2021年12月23-26日的一次强冷空气大风过程进行数值模拟试验,并采用风机的观测资料进行误差检验,对此次大风过程的模式模拟能力进行评估。结果表明,STMAS-WRF模式对此次强冷空气大风过程具有较好的模拟能力,基本模拟出了风速随时间的变化,相关系数均达到0.8以上,均方根误差为2.83~3.86 m·s^(-1),平均绝对百分比误差为21.8%~29.8%。模拟结果表现出了冷空气影响过程中风速减弱增强再减弱的过程,但是对于风速减弱时段大幅度波动的风速,模式模拟普遍还存在偏大的现象。从不同风速段来看,模式对于小风速段和额定风速段的模拟效果较好,对变化风速段的模拟误差较大。通过对预报场和实况场环流的对比分析,发现模式对高层风速模拟效果较好,低层风速预报较实况偏大,可能与模式对于地面摩擦的影响考虑不足有关。 展开更多

Cadmium(Cd)accumulation in rice grain is of health concern.Identifying genes involved in grain Cd accumulation and performing molecular breeding may reduce it.In this study,knockout of OsNRAMP2,a member of the NRAMP family,reduced grain Cd concentrations by more than 38%,and overexpressing OsNRAMP2 increased grain Cd concentrations by more than 50%.Physiological experiments showed that OsNRAMP2 facilitated Cd translocation from root to shoot by positively regulating Cd efflux from the vacuoles.At filling stage,OsNRAMP2 was highly expressed in all tissues except for husk,suggesting its role in Cd remobilization.Changes in OsNRAMP2 expression affected the concentrations of Fe,Mn,Zn,and Cu in grain and also affected rice growth.Phylogenetic analysis showed that the distribution of OsNRAMP2 haplotypes between japonica and indica was different.Among the four haplotypes of OsNRAMP2,Hap 1,with a 6-bp nucleotide insertion in exon 1,had grain Cd concentration at least 45.3%lower than any of the other three haplotypes.Almost all(99.3%)japonica accessions but rare indica accessions(4.44%)from the 3K sequenced rice genomes carry Hap 1 of OsNRAMP2.Our study sheds light on the molecular mechanism of grain Cd accumulation and provides a promising target for low-Cd rice breeding. 展开更多

A new titanium alloy Ti12.5Zr2.5Nb2.5Ta(TZNT) for surgical implant application was synthesized and fully annealed at 700℃for 45 min.The microstructure and the mechanical properties such as tensile properties and fatigue properties were investigated.The results show that TZNT mainly consists of a lot of lamellaα-phase clusters with different orientations distributed in the originalβ-phase grain boundaries and a small amount ofβphases between the lamella a phases.The alloy exhibits better ductility,lower modulus of elasticity,and lower admission strain in comparison with Ti6A14V and Ti6A17Nb,indicating that it has better biomechanical compatibility with human bones.The fatigue limit of TZNT is 333 MPa,at which the specimen has not failed at 10^7 cycles.A large number of striations present in the stable fatigue crack propagation area,and many dimples in the fast fatigue crack propagation area are observed,indicating the ductile fracture of the new alloy. 展开更多

Hot deformation behaviors and microstructure evolution of Ti-3Al-5Mo-4Cr-2Zr-1Fe(Ti-35421)alloy in theβsingle field are investigated by isothermal compression tests on a Gleeble-3500 simulator at temperatures of 820-900°C and strain rates of 0.001-1 s^(-1).The research results show that discontinuous yield phenomenon and rheological softening are affected by the strain rates and deformation temperatures.The critical conditions for dynamic recrystallization and kinetic model of Ti-35421 alloy are determined,and the Arrhenius constitutive model is constructed.The rheological behaviors of Ti-35421 alloys aboveβphase transformation temperature are predicted by the constitutive model accurately.The EBSD analysis proves that the deformation softening is controlled by dynamic recovery and dynamic recrystallization.In addition,continuous dynamic recrystallization is determined during hot deformation,and the calculation model for recrystallization grain sizes is established.Good linear dependency between the experimental and simulated values of recrystallized grain sizes indicates that the present model can be used for the prediction of recrystallized grain size with high accuracy. 展开更多

The primary crystallization of the Ti40Zr25NisCu9Bc18 amorphous alloy was studied by isochronal differential scanning calorimetry (DSC). The activation energy was determined by the Kissinger-Akahim-Sunose method. Trying to analyze the crystallization kinetics of the Ti40Zr25NigCu9Be18 amorphous alloy by two different methods, it was found that the crystallization kinetics did not obey the Johnson-Mehl-Avrami equation. A modified method in consideration of the impingement effect was proposed to perform kinetic analysis of the isochronal crystallization of this alloy. The kinetic parameters were then obtained by the linear fitting method based on the modified kinetic equation. The results show that the isochronal crystallization kinetics of the amorphous Ti40Zr25Ni8CugBe18 alloy is heating rate dependent, and the discrepancy between the Johnson-Mehl-Avrami method and the modified method increases with the increase of heating rate. 展开更多

The endosperm plays essential roles in embryogenesis and seed germination and provides abundant resources for human food and industrial products. Identification of genes regulating the development of the endosperm and elucidation of their functions is of great importance for maize genetics and breeding. This study showed that the genespecific imprinted gene, ETHYLENE-INSENSITIVE 2-like(EIN2-like), is maternally expressed in both endosperm and embryo. The maternally expressed pattern was maintained throughout later seed developmental stages. Bisulfite sequencing using DNA obtained from hybrid endosperm tissues showed that the upstream regions of the alleles of EIN2-like were highly methylated at symmetrical sites(CG and CHG). A differentially methylated region in the upstream part of the maternal allele of EIN2-like was identified and found to be hypomethylated. Expression analysis showed that EIN2-like was highly expressed in the maize endosperm as well as at different stages of cell differentiation(8–12 days after pollination) in the hybrid endosperm. These results suggest that the maternally expressed gene EIN2-like may play crucial roles in the regulation of seed development. 展开更多

Mechanical signal capture without physical contact has emerged as a highly promising research field and attracted tremendous attention due to its prosperous applications in household medical care,lifestyle monitoring and remote operation,offering users high level of safety,convenience and comfort.Moreover,noncontact sensing is ideal to maximize the immersive user experience in the human–machine interaction(HMI),eliminating interference to human activities and mechanical fatigue to the sensor,simultaneously.Herein,we report a self-powered flexible sensor integrated with irradiation cross-linked polypropylene(IXPP)piezoelectret film for noncontact sensing,featuring multi-functions to detect mechanical signals transmitted through solid,liquid and gaseous media and would facilitate their versatile practical applications.The folded-structure configuration of the sensor facilitates the improvement of the noncontact sensing sensitivity.For solid media,such as the rectangular wooden stick used in this study,the sensor can detect mechanical stimulus exerted at a distance of 100 cm.A system detection sensitivity up to 57 pC/kPa with a low detection limit of 0.6 kPa is achieved at a noncontact distance of 10 cm.Even when partly or completely immersed in water,the sensor effectively traces movement signals of human bodies underwater,demonstrating great advantages for non-inductive aquatic fitness training monitoring.Furthermore,due to the low acoustic impedance of piezoelectret film,speech recognition through gaseous medium is also achieved.We further introduce application demonstrations of the developed film sensors to monitor exercise postures and physiological signals without direct contact between human body and the sensor,displaying great potential to be incorporated into future smart electronics.This study commendably expands the application scope of piezoelectret materials,which will have profound implications for exploring novel intelligent human–machine interactions. 展开更多

The mid-infrared optical frequency comb is a powerful tool for gas sensing.In this study,we demonstrate a simple midinfrared dual-comb spectrometer covering 3±4μm in Li Nb O_(3)waveguides.Based on a low-power fiber laser system,the mid-infrared comb is achieved via intra-pulse difference frequency generation in the Li Nb O_(3)waveguide.We construct pre-chirp management before supercontinuum generation to control spatiotemporal alignment for pump and signal pulses.The supercontinuum is directly coupled into a chirped periodically poled Li Nb O_(3)waveguide for the 3±4μm idler generation.A mid-infrared dual-comb spectrometer based on this approach provides a 100 MHz resolution over25 THz coverage.To evaluate the applicability for spectroscopy,we measure the methane spectrum using the dualcomb spectrometer.The measured results are consistent with the HITRAN database,in which the root mean square of the residual is 3.2%.This proposed method is expected to develop integrated and robust mid-infrared dual-comb spectrometers on chip for sensing. 展开更多

Pseudomonas aeruginosa is a ubiquitous and metabolically versatile microorganism naturally found in soil and water.It is also an opportunistic pathogen in plants,insects,animals,and humans.In response to increasing cell density,P.aeruginosa uses two acylhomoserine lactone(AHL)quorum-sensing(QS)signals(i.e.,N-3-oxo-dodecanoyl homoserine lactone[3-oxo-C12-HSL]and Nbutanoyl-homoserine lactone[C4-HsL]),which regulate the expression of hundreds of genes.However,how the biosynthesis of these two QS signals is coordinated remains unknown.We studied the regulation of these two QS signals in the rhizosphere strain PA1201.PA1201 sequentially produced 3-oxo-C12-HSL and C4-HSL at the early and late growth stages,respectively.The highest 3-oxo-C12-HSL-dependent elastase activity was observed at the early stage,while the highest C4-HSL-dependent rhamnolipid production was observed at the late stage.The atypical regulator RsaL played a pivotal role in coordinating 3-oxo-C12-HSL and C4-HSL biosynthesis and QS-associated virulence.RsaL repressed las/transcription by binding the-10 and-35 boxes of the lasl promoter.In contrast,RsaL activated rhll transcription by binding the region encoding the 5'-untranslated region of the rhll mRNA.Further,RsaL repressed its own expression by binding a nucleotide motif located in the-35 box of the rsaL promoter.Thus,RsaL acts as a molecular switch that coordinates the sequential biosynthesis of AHL QS signals and differential virulence in PA1201.Finally,C4-HSL activation by RsaL was independent of the Las and Pseudomonas quinolone signal(PQS)QS signaling systems.Therefore,we propose a new model of the QS regulatory network in PA1201,in which RsaL represents a superior player acting at the top of the hierarchy. 展开更多

We demonstrate an all-polarization-maintaining(APM)fiber mode-locked laser based on nonlinear polarization evolution(NPE).A well-designed Sagnac fiber loop is employed to establish the nonlinear polarization evolution process in a polarization beam splitter(PBS)figure-8 fiber laser.Nonlinear loss curves are calculated to verify the saturable absorption characteristic of this NPE-based APM oscillator.Then,we simulate the pulse propagation process in the cavity to demonstrate the pulse mode-locked formation.Finally,we also design a realizable compact scheme to further reduce noise disturbances,achieving a 101-fs mode-locked pulse train with a 0.3-mrad integrated phase noise and a 0.006%integrated relative intensity noise(RIN).This figure-8 fiber laser provides a new scheme for compacting low-noise compact APM fiber lasers based on the NPE mode-locked mechanism. 展开更多

Protein citrullination,including histone H1 and H3 citrullination,is important for transcriptional regulation,DNA damage response,and pluripotency of embryonic stem cells(ESCs).Tripartite motif containing 28(Trim28),an embryonic development regulator involved in ESC self-renewal,has recently been identified as a novel substrate for citrullination by Padi4.However,the physiological functions of Trim28 citrullination and its role in regulating the chromatin structure and gene transcription of ESCs remain unknown.In this paper,we show that Trim28 is specifically citrullinated in mouse ESCs(m ESCs),and that the loss of Trim28 citrullination induces loss of pluripotency.Mechanistically,Trim28 citrullination enhances the interaction of Trim28with Smarcad1 and prevents chromatin condensation.Additionally,Trim28 citrullination regulates m ESC pluripotency by promoting transcription of Nanog and Klf4 which it does by increasing the enrichment of H3K27ac and H3K4me3 and decreasing the enrichment of H3K9me3 in the transcriptional regulatory region.Thus,our findings suggest that Trim28citrullination is the key for the epigenetic activation of pluripotency genes and pluripotency maintenance of ESCs.Together,these results uncover a role Trim28 citrullination plays in pluripotency regulation and provide novel insight into how citrullination of proteins other than histones regulates chromatin compaction. 展开更多
