Based o...Microorganisms are fundamental for the functioning of marine ecosystems and are involved in the decomposition of organic matter, transformation of nutrients and circulation of biologically-important chemicals. Based on the complexity of the natural geographic characteristics of the Changjiang River Estuary, the geographic distribution of sedimentary microorganisms and the causes of this distribution are largely unexplored. In this work, the surface sediment samples from the adjacent sea area of the Changjiang River Estuary were collected. Their prokaryotic diversity was examined by high-throughput sequencing technology, and the environmental factors of the bacterial community were investigated. The results indicated that the distribution of prokaryotic communities in the sediments of the study areas showed obvious spatial heterogeneity. The sampling sequences divided the sample regions into three distinct clusters. Each geographic region had a unique community structure, although Proteobacteria, Bacteroidota, Desulfobacterota, Acidobacteriota, and Actinobacteriota all existed in these three branches. Canonical correspondence analysis demonstrated that prokaryotic diversity and community distribution were significantly correlated with the geographic location of sediment, seawater depth, and in particular, nutrient content(e.g., total phosphorus, total organic carbon and dissolved oxygen). Moreover, it was found for the first time that the metal ions obviously affected the composition and distribution of the prokaryotic community in this area. In general, this work provides new insights into the structural characteristics and driving factors of prokaryotic communities under the background of the ever-changing Changjiang River Estuary.展开更多

In this work, we discuss the instability of THz plasma waves in the channel of FETs with spin and quantum effects under non-ideal boundary conditions. We obtain a linear dispersion relation by using the hydrodynamic equation, Maxwell equation and spin equation. The influence of source capacitance, drain capacitance, spin effects, quantum effects and channel width on the instability of THz plasma waves under the non-ideal boundary conditions is investigated in great detail. The results of numerical simulation show that the THz plasma wave is unstable when the drain capacitance is smaller than the source capacitance;the oscillation frequency with asymmetric boundary conditions is smaller than that under non-ideal boundary conditions;the instability gain of THz plasma waves becomes lower under non-ideal boundary conditions. This finding provides a new idea for finding efficient THz radiation sources and opens up a new mechanism for the development of THz technology.展开更多
Sesquiterpenes are the major pharmacodynamic components of agarwood,a precious traditional Chinese medicine obtained from the resinous portions of Aquilaria sinensis trees that form in response to environmental stress...Sesquiterpenes are the major pharmacodynamic components of agarwood,a precious traditional Chinese medicine obtained from the resinous portions of Aquilaria sinensis trees that form in response to environmental stressors.To characterize the sesquiterpene synthases responsible for sesquiterpene production in A.sinensis,a bioinformatics analysis of the genome of A.sinensis identifi ed six new terpene synthase genes,and 16 sesquiterpene synthase genes were identifi ed as type TPS-a in a phylogenetic analysis.The expression patterns for eight of the sesquiterpene synthase genes after treatment with various hormones or hydrogen peroxide were analyzed by real-time quantitative PCR.The results suggest that 100μM methyl jasmonate,ethephon,(±)-abscisic acid or hydrogen peroxide could be eff ective short-term eff ectors to increase the expression of sesquiterpene synthase genes,while 1 mM methyl salicylate may have long-term eff ects on increasing the expression of specifi c sesquiterpene synthase genes(e.g.,As-SesTPS,AsVS,AsTPS12 and AsTPS29).The expression changes in these genes under various conditions refl ected their specifi c roles during abiotic or biotic stresses.Heterologous expression of a novel A.sinensis sesquiterpene synthase gene,AsTPS2,in Escherichia coli produced a major humulene product,so AsTPS2 is renamed AsHS1.AsHS1 is diff erent from ASS1,AsSesTPS,and AsVS,for mainly producingα-humulene.Based on the predicted space conformation of the AsHS1 model,the small ligand molecule may bind to the free amino acid by hydrogen bonding for the catalytic function of the enzyme,while the substrate farnesyl diphosphate(FPP)probably binds to the free amino acid on one side of the RxR motif.Arg450,Asp453,Asp454,Thr457,and Glu461 from the NSE/DTE motif and D307 and D311 from the DDxxD motif were found to form a polar interaction with two Mg^(2+)clusters by docking.The Mg^(2+)-bound DDxxD and NSE/DTE motifs and the free RXR motif are jointly directed into the catalytic pocket of AsHS1.Comparison of the tertiary structural models of AsHS1 with ASS1 showed that they diff ered in structures in several positions,such as surrounding the secondary catalytic pocket,which may lead to diff erences in catalytic products.Based on the results,biosynthetic pathways for specifi c sesquiterpenes such asα-humulene in A.sinensis are proposed.This study provides novel insights into the functions of the sesquiterpene synthases of A.sinensis and enriches knowledge on agarwood formation.展开更多

Evasion of programmed cell death(apoptosis) is a hallmark of cancer cells and direct induction of apoptosis by targeting the pro-survival BCL2 family proteins represents a promising therapeutic strategy for cancer treatment. Therefore, understanding the molecular mechanisms underpinning cancer cell survival could provide a molecular basis for potential therapeutic interventions. Method: Here we explored the role of BCL2L1 and the encoded anti-apoptotic BCL-XL in GC. Using Droplet Digital PCR(ddPCR) technology to investigate the DNA amplification of BCL2L1 in GC samples and GC cell lines, the sensitivity of GC cell lines to selective BCL-XL inhibitors A1155463 and A1331852, pan-inhibitor ABT-263, and VHL-based PROTAC-BCL-XL was analyzed using(CellTiter-Glo) CTG assay in vitro. Western Blot(WB) was used to detect the protein expression of BCL2 family members in GC cell lines and the manner in which PROTAC-BCL-XL kills GC cells. Coimmunoprecipitation(Co-IP) was used to investigate the mechanism of A1331852 and ABT-263 kills GC cell lines. DDPCR, WB, and real-time PCR(RTPCR) were used to investigate the correlation between DNA, RNA, protein levels, and drug activity. Results: The functional assay showed that a subset of GC cell lines relies on BCL-XL for survival. In gastric cancer cell lines, BCL-XL inhibitors A1155463 and A1331852 are more sensitive than the pan BCL2 family inhibitor ABT-263, indicating that ABT-263 is not an optimal inhibitor of BCL-XL. VHL-based PROTAC-BCL-XL DT2216 appears to be active in GC cells. DT2216 induces apoptosis of gastric cancer cells in a time-and dose-dependent manner through the proteasome pathway. Statistical analysis showed that the BCL-XL protein level predicts the response of GC cells to BCL-XL targeting therapy and BCL2L1 gene CNVs do not reliably predict BCL-XL expression.Conclusion: We identified BCL-XL as a promising therapeutic target in a subset of GC cases with high levels of BCL-XL protein expression. Functionally, we demonstrated that both selective BCL-XL inhibitors and VHL-based PROTAC BCL-XL can potently kill GC cells that are reliant on BCL-XL for survival. However, we found that BCL2L1 copy number variations(CNVs) cannot reliably predict BCL-XL expression, but the BCL-XL protein level serves as a useful biomarker for predicting the sensitivity of GC cells to BCL-XL-targeting compounds. Taken together, our study pinpointed BCL-XL as potential druggable target for specific subsets of GC.展开更多
High quality Nb films were successfully prepared on both flexible polyimide(PI)and rigid Al2O3substrates and their transport properties were systematically studied at various applied currents,external magnetic fields,...High quality Nb films were successfully prepared on both flexible polyimide(PI)and rigid Al2O3substrates and their transport properties were systematically studied at various applied currents,external magnetic fields,and sample orientations.It is found that a curved Nb/PI film exhibits quite different superconducting transition and vortex dynamics compared to the flat Nb/Al2O3film.For the curved Nb/PI film,smooth superconducting transitions were obtained at low currents,while unexpected cascade structures were revealed in theρ(T)curves at high currents.We attribute this phenomenon to the gradient distribution of vortex density together with a variation of superconductivity along the curved film.In addition,reentrant superconductivity was induced in the curved Nb/PI thin film by properly choosing the measurement conditions.We attribute this effect to the vortex pinning from both in-plane vortices and out-of-plane vortices.This work reveals the complex transport properties of curved superconducting thin films,providing important insights for further theoretical investigations and practical developments of flexible superconductors.展开更多

Increased evidence has shown that hydrogen sulfide(H_(2)S),a novel gasotransmitter,could enhance drought resistance in plants by inducing stomatal closure,with concurrent enhancement of photosynthetic efficiency,but l...Increased evidence has shown that hydrogen sulfide(H_(2)S),a novel gasotransmitter,could enhance drought resistance in plants by inducing stomatal closure,with concurrent enhancement of photosynthetic efficiency,but little is known about the mechanism behind this contradictory phenomenon.This study examined the regulating mechanism of H_(2)S in response to drought stress fromstomatal and non-stomatal factors in Chinese cabbage.The results showed that exogenous H_(2)S could increase the accumulation of photosynthetic pigments and alleviate the damage caused by drought stress.It also regulated the expression in transcriptional level and the activity of ribulose 1,5-bisphosphate carboxylase/oxygenase(BrRuBisCO)under drought stress.The large subunit of BrRuBisCO was found to be modified by S-sulfhydration,which might be the reason for its increased enzyme activity.The fluxes of Cl^(−),K^(+),and H^(+)in the guard cells were detected by non-invasive micro-test techniques while under drought stress.The results indicated that H_(2)S signaling induced a transmembrane Cl^(−)and H^(+)efflux and inhibited K^(+)influx,and the Cl^(−)channel was the main responders for H_(2)S-regulated stomatal movement.In conclusion,H_(2)S signal not only activated the ion channel proteins located in the guard cell membrane to induce stomatal closure,but also regulated the transcriptional expression and the activity of RuBisCO,a non-stomatal factor to enhance the photosynthetic efficiency of leaves.There is therefore a beneficial balance between the regulation of H_(2)S signaling on stomatal factors and non-stomatal factors due to drought stress,which needs to be better understood to apply it practically to increase crop yields.展开更多
[Objectives]Based on UPLC-Q-TOF-MS/MS,network pharmacology and molecular docking techniques,the mechanism of Euphorbia peplus in the treatment of Alzheimer s disease(AD)was studied.[Methods]The UPLC-Q-TOF-MS/MS techni...[Objectives]Based on UPLC-Q-TOF-MS/MS,network pharmacology and molecular docking techniques,the mechanism of Euphorbia peplus in the treatment of Alzheimer s disease(AD)was studied.[Methods]The UPLC-Q-TOF-MS/MS technique was used to rapidly analyze the chemical components of E.peplus.Active components and potential targets of E.peplus were retrieved from TCMSP and Swiss Target Prediction database,and AD targets were screened using GeneCards database.The targets of E.peplus in the treatment of AD were obtained.The PPI network was constructed using String platform,and the network topology of Cytoscape software was used to compute and screen key targets,and the GO and KEGG pathway enrichment analysis was carried out in Metascape database to construct the"component-target-pathway-disease"network.Molecular docking was used to predict the binding properties of active ingredients and targets.[Results]The results of UPLC-Q-TOF-MS/MS showed that 83 compounds were identified from E.peplus,including 19 terpenoids,10 phenolic acids and phenols,16 flavonoids,2 phenylpropanoids,4 coumarins,1 alkaloid,1 anthraquinone and 30 other compounds.The results of network pharmacological analysis showed that 82 active ingredients were screened,and 279 common targets were identified for the treatment of AD,among which the key targets were ALB(albumin),GAPDH(glyceraldehyde triphosphate dehydrogenase),TNF(tumor necrosis factor),AKT1(serine/threonine protein kinase 1),and IL6(interleukin-6).KEGG enrichment analysis showed that key signaling pathways include cancer pathways,lipid and atherosclerosis,Alzheimer s disease,insulin resistance,serotonergic synapses,calcium signaling pathway,cAMP signaling pathway and other signaling pathways.Molecular docking results showed that 14-deoxyandrographolide,dehydroandrographolide,licochalcone B,apigenin and naringenin may be the key components of E.peplus in the treatment of AD.[Conclusions]The results suggest that E.peplus can be used to treat Alzheimer s disease through multi-component,multi-target and multi-pathway.展开更多
A rare large-scale hail process in Yantai and Weihai City in the autumn of 2021 was analyzed based on the surface meteorology,high-altitude observation and S-band radar detection data.The results showed that this proc...A rare large-scale hail process in Yantai and Weihai City in the autumn of 2021 was analyzed based on the surface meteorology,high-altitude observation and S-band radar detection data.The results showed that this process was influenced by shallow trough,and strong upper-level jet stream in the middle and upper levels guided the intrusion of mid-level cold air into the warm and humid environment at the lower level.The shear lines and trunk lines in the lower level also provided certain triggering conditions for uplift,brewing conditions suitable for the occurrence and development of strong convection.The vertical wind shear was unusually large,and vertical wind shear was up to 10 m/s at 0-1 km,20 m/s at 0-3 km,and 32 m/s at 0-6 km,which was conducive to the growth of the storm.In this process,isolated multi-cell thunderstorm was generated at the beginning,and then it developed and strengthened continuously,moving to the southeast.In the later stage,a convection cell was born at the boundary between Yantai and Weihai,and developed into supercell storm.The storm lasted for a long time,and continued to develop and strengthen as it moved eastward,affecting most areas of Weihai from northwest to southeast.In the development stage of the storm,the radar reflectivity factor had obvious characteristics of hook echo and three-body scatter spike,and there was echo overhang and other hail echo features in the middle layer.Weak mesocyclone appeared at a low elevation,and there was the potential for the occurrence of tornado.展开更多
<strong>Objective:</strong> To assess the association between circulating C-reactive protein (CRP), and CRP polymorphisms in the diesel engine exhaust (DEE)-exposed workers. <strong>Methods:</stro...<strong>Objective:</strong> To assess the association between circulating C-reactive protein (CRP), and CRP polymorphisms in the diesel engine exhaust (DEE)-exposed workers. <strong>Methods:</strong> In 137 DEE-exposed workers and 127 unexposed comparable control workers, six urinary mono-hydroxylated polycyclic aromatic hydrocarbons (OH-PAHs) and serum CRP levels were assayed. Genotyping of four CRP single-nucleotide polymorphisms (SNPs) was measured. <strong>Results: </strong>Serum CRP levels increased in exposed versus control workers (all p < 0.001). In the DEE-exposed workers, two CRP polymorphisms were associated with serum CRP levels, the subjects of rs1205 TT genotype had lower serum CRP levels (p < 0.05 compared to TC or CC). <strong>Conclusions: </strong>Our findings suggest that polymorphisms in CRP and circulating CRP involved in the inflammatory process may play significant roles in human sensitivity to lung function injury caused by DEE exposure. This study will help investigate the underlying mechanisms of adverse respiratory effects induced by DEE.展开更多

Methods: 120 breast cancer patients treated by dual-targeted therapy with macromolecular monoclonal antibodies were managed by a cloud platform from March to November 2019. Comprehensive online management included consultation about drugs and side effects and frequently asked questions in the dual-targeted therapy with macromolecular monoclonal antibodies. Results: In the consultation about drugs and side effects, there were five patients with fever, neutrophil, cough, and fatigue;24 with diarrhea;25 with nausea;11 with oral mucosal inflammation;10 with rashes and dry skin;8 with insomnia;and 1 with palpitation. Moreover, 110 patients with anxiety about the missed or delayed treatment were properly handled. Conclusion: The comprehensive online management of dual-targeted therapy with macromolecular monoclonal antibodies based on the cloud platform is helpful to satisfy the at-home breast cancer patients’ needs, ensure the continuity of dual-targeted therapy with macromolecular monoclonal antibodies for breast cancer patients, prevent misinformation, alleviate patients’ negative psychological emotions, and reduce patients’ economic losses. The online cloud platform integrated management model is crucial for managing patients with breast cancer treated by dual-targeted therapy.展开更多

The purpose of this study is to evaluate the role of manual lymphatic drainage (MLD) in breast cancer-related lymphedema treatment. Methods: The electronic databases of EMBASE, PubMed, Web of Science, and The Cochrane Library were searched to find English articles on MLD which were published before January 2020. After two evaluators selected the studies and independently evaluated literature quality, meta-analysis was carried out with RevMan 5.3 software. The outcome index of lymphedema treatment changed in edema volume. Results: The study included six RCTs of 364 patients and the meta-analysis showed no significant difference in the effect of MLD for BCRL compared with other treatments (mean difference, 3.76;95% confidence interval, -35.09 to 42.62;Z, 0.19;p = 0.850). Conclusion: MLD can relieve the body tissue, rapidly improve local condition, and enhance complete decongestive therapy (CDT) efficacy. MLD can prevent BCRL and improve the symptoms of stage I lymphedema. It should be widely applied to prevent BCRL from entering an irreversible state.展开更多

DESIGN,TIME AND SETTING:A completely randomized,controlled,animal experiment was performed at the Experimental Animal Center and Central Laboratory in the Second Hospital of Jilin University between 2007 and 2008. MATERIALS:Magnesium sulfate(25%) was supplied by Tianjin Pharmaceutical Jiaozuo,China. Nitroglycerin was provided by Shanxi Kangbao Biological,China.Substance P primer sequence was synthesized by TaKaRa Biotechnology(Dalian),China. METHODS:A total of 36 healthy,adult,Wistar rats were randomly assigned to 6 groups:control, migraine,low- and high-dose magnesium sulfate treated,and low- and high-dose magnesium sulfate control,with 6 rats in each group.Migraines were induced by subcutaneous injection of 10 mg/kg nitroglycerin in the migraine and low- and high-dose magnesium sulfate treated groups, and 2 mL/kg physiological saline was administered to rats in the control and low- and high-dose of magnesium sulfate control groups.Five minutes following administration,rats in low-dose groups were intraperitoneally injected with 100 mg/kg magnesium sulfate,while those in high-dose groups were injected with 300 mg/kg magnesium sulfate.No interventions were administered to the control and migraine groups. MAIN OUTCOME MEASURES:At 2 hours after nitroglycerin injection,substance P mRNA expression in the rat midbrain was measured by real-time quantitative polymerase chain reaction.At 60-90 minutes after nitroglycerin injection,behavioral changes of pain were analyzed in the experimental rats. RESULTS:The migraine group exhibited significantly lower levels of substance P mRNA expression compared with the control group(P<0.05).Following magnesium sulfate injection,substance P mRNA expression increased,compared with the migraine and control groups(P<0.05).In the low-and high-dose magnesium sulfate treated groups,pain behavior was remarkably ameliorated, compared with the migraine group(P<0.05),particularly with the high-dose injection(P<0.05). CONCLUSION:Magnesium relieved pain behaviors in a rat migraine model in a dose-dependent manner,and the therapeutic effect was achieved in conjunction with increased substance P expression in the midbrain.展开更多
To improve the productivity of poplar plantations, a field experiment of split-plot design with four tree spacings and three poplar clones was established, and four soil enzyme activities and microbial biomass were mo...To improve the productivity of poplar plantations, a field experiment of split-plot design with four tree spacings and three poplar clones was established, and four soil enzyme activities and microbial biomass were monitored in the trial.Soil enzyme activities, in most cases,were significantly higher in topsoil(0–10 cm) than in lower horizons(10–20 cm).Soil cellulase, catalase and protease activities during the growing season were higher than during the non-growing season, while invertase activity followed the opposite trend.Soil invertase, cellulase and catalase activities varied by poplar clone but soil protease activity did not.Cellulase and protease activities in the plantation at 5×5 m spacing were significantly higher than in the other spacings.The highest catalase activity was recorded at 6×6 m spacing.At the same planting density, invertase activity was greater in square spacings than in rectangular spacings.Soil microbial biomass was also significantly affected by seedling spacing and poplar clone.The mean soil MBC was significantly lower in topsoil than in the lower horizon, while MBN showed the opposite pattern.Significantly positive correlations were observed among soil cellulase, protease and catalase activities(p<0.01), whereas soil invertase activity was negatively and significantly correlated with cellulase, protease and catalase activities(p<0.01).Soil microbial biomass and enzyme activities were not correlated except for a significantly negative correlation between soil MBC and catalase activities.Variations in soil enzyme activity and microbial biomass in different poplar plantations suggest that genotype and planting spacing should be considered when modeling soil nutrient dynamics and managing for long-term site productivity.展开更多

文摘Water jets are widely used in seabed scouring and desilting applications.In the present work,dedicated tests have been conducted using a jet scour model test platform and sand beds containing grains with different sizes.The FLOW-3D simulation software has also been used to tackle the problem form a numerical point of view.The boundary conditions of the simulation have been optimized to reduce the gap between the numerical results and the outcomes of the experimental tests.Scour area calculation has been based on a RNG k-εTurbulence model.Moreover,the FAVOR(Fractional Area Volume Obstacle Representation)technology has been selected for grid division to make the simulation results more accurate.Jet scours with inclination angles of 10–20°have been simulated.It has been found that jet scouring is greatly affected by the environmental water flow,that is,water flow can weaken the scouring capacity of the jets.

文摘Microorganisms are fundamental for the functioning of marine ecosystems and are involved in the decomposition of organic matter, transformation of nutrients and circulation of biologically-important chemicals. Based on the complexity of the natural geographic characteristics of the Changjiang River Estuary, the geographic distribution of sedimentary microorganisms and the causes of this distribution are largely unexplored. In this work, the surface sediment samples from the adjacent sea area of the Changjiang River Estuary were collected. Their prokaryotic diversity was examined by high-throughput sequencing technology, and the environmental factors of the bacterial community were investigated. The results indicated that the distribution of prokaryotic communities in the sediments of the study areas showed obvious spatial heterogeneity. The sampling sequences divided the sample regions into three distinct clusters. Each geographic region had a unique community structure, although Proteobacteria, Bacteroidota, Desulfobacterota, Acidobacteriota, and Actinobacteriota all existed in these three branches. Canonical correspondence analysis demonstrated that prokaryotic diversity and community distribution were significantly correlated with the geographic location of sediment, seawater depth, and in particular, nutrient content(e.g., total phosphorus, total organic carbon and dissolved oxygen). Moreover, it was found for the first time that the metal ions obviously affected the composition and distribution of the prokaryotic community in this area. In general, this work provides new insights into the structural characteristics and driving factors of prokaryotic communities under the background of the ever-changing Changjiang River Estuary.

文摘Terahertz(THz) radiation can be generated due to the instability of THz plasma waves in field-effect transistors(FETs). In this work, we discuss the instability of THz plasma waves in the channel of FETs with spin and quantum effects under non-ideal boundary conditions. We obtain a linear dispersion relation by using the hydrodynamic equation, Maxwell equation and spin equation. The influence of source capacitance, drain capacitance, spin effects, quantum effects and channel width on the instability of THz plasma waves under the non-ideal boundary conditions is investigated in great detail. The results of numerical simulation show that the THz plasma wave is unstable when the drain capacitance is smaller than the source capacitance;the oscillation frequency with asymmetric boundary conditions is smaller than that under non-ideal boundary conditions;the instability gain of THz plasma waves becomes lower under non-ideal boundary conditions. This finding provides a new idea for finding efficient THz radiation sources and opens up a new mechanism for the development of THz technology.

文摘Sesquiterpenes are the major pharmacodynamic components of agarwood,a precious traditional Chinese medicine obtained from the resinous portions of Aquilaria sinensis trees that form in response to environmental stressors.To characterize the sesquiterpene synthases responsible for sesquiterpene production in A.sinensis,a bioinformatics analysis of the genome of A.sinensis identifi ed six new terpene synthase genes,and 16 sesquiterpene synthase genes were identifi ed as type TPS-a in a phylogenetic analysis.The expression patterns for eight of the sesquiterpene synthase genes after treatment with various hormones or hydrogen peroxide were analyzed by real-time quantitative PCR.The results suggest that 100μM methyl jasmonate,ethephon,(±)-abscisic acid or hydrogen peroxide could be eff ective short-term eff ectors to increase the expression of sesquiterpene synthase genes,while 1 mM methyl salicylate may have long-term eff ects on increasing the expression of specifi c sesquiterpene synthase genes(e.g.,As-SesTPS,AsVS,AsTPS12 and AsTPS29).The expression changes in these genes under various conditions refl ected their specifi c roles during abiotic or biotic stresses.Heterologous expression of a novel A.sinensis sesquiterpene synthase gene,AsTPS2,in Escherichia coli produced a major humulene product,so AsTPS2 is renamed AsHS1.AsHS1 is diff erent from ASS1,AsSesTPS,and AsVS,for mainly producingα-humulene.Based on the predicted space conformation of the AsHS1 model,the small ligand molecule may bind to the free amino acid by hydrogen bonding for the catalytic function of the enzyme,while the substrate farnesyl diphosphate(FPP)probably binds to the free amino acid on one side of the RxR motif.Arg450,Asp453,Asp454,Thr457,and Glu461 from the NSE/DTE motif and D307 and D311 from the DDxxD motif were found to form a polar interaction with two Mg^(2+)clusters by docking.The Mg^(2+)-bound DDxxD and NSE/DTE motifs and the free RXR motif are jointly directed into the catalytic pocket of AsHS1.Comparison of the tertiary structural models of AsHS1 with ASS1 showed that they diff ered in structures in several positions,such as surrounding the secondary catalytic pocket,which may lead to diff erences in catalytic products.Based on the results,biosynthetic pathways for specifi c sesquiterpenes such asα-humulene in A.sinensis are proposed.This study provides novel insights into the functions of the sesquiterpene synthases of A.sinensis and enriches knowledge on agarwood formation.

文摘In this paper,the evaluations of metal ablation processes under high temperature,i.e.,the Al plate ablated by a laser and a heat carrier and the reactor pressure vessel ablated by a core melt,are studied by a novel peridynamic method.Above all,the peridynamic formulation for the heat conduction problem is obtained by Taylor’s expansion technique.Then,a simple and efficient moving boundary model in the peridynamic framework is proposed to handle the variable geometries,in which the ablated states of material points are described by an additional scalar field.Next,due to the automatic non-interpenetration properties of peridynamic method,a contact algorithm is established to determine the contact relationship between the ablated system and the additional heat carrier.In addition,the corresponding computational procedure is listed in detail.Finally,several numerical examples are carried out and the results verify the validity and accuracy of the present method.
文摘Background: New therapeutic targets are needed to improve the outcomes for gastric cancer(GC) patients with advanced disease. Evasion of programmed cell death(apoptosis) is a hallmark of cancer cells and direct induction of apoptosis by targeting the pro-survival BCL2 family proteins represents a promising therapeutic strategy for cancer treatment. Therefore, understanding the molecular mechanisms underpinning cancer cell survival could provide a molecular basis for potential therapeutic interventions. Method: Here we explored the role of BCL2L1 and the encoded anti-apoptotic BCL-XL in GC. Using Droplet Digital PCR(ddPCR) technology to investigate the DNA amplification of BCL2L1 in GC samples and GC cell lines, the sensitivity of GC cell lines to selective BCL-XL inhibitors A1155463 and A1331852, pan-inhibitor ABT-263, and VHL-based PROTAC-BCL-XL was analyzed using(CellTiter-Glo) CTG assay in vitro. Western Blot(WB) was used to detect the protein expression of BCL2 family members in GC cell lines and the manner in which PROTAC-BCL-XL kills GC cells. Coimmunoprecipitation(Co-IP) was used to investigate the mechanism of A1331852 and ABT-263 kills GC cell lines. DDPCR, WB, and real-time PCR(RTPCR) were used to investigate the correlation between DNA, RNA, protein levels, and drug activity. Results: The functional assay showed that a subset of GC cell lines relies on BCL-XL for survival. In gastric cancer cell lines, BCL-XL inhibitors A1155463 and A1331852 are more sensitive than the pan BCL2 family inhibitor ABT-263, indicating that ABT-263 is not an optimal inhibitor of BCL-XL. VHL-based PROTAC-BCL-XL DT2216 appears to be active in GC cells. DT2216 induces apoptosis of gastric cancer cells in a time-and dose-dependent manner through the proteasome pathway. Statistical analysis showed that the BCL-XL protein level predicts the response of GC cells to BCL-XL targeting therapy and BCL2L1 gene CNVs do not reliably predict BCL-XL expression.Conclusion: We identified BCL-XL as a promising therapeutic target in a subset of GC cases with high levels of BCL-XL protein expression. Functionally, we demonstrated that both selective BCL-XL inhibitors and VHL-based PROTAC BCL-XL can potently kill GC cells that are reliant on BCL-XL for survival. However, we found that BCL2L1 copy number variations(CNVs) cannot reliably predict BCL-XL expression, but the BCL-XL protein level serves as a useful biomarker for predicting the sensitivity of GC cells to BCL-XL-targeting compounds. Taken together, our study pinpointed BCL-XL as potential druggable target for specific subsets of GC.

文摘High quality Nb films were successfully prepared on both flexible polyimide(PI)and rigid Al2O3substrates and their transport properties were systematically studied at various applied currents,external magnetic fields,and sample orientations.It is found that a curved Nb/PI film exhibits quite different superconducting transition and vortex dynamics compared to the flat Nb/Al2O3film.For the curved Nb/PI film,smooth superconducting transitions were obtained at low currents,while unexpected cascade structures were revealed in theρ(T)curves at high currents.We attribute this phenomenon to the gradient distribution of vortex density together with a variation of superconductivity along the curved film.In addition,reentrant superconductivity was induced in the curved Nb/PI thin film by properly choosing the measurement conditions.We attribute this effect to the vortex pinning from both in-plane vortices and out-of-plane vortices.This work reveals the complex transport properties of curved superconducting thin films,providing important insights for further theoretical investigations and practical developments of flexible superconductors.

文摘Themulti-skill resource-constrained project scheduling problem(MS-RCPSP)is a significantmanagement science problem that extends from the resource-constrained project scheduling problem(RCPSP)and is integrated with a real project and production environment.To solve MS-RCPSP,it is an efficient method to use dispatching rules combined with a parallel scheduling mechanism to generate a scheduling scheme.This paper proposes an improved gene expression programming(IGEP)approach to explore newly dispatching rules that can broadly solve MS-RCPSP.A new backward traversal decoding mechanism,and several neighborhood operators are applied in IGEP.The backward traversal decoding mechanism dramatically reduces the space complexity in the decoding process,and improves the algorithm’s performance.Several neighborhood operators improve the exploration of the potential search space.The experiment takes the intelligent multi-objective project scheduling environment(iMOPSE)benchmark dataset as the training set and testing set of IGEP.Ten newly dispatching rules are discovered and extracted by IGEP,and eight out of ten are superior to other typical dispatching rules.
文摘Although previous RNA sequencing methods have been widely used in orthopedic research and have provided ideas for therapeutic strategies,the specific mechanisms of some orthopedic disorders,including osteoarthritis,lumbar disc herniation,rheumatoid arthritis,fractures,tendon injuries,spinal cord injury,heterotopic ossification,and osteosarcoma,require further elucidation.The emergence of the single-cell RNA sequencing(sc RNA-seq)technique has introduced a new era of research on these topics,as this method provides information regarding cellular heterogeneity,new cell subtypes,functions of novel subclusters,potential molecular mechanisms,cell-fate transitions,and cell-cell interactions that are involved in the development of orthopedic diseases.Here,we summarize the cell subpopulations,genes,and underlying mechanisms involved in the development of orthopedic diseases identified by sc RNA-seq,improving our understanding of the pathology of these diseases and providing new insights into therapeutic approaches.

文摘Increased evidence has shown that hydrogen sulfide(H_(2)S),a novel gasotransmitter,could enhance drought resistance in plants by inducing stomatal closure,with concurrent enhancement of photosynthetic efficiency,but little is known about the mechanism behind this contradictory phenomenon.This study examined the regulating mechanism of H_(2)S in response to drought stress fromstomatal and non-stomatal factors in Chinese cabbage.The results showed that exogenous H_(2)S could increase the accumulation of photosynthetic pigments and alleviate the damage caused by drought stress.It also regulated the expression in transcriptional level and the activity of ribulose 1,5-bisphosphate carboxylase/oxygenase(BrRuBisCO)under drought stress.The large subunit of BrRuBisCO was found to be modified by S-sulfhydration,which might be the reason for its increased enzyme activity.The fluxes of Cl^(−),K^(+),and H^(+)in the guard cells were detected by non-invasive micro-test techniques while under drought stress.The results indicated that H_(2)S signaling induced a transmembrane Cl^(−)and H^(+)efflux and inhibited K^(+)influx,and the Cl^(−)channel was the main responders for H_(2)S-regulated stomatal movement.In conclusion,H_(2)S signal not only activated the ion channel proteins located in the guard cell membrane to induce stomatal closure,but also regulated the transcriptional expression and the activity of RuBisCO,a non-stomatal factor to enhance the photosynthetic efficiency of leaves.There is therefore a beneficial balance between the regulation of H_(2)S signaling on stomatal factors and non-stomatal factors due to drought stress,which needs to be better understood to apply it practically to increase crop yields.

文摘[Objectives]Based on UPLC-Q-TOF-MS/MS,network pharmacology and molecular docking techniques,the mechanism of Euphorbia peplus in the treatment of Alzheimer s disease(AD)was studied.[Methods]The UPLC-Q-TOF-MS/MS technique was used to rapidly analyze the chemical components of E.peplus.Active components and potential targets of E.peplus were retrieved from TCMSP and Swiss Target Prediction database,and AD targets were screened using GeneCards database.The targets of E.peplus in the treatment of AD were obtained.The PPI network was constructed using String platform,and the network topology of Cytoscape software was used to compute and screen key targets,and the GO and KEGG pathway enrichment analysis was carried out in Metascape database to construct the"component-target-pathway-disease"network.Molecular docking was used to predict the binding properties of active ingredients and targets.[Results]The results of UPLC-Q-TOF-MS/MS showed that 83 compounds were identified from E.peplus,including 19 terpenoids,10 phenolic acids and phenols,16 flavonoids,2 phenylpropanoids,4 coumarins,1 alkaloid,1 anthraquinone and 30 other compounds.The results of network pharmacological analysis showed that 82 active ingredients were screened,and 279 common targets were identified for the treatment of AD,among which the key targets were ALB(albumin),GAPDH(glyceraldehyde triphosphate dehydrogenase),TNF(tumor necrosis factor),AKT1(serine/threonine protein kinase 1),and IL6(interleukin-6).KEGG enrichment analysis showed that key signaling pathways include cancer pathways,lipid and atherosclerosis,Alzheimer s disease,insulin resistance,serotonergic synapses,calcium signaling pathway,cAMP signaling pathway and other signaling pathways.Molecular docking results showed that 14-deoxyandrographolide,dehydroandrographolide,licochalcone B,apigenin and naringenin may be the key components of E.peplus in the treatment of AD.[Conclusions]The results suggest that E.peplus can be used to treat Alzheimer s disease through multi-component,multi-target and multi-pathway.

文摘A rare large-scale hail process in Yantai and Weihai City in the autumn of 2021 was analyzed based on the surface meteorology,high-altitude observation and S-band radar detection data.The results showed that this process was influenced by shallow trough,and strong upper-level jet stream in the middle and upper levels guided the intrusion of mid-level cold air into the warm and humid environment at the lower level.The shear lines and trunk lines in the lower level also provided certain triggering conditions for uplift,brewing conditions suitable for the occurrence and development of strong convection.The vertical wind shear was unusually large,and vertical wind shear was up to 10 m/s at 0-1 km,20 m/s at 0-3 km,and 32 m/s at 0-6 km,which was conducive to the growth of the storm.In this process,isolated multi-cell thunderstorm was generated at the beginning,and then it developed and strengthened continuously,moving to the southeast.In the later stage,a convection cell was born at the boundary between Yantai and Weihai,and developed into supercell storm.The storm lasted for a long time,and continued to develop and strengthen as it moved eastward,affecting most areas of Weihai from northwest to southeast.In the development stage of the storm,the radar reflectivity factor had obvious characteristics of hook echo and three-body scatter spike,and there was echo overhang and other hail echo features in the middle layer.Weak mesocyclone appeared at a low elevation,and there was the potential for the occurrence of tornado.
文摘<strong>Objective:</strong> To assess the association between circulating C-reactive protein (CRP), and CRP polymorphisms in the diesel engine exhaust (DEE)-exposed workers. <strong>Methods:</strong> In 137 DEE-exposed workers and 127 unexposed comparable control workers, six urinary mono-hydroxylated polycyclic aromatic hydrocarbons (OH-PAHs) and serum CRP levels were assayed. Genotyping of four CRP single-nucleotide polymorphisms (SNPs) was measured. <strong>Results: </strong>Serum CRP levels increased in exposed versus control workers (all p < 0.001). In the DEE-exposed workers, two CRP polymorphisms were associated with serum CRP levels, the subjects of rs1205 TT genotype had lower serum CRP levels (p < 0.05 compared to TC or CC). <strong>Conclusions: </strong>Our findings suggest that polymorphisms in CRP and circulating CRP involved in the inflammatory process may play significant roles in human sensitivity to lung function injury caused by DEE exposure. This study will help investigate the underlying mechanisms of adverse respiratory effects induced by DEE.
文摘Objective: The purpose of the study was to explore the application effects of the cloud platform-based comprehensive online management for breast cancer patients using dual-targeted therapy with macromolecular monoclonal antibodies. Methods: 120 breast cancer patients treated by dual-targeted therapy with macromolecular monoclonal antibodies were managed by a cloud platform from March to November 2019. Comprehensive online management included consultation about drugs and side effects and frequently asked questions in the dual-targeted therapy with macromolecular monoclonal antibodies. Results: In the consultation about drugs and side effects, there were five patients with fever, neutrophil, cough, and fatigue;24 with diarrhea;25 with nausea;11 with oral mucosal inflammation;10 with rashes and dry skin;8 with insomnia;and 1 with palpitation. Moreover, 110 patients with anxiety about the missed or delayed treatment were properly handled. Conclusion: The comprehensive online management of dual-targeted therapy with macromolecular monoclonal antibodies based on the cloud platform is helpful to satisfy the at-home breast cancer patients’ needs, ensure the continuity of dual-targeted therapy with macromolecular monoclonal antibodies for breast cancer patients, prevent misinformation, alleviate patients’ negative psychological emotions, and reduce patients’ economic losses. The online cloud platform integrated management model is crucial for managing patients with breast cancer treated by dual-targeted therapy.
文摘Purpose: The effect of manual lymphatic drainage in patients with breast cancer is controversial. The purpose of this study is to evaluate the role of manual lymphatic drainage (MLD) in breast cancer-related lymphedema treatment. Methods: The electronic databases of EMBASE, PubMed, Web of Science, and The Cochrane Library were searched to find English articles on MLD which were published before January 2020. After two evaluators selected the studies and independently evaluated literature quality, meta-analysis was carried out with RevMan 5.3 software. The outcome index of lymphedema treatment changed in edema volume. Results: The study included six RCTs of 364 patients and the meta-analysis showed no significant difference in the effect of MLD for BCRL compared with other treatments (mean difference, 3.76;95% confidence interval, -35.09 to 42.62;Z, 0.19;p = 0.850). Conclusion: MLD can relieve the body tissue, rapidly improve local condition, and enhance complete decongestive therapy (CDT) efficacy. MLD can prevent BCRL and improve the symptoms of stage I lymphedema. It should be widely applied to prevent BCRL from entering an irreversible state.

文摘According to the characteristic of cruise missiles,navigation point setting is simplified,and the principle of route planning for saturation attack and a concept of reference route are put forward.With the help of the shortest-tangent idea in route-planning and the algorithm of back reasoning from targets,a reference route algorithm is built on the shortest range and threat avoidance.Then a route-flight-time algorithm is built on navigation points.Based on the conditions of multi-direction saturation attack,a route planning algorithm of multi-direction saturation attack is built on reference route,route-flight-time,and impact azimuth.Simulation results show that the algorithm can realize missiles fired in a salvo launch reaching the target simultaneously from different directions while avoiding threat.

DESIGN,TIME AND SETTING:A completely randomized,controlled,animal experiment was performed at the Experimental Animal Center and Central Laboratory in the Second Hospital of Jilin University between 2007 and 2008. MATERIALS:Magnesium sulfate(25%) was supplied by Tianjin Pharmaceutical Jiaozuo,China. Nitroglycerin was provided by Shanxi Kangbao Biological,China.Substance P primer sequence was synthesized by TaKaRa Biotechnology(Dalian),China. METHODS:A total of 36 healthy,adult,Wistar rats were randomly assigned to 6 groups:control, migraine,low- and high-dose magnesium sulfate treated,and low- and high-dose magnesium sulfate control,with 6 rats in each group.Migraines were induced by subcutaneous injection of 10 mg/kg nitroglycerin in the migraine and low- and high-dose magnesium sulfate treated groups, and 2 mL/kg physiological saline was administered to rats in the control and low- and high-dose of magnesium sulfate control groups.Five minutes following administration,rats in low-dose groups were intraperitoneally injected with 100 mg/kg magnesium sulfate,while those in high-dose groups were injected with 300 mg/kg magnesium sulfate.No interventions were administered to the control and migraine groups. MAIN OUTCOME MEASURES:At 2 hours after nitroglycerin injection,substance P mRNA expression in the rat midbrain was measured by real-time quantitative polymerase chain reaction.At 60-90 minutes after nitroglycerin injection,behavioral changes of pain were analyzed in the experimental rats. RESULTS:The migraine group exhibited significantly lower levels of substance P mRNA expression compared with the control group(P<0.05).Following magnesium sulfate injection,substance P mRNA expression increased,compared with the migraine and control groups(P<0.05).In the low-and high-dose magnesium sulfate treated groups,pain behavior was remarkably ameliorated, compared with the migraine group(P<0.05),particularly with the high-dose injection(P<0.05). CONCLUSION:Magnesium relieved pain behaviors in a rat migraine model in a dose-dependent manner,and the therapeutic effect was achieved in conjunction with increased substance P expression in the midbrain.

文摘To improve the productivity of poplar plantations, a field experiment of split-plot design with four tree spacings and three poplar clones was established, and four soil enzyme activities and microbial biomass were monitored in the trial.Soil enzyme activities, in most cases,were significantly higher in topsoil(0–10 cm) than in lower horizons(10–20 cm).Soil cellulase, catalase and protease activities during the growing season were higher than during the non-growing season, while invertase activity followed the opposite trend.Soil invertase, cellulase and catalase activities varied by poplar clone but soil protease activity did not.Cellulase and protease activities in the plantation at 5×5 m spacing were significantly higher than in the other spacings.The highest catalase activity was recorded at 6×6 m spacing.At the same planting density, invertase activity was greater in square spacings than in rectangular spacings.Soil microbial biomass was also significantly affected by seedling spacing and poplar clone.The mean soil MBC was significantly lower in topsoil than in the lower horizon, while MBN showed the opposite pattern.Significantly positive correlations were observed among soil cellulase, protease and catalase activities(p<0.01), whereas soil invertase activity was negatively and significantly correlated with cellulase, protease and catalase activities(p<0.01).Soil microbial biomass and enzyme activities were not correlated except for a significantly negative correlation between soil MBC and catalase activities.Variations in soil enzyme activity and microbial biomass in different poplar plantations suggest that genotype and planting spacing should be considered when modeling soil nutrient dynamics and managing for long-term site productivity.
