Objective:To evaluate the hepatoprotective and immunotherapeutic effects of aqueous extract of turmeric rhizome in CCl_4 intoxicated Swiss albino mice.Methods:first group of mice(n=5) received CCl_4 treatment at a dose of 0.5 mL/kg bw(i.p.) for 7 days.Second group was fed orally the aqueous extract of turmeric at a dose of 50 mg/kg bw for IS days.The third group was given both the turmeric extract(for 15 days,orally) and CCl_4(for last 7 days,i.p.).The fourth group was kept as a control.To study the liver function,the transaminase enzymes(SGOT and SGPT) and bilirubin level were measured in the serum of respective groups.For assaying the immunotherapeutic action of Curcuma longa(C.longa),non specific host response parameters like morphological alteration,phagocytosis,nitric oxide release,myeloperoxidase release and intracellular killing capacity of peritoneal macrophages were studied from the respective groups.Results:The result of present study suggested that CCl_4 administration increased the level of SCOT and SGPT and bilirubin level in serum.However,the aqueous extract of turmeric reduced the level of SGOT, SCFT and bilirubin in CCl_4 intoxicated mice.Apart from damaging the liver system,CCl_4 also reduced non specific host response parameters like morphological alteration,phagocytosis, nitric oxide release,myeloperoxidase release and intracellular killing capacity of peritoneal macrophages.Administration of aqueous extract of C.longa offered significant protection from these damaging actions of CCl_4 on the non specific host response in the peritoneal macrophages of CCl_4 intoxicated mice.Conclusions:In conclusion,the present study suggests that C.longa has immunotherapeutic properties along with its ability to ameliorate hepatotoxicity. 展开更多

The aim of this study is to assess the adverse effects of lead, a well-documented non-essential element that occurs naturally in the environment, on Channa punctatus, in relation with ROS production and oxidative stress. Fishes were sampled, acclimatized and kept treated or untreated with lead (9.43 mg/L) under observation for 4 days. At day 4, respiratory burst activity, lipid peroxidation activity and superoxide dismutase level increased significantly in treated group as compared to the control. On the contrary, catalase, glutathione S-transferase, glutathione peroxidase, glutathione reductase and reduced glutathione activity decreased on treatment with lead. These results suggest that heavy metal like lead induces oxidative stress, influences the antioxidant defense system and may lead to physiological disorders rendering the health and survival of exposed fish to a compromised state. 展开更多

Studies of lead toxicity on inflammatory damage and innate immune functions in testicular macrophages of male Swiss albino mice

Various scientific studies have been carried out on the toxic effects of lead on immune function;however, uncertainties still persist. The present study addresses the immunomodulatory effects of testicular macrophages caused due to lead exposure (in-vivo). In mice, lead has a primary effect on the testes, and acts at all levels of the reproductive as well as the testicular immune response. The study was undertaken to investigate the effects of lead acetate on immune system in Swiss albino mice and the underlying mechanism of pro-inflammatory cytokine action. The immunomodulatory effects of lead are complex and appear to involve multiple pathways, not all of which are fully understood. Testicular macrophage dysfunctions by lead was studied by calculating morphological alteration and associated cell functions of innate immunity followed by estimation of pro-inflammatory cytokine release (TNF-α). The present work shows that lead is responsible for a significant morphological alteration, reducing cell function in testicular macrophages probably by increasing oxidative damage whereas increase in TNF-α release was also observed which increased inflammation. Results demonstrated that lead intoxication leads to increase in pro-inflammatory response. TNF-α is probably unable to bind with the surface receptor in testicular macrophages because of altered morphology which reduces cell function and ultimately causes reproductive dysfunction. 展开更多

The use of the optoelectronic properties of noble metal nanoparticles has given a new dimension to the realm of cancer diagnosis and therapy(theranostics).The multifunctional catalytic properties and surface plasmon resonance of noble metal nanoparticles(NPs)along with their facile surface chemistry gives them an edge in the field of biology,including for immunotherapeutic applications.In a previous study,we reported the anticancer efficacy of silver NPs coated with murine serum albumin against fibrosarcoma in an in vivo mouse model,highlighting their role in eliciting oxystress and immune interference.In addition to addressing the bioavailability and toxicity parameters,our study reported that there was a significant reduction in the size,as well as a delay in the incidence of tumors with AgNP-MSA treatment.Transmission electron micrographs revealed AgNP-MSA uptake by sentinel immune cells associated with the tumor along with the downregulation of pro-tumorigenic inflammatory cytokines,such as TNF-α,IL-6 and IL-1β. 展开更多
