BACKGROUND Glucose imbalance is common in total parenteral nutrition(TPN).Hypoglycemia seems to be less frequent than hyperglycemia,but it influences the clinical outcome to a greater extent.Therefore,it should be eff...BACKGROUND Glucose imbalance is common in total parenteral nutrition(TPN).Hypoglycemia seems to be less frequent than hyperglycemia,but it influences the clinical outcome to a greater extent.Therefore,it should be effectively prevented and treated.However,there is no relevant report on how to treat hypoglycemia caused by TPN in patients with liver cell injury.CASE SUMMARY We present three patients with liver cell injury who developed severe hypo-glycemia during or after TPN infusion.The causes of severe hypoglycemia and glucose-raising strategies were discussed.According to the physiological charac-teristics of the hepatocellular injury,the ratio of nutrition components prescribed in TPN was appropriately adjusted for the three cases.We simultaneously reduced the dose of insulin and fat emulsion,and increased the dose of glucose in TPN.The blood glucose level was restored to normal range and clinical symptoms were eliminated.CONCLUSION When hypoglycemia occurs during or after TPN in patients with hepatocellular injury,physicians need to simultaneously reduce insulin and fat emulsion,and increase glucose,and correct severe hypoglycemia in time to reduce its adverse consequences.展开更多
Objective:To investigate the therapeutic effect of Biling Weitong Granules combined with oryz-aspergillus enzyme and pancreatin tablets on patients with reflux esophagitis with functional dyspepsia.Methods:Sixty patie...Objective:To investigate the therapeutic effect of Biling Weitong Granules combined with oryz-aspergillus enzyme and pancreatin tablets on patients with reflux esophagitis with functional dyspepsia.Methods:Sixty patients diagnosed with reflux esophagitis with functional dyspepsia who were admitted to the Affiliated Hospital of Hebei University between June 2020 and June 2023 were selected and divided into two groups:the control group and the observation group,each consisting of 30 cases.The control group received oryz-aspergillus enzyme and pancreatin tablets only,while the observation group received Biling Weitong Granules in addition to the tablets.The clinical efficacy,Chinese medicine syndrome points,esophageal kinetic indexes,gastrointestinal hormone levels,and therapeutic safety of both groups were evaluated.Results:The total efficiency of the observation group reached 93.33%,significantly higher than the 73.33%of the control group(P<0.05).After treatment,patients in the observation group exhibited significantly lower scores for Chinese medicine symptoms such as early satiety,belching,abdominal distension,abdominal pain,and loss of appetite compared to the control group(P<0.05).Furthermore,the observation group showed significantly higher upper esophageal sphincter pressure,lower esophageal sphincter pressure,and distal esophageal contraction scores compared to the control group(P<0.05).Additionally,levels of gastric motility hormone,vasoactive intestinal peptide,and gastrin were significantly higher in the observation group compared to the control group(P<0.05).Throughout the treatment period,there was no significant difference in the incidence of adverse reactions between the two groups,indicating comparable safety of the two treatment modalities(P>0.05).Conclusion:The combination of Biling Weitong Granules with oryz-aspergillus enzyme and pancreatin tablets demonstrates significant efficacy in the treatment of reflux esophagitis with functional dyspepsia,with a better safety profile.This finding warrants further clinical promotion.展开更多

Proteomic characterization of plasma is critical for the development of novel pharmacodynamic biomarkers.However,the vast dynamic range renders the profiling of proteomes extremely challenging.Here,we synthesized zeol...Proteomic characterization of plasma is critical for the development of novel pharmacodynamic biomarkers.However,the vast dynamic range renders the profiling of proteomes extremely challenging.Here,we synthesized zeolite NaY and developed a simple and rapid method to achieve comprehensive and deep profiling of the plasma proteome using the plasma protein corona formed on zeolite NaY.Specifically,zeolite NaY and plasma were co-incubated to form plasma protein corona on zeolite NaY(NaY-PPC),followed by conventional protein identification using liquid chromatography-tandem mass spectrometry.NaY was able to significantly enhance the detection of low-abundance plasma proteins,minimizing the“masking”effect caused by high-abundance proteins.The relative abundance of middleand low-abundance proteins increased substantially from 2.54%to 54.41%,and the top 20 highabundance proteins decreased from 83.63%to 25.77%.Notably,our method can quantify approximately 4000 plasma proteins with sensitivity up to pg/mL,compared to only about 600 proteins identified from untreated plasma samples.A pilot study based on plasma samples from 30 lung adenocarcinoma patients and 15 healthy subjects demonstrated that our method could successfully distinguish between healthy and disease states.In summary,this work provides an advantageous tool for the exploration of plasma proteomics and its translational applications.展开更多
AIM:To compare the clinical outcomes of wavefront guided femtosecond LASIK(WFG LASIK)and conventional femtosecond LASIK(NWFG LASIK)in eyes with myopia and myopia astigmatism.METHODS:This was a retrospective,nonrandomi...AIM:To compare the clinical outcomes of wavefront guided femtosecond LASIK(WFG LASIK)and conventional femtosecond LASIK(NWFG LASIK)in eyes with myopia and myopia astigmatism.METHODS:This was a retrospective,nonrandomized,comparative investigation enrolling 236 eyes of 122 patients(18-50 y)with low&moderate and high myopia.The WFG group including 97 eyes(50 patients)undergone WFG LASIK and the NWFG group including 139 eyes(72 patients)undergone conventional LASIK.Mean efficacy index,high order aberrations(HOAs),pupil size and the quality of visual questionnaire were evaluated 6 mo postoperatively.RESULTS:There is no difference between WFG group(-0.054±0.049 in logMAR)and NWFG group(-0.040±0.056)in uncorrected distance visual acuity(UDVA)postoperatively.The myopia astigmatism is higher in WFG group than that in NWFG group(P<0.05).However,the mean efficacy index(MEI)in the WFG group(1.09±0.106)is better than that in the NWFG group(1.036±0.124;P<0.001).Increased HOAs were observed in NWFG group(0.30±0.196)than that in WFG group(0.146±0.188;P<0.001).The pupil size is larger in WFG group(5.15±0.76 mm)than that in NWFG group(4.32±0.52 mm).The patients are satisfied with the clinical surgery,yet WFG group showed better visual quality using the questionnaire survey.Meanwhile,high myopia would result in worse MEI,HOAs and visual quality than low&moderate myopia.CONCLUSION:WFG and NWFG FS-LASIK are both effective and safe procedures to correct low&moderate and high myopia,but WFG FS-LASIK gives a better postoperative MEI,aberrometric control and predictable outcome.Meanwhile,WFG FS-LASIK is better than NWFG FS-LASIK in correction of myopia astigmatism.Low&moderate myopia allow better clinical outcomes than high myopia using any surgical method.展开更多
BACKGROUND Very few studies have been published on the hemodynamic changes associated with spinal anesthesia induced with ropivacaine during cesarean deliveries in preeclamptic women.AIM To record and analyze hemodyna...BACKGROUND Very few studies have been published on the hemodynamic changes associated with spinal anesthesia induced with ropivacaine during cesarean deliveries in preeclamptic women.AIM To record and analyze hemodynamic data in women with preeclampsia undergoing cesarean delivery after spinal anesthesia induced with ropivacaine.METHODS Ten eligible women with preeclampsia were enrolled in this prospective observational study.Spinal anesthesia was performed with 2.4 mL of 0.5%ropivacaine.Hemodynamic changes were then analyzed at multiple time points.The hemodynamic responses to vasopressor interventions and uterotonic agents,as well as maternal and neonatal outcomes were also recorded.RESULTS Stable hemodynamic trends were observed in this study.Cardiac output(CO)and stroke volume increased mildly during surgery.In contrast,mean arterial pressure and systemic vascular resistance showed a moderate decrease from induction until the end of surgery.Central venous pressure dramatically increased after delivery.Oxytocin administration was associated with the most significant hemodynamic fluctuations during surgery,namely,an increase in CO and heart rate.Phenylephrine intervention was only required in three patients,and caused an increase in mean arterial pressure and systemic vascular resistance along with a decrease in heart rate,stroke volume,and CO.No maternal and neonatal complications were observed during this study,except transient episodes of hypotension.CONCLUSION Spinal anesthesia for caesarian delivery with ropivacaine in women with preeclampsia is linked to modest hemodynamic changes of no clinical significance in this study.Careful cardiovascular monitoring is still recommended,particularly after the delivery of the fetus or the use of oxytocin.展开更多

The sensitivities of soil respiration components on changing climate patterns are currently not fully understood. We used trench and isotopic methods to separate total soil respiration into autotrophic(RA) and heterotrophic components(R_H). This study was undertaken on a Robinia pseudoacacia L. plantation in the southern Taihang Mountains, China. The fractionation of soil^(13)CO_2 was analyzed by comparing the δ^(13) C of soil CO_2 extracted from buried steel tubes with results from Gas Vapor Probe Kits at a depth of 50 cm at the preliminary test(2.03%). The results showed that the contribution of autotrophic respiration(f R_A) increased with increasing soil depth.The contribution of heterotrophic respiration(f R_H) declined with increasing soil depth. The contribution of autotrophic respiration was similar whether estimated by the trench method(f RA, 23.50%) or by the isotopic method in which a difference in value of^(13) C between soil and plant prevailed in the natural state(R_C, 21.03%). The experimental error produced by the trench method was insignificant as compared with that produced by the isotopic method, providing a technical basis for further investigations.展开更多
The human gut microbiome has primarily been studied through the use of fecal samples,a practice that has generated vital knowledge on the composition and functional capacities of gastrointestinal microbial communities...The human gut microbiome has primarily been studied through the use of fecal samples,a practice that has generated vital knowledge on the composition and functional capacities of gastrointestinal microbial communities.However,this reliance on fecal materials limits the investigation of microbial dynamics in other locations along the gastrointestinal tract(in situ),and the infrequent availability of fecal samples prevents analysis at finer temporal scales(e.g.,hours).In our study,we utilized colonic transendoscopic enteral tubing,a technology originally developed for fecal microbiota transplantation,to sample the ileocecal microbiome twice daily;metagenomic and metatranscriptomic analyses were then conducted on these samples.A total of 43 ileocecal and 28 urine and fecal samples were collected from five healthy volunteers.The ileocecal and fecal microbiomes,as profiled in the five volunteers,were found to be similar in metagenomic profiling,yet their active genes(metatranscriptome)were found to be highly distinct.Both microbiomes were perturbed after laxative exposure;over time,they exhibited reduced dissimilarity to their pre-treatment state,thereby demonstrating resilience as an innate property of the gut microbiome,although they did not fully recover within our observation time window.Sampling of the ileocecal microbiome during the day and at night revealed the existence of diurnal rhythms in a series of bacterial species and functional pathways,particularly those related to short-chain fatty acid production,such as Propionibacterium acnes and coenzyme A biosynthesis Ⅱ.Autocorrelation analysis and fluctuations decomposition further indicated the significant periodicity of the diurnal oscillations.Metabolomic profiling in the fecal and urine samples mirrored the perturbance and recovery in the gut microbiome,indicating the crucial contribution of the gut microbiome to many key metabolites involved in host health.This study provides novel insights into the human gut microbiome and its inner resilience and diurnal rhythms,as well as the potential consequences of these to the host.展开更多

In the water transfer tunnels from the East River to Shenzhen and Hong Kong, golden mussels attach to the walls of pipelines and gates, causing serious biofouling, increased flow resistance, and resulted in corrosion of the tunnel wall. Golden mussel has very high environmental adaptability and may colonize habitats with low dissolved oxygen and a wide range of trophic levels. The colonization process of the species on solid surface was studied in the Xizhijiang River, a tributary of the East River and the main water resource of Shenzhen from March 2010 to April 2011. The results showed that the golden mussel completed three generations and reproduced six cohorts per year in the tropic zone. Water temperature was the controlling factor for the growth rate and maturity of each cohort. Based on the results, an ecological method for controlling the invasion of golden mussels in water transfer tunnels was proposed.展开更多

Whole-exome sequencing(WES), coupling the Agilent whole-exome capture system to the Illumina Hi Seq 2000 DNA sequencing platform was used to search the specific gene mutation in 3 affected family members and 1unaffected member. After a suggested variant was found through the data analysis, the putative mutation was validated by Sanger DNA sequencing of samples from all available family members.· RESULTS: The results of both WES and Sanger sequencing revealed a novel nonsense mutation c.C766T(p.Q256X) within exon 5 of CRX gene which was pathogenic for ad CORD in this family. The mutation could affect photoreceptor-specific gene expression with a dominant-negative effect and resulted in loss of the OTX tail, thus the mutant protein occupies the CRX-binding site in target promoters without establishing an interaction and, consequently, may block transactivation.·CONCLUSION: All modes of Mendelian inheritance in CORD have been observed, and genetic heterogeneity is a hallmark of CORD. Therefore, conventional genetic diagnosis of CORD would be time-consuming and laborintensive. Our study indicated the robustness and cost-effectiveness of WES in the genetic diagnosis of CORD.展开更多

The differences in serum HP-IgG and anti-HP-CagA-IgG contents as well as lipid metabolism index, platelet function index and inflammatory mediator contents were compared between the two groups, and correlation analysis was used to determine the inner link of serum HP-IgG and anti-HP-CagA-IgG contents with coronary artery lesions in patients with coronary heart disease. Results:Serum HP-IgG and anti-HP-CagA-IgG contents of CHD group were higher than those of control group;peripheral blood lipid metabolism index TC content was higher than that of control group whereas HDL-C and apoA1 contents were lower than those of control group;serum platelet function indexes P-selectin, PAF and PDGF contents were higher than those of control group;serum inflammatory mediators hs-CRP, IL-8, IL-17 and TNF-α contents were higher than those of control group. Pearson correlation analysis showed that serum HP-IgG and anti-HP-CagA-IgG contents in patients with coronary heart disease were closely related to the degree of coronary artery lesions. Conclusion: Serum HP-IgG and anti-HP-CagA-IgG contents were extremely high in patients with coronary heart disease, and the specific contents are closely related to coronary artery lesions.展开更多
With the advancement of network communication technology,network traffic shows explosive growth.Consequently,network attacks occur frequently.Network intrusion detection systems are still the primary means of detectin...With the advancement of network communication technology,network traffic shows explosive growth.Consequently,network attacks occur frequently.Network intrusion detection systems are still the primary means of detecting attacks.However,two challenges continue to stymie the development of a viable network intrusion detection system:imbalanced training data and new undiscovered attacks.Therefore,this study proposes a unique deep learning-based intrusion detection method.We use two independent in-memory autoencoders trained on regular network traffic and attacks to capture the dynamic relationship between traffic features in the presence of unbalanced training data.Then the original data is fed into the triplet network by forming a triplet with the data reconstructed from the two encoders to train.Finally,the distance relationship between the triples determines whether the traffic is an attack.In addition,to improve the accuracy of detecting unknown attacks,this research proposes an improved triplet loss function that is used to pull the distances of the same class closer while pushing the distances belonging to different classes farther in the learned feature space.The proposed approach’s effectiveness,stability,and significance are evaluated against advanced models on the Android Adware and General Malware Dataset(AAGM17),Knowledge Discovery and Data Mining Cup 1999(KDDCUP99),Canadian Institute for Cybersecurity Group’s Intrusion Detection Evaluation Dataset(CICIDS2017),UNSW-NB15,Network Security Lab-Knowledge Discovery and Data Mining(NSL-KDD)datasets.The achieved results confirmed the superiority of the proposed method for the task of network intrusion detection.展开更多
Cryobioprinting has tremendous potential to solve problems to do with lack of shelf availability in traditional bioprinting by combining extrusion bioprinting and cryopreservation.In order to ensure the viability of c...Cryobioprinting has tremendous potential to solve problems to do with lack of shelf availability in traditional bioprinting by combining extrusion bioprinting and cryopreservation.In order to ensure the viability of cells in the frozen state and avoid the possible toxicity of dimethyl sulfoxide(DMSO),DMSO-free bioink design is critical for achieving successful cryobioprinting.A nontoxic gelatin methacryloyl-based bioink used in cryobioprinting is composed of cryoprotective agents(CPAs)and a buffer solution.The selection and ratio of CPAs in the bioink directly affect the survival of cells in the frozen state.However,the development of universal and efficient cryoprotective bioinks requires extensive experimentation.We first compared two commonly used CPA formulations via experiments in this study.Results show that the effect of using ethylene glycol as the permeable CPA was 6.07%better than that of glycerol.Two datasets were obtained and four machinelearning models were established to predict experimental outcomes.The predictive powers of multiple linear regression(MLR),decision tree(DT),random forest(RF),and artificial neural network(ANN)approaches were compared,suggesting an order of ANN>RF>DT>MLR.The final selected ANN model was then applied to another dataset.Results reveal that this machine-learning method can accurately predict the effects of cryoprotective bioinks composed of different CPAs.Outcomes also suggest that the formulations presented here have universality.Our findings are likely to greatly accelerate research and development on the use of bioinks for cryobioprinting.展开更多
This paper describes the realization of a homogeneous dielectric barrier discharge(DBD)in argon at atmospheric pressure.The effect of the morphology of the dielectric surface(especially the dielectric surface covered ...This paper describes the realization of a homogeneous dielectric barrier discharge(DBD)in argon at atmospheric pressure.The effect of the morphology of the dielectric surface(especially the dielectric surface covered by hollow ceramic beads(99%Al_(2)O_(3))with different diameters)on discharge is investigated.With different dielectrics,the argon DBD presents two discharge modes:a filamentary mode and a homogeneous mode.Fast photography shows that the filamentary mode operates in a streamer discharge,and the homogeneous mode operates in a Townsend discharge regime.It is found that a homogeneous discharge can be generated within a certain voltage range.The voltage amplitude range decreases,and the breakdown voltage increases with the increase in the mean diameter of the ceramic beads.Waveforms of the total current and optical emission signal present stochastic pulses per half voltage cycle for the filamentary mode,whereas there is one single hump per half voltage cycle for the homogeneous mode.In the homogeneous mode,the intensity of the optical emission decreases with the mean diameter of the ceramic beads.The optical emission spectrum is mainly composed of atomic lines of argon and the second positive system of molecular nitrogen.It reveals that the electron density decreases with the increasing mean diameter of the ceramic beads.The vibrational temperature increases with the increasing mean diameter of the ceramic beads.It is believed that a large number of microdischarges are formed,and smaller ceramic beads have a larger activation surface area and more point discharge.Electrons liberated in the shallow well and electrons generated from microdischarges can increase the secondary electron emission coefficient of the cathode and provide initial electrons for discharge continuously.Therefore,the breakdown electric field is reduced,which contributes to easier generation of homogeneous discharge.This is confirmed by the simulation results.展开更多
Leopard coral groupers(Plectropomus leopardus),commercially bred in South China,are a significant economical fish species.In this study,by means of quantitative real-time PCR(qRT-PCR)technology,we assessed the stabili...Leopard coral groupers(Plectropomus leopardus),commercially bred in South China,are a significant economical fish species.In this study,by means of quantitative real-time PCR(qRT-PCR)technology,we assessed the stability of six common reference genes expression and selected the appropriate reference genes in leopard coral groupers with or without Vibrio harveyi stimulation at different time points.These data produced by qRT-PCR was handled via the geNorm,NormFinder,and BestKeeper software.The results revealed all the examined reference genes had manifest tissue-specific expression in different tissues.Prior to V.harveyi stimulation,RPL13 gene was the appropriate reference gene among eleven tissue types(blood,spleen,hepatopancreas,kidney,stomach,gill,heart,skin,muscle,intestine,brain)in leopard coral groupers.Under V.harveyi stimulation,the most reliable reference genes varied from tissue to tissue and were closely hinged upon different time points.At 6-h post-bacterial injection,the appropriate reference genes in hepatopancreas,spleen,kidney,and gill were Actin,B2M,UBCE,and Actin,respectively.At 9-and 12-h post-bacterial injection,the appropriate reference genes in hepatopancreas,spleen,kidney,and gill were RPL13,Actin,Actin,and Actin,respectively.If one reference gene was preferable,RPL13,Actin,Actin,and Actin could be selected as the reference gene in hepatopancreas,spleen,kidney,and gill of leopard coral groupers after V.harveyi infection,respectively.Expression profiles of two target genes(IL-6 and NK-lysin)were used to further validate reliability of these selected appropriate candidates.This study will lay a solid foundation for the future research on qRTPCR analysis of gene expression in leopard coral groupers.展开更多
文摘BACKGROUND Glucose imbalance is common in total parenteral nutrition(TPN).Hypoglycemia seems to be less frequent than hyperglycemia,but it influences the clinical outcome to a greater extent.Therefore,it should be effectively prevented and treated.However,there is no relevant report on how to treat hypoglycemia caused by TPN in patients with liver cell injury.CASE SUMMARY We present three patients with liver cell injury who developed severe hypo-glycemia during or after TPN infusion.The causes of severe hypoglycemia and glucose-raising strategies were discussed.According to the physiological charac-teristics of the hepatocellular injury,the ratio of nutrition components prescribed in TPN was appropriately adjusted for the three cases.We simultaneously reduced the dose of insulin and fat emulsion,and increased the dose of glucose in TPN.The blood glucose level was restored to normal range and clinical symptoms were eliminated.CONCLUSION When hypoglycemia occurs during or after TPN in patients with hepatocellular injury,physicians need to simultaneously reduce insulin and fat emulsion,and increase glucose,and correct severe hypoglycemia in time to reduce its adverse consequences.
文摘Objective:To investigate the therapeutic effect of Biling Weitong Granules combined with oryz-aspergillus enzyme and pancreatin tablets on patients with reflux esophagitis with functional dyspepsia.Methods:Sixty patients diagnosed with reflux esophagitis with functional dyspepsia who were admitted to the Affiliated Hospital of Hebei University between June 2020 and June 2023 were selected and divided into two groups:the control group and the observation group,each consisting of 30 cases.The control group received oryz-aspergillus enzyme and pancreatin tablets only,while the observation group received Biling Weitong Granules in addition to the tablets.The clinical efficacy,Chinese medicine syndrome points,esophageal kinetic indexes,gastrointestinal hormone levels,and therapeutic safety of both groups were evaluated.Results:The total efficiency of the observation group reached 93.33%,significantly higher than the 73.33%of the control group(P<0.05).After treatment,patients in the observation group exhibited significantly lower scores for Chinese medicine symptoms such as early satiety,belching,abdominal distension,abdominal pain,and loss of appetite compared to the control group(P<0.05).Furthermore,the observation group showed significantly higher upper esophageal sphincter pressure,lower esophageal sphincter pressure,and distal esophageal contraction scores compared to the control group(P<0.05).Additionally,levels of gastric motility hormone,vasoactive intestinal peptide,and gastrin were significantly higher in the observation group compared to the control group(P<0.05).Throughout the treatment period,there was no significant difference in the incidence of adverse reactions between the two groups,indicating comparable safety of the two treatment modalities(P>0.05).Conclusion:The combination of Biling Weitong Granules with oryz-aspergillus enzyme and pancreatin tablets demonstrates significant efficacy in the treatment of reflux esophagitis with functional dyspepsia,with a better safety profile.This finding warrants further clinical promotion.

文摘Traditional microtubule inhibitors fail to significantly enhance+e effect of colorectal cancer;hence,new and efficient strategies are necessary.In+is study,a supramolecular nanoreactor(DOC@TA-Fe^(3+))based on tannic acid(TA),iron ion(Fe^(3+)),and docetaxel(DOC)wi+microtubule inhibition,reactive oxygen species(ROS)generation,and gluta+ione peroxidase 4(GPX4)inhibition,is prepared for ferroptosis/apoptosis treatment.After internalization by CT26 cells,+e DOC@TA-Fe^(3+)nanoreactor escapes from+e lysosomes to release payloads.+e subsequent Fe^(3+)/Fe^(2+)conversion mediated by TA reducibility can trigger+e Fenton reaction to enhance+e ROS concentration.Additionally,Fe^(3+)can consume gluta+ione to repress+e activity of GPX4 to induce ferroptosis.Meanwhile,+e released DOC controls microtubule dynamics to activate+e apoptosis pa+way.+e superior in vivo antitumor efficacy of DOC@TA-Fe^(3+)nanoreactor in terms of tumor grow+inhibition and improved survival is verified in CT26 tumor-bearing mouse model.+erefore,+e nanoreactor can act as an effective apoptosis and ferroptosis inducer for application in colorectal cancer+erapy.

文摘Proteomic characterization of plasma is critical for the development of novel pharmacodynamic biomarkers.However,the vast dynamic range renders the profiling of proteomes extremely challenging.Here,we synthesized zeolite NaY and developed a simple and rapid method to achieve comprehensive and deep profiling of the plasma proteome using the plasma protein corona formed on zeolite NaY.Specifically,zeolite NaY and plasma were co-incubated to form plasma protein corona on zeolite NaY(NaY-PPC),followed by conventional protein identification using liquid chromatography-tandem mass spectrometry.NaY was able to significantly enhance the detection of low-abundance plasma proteins,minimizing the“masking”effect caused by high-abundance proteins.The relative abundance of middleand low-abundance proteins increased substantially from 2.54%to 54.41%,and the top 20 highabundance proteins decreased from 83.63%to 25.77%.Notably,our method can quantify approximately 4000 plasma proteins with sensitivity up to pg/mL,compared to only about 600 proteins identified from untreated plasma samples.A pilot study based on plasma samples from 30 lung adenocarcinoma patients and 15 healthy subjects demonstrated that our method could successfully distinguish between healthy and disease states.In summary,this work provides an advantageous tool for the exploration of plasma proteomics and its translational applications.

文摘AIM:To compare the clinical outcomes of wavefront guided femtosecond LASIK(WFG LASIK)and conventional femtosecond LASIK(NWFG LASIK)in eyes with myopia and myopia astigmatism.METHODS:This was a retrospective,nonrandomized,comparative investigation enrolling 236 eyes of 122 patients(18-50 y)with low&moderate and high myopia.The WFG group including 97 eyes(50 patients)undergone WFG LASIK and the NWFG group including 139 eyes(72 patients)undergone conventional LASIK.Mean efficacy index,high order aberrations(HOAs),pupil size and the quality of visual questionnaire were evaluated 6 mo postoperatively.RESULTS:There is no difference between WFG group(-0.054±0.049 in logMAR)and NWFG group(-0.040±0.056)in uncorrected distance visual acuity(UDVA)postoperatively.The myopia astigmatism is higher in WFG group than that in NWFG group(P<0.05).However,the mean efficacy index(MEI)in the WFG group(1.09±0.106)is better than that in the NWFG group(1.036±0.124;P<0.001).Increased HOAs were observed in NWFG group(0.30±0.196)than that in WFG group(0.146±0.188;P<0.001).The pupil size is larger in WFG group(5.15±0.76 mm)than that in NWFG group(4.32±0.52 mm).The patients are satisfied with the clinical surgery,yet WFG group showed better visual quality using the questionnaire survey.Meanwhile,high myopia would result in worse MEI,HOAs and visual quality than low&moderate myopia.CONCLUSION:WFG and NWFG FS-LASIK are both effective and safe procedures to correct low&moderate and high myopia,but WFG FS-LASIK gives a better postoperative MEI,aberrometric control and predictable outcome.Meanwhile,WFG FS-LASIK is better than NWFG FS-LASIK in correction of myopia astigmatism.Low&moderate myopia allow better clinical outcomes than high myopia using any surgical method.

文摘BACKGROUND Very few studies have been published on the hemodynamic changes associated with spinal anesthesia induced with ropivacaine during cesarean deliveries in preeclamptic women.AIM To record and analyze hemodynamic data in women with preeclampsia undergoing cesarean delivery after spinal anesthesia induced with ropivacaine.METHODS Ten eligible women with preeclampsia were enrolled in this prospective observational study.Spinal anesthesia was performed with 2.4 mL of 0.5%ropivacaine.Hemodynamic changes were then analyzed at multiple time points.The hemodynamic responses to vasopressor interventions and uterotonic agents,as well as maternal and neonatal outcomes were also recorded.RESULTS Stable hemodynamic trends were observed in this study.Cardiac output(CO)and stroke volume increased mildly during surgery.In contrast,mean arterial pressure and systemic vascular resistance showed a moderate decrease from induction until the end of surgery.Central venous pressure dramatically increased after delivery.Oxytocin administration was associated with the most significant hemodynamic fluctuations during surgery,namely,an increase in CO and heart rate.Phenylephrine intervention was only required in three patients,and caused an increase in mean arterial pressure and systemic vascular resistance along with a decrease in heart rate,stroke volume,and CO.No maternal and neonatal complications were observed during this study,except transient episodes of hypotension.CONCLUSION Spinal anesthesia for caesarian delivery with ropivacaine in women with preeclampsia is linked to modest hemodynamic changes of no clinical significance in this study.Careful cardiovascular monitoring is still recommended,particularly after the delivery of the fetus or the use of oxytocin.

The existence of multiple solutions and its condi- tions were established by taking into account exponentially small terms in matched asymptotic expansion. The correctness of the analytical predictions was verified by numerical results.

文摘Partitioning the respiratory components of soil surface CO_2 efflux is important in understanding carbon turnover and in identifying the soil carbon sink/source function in response to land-use change. The sensitivities of soil respiration components on changing climate patterns are currently not fully understood. We used trench and isotopic methods to separate total soil respiration into autotrophic(RA) and heterotrophic components(R_H). This study was undertaken on a Robinia pseudoacacia L. plantation in the southern Taihang Mountains, China. The fractionation of soil^(13)CO_2 was analyzed by comparing the δ^(13) C of soil CO_2 extracted from buried steel tubes with results from Gas Vapor Probe Kits at a depth of 50 cm at the preliminary test(2.03%). The results showed that the contribution of autotrophic respiration(f R_A) increased with increasing soil depth.The contribution of heterotrophic respiration(f R_H) declined with increasing soil depth. The contribution of autotrophic respiration was similar whether estimated by the trench method(f RA, 23.50%) or by the isotopic method in which a difference in value of^(13) C between soil and plant prevailed in the natural state(R_C, 21.03%). The experimental error produced by the trench method was insignificant as compared with that produced by the isotopic method, providing a technical basis for further investigations.

文摘The human gut microbiome has primarily been studied through the use of fecal samples,a practice that has generated vital knowledge on the composition and functional capacities of gastrointestinal microbial communities.However,this reliance on fecal materials limits the investigation of microbial dynamics in other locations along the gastrointestinal tract(in situ),and the infrequent availability of fecal samples prevents analysis at finer temporal scales(e.g.,hours).In our study,we utilized colonic transendoscopic enteral tubing,a technology originally developed for fecal microbiota transplantation,to sample the ileocecal microbiome twice daily;metagenomic and metatranscriptomic analyses were then conducted on these samples.A total of 43 ileocecal and 28 urine and fecal samples were collected from five healthy volunteers.The ileocecal and fecal microbiomes,as profiled in the five volunteers,were found to be similar in metagenomic profiling,yet their active genes(metatranscriptome)were found to be highly distinct.Both microbiomes were perturbed after laxative exposure;over time,they exhibited reduced dissimilarity to their pre-treatment state,thereby demonstrating resilience as an innate property of the gut microbiome,although they did not fully recover within our observation time window.Sampling of the ileocecal microbiome during the day and at night revealed the existence of diurnal rhythms in a series of bacterial species and functional pathways,particularly those related to short-chain fatty acid production,such as Propionibacterium acnes and coenzyme A biosynthesis Ⅱ.Autocorrelation analysis and fluctuations decomposition further indicated the significant periodicity of the diurnal oscillations.Metabolomic profiling in the fecal and urine samples mirrored the perturbance and recovery in the gut microbiome,indicating the crucial contribution of the gut microbiome to many key metabolites involved in host health.This study provides novel insights into the human gut microbiome and its inner resilience and diurnal rhythms,as well as the potential consequences of these to the host.
文摘Golden mussel Limnoperna fortunei(Dunker 1857) is a filter-collector species of fresh water mussel originating from southern China. In the water transfer tunnels from the East River to Shenzhen and Hong Kong, golden mussels attach to the walls of pipelines and gates, causing serious biofouling, increased flow resistance, and resulted in corrosion of the tunnel wall. Golden mussel has very high environmental adaptability and may colonize habitats with low dissolved oxygen and a wide range of trophic levels. The colonization process of the species on solid surface was studied in the Xizhijiang River, a tributary of the East River and the main water resource of Shenzhen from March 2010 to April 2011. The results showed that the golden mussel completed three generations and reproduced six cohorts per year in the tropic zone. Water temperature was the controlling factor for the growth rate and maturity of each cohort. Based on the results, an ecological method for controlling the invasion of golden mussels in water transfer tunnels was proposed.

文摘AIM: To identify the disease-causing gene mutation in a Chinese pedigree with autosomal dominant cone-rod dystrophy(ad CORD).· METHODS: A southern Chinese ad CORD pedigree including 9 affected individuals was studied. Whole-exome sequencing(WES), coupling the Agilent whole-exome capture system to the Illumina Hi Seq 2000 DNA sequencing platform was used to search the specific gene mutation in 3 affected family members and 1unaffected member. After a suggested variant was found through the data analysis, the putative mutation was validated by Sanger DNA sequencing of samples from all available family members.· RESULTS: The results of both WES and Sanger sequencing revealed a novel nonsense mutation c.C766T(p.Q256X) within exon 5 of CRX gene which was pathogenic for ad CORD in this family. The mutation could affect photoreceptor-specific gene expression with a dominant-negative effect and resulted in loss of the OTX tail, thus the mutant protein occupies the CRX-binding site in target promoters without establishing an interaction and, consequently, may block transactivation.·CONCLUSION: All modes of Mendelian inheritance in CORD have been observed, and genetic heterogeneity is a hallmark of CORD. Therefore, conventional genetic diagnosis of CORD would be time-consuming and laborintensive. Our study indicated the robustness and cost-effectiveness of WES in the genetic diagnosis of CORD.
文摘Herein,we report a novel sensor to detect trypsin using a purpose-designed fluorescein-labelled peptide with negatively charged carbon nanoparticles(CNPs)modified by acid oxidation.The fluorescence of the fluorescein-labelled peptide was quenched by CNPs.The sensor reacted with trypsin to cleave the peptide,resulting in the release of the dye moiety and a substantial increase in fluorescence intensity,which was dose-and time-dependent,and trypsin could be quantified accordingly.Correspondingly,the biosensor has led to the development of a convenient and efficient fluorescent method to measure trypsin activity,with a detection limit of 0.7 mg/mL.The method allows rapid determination of trypsin activity in the normal and acute pancreatitis range,suitable for point-of-care testing.Furthermore,the applicability of the method has been demonstrated by detecting trypsin in spiked urine samples.
文摘Objective: To investigate the correlation of serum Hp-specific antibody and anti-HP-CagA antibody with coronary artery lesions in patients with coronary heart disease. Methods: A total of 119 patients with coronary heart disease were enrolled in coronary heart disease group and 100 healthy subjects were enrolled in normal control group. The differences in serum HP-IgG and anti-HP-CagA-IgG contents as well as lipid metabolism index, platelet function index and inflammatory mediator contents were compared between the two groups, and correlation analysis was used to determine the inner link of serum HP-IgG and anti-HP-CagA-IgG contents with coronary artery lesions in patients with coronary heart disease. Results:Serum HP-IgG and anti-HP-CagA-IgG contents of CHD group were higher than those of control group;peripheral blood lipid metabolism index TC content was higher than that of control group whereas HDL-C and apoA1 contents were lower than those of control group;serum platelet function indexes P-selectin, PAF and PDGF contents were higher than those of control group;serum inflammatory mediators hs-CRP, IL-8, IL-17 and TNF-α contents were higher than those of control group. Pearson correlation analysis showed that serum HP-IgG and anti-HP-CagA-IgG contents in patients with coronary heart disease were closely related to the degree of coronary artery lesions. Conclusion: Serum HP-IgG and anti-HP-CagA-IgG contents were extremely high in patients with coronary heart disease, and the specific contents are closely related to coronary artery lesions.

文摘With the advancement of network communication technology,network traffic shows explosive growth.Consequently,network attacks occur frequently.Network intrusion detection systems are still the primary means of detecting attacks.However,two challenges continue to stymie the development of a viable network intrusion detection system:imbalanced training data and new undiscovered attacks.Therefore,this study proposes a unique deep learning-based intrusion detection method.We use two independent in-memory autoencoders trained on regular network traffic and attacks to capture the dynamic relationship between traffic features in the presence of unbalanced training data.Then the original data is fed into the triplet network by forming a triplet with the data reconstructed from the two encoders to train.Finally,the distance relationship between the triples determines whether the traffic is an attack.In addition,to improve the accuracy of detecting unknown attacks,this research proposes an improved triplet loss function that is used to pull the distances of the same class closer while pushing the distances belonging to different classes farther in the learned feature space.The proposed approach’s effectiveness,stability,and significance are evaluated against advanced models on the Android Adware and General Malware Dataset(AAGM17),Knowledge Discovery and Data Mining Cup 1999(KDDCUP99),Canadian Institute for Cybersecurity Group’s Intrusion Detection Evaluation Dataset(CICIDS2017),UNSW-NB15,Network Security Lab-Knowledge Discovery and Data Mining(NSL-KDD)datasets.The achieved results confirmed the superiority of the proposed method for the task of network intrusion detection.

文摘Cryobioprinting has tremendous potential to solve problems to do with lack of shelf availability in traditional bioprinting by combining extrusion bioprinting and cryopreservation.In order to ensure the viability of cells in the frozen state and avoid the possible toxicity of dimethyl sulfoxide(DMSO),DMSO-free bioink design is critical for achieving successful cryobioprinting.A nontoxic gelatin methacryloyl-based bioink used in cryobioprinting is composed of cryoprotective agents(CPAs)and a buffer solution.The selection and ratio of CPAs in the bioink directly affect the survival of cells in the frozen state.However,the development of universal and efficient cryoprotective bioinks requires extensive experimentation.We first compared two commonly used CPA formulations via experiments in this study.Results show that the effect of using ethylene glycol as the permeable CPA was 6.07%better than that of glycerol.Two datasets were obtained and four machinelearning models were established to predict experimental outcomes.The predictive powers of multiple linear regression(MLR),decision tree(DT),random forest(RF),and artificial neural network(ANN)approaches were compared,suggesting an order of ANN>RF>DT>MLR.The final selected ANN model was then applied to another dataset.Results reveal that this machine-learning method can accurately predict the effects of cryoprotective bioinks composed of different CPAs.Outcomes also suggest that the formulations presented here have universality.Our findings are likely to greatly accelerate research and development on the use of bioinks for cryobioprinting.

文摘This paper describes the realization of a homogeneous dielectric barrier discharge(DBD)in argon at atmospheric pressure.The effect of the morphology of the dielectric surface(especially the dielectric surface covered by hollow ceramic beads(99%Al_(2)O_(3))with different diameters)on discharge is investigated.With different dielectrics,the argon DBD presents two discharge modes:a filamentary mode and a homogeneous mode.Fast photography shows that the filamentary mode operates in a streamer discharge,and the homogeneous mode operates in a Townsend discharge regime.It is found that a homogeneous discharge can be generated within a certain voltage range.The voltage amplitude range decreases,and the breakdown voltage increases with the increase in the mean diameter of the ceramic beads.Waveforms of the total current and optical emission signal present stochastic pulses per half voltage cycle for the filamentary mode,whereas there is one single hump per half voltage cycle for the homogeneous mode.In the homogeneous mode,the intensity of the optical emission decreases with the mean diameter of the ceramic beads.The optical emission spectrum is mainly composed of atomic lines of argon and the second positive system of molecular nitrogen.It reveals that the electron density decreases with the increasing mean diameter of the ceramic beads.The vibrational temperature increases with the increasing mean diameter of the ceramic beads.It is believed that a large number of microdischarges are formed,and smaller ceramic beads have a larger activation surface area and more point discharge.Electrons liberated in the shallow well and electrons generated from microdischarges can increase the secondary electron emission coefficient of the cathode and provide initial electrons for discharge continuously.Therefore,the breakdown electric field is reduced,which contributes to easier generation of homogeneous discharge.This is confirmed by the simulation results.

文摘Leopard coral groupers(Plectropomus leopardus),commercially bred in South China,are a significant economical fish species.In this study,by means of quantitative real-time PCR(qRT-PCR)technology,we assessed the stability of six common reference genes expression and selected the appropriate reference genes in leopard coral groupers with or without Vibrio harveyi stimulation at different time points.These data produced by qRT-PCR was handled via the geNorm,NormFinder,and BestKeeper software.The results revealed all the examined reference genes had manifest tissue-specific expression in different tissues.Prior to V.harveyi stimulation,RPL13 gene was the appropriate reference gene among eleven tissue types(blood,spleen,hepatopancreas,kidney,stomach,gill,heart,skin,muscle,intestine,brain)in leopard coral groupers.Under V.harveyi stimulation,the most reliable reference genes varied from tissue to tissue and were closely hinged upon different time points.At 6-h post-bacterial injection,the appropriate reference genes in hepatopancreas,spleen,kidney,and gill were Actin,B2M,UBCE,and Actin,respectively.At 9-and 12-h post-bacterial injection,the appropriate reference genes in hepatopancreas,spleen,kidney,and gill were RPL13,Actin,Actin,and Actin,respectively.If one reference gene was preferable,RPL13,Actin,Actin,and Actin could be selected as the reference gene in hepatopancreas,spleen,kidney,and gill of leopard coral groupers after V.harveyi infection,respectively.Expression profiles of two target genes(IL-6 and NK-lysin)were used to further validate reliability of these selected appropriate candidates.This study will lay a solid foundation for the future research on qRTPCR analysis of gene expression in leopard coral groupers.
