While these platforms offer avenues for self-expression and community support, they concurrently harbor negative impacts, fostering antisocial behaviors like phishing, impersonation, hate speech, cyberbullying, cyberstalking, cyberterrorism, fake news propagation, spamming, and fraud. Notably, individuals also leverage these platforms to connect with authorities and seek aid during disasters. The overarching objective of this research is to address the dual nature of network platforms by proposing innovative methodologies aimed at enhancing their positive aspects and mitigating their negative repercussions. To achieve this, the study introduces a weight learning method grounded in multi-linear attribute ranking. This approach serves to evaluate the significance of attribute combinations across all feature spaces. Additionally, a novel clustering method based on tensors is proposed to elevate the quality of clustering while effectively distinguishing selected features. The methodology incorporates a weighted average similarity matrix and optionally integrates weighted Euclidean distance, contributing to a more nuanced understanding of attribute importance. The analysis of the proposed methods yields significant findings. The weight learning method proves instrumental in discerning the importance of attribute combinations, shedding light on key aspects within feature spaces. Simultaneously, the clustering method based on tensors exhibits improved efficacy in enhancing clustering quality and feature distinction. This not only advances our understanding of attribute importance but also paves the way for more nuanced data analysis methodologies. In conclusion, this research underscores the pivotal role of network platforms in contemporary society, emphasizing their potential for both positive contributions and adverse consequences. The proposed methodologies offer novel approaches to address these dualities, providing a foundation for future research and practical applications. Ultimately, this study contributes to the ongoing discourse on optimizing the utility of network platforms while minimizing their negative impacts.展开更多

In this study, the mechanisms of wild pink bayberry free phenolic extract(WPBFE)inhibiting the proliferation and inducing apoptotic of MDA-MB-231 breast cancer cells was examined. The main phenolic acids and flavonols in WPBFE were gallic acid((18.83 ± 0.44)μg/g FW)and myricetin((1.52 ± 0.05)μg/g FW), respectively. The maximum inhibition rate of WPBFE at non-cytotoxicity dose(below 80 mg/mL)was 81%. Western blotting analysis showed that WPBFE could cause the arrest of cell cycle in G0/G1 phase by down-regulating expression levels of PCNA, CDK4, cyclin D1 and up-regulating the expression level of p21. Meanwhile, WPBFE induced apoptosis through initiating the mitochondrial death pathway by up-regulating cleaved caspase-3 and enhancing the ratio of Bax/Bcl-2, with the maximum expression levels of 1.29 and 2.03 folds that of control group, respectively. Further study of the upstream protein, we found that WPBFE down-regulated TRAF2, while upregulated p-ASK1, p-p38 and p-p53. Furthermore, WPBFE could down-regulate the expression of p-PI3K and p-Akt. These observations indicated that WPBFE might play an anticancer role through regulating the p38 MAPK together with PI3K/Akt pathway.展开更多

Ultimately, this study contributes to the ongoing discourse on optimizing the utility of network platforms while minimizing their negative impacts.

文摘AIM:To evaluate the regulation of the aberrant expression of collagen typeⅣalpha 1 chain(COL4A1)in the development of age-related cataract(ARC).METHODS:Quantitative reverse transcription-polymerase chain reaction(qRT-PCR)and Western blot analysis were employed to evaluate the expression of COL4A1 in ARC patients and healthy controls. The proliferation, apoptosis, cell cycle and epithelial-mesenchymal transition(EMT)of human lens epithelial cell(HLE-B3)were further analyzed under the condition of COL4A1 gene silence.Alteration of gene expression at mRNA level after knockdown COL4A1 were also evaluated by qRT-PCR on HLE-B3 cells.RESULTS:The aberrant expression of COL4A1 was identified a clinically associated with the ARC.Silencing of COL4A1 promoted the apoptosis and inhibited the proliferation of HLE-B3 by blocking the cell cycle.Moreover,COL4A1 gene silence didn’t affect the cytoskeleton of HLE-B3 but down-regulated the Collagen typeⅣAlpha 2 Chain(COL4A2),paired box 6(PAX6),procollagen-lysine 2-oxoglutarate 5-dioxygenases 1(PLOD1)and procollagenlysine 2-oxoglutarate 5-dioxygenases 2(PLOD2)expression levels in HLE-B3 cells.Silencing the COL4A1 gene induced EMT of the HLE-B3 cells by promoting the transforming growth factor beta(TGF-β)expression.CONCLUSION:Silencing of COL4A1 induces S-phase arrest, also inhibits the proliferation and enhance HLE-B3 apoptosis and EMT,and down-regulates the expression of COL4A2,PAX6,PLOD1 and PLOD2.Thus,the expression alteration of COL4A1 may play a critical role in the pathogenesis of ARC.

文摘Domestic cats and dogs are carnivores that have evolved differentially in the nutrition and metabolism of amino acids. This article highlights both proteinogenic and nonproteinogenic amino acids.Dogs inadequately synthesize citrulline(the precursor of arginine)from glutamine, glutamate, and proline in the small intestine.Although most breeds of dogs have potential for adequately converting cysteine into taurine in the liver,a small proportion(1.3%-2.5%)of the Newfoundland dogs fed commercially available balanced diets exhibit a deficiency of taurine possibly due to gene mutations.Certain breeds of dogs(e.g.,golden retrievers)are more prone to taurine deficiency possibly due to lower hepatic activities of cysteine dioxygenase and cysteine sulfinate decarboxylase.De novo synthesis of arginine and taurine is very limited in cats. Thus, concentrations of both taurine and arginine in feline milk are the greatest among domestic mammals.Compared with dogs, cats have greater endogenous nitrogen losses and higher dietary requirements for many amino acids(e.g.,arginine,taurine,cysteine,and tyrosine),and are less sensitive to amino acid imbalances and antagonisms.Throughout adulthood, cats and dogs may lose 34%and 21%of their lean body mass, respectively.Adequate intakes of high-quality protein(i.e.,32%and 40%animal protein in diets of aging dogs and cats, respectively;dry matter basis)are recommended to alleviate aging-associated reductions in the mass and function of skeletal muscles and bones.Pet-food grade animal-sourced foodstuffs are excellent sources of both proteinogenic amino acids and taurine for cats and dogs, and can help to optimize their growth, development, and health.

文摘Background:In China, cage systems with a high space utilization have gradually replaced ground litter systems, but the disease incidence of chickens in cages is higher.Broilers in the ground litter pens may be stimulated by more environmental microbes during the growth process and show strong immune function and status, but knowledge of which microbes and their metabolites play an immunomodulatory role is still limited. This study aimed to explore the differences and correlations in the immune function, gut microbiota and metabolites and the importance of gut microbiota of broilers raised in cages and ground litter pens. Methods:The experiment involved a 2×2 factorial arrangement, with rearing systems(cages or ground litter pens)and antibiotic treatment(with or without broad-spectrum antibiotics in drinking water)as factors. Results:The results showed that, compared with the cage group, the ground litter broilers had stronger nonspecific immune function(Macrophages%and NO in blood),humoral immune function(IgG in blood,LPS stimulation index in ileum)and cellular immune function(T%,Tc%,ConA stimulation index and cytokines in blood).Antibiotic(ABX)treat-ment significantly reduced nonspecific immune function(Macrophages%and NO in blood,iNOS and Mucin2 mRNA expression in ileum),humoral immune function(IgG in blood and sIgA in ileum)and cellular immune function(T%and cytokines in blood,Th and Tc ratio,TLRs and cytokines mRNA expression in ileum).Furthermore, the ground litter broil-ers had higherαdiversity of microbiota in ileum. The relative abundance of Staphylococcus,Jeotgalicoccus,Jeotgalibaca and Pediococcus in the ileum of ground litter broilers were higher.ABX treatment significantly reduced theαdiversity of ileal microbiota, with less Chloroplast and Mitochondria. In addition, the levels of acetic acid, isobutyric acid, kynurenic acid and allolithocholic acid in the ileum of ground litter broilers were higher.Spearman correlation analysis showed that Jeotgalibaca,Pediococcus, acetic acid, kynurenic acid and allolithocholic acid were related to the immune function. Conclusions:There were more potential pathogens, litter breeding bacteria,short-chain fatty acids, kynurenine, allolithocholic acid and tryptophan metabolites in the ileum of broilers in ground litter pens, which may be the reason for its stronger immune function and status.

文摘Background:Patients with pancreatic ductal adenocarcinoma(PDAC)who undergo surgical resection and receive effective chemotherapy have the best chance for longterm survival.Unfortunately, because of the heterogeneity of pancreatic cancer,it is difficult to find a personalized treatment strategy for patients.Organoids are ideal preclinical models for personalized medicine. Therefore,we explore the cultivation conditions and construction methods of PDAC organoid models to screen the individualized therapy strategy. Methods:Fresh PDAC tissues from surgical resection were collected and digested with digestive enzymes;then the tumor cells were embedded in Matrigel with a suitable medium to establish the PDAC organoid models. The genetic stability of the organoids was analyzed using whole exon sequencing;hematoxylin and eosin staining and immunohistochemistry of organoids were performed to analyze their consistency with the pathological morphology of the patient's tumor tissue; After 2 days of organoid culture,we selected four commonly used clinical chemotherapy drugs for single or combined treatment to analyze drug sensitivity. Results:Two cases of PDAC organoid models were successfully established, and the results of their pathological characteristics and exome sequencing were consistent with those of the patient's tumor tissue. Both PDAC organoids showed more sensitivity to gemcitabine and cisplatin, and the combined treatment was more effective than monotherapy. Conclusion:Both organoids better retained the pathological characteristics, genomic stability, and heterogeneity with the original tumor.Individual PDAC organoids exhibited different sensitivities to the same drugs. Thus, this study provided ideal experimental models for screening individualized therapy strategy for patients with PDAC.

文摘Based on the underground powerhouse of Shuangjiangkou hydropower station,Octree theory is adopted to define the indices of the microseismic(MS)spatial aggregation degree and the deviation values of MS count and energy. The relationship between the MS multiple parameters and surrounding rock mass instability is established from three aspects:time, space, and strength.Supplemented by the center frequency of the signal evolution characteristics,A fuzzy comprehensive evaluation model and the evolution trend of the MS event center frequency are constructed to quantitatively describe the early warning state of the surrounding rock mass instability. The results show that the multilevel tree structure and voxels generated based on the Octree theory fit relatively well with the set of MS points in threedimensional space. The fuzzy comprehensive evaluation model based on MS spatial aggregation and MS count and energy deviation values enables three-dimensional visualization of the potential damage area and damage extent of the surrounding rock mass. The warning time and potential damage zone quantified are highly consistent with the characteristics of MS precursors, with wide recognition and field investigation results, which fully validate the rationality and applicability of the proposed method. These findings can provide references for the early warning of surrounding rock mass instability in similar underground engineering.

文摘Background:Delirium is a neurocognitive disorder characterized by an abrupt decline in attention, awareness, and cognition after surgical/illness-induced stressors on the brain. There is now an increasing focus on how cardiovascular health interacts with neurocognitive disorders given their overlapping risk factors and links to subsequent dementia and mortality. One common indicator for cardiovascular health is the heart rate response/recovery(HRR)to exercise, but how this relates to future delirium is unknown. Methods:Electrocardiogram data were examined in 38,740 middle-to older-aged UK Biobank participants(mean age=58.1 years,range:40-72 years;47.3%males)who completed a standardized submaximal exercise stress test(15-s baseline,6-min exercise, and 1-min recovery)and required hospitalization during follow-up.An HRR index was derived as the product of the heart rate(HR)responses during exercise(peak/resting HRs)and recovery(peak/recovery HRs)and categorized into low/average/high groups as the bottom quartile/middle 2 quartiles/top quartile, respectively.Associations between 3 HRR groups and new-onset delirium were investigated using Cox proportional hazards models and a2-year landmark analysis to minimize reverse causation.Sociodemographic factors, lifestyle factors/physical activity, cardiovascular risk, comorbidities, cognition, and maximal workload achieved were included as covariates. Results:During a median follow-up period of 11 years,348 participants(9/1000)newly developed delirium.Compared with the high HRR group(16/1000),the risk for delirium was almost doubled in those with low HRR(hazard ratio=1.90,95%)confidence interval(95%CI):1.30-2.79,p=0.001)and average HRR(hazard ratio=1.54,95%CI:1.07-2.22,p=0.020)).Low HRR was equivalent to being 6 years older,a current smoker,or>3 additional cardiovascular disease risks. Results were robust in sensitivity analysis, but the risk appeared larger in those with better cognition and when only postoperative delirium was considered(n=147;hazard ratio=2.66,95%CI:1.46-4.85,p=0.001).Conclusion:HRR during submaximal exercise is associated with future risk for delirium.Given that HRR is potentially modifiable,it may prove useful for neurological risk stratification alongside traditional cardiovascular risk factors.

文摘Polyphenol-rich foods have been shown to be good for cancer prevention as powerful antioxidants. These observations indicated that WPBFE might play an anticancer role through regulating the p38 MAPK together with PI3K/Akt pathway.

文摘BACKGROUND Bronchiolar adenoma(BA)and ciliated muconodular papillary tumor are rare tumors that have bilayered cell proliferation and continuous expression of p40 and CK5/6 in the basal cell layer.Diagnosis is difficult because of the limited knowledge of these tumors and their morphological similarities to malignant tumors, including invasive mucinous adenocarcinoma, especially based on the histopathology of intraoperative frozen sections. These tumors are now considered to be benign neoplasms, with malignant transformation reported in only a few cases.CASE SUMMARY A 57-year-old woman presented with a 17.0 mm×7.0 mm nodule in the lower lobe of the left lung.Hematoxylin-eosin staining and immunohistochemistry of a surgical specimen were performed. The tumor consisted of a BA area and a mucinous adenocarcinoma(MA)area. In the BA area, the tumor had a bilayered structure of luminal cells and basal cells. The basal cells were positive for CK5/6 and p40,but the MA area was negative for these biomarkers. The Ki-67 proliferation index was low(1%-2%).The patient was diagnosed with BA accompanied by MA,and had a favorable outcome.CONCLUSION The present study indicated that BA may be carcinogenic, and suggests that clinicians should be aware of its potential for malignant transformation.

文摘We report a highly efficient three-dimensional degenerated Raman sideband cooling(3D dRSC)that enhances the loading of a magnetically levitated optical dipole trap, and observe the strong atom loss due to the three-body recombination. The 3D dRSC is implemented to obtain 5×10^(7)Cs atoms with the temperature of~480 nK.The cold temperature enables 1.8×10^(7)atoms loaded into a crossed dipole trap with an optimized excessive levitation magnetic gradient.Compared to the loading of atoms from a bare magneto-optical trap or the gray-molasses cooling, there is a significant increase in the number of atoms loaded into the optical dipole trap.We derive for the three-body recombination coefficient of L_(3)=7.73×10^(-25)cm^(6)/s by analyzing the strong atom loss at a large scattering length of 1418 Bohr radius, and discover the transition from the strong three-body loss to the dominant one-body loss. Our result indicates that the lifetime of atoms in the optical dipole trap is finally decided by the one-body loss after the initial strong three-body loss.

文摘AIM:To study microRNAs(miRNAs)and their potential effects in high glucose-induced human retinal pigment epithelial cell damage.METHODS:We screened the GSE52233 miRNA expression dataset for differentially expressed miRNAs(DEMs).The target genes of the top 10 DEMs were predicted using miRWalk 2.0 database, followed by function enrichment and protein-protein interaction analysis.miRNA expression was determined in the human retinal pigment epithelial cell line ARPE-19 treated with high glucose(HG)by quantitative reverse transcription-polymerase chain reaction(qRT-PCR).Cell proliferation was determined using cell counting kit(CCK)-8 assay.Cell cycle, apoptosis, and reactive oxygen species(ROS)levels were determined by flow cytometry. The direct interaction between miRNA and targets was validated using dual-luciferase reporter assay.RESULTS:Thirty-nine DEMs were screened, and we predicted 125 miRNA-mRNA pairs for the top 10 DEMs,including 119 target genes of seven DEMs such as miR-346,which was upregulated in diabetic retinopathy(DR).miR-346 target genes were substantially enriched in the regulation of intracellular transport and retinoic acidinducible gene I(RIG-I)-like receptor signaling pathway.Expression of three upregulated and downregulated miRNAs were verified by qRT-PCR in HG-treated ARPE-19 cells.Expression of miR-346 was elevated in HG treated ARPE-19 cells in a dose-dependent manner.HG inhibited cell proliferation and induced apoptosis, which were partly reversed by transfecting an miR-346 inhibitor, which even decreased the ROS levels elevated due to HG.Argonaute 2(AGO2)was a target of miR-346.CONCLUSION:miR-346 is a key miRNA and plays an important role in HG-induced damage in human retinal pigment epithelial cells.

文摘The correlation between microbiota plays a vital role in the progression of periodontal disease. This study investigated the in situ interaction networks between periodontal pathogens in periodontal and peri-implant disease. We used quantitative real-time polymerase chain reaction and Pearson’s correlation coefficients to quantify the copy numbers and correlations of four oral core species—Fusobacterium nucleatum,Porphyromonas gingivalis,Prevotella intermedia, and Streptococcus gordonii—from 80 subgingival sites(healthy and with periodontitis or gingivitis)in patients with periodontitis, and 68 subgingival sites(healthy and with periodontitis,gingivitis,peri-implantitis,or peri-implant mucositis)in patients with implants. The highest bacterial counts were observed for Porphyromonas gingivalis and Prevotella intermedia at all the sites.Within the same cohorts, the bacterial loads were greater at diseased sites than at healthy sites.Bacterial counts did not differ among clinical sites in the same group(P>0.05)but differed between periodontitis and peri-implant mucositis sites in the two groups.Porphyromonas gingivalis,F.nucleatum, and Prevotella intermedia had strong correlations at gingivitis and healthy sites and moderate correlations at periodontitis sites in patients with periodontitis. In patients with implants,Prevotella intermedia,F.nucleatum, and S.gordonii had strong correlations only at peri-implantitis sites. Also, based on metagenomic analysis,F.nucleatum and Prevotella intermedia were significantly correlated at the subgingival plaque in peri-implantitis and periodontitis samples. Our results suggest that variations in microbe-microbe interactions in subgingival plaque reflect changes in the progression of periodontal disease, providing a new perspective for understanding the mechanisms of periodontitis and peri-implantitis.
