β-Sitosterol is a type of phytosterol that occurs naturally in plants.Previous studies have shown that it has anti-oxidant,anti-hyperlipidemic,anti-inflammatory,immunomodulatory,and anti-tumor effects,but it is unkno...β-Sitosterol is a type of phytosterol that occurs naturally in plants.Previous studies have shown that it has anti-oxidant,anti-hyperlipidemic,anti-inflammatory,immunomodulatory,and anti-tumor effects,but it is unknown whetherβ-sitosterol treatment reduces the effects of ischemic stroke.Here we found that,in a mouse model of ischemic stroke induced by middle cerebral artery occlusion,β-sitosterol reduced the volume of cerebral infarction and brain edema,reduced neuronal apoptosis in brain tissue,and alleviated neurological dysfunction;moreover,β-sitosterol increased the activity of oxygen-and glucose-deprived cerebral cortex neurons and reduced apoptosis.Further investigation showed that the neuroprotective effects ofβ-sitosterol may be related to inhibition of endoplasmic reticulum stress caused by intracellular cholesterol accumulation after ischemic stroke.In addition,β-sitosterol showed high affinity for NPC1L1,a key transporter of cholesterol,and antagonized its activity.In conclusion,β-sitosterol may help treat ischemic stroke by inhibiting neuronal intracellular cholesterol overload/endoplasmic reticulum stress/apoptosis signaling pathways.展开更多
Due to their high reliability and cost-efficiency,submarine pipelines are widely used in offshore oil and gas resource engineering.Due to the interaction of waves,currents,seabed,and pipeline structures,the soil aroun...Due to their high reliability and cost-efficiency,submarine pipelines are widely used in offshore oil and gas resource engineering.Due to the interaction of waves,currents,seabed,and pipeline structures,the soil around submarine pipelines is prone to local scour,severely affecting their operational safety.With the Yellow River Delta as the research area and based on the renormalized group(RNG)k-εturbulence model and Stokes fifth-order wave theory,this study solves the Navier-Stokes(N-S)equation using the finite difference method.The volume of fluid(VOF)method is used to describe the fluid-free surface,and a threedimensional numerical model of currents and waves-submarine pipeline-silty sandy seabed is established.The rationality of the numerical model is verified using a self-built waveflow flume.On this basis,in this study,the local scour development and characteristics of submarine pipelines in the Yellow River Delta silty sandy seabed in the prototype environment are explored and the influence of the presence of pipelines on hydrodynamic features such as surrounding flow field,shear stress,and turbulence intensity is analyzed.The results indicate that(1)local scour around submarine pipelines can be divided into three stages:rapid scour,slow scour,and stable scour.The maximum scour depth occurs directly below the pipeline,and the shape of the scour pits is asymmetric.(2)As the water depth decreases and the pipeline suspension height increases,the scour becomes more intense.(3)When currents go through a pipeline,a clear stagnation point is formed in front of the pipeline,and the flow velocity is positively correlated with the depth of scour.This study can provide a valuable reference for the protection of submarine pipelines in this area.展开更多
PD-1/PD-L1 inhibitors have emerged as standard treatments for advanced solid tumors;however,challenges such as a low overall response rate and systemic side effects impede their implementation.Hypoxia drives the remod...PD-1/PD-L1 inhibitors have emerged as standard treatments for advanced solid tumors;however,challenges such as a low overall response rate and systemic side effects impede their implementation.Hypoxia drives the remodeling of the tumor microenvironment,which is a leading reason for the failure of immunotherapies.Despite some reported strategies to alleviate hypoxia,their individual limitations constrain further improvements.Herein,a novel two-pronged strategy is pre-sented to efficiently address hypoxia by simultaneously adopting atovaquone(ATO,inhibiting oxygen consumption)and oxyhemoglobin(HbO2,directly supplement-ing oxygen)within a multifunctional aggregate termed NPs-aPD-1/HbO2/ATO.In addition to eliminating hypoxia with these two components,this smart aggre-gate also includes albumin and an ROS-responsive cross-linker as a controlled release scaffold,along with PD-1 antibody(aPD-1)for immunotherapy.Intriguingly,NPs-aPD-1/HbO2/ATO demonstrates exceptional tumor targeting in vivo,exhibit-ing≈4.2 fold higher accumulation in tumors than in the liver.Consequently,this aggregate not only effectively mitigates hypoxia and significantly assists aPD-1 immunotherapy but also simultaneously resolves the targeting and systemic toxicity issues associated with individual administration of each component.This study pro-poses substantial implications for drug-targeted delivery,addressing tumor hypoxia and advancing immunotherapy,providing valuable insights for advancing cancer treatment strategies.展开更多

The loss of a toothhas lifelong consequences for children and teenagers, but the maintenance of pulphealth and the calcific healing of multiple root fractures are rarely reported in theliterature.CASE SUMMARY This case reports healing of a permanent tooth with complicated crown–root andadditional root fractures, in which pulp health was maintained. A 10-year-old girlfell and fractured the root of her maxillary left central incisor at the cervical level.After the coronal fragment was repositioned, the tooth was splinted until thetooth was no longer mobile, 2 years later. Eight years after treatment, the toothhas remained asymptomatic with vital pulp and localized gingival overgrowth.Cone-beam computed tomography revealed not only calcified healing of the CRFbut also spontaneous healing in an additional undiagnosed root fracture. Thefracture line on the enamel could not be healed by hard tissue and formed agroove in the cervical crown. It was speculated that the groove was related to thelocalized gingival overgrowth.CONCLUSION This case provides a clinical perspective of the treatment of a tooth with acomplicated CRF and an additional root fracture.展开更多
[Objectives]This study was conducted to explore rapid and large-scale screening and detection of peste des petits ruminants(PPR),so as to provide important technical means for prevention,control and purification of PP...[Objectives]This study was conducted to explore rapid and large-scale screening and detection of peste des petits ruminants(PPR),so as to provide important technical means for prevention,control and purification of PPR.[Methods]Soluble N protein and NH fusion protein were successfully obtained in an Escherichia coli expression system by optimizing E.coli codon and expression conditions.Furthermore,based on purified soluble N protein and NH fusion protein,a double-antigen sandwich time-resolved fluorescence immunoassay method for detection of peste des petits ruminants virus(PPRV)was established.[Results]The method has high sensitivity and specificity and can specifically detect the antibody against PPRV in sheep serum,and it has no cross reaction with other related diseases.The method was used to detect 292 clinical samples,and compared with French IDVET competition ELISA kit.The coincidence rates of positive samples and negative samples from the two kinds of test kits were 92.47%and 97.26%,respectively,and the overall coincidence rate was 94.86%.The intra-group and inter-group coefficients of variation in the repeatability test were less than 10%.[Conclusions]Compared with the traditional ELISA method,the double-antigen sandwich time-resolved fluorescence immunoassay for detection of PPRV has equivalent sensitivity and specificity,and simple and rapid operation,and thus high application and popularization value.展开更多

Methods: Expression of CDH11 in...Objective: The main purpose was to investigate expression of CDH11 in pancreatic tissues and analyze its relations with clinical characteristics of patients with pancreatic cancer (PC). Methods: Expression of CDH11 in cancerous tissues and cancer adjacent tissues were detected by immunohistochemistry staining, and their associations with clinical characteristics were analyzed. The text mining methods were used to assist the study of the effect of CDH11 on prognosis. Results: A total of 79 patients with PC were enrolled in the study, including 51 (64.6%) men and 28 (35.4%) women with a median age of 62 (41 - 84). The CDH11 expression in cancerous tissues was higher than that in adjacent tissues and the expression of CDH11 in both tissues was positively correlated (P 0.999;stage II vs III & IV, P = 0.308). A higher level of CDH11 expression correlated with worse overall survival (OS) time (P = 0.015). Conclusion: CDH11 may be involved in the development of early PC and lead to poor prognosis and could be a new target molecule for early diagnosis and treatment of PC.展开更多
