Objective:To explore the role of cyclophilin A(CyPA)in sensitization of lung adenocarcinoma to radiotherapy using CRISPR/Cas9 technology.Methods:A CyPA knockout human lung adenocarcinoma cell line H1975 was established by CRISPR/Cas9 technology.Groups included a control group(wildtype),CyPA knockout group,traditional Chinese medicine(TCM)extract with Fuzhengzengxiao decoction group,and TCM extract with Fuzhengzengxiao decoction t CyPA knockout group.Each group was exposed to radiation at doses of 0,2,4,6,and 8 Gy.After 24 h,MTT assays were used to determine the survival rate of lung cancer cells and calculate radiosensitivity.The qPCR was used to measure mRNA expression of DDIT3,CDKN1A,and CDC25A associated with DNA damage repair.Results:Without irradiation,Fuzhengzengxiao decoction reduced the survival rate of lung adenocarcinoma cells(P<.0001).After irradiation,TCM extract with Fuzhengzengxiao decoction,CyPA knockout,and TCM extract with Fuzhengzengxiao decoction t CyPA knockout groups had reduced survival rates(P<.0001)and radiosensitivity was increased significantly.Expression of DDIT3,CDKN1A,and CDC25A was upregulated after knockout of CyPA(P<.0001).Expression of DDIT3 and CDC25A was increased after irradiation in wildtype cells treated with TCM extract with Fuzhengzengxiao decoction(DDIT3,P<.0001;CDC25A,P紏.0059).The TCM extract with Fuzhengzengxiao decoction t CyPA knockout group also had increased expression of DDIT3 and CDC25A after irradiation(P<.0001).Conclusion:Fuzhengzengxiao decoction significantly decreases the survival rate of lung cancer cells,and its mechanism may be related to radiosensitization by decreasing expression of CyPA and inducing G1/S cell cycle arrest. 展开更多

The biomedical model,which limits itself to finding the attributions of organic disease,is challenged by gastrointestinal(GI)symptoms.Simultaneously,physicians'attribution of GI symptoms to underlying psychological issues is not readily accepted by patients and can negatively affect the clinical rapport between doctor and patient.In reality,psychosocial aspects are involved in many functional disorders and organic diseases,not just in mental disorders.Time is overdue for gastroenterologists to recognise the inadequacy and limitations of conventional gastroenterology and consider the role of psychological,social and biological variables throughout the entire clinical course of the illness,as is shown in George Engel’s model.This review discusses the following:(1)the current challenges of using the conventional clinical model for both functional and organic GI illness,(2)the inadequacy and limitations of explaining GI symptoms simply as psychological disorders,(3)the exploration of the symptom-centred,stepped reattribution clinical model,(4)the clarification of psychosomatic medical concepts for use in gastroenterology,and(5)the significance of a systematic and interdisciplinary framework for a comprehensive psychosomatic model in gastroenterology. 展开更多

The effect of methylseleninic acid on paclitaxel efficacy in A2780 ovarian cancer cells

Objective:The role of methylseleninic acid (MSeA), a selenium compound, has been documented in cancer chemoprevention. However, the therapeutic effect of MSeA in combination with paclitaxel, a chemotherapeutic agent used to treat ovarian cancer, is unknown. In this study, we investigated the effect of combination treatment of MSeA and paclitaxel against ovarian cancer cells. Methods:Ovarian cancer cells(A2780) were treated with different concentrations of MSeA, paclitaxel alone or in combination. The individual and combined concentrations of drugs that achieved certain cells growth/death were determined using a sulforhodamine B(SRB) assay. Drug effects on cell viability were further confirmed using floating cell count and trypan blue exclusion assay. The mean Values ± standard deviation were calculated and compared between treatment groups using unpaired t test. Results: The concentration of paclitaxel alone that inhibited 50% of cell growth(IC50) was 0.5 μmol/L. This concentration increased to 1.2 μmol/L when paclitaxel was given in sequential combination with MSeA. The number of dead cells after the combination treatment did not show a significance increase when compared with drug alone. Conclusion:Pretreatment with MSeA did not enhance the paclitaxel effect against A2780 ovarian cancer cells. 展开更多

The development of lithium-metal batteries(LMBs)is seriously restricted by the out-of-control dendrites growth and infinite volume expansion.Herein,a pervasive organic-inorganic layer construction strategy is reported for the composite lithium metal anode with congener-derived organic-inorganic solid electrolyte interphase(SEI).In this strategy,the organic-inorganic Ag@polydopamine(Ag@PDA)layer is coated on the arbitrary substrates by a simple two-step method.The thin and stable congener-derived SEI is insitu formed with fewer inorganic components and more organic components during charging/discharging.The polydopamine with sufficient adhesion groups and lithiophilic Ag layer realize near-zero nucleation overpotential during lithium deposition.The low interface resistance and stable lithium deposition are achieved.Moreover,the practical areal and volumetric capacities of the composite anode with three-dimensional copper(3DCu)as the substrate are 10 mAh/cm^(2)and 1538 mAh/cm^3(vs.the mass of anode).The symmetrical cell shows very low polarization voltage(10 mV)and more than 2500 h cycles life at 1 mA/cm^(2)(1 mAh/cm^(2)).The LiNi_(0.8)Co_(0.1)Mn_(0.1)O_(2)(NCM811)-based full cells show improved capacity retention(82%)after 100 cycles at 0.5 C.The modified lithiophilic anode with congener-derived interphase provides a promising strategy to realize the next-generation dendrite-free LMBs. 展开更多

Organophosphate nerve agents(OPNAs)are lethal chemical weapons(CWs)which hydrolyze rapidly to alkyl phosphate compounds in the environment.The identification of alkyl phosphate compounds in the environment is critical for the verification of OPNAs.Currently,due to the complex sample preparation and long detection process for organophosphate nerve agent hydrolysis products(OPNAHPs),their high-throughput determination is difficult.To simplify sample preparation and detection of OPNAHPs from environmental samples,a quick,easy,cheap,effective,rugged and safe(Qu ECh ERS)method for the simultaneous purification of the five OPNAHPs in environmental samples combined with detection through direct analysis in real-time ionization source coupled with tandem mass spectrometer(DART-MS/MS)was developed.An internal standard method was applied for the quantification.The method exhibited a good linearity(R^(2)>0.991)in the range of 1.00 ng/m L-4.00μg/m L.The limits of detection and limits of quantification were 1.00-7.00 ng/m L and 10.0-20.0 ng/m L,respectively.The recoveries of OPNAHPs in environmental samples were between 65.3%and 107%,with relative standard deviations ranged from 1.6%to 19%.In addition,the sample preparation and analysis process for OPNAHPs could be completed in less than 10 min.The results demonstrated that the modified Qu ECh ERS method could quickly and efficiently remove the matrix of environmental samples.Combined with DART-MS/MS,an accurate and high-throughput detection method of OPNAHPs in environmental samples was provided.The developed method has great potential in the fields of early warning,clinical treatment and environmental decontamination. 展开更多

We experimentally demonstrate for the first time, to the best of our knowledge, an all-fiber passively mode-locked laser operation based on the nonlinear multimode interference of step-index multimode fiber. Such a structure couples the light in and out of the multimode fiber via single-mode fibers, and its physical mechanisms for saturable absorption have been analyzed theoretically based on the third-order nonlinear Kerr effect of multimode fiber. Using the nonlinear multimode interference structure with 48.8 mm length step-index multimode fiber, the modulation depth has been measured to be ~5%. The passively mode-locked laser output pulses have a central wavelength of 1596.66 nm, bandwidth of 2.18 nm, pulsewidth of ~625 fs, and fundamental repetition rate of8.726 MHz. Furthermore, the influence of total cavity dispersion on the optical spectrum, pulse width, and output power is investigated systematically by adding different lengths of single-mode fiber and dispersion compensation fiber in the laser cavity. 展开更多
