The solar flare is one of the most violent explosions,and can disturb the near-Earth space weather.Except for commonly single-peaked solar flares in soft X-ray,some special flares show intriguing a two-peak feature that is deserved much more attentions.Here,we reported a confined two-peaked solar flare and analyzed the associated eruptions using high-quality observations from Educational Adaptive-optics Solar Telescope and Solar Dynamics Observatory.Before the flare,a magnetic flux rope(MFR)formed through partially tether-cutting reconnection between two sheared arches.The flare occurred after the MFR eruption that was confined by the overlying strong field.Interestingly,a small underlying filament immediately erupted,which was possibly destabilized by the flare ribbon.The successive eruptions were confirmed by the analysis of the emission measure and the reconnection fluxes.Therefore,we suggest that the two peaks of the confined solar flare are corresponding to two episodes of magnetic reconnection during the successive eruptions of the MFR and the underlying filament. 展开更多

Rock fracture mechanics and accurate characterization of rock fracture are crucial for understanding a variety of phenomena interested in geological engineering and geoscience.These phenomena range from very large-scale asymmetrical fault structures to the scale of engineering projects and laboratory-scale rock fracture tests.Comprehensive study can involve mechanical modeling,site or post-mortem investigations,and inspection on the point cloud of the source locations in the form of earthquake,microseismicity,or acoustic emission.This study presents a comprehensive data analysis on characterizing the forming of the asymmetrical damage zone around a laboratory mixed-mode rock fracture.We substantiate the presence of asymmetrical damage through qualitative analysis and demonstrate that measurement uncertainties cannot solely explain the observed asymmetry.The implications of this demonstration can be manifold.On a larger scale,it solidifies a mechanical model used for explaining the contribution of aseismic mechanisms to asymmetrical fault structures.On a laboratory scale,it exemplifies an alternative approach to understanding the observational difference between the source location and the in situ or post-mortem inspection on the rock fracture path.The mechanical model and the data analysis can be informative to the interpretations of other engineering practices as well,but may face different types of challenges. 展开更多

Genomic selection(GS)has been widely used in livestock,which greatly accelerated the genetic progress of complex traits.The population size was one of the significant factors affecting the prediction accuracy,while it was limited by the purebred population.Compared to directly combining two uncorrelated purebred populations to extend the reference population size,it might be more meaningful to incorporate the correlated crossbreds into reference population for genomic prediction.In this study,we simulated purebred offspring(PAS and PBS)and crossbred offspring(CAB)base on real genotype data of two base purebred populations(PA and PB),to evaluate the performance of genomic selection on purebred while incorporating crossbred information.The results showed that selecting key crossbred individuals via maximizing the expected genetic relationship(REL)was better than the other methods(individuals closet or farthest to the purebred population,CP/FP)in term of the prediction accuracy.Furthermore,the prediction accuracy of reference populations combining PA and CAB was significantly better only based on PA,which was similar to combine PA and PAS.Moreover,the rank correlation between the multiple of the increased relationship(MIR)and reliability improvement was 0.60-0.70.But for individuals with low correlation(Cor(Pi,PA or B),the reliability improvement was significantly lower than other individuals.Our findings suggested that incorporating crossbred into purebred population could improve the performance of genetic prediction compared with using the purebred population only.The genetic relationship between purebred and crossbred population is a key factor determining the increased reliability while incorporating crossbred population in the genomic prediction on pure bred individuals. 展开更多

The northern South China Sea,including the Zhujiangkou Basin and the Beibuwan Basin,developed high-quality lacustrine source rocks during the Eocene rifting period.These source rocks are vital for hydrocarbon generation in the northern South China Sea.The ZhuⅠdepression in the Zhujiangkou Basin and the Beibuwan Basin typically exhibit high abundance of C_(30)4-methyl steranes.However,shales in the Eocene Wenchang Formation in the ZhuⅢdepression of the Zhujiangkou Basin contains lower quantities of high-quality lacustrine source rocks with 4-methyl steranes,which often co-elute with some pentacyclic triterpanes in gas chromatography-mass spectrometry(GC-MS).Therefore,the single 4-methylsterane parameter based on GC-MS cannot accurately distinguish organic source in the deep to semi-deep water lacustrine source rocks of the Wenchang Formation from other source rocks,thus impeding the recognition of their contributions to petroleum reservoirs.In this study,GC-MS of aliphatic hydrocarbons,palynofacies and algal identification,as well as stable carbon isotope data of organic matter were used to identify the algal species and construct the paleoclimate during deposition of the Wenchang Formation source rocks in the ZhuⅢdepression of the Zhujiangkou Basin.It is suggested that during the Wenchang Formation period,freshwater green algae prevailed in the lake,which is likely account for the relatively low content of 4-methyl steranes in the high-quality lacustrine source rocks.Controlled by the algal species,it is proposed that the content of C_(30)tetracyclic polyprenoids(TPP)can better indicate the quality of the Wenchang source rocks than C_(30)4-methyl steranes.Consequently,a relationship between the TPP index and the quality of the lacustrine source rocks in the Wenchang Formation of the ZhuⅢdepression was established.A higher TPP index indicates higher organic matter abundance and hydrogen index of the lacustrine source rocks.When applied to the origin analysis of oils in the ZhuⅢdepression,it is believed that the organic-rich deep lacustrine source rocks in the Wenchang Formation made great contribution to the transitional zone crude oils in the Wenchang A and Wenchang B depressions. 展开更多

The Educational Adaptive-optics Solar Telescope(EAST)at the Shanghai Astronomy Museum has been running routine astronomical observations since 2021.It is a 65-cm-aperture Gregorian solar telescope for scientific education,outreach,and research.The telescope system is designed in an“open”format so that the solar tower architecture can be integrated with it,and visitors can watch the observations live from inside the tower.Equipped with adaptive optics,a high-resolution imaging system,and an integral field unit spectro-imaging system,this telescope can obtain high-resolution solar images in the TiO and Hαbands,and perform spectral image reconstruction using 400 optical fibers at selected wavelengths.It can be used not only in public education and scientific outreach but also in solar physics research. 展开更多

AIM: To detect linc00675 expression in pancreatic ductal adenocarcinoma(PDAC),to analyze the relationship between the expression level of linc00675 and the clinical pathological characteristics,to explore the biological functions of linc00675,and to determine whether linc00675 has independent prognostic value in PDAC.METHODS: We studied linc00675 expression among eight histologically confirmed PDAC tissue samples and four chronic pancreatitis tissue samples through microarray screening. RT-q PCR was conducted to further investigate linc00675 expression in PDAC cell lines as well as archived tissues from a large cohort of PDAC patients. The correlations between the level of lnc00675 and clinicopathological characteristics and survival in patients with pancreatic cancer were evaluated using Correlation analysis. Univariate andmultivariate analyses were conducted to predict whether lnc00675 expression is an independent prognostic and recurrence factor in patients with pancreatic cancer. After downregulating the expression of linc00675 through si RNA,MTT assay,flow cytometry,transwell assay and Western blot were used to explore the biological function of linc00675 in proliferation,invasion,and cell cycle progression of pancreatic cancer cells. The relative molecular expression levels of epithelial-mesenchymal transition were determined by reverse transcription-polymerase chain reaction(RT-PCR) and Western blot.RESULTS: The expression of Linc00675 in PDAC tissue samples was shown to be 672 times that in chronic pancreatitis tissue samples by microarray screening(P = 3.69 × 10-5). This finding was confirmed in tumor tissues from 90 patients with PDAC compared with adjacent normal tissue samples by quantitative RTPCR. We found that linc00675 overexpression positively correlated with lymph node metastasis(P = 0.005),perineural invasion(P = 0.006),and poor survival(P < 0.001). Univariate and multivariate analyses showed that linc00675 expression served as an independent predictor of overall survival(P = 0.009). Additionally,receiver operating characteristic curve analysis showed that high linc00675 might serve as a predictor of tumor progression within 6 mo to a year after surgery. In vitro functional analysis demonstrated that knockdown of linc00675 attenuated pancreatic cancer cell proliferation and invasion as well as induced S phase arrest. Suppression of linc00675 in pancreatic cancer cells resulted can reverse the progress of epithelialmesenchymal transition.CONCLUSION: Linc00675 may function as an oncogene during PDAC development,and its expression is an independent predictor of unfavorable prognosis in patients with PDAC. 展开更多

To establish and validate a method for cell irradiation in 96-well and 6-well plates using a linear accelerator, three irradiation methods(G0 B0 F40,G0 B1.5 F40, and G180 B1.5 F40) were designed to irradiate cell culture plasticware simulated with RW3 slab phantom and polystyrene. The difference between the actual physical measured dose and the preset dose was compared among the three methods under the preparatory conditions of 2, 4, 6, 8, and 10 Gy. MDA-MB-231 cells were analyzed by using a cell proliferation assay and a clonogenic assay to verify the difference between the three cell irradiation methods on cell radiosensitivity. For each preset dose, the difference between the actual measured dose and the preset dose was the lowest for Method G0 B1.5 F40, the second lowest for Method G180 B1.5 F40, and the maximum for Method GOB0 F40. The ranges of the differences were-0.28 to 0.02%,-2.17 to-1.80%, and-4.92 to-4.55%, and 0.31 to-0.12%,-3.42 to-2.86%, and-7.31 to-6.92%,respectively, for 96-well and 6-well plates. The cell culture experiments proved that Method G0 B1.5 F40 was an accurate, effective, simple, and practical irradiation method. The most accurate and effective cell irradiation method should always be used, as it will reduce dose differences and instability factors and provide improved accuracy and comparability for laboratories researching cellular radiosensitivity. 展开更多

Objective:The purpose of this study was to observe the anti-cancer activity of Tonglian decoction(TD)on esophageal cancer(EC)cells in vitro,and to elucidate the related molecular mechanisms in the cell cycle and PI3K/Akt signaling pathway.Methods:EC9706 cells were cultured in RPMI 1640 medium supplemented with 10%calf serum at 37C in a 5%CO2 incubator.The cells were treated with rat serum containing TD or the serum of rats administered Xiaoaiping as a positive control drug.Cell proliferation was assessed by methylthiazolyldiphenyl-tetrazolium bromide assays.Cell morphology was observed under a microscope.The cell cycle was examined by flow cytometry.Protein expression in the PI3K/Akt signaling pathway was measured by western blotting.Results:TD mainly inhibited cell proliferation.Concentrations of 50%cell inhibition by rat serum containing TD or Xiaoaiping were 73.6 and 153.8 mL/mL,respectively.TD also influenced cell morphology characterized by small shrunken cells.Cell colonies became small and the cell proliferation rate was slower.In cell cycle analysis,the percentage of cells in S phase was decreased significantly by TD and Xiaoaiping compared with the blank control group(P<.05).Western blotting showed that serum containing TD strongly downregulated EGFR,PI3K,Akt,p-Akt,and mTOR expression compared with the blank control group(P<.05).Conclusion:TD could inhibit EC9706 carcinoma cell proliferation by blocking the cell cycle progression in S phase.The possible mechanism was inhibition of multiple targets in the PI3K/Akt signaling pathway by TD. 展开更多

In order to improve the availability of regional model precipitation forecast, this project intends to use the measured heavy rainfall data of dense automatic stations to carry out historical precipitation in the high resolution: the Severe Weather Automatic Nowcast System (SWAN) quantitative precipitation forecast and the High-Resolution Rapid Refresh (HRRR) regional numerical model precipitation forecast in short-term nowcasting aging. Based on the error analysis, the grid fusion technology is used to establish the measured rainfall, HRRR regional model precipitation forecast, and optical flow radar quantitative precipitation forecast (QPF) three-source fusion correction scheme, comprehensively integrate the revised forecasting effect, adjust the fusion correction parameters, establish an optimal correction plan, generate a frozen rolling update revised product based on measured dense data and short-term forecast, and put it into business operation, and perform real-time effect rolling test evaluation on the forecast product. 展开更多

Objective:To observe the effect of tetradecylene naphthalene soft capsule combined with Yishen Gubenyutong Decoction on bone metabolism and bone turnover in patients with osteoporosis and its clinical efficacy.Methods:A total of 100 patients with primary osteoporosis(all were treated by our hospital from February 2017 to June 2018)were divided into two groups.The control group(50 cases)was given to menatetrenone capsules.Treatment,observation group(50 cases)combined with Yishen Gubenqiaotongtong on the basis of the treatment of the control group,6 months for a course of treatment.The bone metabolic markers,bone mineral density changes and clinical efficacy were measured before and after treatment.Results:The total effective rate of the observation group was higher than that of the control group.After treatment,the observation group showed serum calcium(Ca),serum alkaline phosphatase(ALP)and typeⅠcollagen.The level of cross-linked C-terminal peptide(CTX-1)was lower than that of the control group.After treatment,the osteocalcin(OC)in the observation group was higher than that in the control group,and the bone mineral density(BMD)of lumbar vertebrae L2-4,femoral neck and femur trochanter in the observation group were higher than those in the control group.Conclusion:Menatetrenone soft capsule combined with Yishen Gubenyutong Decoction has good clinical effect on osteoporosis patients and it is worth further promotion in clinic. 展开更多

The sedimentary system of Kalimantan has undergone significant development since the Oligocene.Previous research have largely ignored the capacity of the Cretaceous–Eocene sediments to produce hydrocarbons,focusing instead primarily on the Oligocene–Miocene coal as the principal source rocks.Shales and coals from the outcrops in the northern margin of Kalimantan were analyzed with palynological and geochemical methods to characterize the palaeoenvironmental and palaeoecological differences between the Cretaceous–Eocene and the Oligocene–Miocene samples.The high proportion of Cheirolepidoaceae,Schizaeoisporites and Ephedripites in the pollen assemblage from the Cretaceous–Eocene outcrops reflects an arid tropical/subtropical climate.The relatively low abundances of gymnosperm-derived biomarkers including isopimarane,β-phyllocladane,β-kaurane,suggest the gymnosperm features in flora.High C_(27)/C_(29)ααα20R sterane ratios,(C_(19)–C_(29))tricyclic terpanes/C_(30)αβhopane and extremely low oleanane/C_(30)αβhopane,bicadinane T/C_(30)αβhopane,and diterpenoid abundance indicate that there was a dominance of algae relative to higher plants in the organic matter.The gymnosperm-derived biomarkers,including isopimarane,β-phyllocladane,β-kaurane,suggest that palaeovegetation during this period was dominated by gymnosperms.The saline and reducing conditions in the bathyal and abysmal sea,evidenced by rather low Pr/Ph and high Gammarerane index,are beneficial for the preservation of hydrogen-rich organic matter.It is presumed that the Cretaceous–Eocene shales had great hydrocarbon generation potential in the southern South China Sea.During the period of Oligocene to Miocene in the Zengmu Basin and the Baram-Sabah Basin,the climate changed to a dominant humid and warm condition,which is corroborated by abundant pollen of Florschuetzia and Magnastriatites hawardi.Low C_(27)/C_(29)ααα20R sterane ratios,(C_(19)–C_(29))tricyclic terpanes/C_(30)αβhopane,and high oleanane/C_(30)αβhopane,bicadinane T/C_(30)αβhopane suggest that the palaeovegetation was dominated by angiosperms including the mangrove plants.The extremely abundant higher plants provide ample terrigenous organic matter for the formation of coal-measures in delta facies.The low gammacerane index and high Pr/Ph indicate the fresh and sub-oxic water in delta-neriticabysmal faces,which is not beneficial for the accumulation of hydrogen-rich organic matter.Thus,the Oligocene–Miocene marine argillaceous rocks can be potential sources of natural gas. 展开更多

BACKGROUND Gestational diabetes mellitus(GDM)is a condition characterized by high blood sugar levels during pregnancy.The prevalence of GDM is on the rise globally,and this trend is particularly evident in China,which has emerged as a significant issue impacting the well-being of expectant mothers and their fetuses.Identifying and addressing GDM in a timely manner is crucial for maintaining the health of both expectant mothers and their developing fetuses.Therefore,this study aims to establish a risk prediction model for GDM and explore the effects of serum ferritin,blood glucose,and body mass index(BMI)on the occurrence of GDM.AIM To develop a risk prediction model to analyze factors leading to GDM,and evaluate its efficiency for early prevention.METHODS The clinical data of 406 pregnant women who underwent routine prenatal examination in Fujian Maternity and Child Health Hospital from April 2020 to December 2022 were retrospectively analyzed.According to whether GDM occurred,they were divided into two groups to analyze the related factors affecting GDM.Then,according to the weight of the relevant risk factors,the training set and the verification set were divided at a ratio of 7:3.Subsequently,a risk prediction model was established using logistic regression and random forest models,and the model was evaluated and verified.RESULTS Pre-pregnancy BMI,previous history of GDM or macrosomia,hypertension,hemoglobin(Hb)level,triglyceride level,family history of diabetes,serum ferritin,and fasting blood glucose levels during early pregnancy were determined.These factors were found to have a significant impact on the development of GDM(P<0.05).According to the nomogram model’s prediction of GDM in pregnancy,the area under the curve(AUC)was determined to be 0.883[95%confidence interval(CI):0.846-0.921],and the sensitivity and specificity were 74.1%and 87.6%,respectively.The top five variables in the random forest model for predicting the occurrence of GDM were serum ferritin,fasting blood glucose in early pregnancy,pre-pregnancy BMI,Hb level and triglyceride level.The random forest model achieved an AUC of 0.950(95%CI:0.927-0.973),the sensitivity was 84.8%,and the specificity was 91.4%.The Delong test showed that the AUC value of the random forest model was higher than that of the decision tree model(P<0.05).CONCLUSION The random forest model is superior to the nomogram model in predicting the risk of GDM.This method is helpful for early diagnosis and appropriate intervention of GDM. 展开更多

Chemotherapeutics can induce immunogenic cell death(ICD)in tumor cells,offering new possibilities for cancer therapy.However,the efficiency of the immune response generated is insufficient due to the inhibitory nature of the tumor microenvironment(TME).Here,we developed a pH/reactive oxygen species(ROS)dual-response system to enhance chemoimmunotherapy for melanoma.The system productively accumulated in tumors by specific binding of phenylboronic acid(PBA)to sialic acids(SA).The nanoparticles(NPs)rapidly swelled and released quercetin(QUE)and doxorubicin(DOX)upon the stimulation of tumor microenvironment(TME).The in vitro and in vivo results consistently demonstrated that the NPs improved anti-tumor efficacy and prolonged survival of mice,significantly enhancing the effects of the combination.Our study revealed DOX was an ICD inducer,stimulating immune responses and promoting maturation of dendritic cells(DCs).Additionally,QUE served as a TME regulator by inhibiting the cyclooxygenase-2(COX2)-prostaglandin E2(PGE2)axis,which influenced various immune cells,including increasing cytotoxic T cells(CLTs)infiltration,promoting M1 macrophage polarization,and reducing regulatory T cells(Tregs)infiltration.The combination synergistically facilitated chemoimmunotherapy efficacy by remodeling the immunosuppressive microenvironment.This work presents a promising strategy to increase anti-tumor efficiency of chemotherapeutic agents. 展开更多

目的探讨胸腹腔镜Ivor-Lewis术在食管中下段癌手术治疗中的临床应用。方法回顾分析2014年3月—2016年5月行Ivor-Lewis术治疗胸中下段食管癌的148例患者的临床资料。按手术方式分为胸腹腔镜Ivor-Lewis组(腔镜组,80例)或开放Ivor-Lewis组... 目的探讨胸腹腔镜Ivor-Lewis术在食管中下段癌手术治疗中的临床应用。方法回顾分析2014年3月—2016年5月行Ivor-Lewis术治疗胸中下段食管癌的148例患者的临床资料。按手术方式分为胸腹腔镜Ivor-Lewis组(腔镜组,80例)或开放Ivor-Lewis组(开放组,68例)。两组患者术前一般资料比较,差异无统计学意义(P>0.05)。比较两组患者围手术期情况及2年随访结果。结果与开放组相比,腔镜组淋巴结清扫枚数多、出血量少,术后住院时间缩短,总并发症减少,其中以肺部并发症减少为主;腔镜组总手术时间长,住院费用增加(P <0.05),其中主要为胸部手术时间长。随访2年,两组患者预后(吻合口狭窄、膈疝、局部复发/转移、死亡等)比较,差异无统计学意义(P>0.05)。结论胸腹腔镜Ivor-Lewis术在肿瘤根治性、术中出血控制、术后恢复等方面具有优势,近期并发症少,且远期效果可靠,值得临床推广。 展开更多

为解决揉碎玉米秸秆螺旋输送过程中的生产率低、功耗大、易堵塞及机件磨损严重等问题,该文设计了一种螺旋-气力耦合输送装置,并以揉碎玉米秸秆为原料开展了试验研究。螺旋-气力耦合输送装置主要由螺旋输送装置和气力辅助输送系统组成。... 为解决揉碎玉米秸秆螺旋输送过程中的生产率低、功耗大、易堵塞及机件磨损严重等问题,该文设计了一种螺旋-气力耦合输送装置,并以揉碎玉米秸秆为原料开展了试验研究。螺旋-气力耦合输送装置主要由螺旋输送装置和气力辅助输送系统组成。其中气力辅助输送系统主要由喷射角度可调的Y型喷嘴座、喷嘴、最大出气压力为1.6 MPa的空气压缩机、直径为10 mm的PPR(polypropylene random)管和15°弯管及压力表等组成。螺旋输送装置主要由机壳、螺旋叶片和中心轴等组成,其关键参数为:螺旋叶片外径为250 mm,中心轴直径为60 mm,螺距为335 mm,螺旋槽用U型机壳。以比功耗、轴向推力、螺旋叶片及机壳各部位所受压力作为输送性能指标,对施加气流前后各部位所受压力进行测试。结果表明,当螺距为335 mm、螺旋轴转速为100 r/min、喂入量为70 kg/min、气流速度为10~50 m/s时,随着气流速度的增大,输送装置的比功耗先减小后增大。当气流速度为20 m/s时比功耗最小,为10.78 W/kg,比无气流时的比功耗减小了8.3%,轴向推力、叶片和机壳各部位所受压力随着气流速度的增大而减小,且均小于不加气流时的值。 展开更多
