In the nondestructive testing and evaluation area,magnetic major hysteresis loop measurement technology are widely applied for ferromagnetic material evaluation.However the characterization ability of major hysteresis loop measurement technology greatly varies as the evaluated target properties.To solve this limitation,magnetic minor hysteresis loops,which reflect the responses of ferromagnetic material magnetization in a systematic way,is recommend.Inspired by plenty of information carried by the minor loops,the sensitivity mapping technique was developed to achieve the highest sensitivity of minor-loop parameters to the nondestructively evaluated targets.In this study,for the first time,the sensitivity mapping technique is used to measure the tensile force in a steel strand and evaluate the effective case depth in induction-hardened steel rods.The method and procedures for the sensitivity mapping technique are given before experimental detection.The obtained experimental results indicate that the linear correlation between the induced voltage(or the magnetic induction intensity)and the tensile force(or effective case depth)exists at most of the locations in the cluster of minor loops.The obtained sensitivity maps can be used to optimize the applied magnetic field(or excitation current)and the analyzed locations at the minor loops for achieving the highest sensitivity.For the purpose of tensile force measurement,it is suggested that the strand should be firstly magnetized to the near-saturation state and then restored to the remanent state.In this way,the highest sensitivity is obtained as about 15.26 mV/kN.As for the induction-hardened steel rods,the highest sensitivity of magnetic induction intensity to the effective case depth occurs under low magnetic field conditions and the absolute value of the highest sensitivity is about 0.1110 T/mm.This indicates that if the highest sensitivity is required in the case depth evaluation,the induction-hardened steel rods are only required to be weakly magnetized.The proposed sensitivity mapping technique shows the good performance in the high-sensitivity evaluation of tensile force and case depth in ferromagnetic materials and its application scope can be extended to other nondestructive detection fields. 展开更多

Objective: This study was designed to explore whether inhibition of the extracellular-regulated kinase (ERK) and phosphatidylinositol-3-kinase (PI3K) signaling pathways can inhibit the growth of xenografts of endometrial cancer cell lines with different estrogen receptors (ER) profiles in vivo and to provide preliminary laboratory basis for the probability of endometrial adenocarcinoma treatment with blockage of the two pathways, especially to endometrial cancer with low ER status. Methods: Human endometrial cancer Ishikawa bearing ER and HEC-1Awith low ER status cells were subcutaneously injected into BALB/c nude mice to establish endometrial cancer xenograft tumor models. The effects of PI3K/Akt inhibitor LY294002, MAPK/ERK1/2 inhibitor PD-98059 and their combinations on the growth of the xenograft tumors and apoptotic state of Ishikawa and HEC-1Acells were tested in vivo using the inhibitory rate, the terminal deoxynucleotidyl transferase-mediated nick-end labeling assay, H/E-stain. Western blot analysis was used to detect the alterations of activated ERK (P-ERK) and AKT (P-AKT) during this process. Results: LY294002, a PI3K/Akt pathway inhibitor, induced significant suppression in the growth of both Ishikawa and HEC-1Acell xenograft tumors, concomitant with increased apoptosis in xenografts as evidenced by TUNEL. A similar effect was also observed when the MAPK/ERK1/2 signaling pathway was inhibited by PD98059. Concurrent inhibition of the PI3K/Akt and MAPK/ERK1/2 pathways showed enhanced anti-tumor effects in vivo as indicated by increased apoptosis. At the same time, the levels of P-ERK and P-AKT in both xenograft tumors decreased, and their levels in combination group was the lowest. Conclusions: PD98059, LY294002 and their combinations showed remarkable inhibitory effects on xenograft tumors of endometrial carcinoma cell lines with different expression status of ER in vivo through blockage of PI3K/Akt and MAPK/ERK1/2 signaling pathways. This suggests that targeting these pathways may be an effective therapeutic strategy against endometrial carcinomas, especially for ER-negative cancers which show poor response to endocrinal therapy. 展开更多

老年高尿酸血症患者血清白细胞介素-23、尿酸、单核细胞趋化蛋白-1与肾功能损伤的相关性研究

目的探讨老年高尿酸血症(HUA)患者白细胞介素-23(IL-23)、尿酸(UA)、单核细胞趋化蛋白-1(MCP-1)与肾功能损伤的相关性。方法选取2020年3月至2021年10月我院老年HUA患者112例。收集患者住院期间的临床资料及IL-23、MCP-1水平等实验室指... 目的探讨老年高尿酸血症(HUA)患者白细胞介素-23(IL-23)、尿酸(UA)、单核细胞趋化蛋白-1(MCP-1)与肾功能损伤的相关性。方法选取2020年3月至2021年10月我院老年HUA患者112例。收集患者住院期间的临床资料及IL-23、MCP-1水平等实验室指标。通过Logistic多因素回归分析血清指标与肾功能损伤的关联性,采用Spearman相关系数模型分析血清指标水平与不同程度肾功能损伤的关系。通过受试者工作特征(ROC)曲线评价血清指标预测肾功能损伤的价值。结果与肾功能正常患者相比,肾功能损伤患者的病程更长,合并高血压比例更高,血清的IL-23、UA、MCP-1水平增高,GFR减低(P<0.05)。校正后的血清IL-23、UA、MCP-1水平与肾功能损伤的发生存在关联性(P<0.05)。血清IL-23、UA、MCP-1水平与肾功能损伤的程度亦呈正相关(P<0.05),重度肾功能损伤患者的血清IL-23、UA、MCP-1水平最高(P<0.05)。血清IL-23、UA、MCP-1联合预测肾功能损伤的曲线下面积(AUC)为0.892,大于各单独指标。结论老年HUA患者的血清IL-23、UA、MCP-1水平与肾功能损伤的发生以及损伤程度均呈正相关,3个指标联合对老年HUA患者肾功能损伤的预测价值更大。 展开更多

CT与核酸检测对新型冠状病毒肺炎诊断及病程演变的对比分析 被引量:3

目的对比评估CT与核酸检测对新型冠状病毒肺炎(COVID-19)的诊断效能和病程演变特征。方法回顾性分析2020年1月20日至3月5日郑州大学第一附属医院收治的164例确诊和疑似COVID-19患者的临床资料、CT和核酸检测结果,其中,确诊COVID-19患者7... 目的对比评估CT与核酸检测对新型冠状病毒肺炎(COVID-19)的诊断效能和病程演变特征。方法回顾性分析2020年1月20日至3月5日郑州大学第一附属医院收治的164例确诊和疑似COVID-19患者的临床资料、CT和核酸检测结果,其中,确诊COVID-19患者76例[男性40例、女性36例,年龄25~83岁]、疑似COVID-19患者88例[男性47例、女性41例,年龄21~75岁]。将所有受检者按年龄分为<60岁组(122例)和≥60岁组(42例)。以咽拭子取样核酸检测结果为“金标准”,评估CT对COVID-19的诊断效能。不同年龄组间各诊断效能的比较采用独立样本卡方检验。分析确诊患者自就诊以来的多次CT图像和核酸检测结果,对比COVID-19患者病程演变过程中的临床表现、CT和核酸检测结果的转变规律。结果164例患者中,核酸检测最终确诊阳性76例,阳性率为46.3%;CT诊断阳性143例,阳性率为87.2%。CT诊断的灵敏度、特异度、阳性预测值、阴性预测值和准确率分别为96.1%(73/76)、20.5%(18/88)、51.0%(73/143)、85.7%(18/21)、55.5%(91/164)。CT的各诊断效能在<60岁和≥60岁患者之间的差异均无统计学意义(χ^2=0.051~0.414,均P>0.05)。88例疑似COVID-19患者中,CT诊断阳性率为79.5%(70/88)。76例确诊患者初诊时,8例(10.5%)为无症状亚临床期,7例(9.2%)CT无阳性表现,11例(14.5%)核酸检测结果为阴性[其中9例(81.8%)CT诊断有阳性表现,CT肺炎进展期核酸检测结果转为阳性]。病程演变过程中,CT能清晰显示肺炎的进展和缓解程度,4~8 d后复查,28例CT表现为缓解期的患者中有14例(50.0%)核酸检测结果转阴;9~12 d后复查,46例CT表现为缓解期的患者中有31例(67.4%)核酸检测结果转阴。结论CT在早期COVID-19的筛查和诊断中较核酸检测更灵敏,且能更早地评估COVID-19的病程演变,可作为COVID-19临床诊疗过程中的重要辅助手段。 展开更多

Background:Sepsis is a serious disease caused by infection.Aminophylline has anti-asthma and anti-inflammatory effects.We aimed to explore the safety and effect of aminophylline in sepsis.Methods:We conducted a clinical randomized controlled trial involving 100 patients diagnosed with sepsis within 48 h after intensive care unit(ICU)admission in two sites.All patients were randomized in a 1:1 ratio to receive standard therapy with or without aminophylline.The primary clinical outcome was all-cause mortality at 28 days.Results:From September 27,2018 to February 12,2020,we screened 277 septic patients and eventually enrolled 100 patients,with 50 assigned to the aminophylline group and 50 to the usual-care group.At 28 days,7 of 50 patients(14.0%)in the aminophylline group had died,compared with 16 of 50(32.0%)in the usual-care group(P=0.032).Cox regression showed that the aminophylline group had a lower hazard of death(hazard ratio=0.312,95%confidence interval:0.129–0.753).Compared with the usual-care group,patients in the aminophylline group had a longer survival time(P=0.039 by the log-rank test).The effects of aminophylline on vasopressor dose,oxygenation index,and sequential organ failure assessment score were timedependent with treatment.There were no significant differences in total hospitalization days,ICU hospitalization days,and rates of serious adverse events(all P>0.05).No adverse events were observed in the trial.Conclusions:Aminophylline as an adjunct therapy could significantly reduce the risk of death and prolong the survival time of patients with sepsis.Trial registration:ChiCTR.org.cn,ChiCTR1800019173. 展开更多
