With the rise of probiotics fermentation in food industry,fermented foods have attracted worldwide attention.In this study,protective effects of Rosa roxburghii&edible fungus fermentation broth(REFB)on immune function and gut health in Cyclophosphamide induced immunosuppressed mice were investigated.Results showed that REFB could improve the immune organ index,and promote the proliferation and differentiation of splenic T lymphocytes.In addition,it attenuated intestinal mucosal damage and improved intestinal cellular immunity.REFB administration also up-regulated the expression of IL-4,INF-γ,TNF-α,T-bet and GATA-3 mRNA in small intestine.Furthermore,administration of REFB modulated gut microbiota composition and increased the relative abundance of beneficial genus,such as Bacteroides.It also increased the production of fecal short-chain fatty acids.These indicate that REFB has the potential to improve immunity,alleviate intestinal injury and regulate gut microbiota in immunosuppressed mice. 展开更多

Most scientific investigations regarding inflammatory bowel disease(IBD)pathogenesis or therapeutic strategies use dextran sulfate sodium(DSS)-induced models performed on mice.However,differences between human and animal microbiota may confound the data reproducibility from rodent experiments to clinical trials.In this study,the intervention effects of Bifidobacterium longum NSP001 on DSS-induced colitis were investigated using mice colonized with either native or humanised microbiota.Disorders in disease activity index(DAI),morphology and histology of colon tissue,intestinal permeability,and secretion of MPO,TNF-αand IL-6 were ameliorated by daily intake of live B.longum NSP001 cells in both conventional and humanised colitis mice.But the abnormal thymus index,and colonic production of ZO-1 and iNOS were improved only in colitis mice treated with B.longum NSP001 and humanised microbiome.The accumulation of acetic acid and propionic acid in colon microbiome,and the optimization of primary bile acid biosynthesis and glycerophospholipid metabolism pathways in cecum commensals were likely to explain the beneficial effects of B.longum NSP001.These data revealed that intestinal microbiome baseline would possibly affect the manifestation features of interventions by probiotics or dietary components and highlighted the necessity to include humanised microbiome while investigating potential therapeutic strategies based on rodent models. 展开更多

Dietary flavonoids are abundant in natural plants and possess multiple pharmacological and nutritional activities.In this study,apigenin,luteolin,and baicalein were chosen to evaluate their anti-diabetic effect in high-glucose and dexamethasone induced insulin-resistant(IR)HepG2 cells.All flavonoids improves the glucose consumption and glycogen synthesis abilities in IR-HepG2 cells via activating glucose transporter protein 4(GLUT4)and phosphor-glycogen synthase kinase(GSK-3β).These fl avonoids signifi cantly inhibited the production of reactive oxygen species(ROS)and advanced glycation end-products(AGEs),which were closely related to the suppression of the phosphorylation form of NF-κB and P65.The expression levels of insulin receptor substrate-1(IRS-1),insulin receptor substrate-2(IRS-2)and phosphatidylinositol 3-kinase(PI3K)/protein kinase B(Akt)pathway in IR-HepG2 cells were all partially activated by the fl avonoids,with variable effects.Furthermore,the intracellular metabolic conditions of the fl avonoids were also evaluated. 展开更多

Polysaccharide was a class of macromolecular substance with various bioactive functions.Gut symbiotic microorganisms could utilize the polysaccharides from various sources,thus have important impact on human health.Bacteroides represented one of the dominant colonizers in the human gut.The utilization of polysaccharide by Bacteroides was important for supporting the function and stability of gut microbiota.After the degradation of polysaccharides by Bacteroides,gut microbes could ferment the monosaccharides and oligosaccharides degraded from polysaccharides into some metabolites,such as short-chain fatty acids(SCFAs),amino acids,etc.Among the metabolites,the SCFAs could have beneficial effects on gut health.This review summarized the niches of Bacteroides among gut microbiota,and also described the gene clusters and membrane proteins involved in the utilization processes of polysaccharide by gut Bacteroides.SCFAs could act as energy substrates for intestinal epithelial cells,inhibit histone deacetylases and activate G protein-coupled receptors.In addition,the future perspectives in investigating new degradation pathways for polysaccharide,and using polysaccharides or their metabolites as therapeutic approaches for diseases mediated by the gut dysbiosis were also provided. 展开更多

Furan(C4H4O)has been classified as a possible animal and human carcinogen by many international agencies.The formation of furan in three sugar–glycine models using glucose,fructose,and sucrose was investigated using headspace gas chromatography mass spectrometry method(HSGC–MS)with various dual combinations of three important heat processing conditions,i.e.pH,temperature,and heating time.Results indicated that furan levels from sugar–glycine model systems during the thermal processing can be attributed to selective sugar types,pH,temperature,and heating time.In glucose–glycine and fructose–glycine system,the lowest furan level was detected in acid condition but in sucrose–glycine system furan formed significantly lower(P<0.05)in acidic conditions the lowest furan level was found in alkaline conditions.The furan levels were observed to increase with heating time in all three model systems.Furthermore,less furan was generated in non-reducing sugar system(sucrose)than in reducing sugar system(glucose and fructose).Therefore,they demonstrate the possibility of limiting the formation of furan in heat processed foods by both the careful selection of carbohydrates(i.e.non-reducing sugars and reducing sugars)ingredients and appropriate processing conditions.©2013 Beijing Academy of Food Sciences.Production and hosting by Elsevier B.V.All rights reserved. 展开更多

Background:Pseudoaneurysm with median nerve injury is a serious complication of radial artery puncture.It is very important to summarize the prevention and treatment experience of this complication through case discussion.Case report:A 66-year-old woman was admitted to the hospital because of‘‘paroxysmal chest tightness and suffocation for 5 days.”Coronary angiography was performed.During insertion of the arterial sheath,the patient experienced severe pain in the right forearm,which radiated to the palm.The puncture sheath did not return blood after the sheath core was withdrawn.The sheath was removed and local compression was used to stop bleeding.There was no obvious bleeding at the puncture point,and the compression was removed 6 hours after the procedure.Local swelling and increased tension were seen in the right forearm.At the 1-week follow-up she exhibited swelling,high local tension,small blisters,and bluish-purple skin of the right forearm,with an acceptable right radial artery pulsation.She had severe pain in the affected limb,which radiated to the thumb,index fi nger,and middle fi nger.Case discussion:We discuss the causes of and treatment measures for pseudoaneurysm with median nerve injury caused by radial artery puncture. 展开更多

Diabetes mellitus is a serious threat to human health.Cyclocarya paliurus(Batal.)Iljinskaja(C.paliurus)is one of the traditional herbal medicine and food in China for treating type 2 diabetes,and the C.paliurus polysaccharides(CP)were found to be one of its major functional constituents.This research aimed at investigating the hypoglycemic mechanism for CP.It was found that CP markedly attenuated the symptoms of diabetes,and inhibited the protein expression of Bax,improved the expression of Bcl-2 in pancreas of diabetic rats,normalized hormones secretion and controlled the inflammation which contributed to the regeneration of pancreaticβ-cell and insulin resistance.CP treatment increased the beneficial bacteria genus Ruminococcaceae UCG-005 which was reported to be a key genus for protecting against diabetes,and the fecal short-chain fatty acids levels were elevated.Uric metabolites analysis showed that CP treatment helped to protect with the diabetes by seven significantly improved pathways closely with the nutrition metabolism(amino acids and purine)and energy metabolism(TCA cycle),which could help to build up the intestinal epithelial cell defense for the inflammation associated with the diabetes.Our study highlights the specific mechanism of prebiotics to attenuate diabetes through multi-path of gut microbiota and host metabolism. 展开更多

Objective:This study aimed to estimate the proportion of non-obstructive coronary artery disease(CAD)patients in large percutaneous coronary intervention(PCI)centers in China.Methods:The study was conducted at 6 large PCI centers in China from January 1,2013 to December 31,2015.Demographic and clinical data were collected from medical records,prescription records,and laboratory reports of patients with symptoms of angina who underwent coronary angiography(CAG).Results:A total of 1713 patients were consecutively screened,1600 of whom were included in the study.CAG showed that 300 patients had non-obstructive CAD while 1300 had obstructive CAD.Among the 300 patients with non-obstructive CAD,203 displayed mild coronary stenosis(20%-49%)and 91 had normal coronary status(ie,<20%stenosis).Of the 1300 patients with obstructive CAD,61.6%(801/1300)had typical symptoms of angina,compared with 49.3%(148/300)for patients with nonobstructive CAD.In addition,there were more women than men in the non-obstructive CAD group,whereas the reverse was observed among obstructive CAD patients.Conclusion:The prevalence of non-obstructive CAD in the Chinese population with coronary heart disease was estimated to be approximately 20%.Additionally,typical angina symptoms were correlated with obstructive CAD,whereas female gender was identified as a risk factor for non-obstructive CAD. 展开更多

Polysaccharides from genus Ganoderma have prominent anti-aging effect,but their mechanisms are incompletely clarified.In our previous experiments,Ganoderma atrum polysaccharide(PSG)was exhibited to significantly alleviate senescence and DNA damage of A375 cells.To investigate its underlying mechanism,we conducted RNA-Seq analysis and found that PSG upregulated autophagy and mitochondria pathways compared to the model group.Further experiments showed that PSG relieved mitochondrial dysfunction via inhibiting the accumulation of reactive oxygen species(ROS)and loss of the mitochondrial membrane potential(MMP).Meanwhile,PSG activated autophagy and enhanced lysosomal function through transcription factor EB(TFEB).Interestingly,the pretreatment of autophagy inhibitors bafilomycin A1(Baf A1)reversed the ROS decline induced by PSG,which subsequently increased cellular senescence of A375.In addition,PSG had a similar anti-aging effect on normal fibroblast WI-38.Taken together,PSG might reduce ROS levels through activating autophagy,which in turn suppressed mitochondrial dysfunction and cellular senescence. 展开更多
