Effects of Fungi Fusarium sp. to Rhizosphere Soil and Physiological Characteristics of Camellia oleifera Abel.

[Objectives]To study the effects of fungi Fusarium sp.to rhizosphere soil and physiological characteristics of Camellia oleifera Abel.[Methods]We investigated the effects of Fusarium sp.to rhizosphere soil nutrient element content and metabolites of C.oleifera.C.oleifera was inoculated with the suspension of Fusarium sp.in pot experiments and ammonium-N,available phosphorus,available potassi-um,organic matter,enzymes and pH of rhizosphere soil,MDA content,activity of SOD,POD of C.oleifera leaves were analyzed.[Results]Fusarium sp.stress significantly inhibited soil enzyme activities and significantly reduced available phosphorus content,especially for phospha-tase and sucrase.Antioxidant enzyme activities in C.oleifera tissues showed that Fusarium sp.stress significantly increased MDA and SOD enzyme activities and decreased POD enzyme activity.Especially,SOD enzyme activity was elevated by 53.86%compared with the CK group.In addition,analysis of the content of major metabolites in C.oleifera leaves showed that Fusarium sp.stress significantly reduced the content of total flavonoids,quercetin,isoquercitrin and isoquercitrin in C.oleifera leaves by 7.80%,50.00%and 75.90%,respectively.[Conclusions]Our results are an important step which showed strong resistance of C.oleifera and can give a novel insight for researches on the effects in the rhizosphere soil enzyme,soil nutrient elements and metabolites of C.oleifera under the Fusarium sp.too. 展开更多

Effects of Exogenous Plant Hormones on Growth Status and Secondary Metabolism of Houttuynia cordata Thunb.

[Objectives]To improve the yield and secondary metabolite content of medicinal plants and to further develop and utilize the medicinal and other functions of medicinal plants.[Methods]We used the sterile tissue culture method with Houttuynia cordata Thunb.as the research object.Different concentrations of 1-naphthalene acetic acid(NAA),auxin(indole-3-acetic acid,IAA)and gibberellin acid(GA_(3))were added to the group culture medium of H.cordata to investigate the effects of exogenous plant hormones on plant height,root length,fresh weight,morphological characteristics,four phenolics and 20 volatile compounds.[Results]The results showed that the exogenous plant hormone of 3 mg/L GA_(3)significantly increased plant height by 79.9%over the control;the exogenous plant hormone of 3 mg/L IAA significantly increased root length by 52.6%over the control;and the exogenous plant hormone of 1 mg/L GA_(3)significantly increased fresh weight of single plant by 458.2%over the control.In the treatment group of 1 mg/L NAA,chlorogenic acid content was significantly increased by 52.6%compared with the control;in the treatment group of 1 mg/L IAA,chlorogenic acid,rutin,isodendrin and quercetin content were significantly increased by 109.1%,100.6%,173.8%,and 198.7%compared with the control,respectively;in the treatment of 3 mg/L GA_(3),chlorogenic acid,rutin,isoquercitin,and quercitin content were significantly increased by 65.3%,104.9%,139.0%and 191.2%over the control.In addition,the content of volatile compounds was significantly higher in all H.cordata treated with exogenous plant hormones of 2 mg/L NAA,1 mg/L IAA,and 3 mg/L GA_(3);however,the content of volatile compounds was lower in all of the treatments with 2 mg/L GA_(3).[Conclusions]Different exogenous plant hormones have certain effects on the growth morphology and secondary metabolic content of H.cordata,which provides theoretical basis and technical support for the development and utilization of medicinal plants. 展开更多

Resistance of Microbial Community of Artemisia annua L.to Pathogenic Fungi

[Objectives]This paper was to figure out whether the dominant bacterial community has the role and effect of bacterial community and its defense mechanism against potential pathogenic fungi in Artemisia annua,and thus establish a systematic model of bacteria-fungus-plant.[Methods]Fifty-eight strains of bacteria and one strain of pathogenic fungi,Globisporangium ultimatum,were used for the experiments.These 58 bacterial strains were assembled into a bacterial community,and the bacteria with abundance in the top 1%were reassembled into a dominant bacterial community as measured by 16S rDNA.[Results]The growth of A.annua seedlings inoculated with bacterial communities and pathogenic fungi or dominant bacterial communities and pathogenic fungi was significantly better than that of A.annua seedlings inoculated with pathogenic fungi during in vitro confrontation,which was evident in both enzymatic and non-enzymatic antioxidant assays.[Conclusions]The results suggest that the dominant bacterial community has a crucial role as a representative core microbial community of synthetic bacterial community,which can protect plants by interfering with the growth of phytopathogenic fungi mediated by chemical signals,and can be used as the main synthetic community of biocides to achieve the effect of biocontrol. 展开更多

Effects of GA3 and NAA on Fatty Acid Degradation in Germination and Seedling Growth of Cassia obtusifolia L. Seed 被引量:1

[Objectives]To understand the content changes of fatty acid compositions and seedling growths during seed germinations of Cassia obtusifolia L.[Methods]The fatty acid compositions of germinations and seedling growths of C.obtusifolia seeds treated with different concentrations of gibberellin(GA3)and naphthalene acetic acid(NAA)were analyzed using gas chromatograph-mass spectrometer(GC-MS),and the germination rates,root lengths,plant heights and fresh weights of their seedlings were analyzed.[Results]The contents of eicosenoic acid,α-linoleic acid,arachidic acid,eicosadienoic acid and eicosapentaenoic acid of C.obtusifolia seeds treated with different concentrations of GA3 and NAA decreased with germination time.There were significant differences between the ratios of germinations of C.obtusifolia seeds treated with different concentrations of GA3 and NAA.GA3 and NAA of different ratio treatments could promote seedling heights and fresh weights of C.obtusifolia,but increases of seedling heights and fresh weights were different.Eicosenoic acid,α-linoleic acid,arachidic acid,eicosadienoic acid and eicosapentaenoic acid were remarkably negative correlation with seedling heights and fresh weights.[Conclusions]The fatty acid components of C.obtusifolia seed germination and growth of their seedlings were affected.It indicated that GA3 and NAA treatments can affect fatty acid metabolism,the germination rates,root lengths,plant heights and fresh weights of C.obtusifolia.It is intended to provide a theoretical basis and technological reference for standardized cultivated techniques of C.obtusifolia. 展开更多

[Objectives] To evaluate the phenolic components of Houttuynia cordata Thunb. and their antioxidant activities from different sources. [Methods] Samples of 35 different sources of H. cordata were collected from Guizhou,and polyphenols,total flavonoids,total chlorogenic acid and 7 kinds of flavonoids( xanthol-3-O-glucoside,hyperoside,quercitrin,rutin,quercetin,kaempferol and isorhamnetin),and DPPH free radical scavenging rate of the extracts were determined. Relationship between the contents of phenolics and the DPPH free radical scavenging rate of the extracts were analyzed by SPSS software. [Results] There were some significant differences in polyphenols,total flavonoids,total chlorogenic acid and the seven flavonoids between samples of different habitats,and the contents of polyphenols,total flavonoids and total chlorogenic acid increased linearly with altitude. There was a significant positive correlation between polyphenols,total flavonoids and total chlorogenic acid content and DPPH free radical scavenging rate of extracts. The contents of kaempferol-3-O-glucoside,hyperoside,quercitrin,rutin,quercetin kaempferol and isorhamnetin were positively correlated with the DPPH free radical scavenging rates of the extracts,except for the significant correlation with quercitrin. [Conclusions]The experimental results suggest that the antioxidant capacity of H. cordata is derived from the total contribution of the phenolic components,indicating that these components have overall equivalence of antioxidant capacities. This provides a new research idea for solving the key problems of quality control of traditional Chinese medicine. 展开更多

[Objectives]To study responses of Houttuynia cordata Thunb.to tetracycline(TC)stress in a sterile environment.[Methods]Aseptic H.cordata seedlings were cultured in the medium which added the different concentrations of TC,and then the contents of chlorophyll and malondialdehyde(MDA),and activity of catalase(CAT),peroxidase(POD)and superoxide dismutase(SOD)of H.cordata leaves were measured.[Results]Compared with the CK,the contents of chlorophyll of H.cordata leaves decreased,and the accumulation of MDA,and the activity of CAT,POD and SOD in H.cordata leaves increased when H.cordata was stressed by different concentration of TC.[Conclusions]The growth,metabolism and physiology of H.cordata were affected by the stress of TC.More importantly,this proved an insight into the effect or harm of undegraded antibiotics in the soil and water in the natural environment to plant growth all the time. 展开更多
