“Baihui”(DU20)-penetrating “Qubin”(GB7) acupuncture on blood–brain barrier integrity in rat intracerebral hemorrhage models via the RhoA/ROCK Ⅱ/MLC 2 signaling pathway

Background: Blocking the Rho A/ROCK Ⅱ/MLC 2(Ras homolog gene family member A/Rho kinase Ⅱ/myosin light chain 2) signaling pathway can initiate neuroprotective mechanisms against neurological diseases such as stroke, cerebral ischemia, and subarachnoid hemorrhage. Nevertheless, it is not clear whether and how disrupting the Rho A/ROCK Ⅱ/MLC 2 signaling pathway changes the pathogenic processes of the blood–brain barrier(BBB) after intracerebral hemorrhage(ICH). The present investigation included the injection of rat caudal vein blood into the basal ganglia area to replicate the pathophysiological conditions caused by ICH. Methods: Scalp acupuncture(SA) therapy was performed on rats with ICH at the acupuncture point “Baihui”-penetrating “Qubin,” and the ROCK selective inhibitor fasudil was used as a positive control to evaluate the inhibitory effect of acupuncture on the Rho A/ROCK Ⅱ/MLC 2 signaling pathway. Post-assessments included neurological deficits, brain edema, Evans blue extravasation, Western blot, quantitative polymerase chain reaction, and transmission electron microscope imaging. Results: We found that ROCK Ⅱ acts as a promoter of the Rho A/ROCK Ⅱ/MLC 2 signaling pathway, and its expression increased at 6 h after ICH, peaked at 3 days, and then decreased at 7 days after ICH, but was still higher than the preintervention level. According to some experimental results, although 3 days is the peak, 7 days is the best time point for acupuncture treatment. Starting from 6 h after ICH, the neurovascular structure and endothelial cell morphology around the hematoma began to change. Based on the changes in the promoter ROCK Ⅱ, a 7-day time point was selected as the breakthrough point for treating ICH model rats in the main experiment. The results of this experiment showed that both SA at “Baihui”-penetrating “Qubin” and treatment with fasudil could improve the expression of endothelial-related proteins by inhibiting the Rho A/ROCK Ⅱ/MLC 2 signaling pathway and reduce neurological dysfunction, brain edema, and BBB permeability in rats. Conclusion: This study found that these experimental data indicated that SA at “Baihui”-penetrating “Qubin” could preserve BBB integrity and neurological function recovery after ICH by inhibiting Rho A/ROCK Ⅱ/MLC 2 signaling pathway activation and by regulating endothelial cell–related proteins. 展开更多

To meet the growing emission of water contaminants,the development of new materials that enhance the efficiency of the water treatment system is urgent.Ordered mesoporous materials provide opportunities in environmental processing applications due to their exceptionally high surface areas,large pore sizes,and enough pore volumes.These properties might enhance the performance of materials concerning adsorption/catalysis capability,durability,and stability.In this review,we enumerate the ordered mesoporous materials as adsorbents/catalysts and their modifications in water pollution treatment from the past decade,including heavy metals(Hg^(2+),Pb^(2+),Cd^(2+),Cr^(6+),etc.),toxic anions(nitrate,phosphate,fluoride,etc.),and organic contaminants(organic dyes,antibiotics,etc.).These contributions demonstrate a deep understanding of the synergistic effect between the incorporated framework and homogeneous active centers.Besides,the challenges and perspectives of the future developments of ordered mesoporous materials in wastewater treatment are proposed.This work provides a theoretical basis and complete summary for the application of ordered mesoporous materials in the removal of contaminants from aqueous solutions. 展开更多

新国标下婴幼儿配方奶粉与中国母乳脂质组分的比较研究

以新国标婴幼儿配方奶粉和母乳为研究对象,采取改良Folch法提取乳中粗脂,通过气相色谱,超高效液相色谱串联四级杆飞行时间质谱仪(UPLC-Q-TOF-ESI-MS/MS),高效液相色谱蒸发光检测器(HPLC-ELSD)和高效液相色谱二极管阵列检测器(HPLC-DAD)... 以新国标婴幼儿配方奶粉和母乳为研究对象,采取改良Folch法提取乳中粗脂,通过气相色谱,超高效液相色谱串联四级杆飞行时间质谱仪(UPLC-Q-TOF-ESI-MS/MS),高效液相色谱蒸发光检测器(HPLC-ELSD)和高效液相色谱二极管阵列检测器(HPLC-DAD)分析脂肪酸组成、甘油三酯分子种类、磷脂和胆固醇含量,剖析新国标不同阶段的婴幼儿配方奶粉的特点,并与中国母乳脂质进行比较。结果表明,新国标婴幼儿配方奶粉与母乳中检测到50种甘油三酯,婴幼儿配方奶粉中主要包括长链甘油三酯(如O/P/O,O/P/L)和中链甘油三酯(如Ca/La/M,La/La/M),其中中链甘油三酯(18.034%~22.209%)高于母乳(1.108%);母乳甘油三酯主要为中长链甘油三酯,且大部分为一个中链脂肪酸和两个长链脂肪酸的单中链型甘油三酯。新国标婴幼儿配方奶粉总磷脂含量高于母乳,以磷脂酰乙醇胺和磷脂酰胆碱为主,母乳以鞘磷脂为主。此外,新国标婴幼儿配方奶粉的胆固醇低于母乳,为母乳胆固醇1/3。通过马氏距离分析,新国标婴幼儿配方奶粉的脂质与母乳的相似度得分可达80%以上,其中新国标婴幼儿配方奶粉I段的脂质与母乳的相似度最高。 展开更多

为了开发高营养无麸质荞麦食品,以食物营养质量指数(Index of Nutrition Quality,INQ)和蛋白质体外消化率为指标,采用正交试验法对荞麦粉进行营养复配优化。高INQ粉配方为72%荞麦粉、6%大豆分离蛋白(Soy Protein Isalate,SPI)、黄豆粉12... 为了开发高营养无麸质荞麦食品,以食物营养质量指数(Index of Nutrition Quality,INQ)和蛋白质体外消化率为指标,采用正交试验法对荞麦粉进行营养复配优化。高INQ粉配方为72%荞麦粉、6%大豆分离蛋白(Soy Protein Isalate,SPI)、黄豆粉12%及芝麻粉10%,测得此配方下复配粉的INQ为1.47,体外消化率为91.00%;高消化粉配方为荞麦粉77%、SPI 4%、黄豆粉6%及芝麻粉13%,测得此配方下复配粉的INQ为1.23,体外消化率为95.66%。2种粉的理化特性测定结果显示:高INQ粉拥有最低的回生值和最高的水溶性指数,水溶性最好,且容易形成稳定的糊状物。对荞麦粉及复配粉粥的测定结果表明,高INQ粉粥的冲调稳定性最高,复水性较好,结块率最低,适合粥类产品的开发。 展开更多

Acupuncture is widely used in the treatment of cerebral hemorrhage,and it improves outcomes in experimental animal models and patients.However,the mechanisms underlying the effectiveness of acupuncture treatment for cerebral hemorrhage are still unclear.In this study,a model of intracerebral hemorrhage was produced by injecting 50μL autologous blood into the caudate nucleus in Wistar rats.Acupuncture at Baihui(DU20)and Qubin(GB7)acupoints was performed at a depth of 1.0 inch,12 hours after blood injection,once every 24 hours.The needle was rotated at 200 r/min for 5 minutes,For each 30-minute session,needling at 200 r/min was performed for three sessions,each lasting 5 minutes.For the positive control group,at 6 hours,and 1,2,3 and 7 days after induction of hemorrhage,the rats were intraperitoneally injected with 1 mL aniracetam(0.75 mg/mL),three times a day.The Bederson behavioral test was used to assess palsy in the contralateral limbs.Western blot assay was used to examine the expression levels of Nestin and basic fibroblast growth factor in the basal ganglia.Immunohistochemistry was performed to count the number of Nestin-and glial cell line-derived neurotrophic factor-positive cells in the basal ganglia.Acupuncture effectively reduced hemorrhage and brain edema,elevated the expression levels of Nestin and basic fibroblast growth factor in the basal ganglia,and increased the number of Nestin-and glial cell line-derived neurotrophic factor-positive cells in the basal ganglia.Together,these findings suggest that acupuncture promotes functional recovery after cerebral hemorrhage by increasing the expression of neurotrophic factors.The study was approved by the Committee for Experimental Animals of Heilongjiang Medical Laboratory Animal Center(approval No.2017061001)on June 10,2017. 展开更多

Inflammation plays an important role in nerve defects caused by intracerebral hemorrhage. Repairing brain damage by inhibiting the macrophage-inducible C-type lectin/spleen tyrosine kinase (Mincle/Syk) signaling pathway is a potential new target for treating cerebral hemorrhage. In this study, we aimed to determine whether acupuncture through Baihui (DU20) to Qubin (GBT) is an effective treatment for intracerebral hemorrhage through the Mincle/Syk signaling pathway. An intracerebral hemorrhage rat model was established by autol- ogous blood infusion into the caudate nucleus. Acupuncture through Baihui to Qubin was performed for 30 minutes, once every 12 hours, for a total of three times. Piceatannol (34.62 mg/kg), a Syk inhibitor, was intraperitoneally injected as a control. Modified neurological severity score was used to assess neurological function. Brain water content was measured. Immunohistochemistry and western blot assay were used to detect immunoreactivity and protein expression levels of Minde, Syk, and CARD9. Real-time polymerase chain reaction was used to determine interleukin-1[~ mRNA levels. Hematoxylin-eosin staining was performed to observe histopathological changes. Our re- suits showed that acupuncture through Baihui to Qubin remarkably improved neurological function and brain water content, and inhibited immunoreactivity and expression of Mincle, Syk, CARDg, and interkeukin-1β Moreover, this effect was similar to piceatannol. These find- ings suggest that acupuncture through Baihui to Qubin can improve neurological impairment after cerebral hemorrhage by inhibiting the Mincle/Syk signaling pathway. 展开更多

Baihui(DU20)-penetrating-Qubin(GB7) acupuncture can inhibit inflammatory reactions and activate signaling pathways related to proliferation after intracerebral hemorrhage.However,there is no research showing the relationship between this treatment and cell apoptosis.Rat models of intracerebral hemorrhage were established by injecting 60 μL of autologous blood into the right side of the caudate-putamen.Six hours later,the needle traveled subcutaneously from the Baihui acupoint to Qubin acupoint.The needle was alternately rotated(180 ± 10 turns/min) manually along clockwise and counter-clockwise directions.Stimulation lasted for 7 days,and was performed three times each for 6 minutes with 6-minute intervals between stimulations.Rats intraperitoneally receiving Sonic hedgehog pathway activator,purmorphamine(1 mg/kg per day),served as positive controls.Motor and sensory function were assessed using the Ludmila Belayev test.Extent of pathological changes were measured in the perihemorrhagic penumbra using hematoxylin-eosin staining.Apoptosis was examined by terminal deoxynucleotidyl transferase(Td T)-mediated d UTP nick end labeling assay.Expression of smoothened(Smo) and glioma-associated homolog 1(Gli1) was determined by western blot assay.Our results showed that Baihui-penetrating-Qubin acupuncture promoted recovery of motor and sensory function,reduced the apoptotic cell percentage in the perihemorrhagic penumbra,and up-regulated Smo and Gli1 expression.We conclude that Baihui-penetrating-Qubin acupuncture can mitigate hemorrhage and promote functional recovery of the brain in a rat model of intracerebral hemorrhage,possibly by activating the Sonic hedgehog pathway. 展开更多

Porous C‐I codoped carbon nitride materials were synthesized by in‐situ codoping with iodized ionic liquid followed by post‐thermal treatment in air.The effects of doping content of C‐I codoping with different amounts of ionic liquid on the structural,optical and photocatalytic properties of the samples were investigated.Characterization results show that more compact interlayer sacking can be achieved by post‐thermal treatment.Combined with C‐I codoping by insertion of ionic liquids,much enlarged surface area but optimized sp2 conjugated heterocyclic structure can be found in the catalysts.Optical and energy band analysis results evidence that the light absorptions especially in visible light region are significantly improved.Although the band gap of porous C‐I codoped samples enlarge because of the generation of porous,the negatively shifted conduction band position thermodynamically supplies stronger motivation for water reduction.Photoelectricity tests reveal that the photo‐induced electron density was increased after C‐I codoping modification.Also,the recombination rate of electron‐hole pairs is remarkably inhibited.The catalysts with moderate C‐I codoing content perform sharply enhanced photocatalytic H2 evolution activity under visible light irradiation.A H2 evolution rate of 168.2μmol/h was achieved and it was more than 9.8 times higher than pristine carbon nitride.This study demonstrates a novel non‐metal doping strategy for synthesis and optimization of polymer semiconductor with gratifying photocatalytic H2 evolution performance from water hydrolysis. 展开更多

Photocatalytic H2 production and CO2 reduction have attracted considerable attention for clean energy development.In this work,we designed an efficient photocatalyst by integrating lamellar oxygen-doped carbon nitride(CNO)nanosheets into ZnIn2S4(ZIS)microflowers by a one-step hydrothermal method.A well-fitted 2D hierarchical hybrid heterostructure was fabricated.Under visible light irradiation,the ZIS@CNO composite with 40 wt%CNO(ZC 40%)showed the highest hydrogen evolution rate from water(188.4μmol·h-1),which was approximately 2.1 times higher than those of CNO and ZIS(88.6 and 90.2μmol·h-1,respectively).Furthermore,the selective CO production rates of ZC 40%(12.69μmol·h-1)were 2.2 and 14.0 times higher than those of ZIS(5.85μmol·h-1)and CNO(0.91μmol·h-1),respectively,and the CH4 production rate of ZC 40%was 1.18μmol·h-1.This enhanced photocatalytic activity of CNO@ZIS is due mainly to the formation of a heterostructure that can promote the transfer of photoinduced electrons and holes between CNO and ZIS,thereby efficiently avoiding recombination of electron-hole pairs. 展开更多

Lamellar bone,compactly and ingeniously organized in the hierarchical pattern with 6 ordered scales,is the structural motif of mature bone.Each hierarchical scale exerts an essential role in determining physiological behavior and osteogenic bioactivity of bone.Engineering lamellar bone with full-scale hierarchy remains a longstanding challenge.Herein,using bioskiving and mineralization,we attempt to engineer compact constructs resembling full-scale hierarchy of lamellar bone.Through systematically investigating the effect of mineralization on physicochemical properties and bioactivities of multi-sheeted collagen matrix fabricated by bioskiving,the hierarchical mimicry and hierarchy-property relationship are elucidated.With prolongation of mineralization,hierarchical mimicry and osteogenic bioactivity of constructs are performed in a bidirectional manner,i.e.first rising and then descending,which is supposed to be related with transformation of mineralization mechanism from nonclassical to classical crystallization.Construct mineralized 9 days can accurately mimic each hierarchical scale and efficiently promote osteogenesis.Bioinformatic analysis further reveals that this construct potently activates integrinα5-PI3K/AKT signaling pathway through mechanical and biophysical cues,and thereby repairing critical-sized bone defect.The present study provides a bioinspired strategy for completely resembling complex hierarchy of compact mineralized tissue,and offers a critical research model for in-depth studying the structure-function relationship of bone. 展开更多

目的研究婴幼儿食品中分离的阪崎克罗诺杆菌(C.sakazakii)分子生物学特征。方法对婴幼儿食品中C.sakazakii进行分离鉴定、药物敏感性试验和全基因组测序,利用BioNumerics软件对全基因组数据进行拼接组装,对组装基因组开展多位点序列分型... 目的研究婴幼儿食品中分离的阪崎克罗诺杆菌(C.sakazakii)分子生物学特征。方法对婴幼儿食品中C.sakazakii进行分离鉴定、药物敏感性试验和全基因组测序,利用BioNumerics软件对全基因组数据进行拼接组装,对组装基因组开展多位点序列分型(MLST)、核心基因组多位点序列分型(cgMLST)、毒力基因和耐药基因分析,并与PubMLST数据库中ST型进行比较分析。结果本研究中分离的9株C.sakazakii共分为5个ST型,ST1和ST64型为主要型别,同时也是PubMLST数据库中C.sakazakii的主要型别。3株C.sakazakii携带mcr-9耐药基因,但药敏结果显示所有菌株对包括多粘菌素B和多粘菌素E在内的12种抗生素均敏感。除携带共同毒力基因谱外,ST1、ST64和ST458型携带脂多糖基因gtrB。cgMLST聚类分析显示,9株C.sakazakii呈高度多样性。结论与临床分离株相关的ST1和ST64型是本研究食品分离株中的主要型别,提示C.sakazakii具有潜在的致病性,有必要对婴幼儿食品中C.sakazakii开展连续监测,为预防控制由其引起的食源性疾病提供依据。 展开更多

目的研究食品中分离克罗诺杆菌属种的鉴定方法,旨在建立稳定、可靠的种的鉴定方法。方法本研究选择三种鉴定方法,包括生化鉴定(VITEK 2 Compact)法、基质辅助激光解吸电离飞行时间质谱(MALDI-TOF MS)、全基因组测序,通过比较分析三种方... 目的研究食品中分离克罗诺杆菌属种的鉴定方法,旨在建立稳定、可靠的种的鉴定方法。方法本研究选择三种鉴定方法,包括生化鉴定(VITEK 2 Compact)法、基质辅助激光解吸电离飞行时间质谱(MALDI-TOF MS)、全基因组测序,通过比较分析三种方法对19株克罗诺杆菌属的鉴定结果,阐述三种鉴定方法的优缺点及其适用性。结果本研究中19株克罗诺杆菌属的鉴定结果显示,VITEK 2 Compact对阪崎克罗诺杆菌和丙二酸盐克罗诺杆菌的鉴定准确率只有67%和66%,而MALDI-TOF MS和全基因组测序的鉴定结果一致,因此可以判定MALDI-TOF MS能够快速、准确、高通量对克罗诺杆菌属进行种的鉴定。但是MALDI-TOF MS受限于数据库,不能鉴定到亚种的水平。结论本研究结果提示VITEK 2 Compact法不适用于克罗诺杆菌属种的鉴定;全基因组测序鉴定结果准确可靠;MALDI-TOF MS可以实现快速、高通量、准确的鉴定,仍需要进一步研究克罗诺杆菌属3个亚种的蛋白指纹图谱特点,丰富蛋白指纹图谱数据库,使其能够准确鉴定克罗诺杆菌属的7个种和3个亚种。 展开更多

The rational design and precise synthesis of multifunctional hybrid nanostructures with a tailored active core and a large, dendritic, modified mesoporous structured shell can promote catalysis, energy storage, and biological applications. Here, an oil-water biphase stratification coating strategy has been developed to prepare monodisperse magnetic dendritic mesoporous silica core-shell structured nano- spheres. These sophisticated Fe3O4@SiO2@dendritic-mSiO2 nanospheres feature large dendritic open pores (2.7 and 10.3 nm). Significantly, the silica shells can be converted into dendritic mesoporous aluminosilicate frameworks with unchanged porosity, a Si/Al molar ratio of 14, and remarkably strong acidic sites, through a post-synthesis approach. In addition, the resultant magnetic dendritic mesoporous aluminosilicate nanospheres exhibit outstanding properties and promising application in phosphate removal from wastewater. 展开更多
