We hypothesized that these allografts compounded with both brain-derived neurotrophic factor- and ciliary neurotrophic factor-transfected bone marrow mesenchymal stem cells may demonstrate even better effects in the repair of peripheral nerve injury. We cultured bone marrow mesenchymal stem cells expressing brain-derived neuro- trophic factor and/or ciliary neurotrophic factor and used them to treat sciatic nerve injury in rats. We observed an increase in sciatic functional index, triceps wet weight recovery rate, myelin thickness, number of myelinated nerve fibers, amplitude of motor-evoked potentials and nerve conduction velocity, and a shortened latency of motor-evoked potentials when al- lografts loaded with both neurotrophic factors were used, compared with allografts loaded with just one factor. Thus, the combination of both brain-derived neurotrophic factor and cili- ary neurotrophic factor-transfected bone marrow mesenchymal stem cells can greatly improve nerve injury.展开更多

METHODS:Isolated rat livers were kept in CS in University of Wisconsin Solution for 20 h at 4℃,then exposed to 25 min of warm ischemia(WI)at 37℃ followed by 2 h of warm perfusion(WP)at 37℃with oxygenated(95%oxygen and 5%carbon dioxide) Krebs-Henseleit buffer.Desferrioxamine(DFO),an iron chelator, was added at different stages of storage, ischemia and perfusion:in CS only,in WI only,in WP only, in WI and perfusion,or in all stages.Effluent samples were collected after CS and after WI.Perfusate samples and bile were collected every 30 min(0,0.5,1,1.5 and 2 h)during liver perfusion.Cellular injury was assessed by the determination of lactate dehydrogenase(LDH) and aspartate aminotransferase(AST)in the effluent and perfusate samples.Total iron was analysed in the perfusate samples. After WP,the liver was collected for the determination of liver swelling(wet to dry ratio) and liver morphological examination(hematoxylin and eosin staining). RESULTS:Increased CS time caused increased liver dysfunction during WP.After 2 h of WP,liver injury was indicated by increased release of AST(0.5 h CS:9.4± 2.2 U/g liver vs 20 h CS:45.9±10.8 U/g liver,P<0.05) and LDH(0.5 h CS:59±14 U/g liver vs 20 h CS:297 ±71 U/g liver,P<0.05).There was an associated increase in iron release into the perfusate(0.5 h CS:0.11 ±0.03μmoL/g liver vs 20 h CS:0.58±0.10μmoL/g liver,P<0.05)and reduction in bile flow(0.5 h CS: 194±12μL/g vs 20 h CS:71±8μL/g liver,P<0.05). When DFO was added during WI and WP following 20 h of CS,release of iron into the perfusate was de- creased(DFO absent 0.58±0.10μmoL/g liver vs DFO present 0.31±0.06μmoL/g liver,P<0.05),and liver function substantially improved with decreased release of AST(DFO absent 45.9±10.8 U/g liver vs DFO present 8.1±0.9 U/g liver,P<0.05)and LDH(DFO absent 297±71 U/g liver vs DFO present 56±7 U/g liver,P<0.05),and increased bile flow(DFO absent 71±8μL/g liver vs DFO present 237±36μL/g liver, P<0.05).DFO was also shown to improve liver morphology after WP.Cellular injury(the release of LDH and AST)was significantly reduced with the addition of DFO in CS medium but to a lesser extent compared to the addition of DFO in WP or WI and perfusion. There was no effect on liver swelling or bile flow when DFO was only added to the CS medium. CONCLUSION:DFO added during WI and perfusion decreased liver perfusion injury aggravated by extended CS.展开更多

By applying NIRS with multivariate analysis, it was possible to build variation into the calibration sample set, and the Plackett-Burman design, Box-Behnken design, and a concentrating-diluting method were used to obtain the sample set covered with sufficient fluctuation of process parameters and extended concentration information. NIR data were calibrated to predict the four quality indicators using partial least squares regression(PLSR). In the four calibration models, the root mean squares errors of prediction(RMSEPs) were 1.22 μg/ml, 10.5 μg/ml, 1.43 μg/ml, and 0.433% for lobetyolin, total flavonoids, pigments, and total solid contents, respectively. The results indicated that multi-components quantification of the alcohol precipitation liquid of Codonopsis Radix could be achieved with an NIRS-based method, which offers a useful tool for real-time release testing(RTRT) of intermediates in the manufacture of Codonopsis Radix.展开更多

CONCLUSION:DFO added during WI and perfusion decreased liver perfusion injury aggravated by extended CS.

文摘Prostate cancer(PCa)exhibits epidemiological and molecular heterogeneity.Despite extensive studies of its phenotypic and genetic properties in Western populations, its molecular basis is not clear in Chinese patients. To determine critical molecular characteristics and explore correlations between genomic markers and clinical parameters in Chinese populations,we applied an integrative genetic/transcriptomic assay that combines targeted next-generation sequencing and quantitative real-time PCR(qRT-PCR)on samples from 46 Chinese patients with PCa.Lysine(K)-specific methyltransferase 2D(KMT2D),zinc finger homeobox 3(ZFHX3),A-kinase anchoring protein 9(AKAP9),and GLI family zinc finger 1(GLI1)were frequently mutated in our cohort. Moreover,a clinicopathological analysis showed that RB transcriptional corepressor 1(RB1)deletion was common in patients with a high risk of disease progression.Remarkably, four genomic events,MYC proto-oncogene(MYC)amplification,RB1 deletion,APC regulator of WNT signaling pathway(APC)mutation or deletion, and cyclin-dependent kinase 12(CDK12)mutation,were correlated with poor disease-free survival. In addition,a close link between KMT2D expression and the androgen receptor(AR)signaling pathway was observed both in our cohort and in The Cancer Genome Atlas Prostate Adenocarcinoma(TCGA-PRAD)data.In summary, our results demonstrate the feasibility and benefits of integrative molecular characterization of PCa samples in disease pathology research and personalized medicine.

文摘A near infrared spectroscopy(NIRS) approach was established for quality control of the alcohol precipitation liquid in the manufacture of Codonopsis Radix. The results indicated that multi-components quantification of the alcohol precipitation liquid of Codonopsis Radix could be achieved with an NIRS-based method, which offers a useful tool for real-time release testing(RTRT) of intermediates in the manufacture of Codonopsis Radix.

文摘Objective:Critical process parameters(CPPs)identification is an important step of the implementation of quality by design(Qb D)concept. There are many CPP identification methods, such as risk analysis method, sensitivity analysis method, multiple linear regression method, standard partial regression coefficient(SPRC)method, and so on.The SPRC method can consider multiple process critical quality attributes(CQAs)simultaneously, but the determination of CPP number is subjective. Therefore, new CPP identification method is still required. Methods:The manufacturing process of Astragali Radix extract, which contained water reflux extraction, concentration, and ethanol precipitation, was used as an example.First, the multiple process CQAs were determined to be the yield of pigment, dry matter, sugars, and active ingredients.Second, the potential CPPs were determined by a knowledge organization method.Plackett-Burman designed experiments were then performed.A weighted determination coefficient(R2 w)method was presented to identify CPPs.In this method, the importance of different CQAs was considered.Process parameters were removed one-by-one according to their importance index. The decrease in R2 wwas used to characterize the importance of the removed parameter. If the decrease of R2 wwas less than a preset threshold, the removed parameter was not a CPP.Results:During the manufacturing process of Astragali Radix extract, the potential CPPs determined by the knowledge organization method were water consumption, reflux extraction time, extraction frequency, ethanol content, ethanol consumption, and concentration endpoint.Reflux extraction time, the first ethanol consumption, the second ethanol consumption, and the second ethanol precipitation refrigeration temperature were found to be CPPs using the weighted determination coefficient method with the threshold of 10%.Conclusion:Using the weighted determination coefficient method,CPPs can be determined with all the CQAs considered based on their importance. The determination of CPP number is more objective compared with the SPRC method.
