文摘MetarhizosidesA-G(1-7),seven new polysubstituted phenyl glucosides, were isolated from the extracts of solid rice medium of a marine-derived fungus Metarrhizium anisopliae.Compounds 1-7 all contain a polysubstituted phenyl group and the sugar unit is identified as 4'-O-methyl-β-D-glucopyranose.Their structures were elucidated by NMR spectroscopy and chemical method. These compounds were evaluated for anti-inflammatory activity by using LPS-stimulated murine macrophage RAW 264.7 cells and the cytotoxicities against four human cancer cell lines.
文摘Gaussian network model(GNM)is an efficient method to investigate the structural dynamics of biomolecules. However, the application of GNM on RNAs is not as good as that on proteins, and there is still room to improve the model. In this study, two novel approaches, named the weighted GNM(wGNM)and the force-constant-decayed GNM(fcdGNM),were proposed to enhance the performance of ENM in investigating the structural dynamics of RNAs.In wGNM,the force constant for each spring is weighted by the number of interacting heavy atom pairs between two nucleotides. In fcdGNM,all the pairwise nucleotides were connected by springs and the force constant decayed exponentially with the separate distance of the nucleotide pairs. The performance of these two proposed models was evaluated by using a non-redundant RNA structure database composed of 51 RNA molecules. The calculation results show that both the proposed models outperform the conventional GNM in reproducing the experimental B-factors of RNA structures.Compared with the conventional GNM,the Pearson correlation coefficient between the predicted and experimental B-factors was improved by 9.85%and 6.76%for wGNM and fcdGNM,respectively. Our studies provide two candidate methods for better revealing the dynamical properties encoded in RNA structures.

文摘α-C(sp^(3))-H arylation is an important type of C-H functionalization.Various biologically significant natural products,chemi-cal intermediates, and drugs have been effectively prepared via C-H functionalization.Cyclic carbonyl compounds comprise of cyclic ketones, enones, lactones, and lactams.Theα-C(sp^(3))-H arylation of these compounds have been exhibited high efficiency in forming C(sp^(3))-C(sp^(2))bonds,played a crucial role in organic synthesis, and attracted majority of interests from organic and medicinal communities. This review focused on the most significant advances including methods, mechanism, and applications in total synthesis of natural products in the field ofα-C(sp^(3))-H arylations of cyclic carbonyl compounds in recent years.

文摘The major vascular complications associated with diabetes make the management of diabetic mellitus erectile dysfunction(DMED)a challenging endeavor.Notable factors contributing to DMED include oxidative stress, nitric oxide(NO)/cyclic guanosine monophosphate(cGMP)pathway activation, and apoptosis, while nitro-oleic acid(NO,-OA)has been shown to be beneficial in treating these aspects of this condition. We,herein, investigated the effects and possible mechanisms of NO,-OA on erectile function as assessed in a streptozotocin-induced rat model of diabetes. Our results revealed that the erectile function of DMED rats was significantly impaired compared with that of the control group. However,in response to 4 weeks of NO,-OA treatment, there was an improvement in erectile function. The expression of oxidative stress-related indicators was significantly increased and the NO/cGMP pathway was impaired in the DMED group. The expression of proapoptotic factors was increased, while that of antiapoptotic factors was decreased in the DMED group. Moreover, the cell morphology in the cavernous tissue of the DMED group also changed adversely.NO,-OA treatment significantly reversed all these changes observed in the DMED group. In conclusion,NO,-OA treatment partially improved erectile function in DMED rats through mechanisms that included inhibition of oxidative stress, activation of the NO/cGMP pathway, and a reduction in apoptosis.
