Upland rice shows dryland adaptation in the form of a deeper and denser root system and greater drought resistance than its counterpart,irrigated rice.Our previous study revealed a difference in the frequency of the OsNCED2 gene between upland and irrigated populations.A nonsynonymous mutation(C to T,from irrigated to upland rice)may have led to functional variation fixed by artificial selection,but the exact biological function in dryland adaptation is unclear.In this study,transgenic and association analysis indicated that the domesticated fixed mutation caused functional variation in OsNCED2,increasing ABA levels,root development,and drought tolerance in upland rice under dryland conditions.OsNCED2-overexpressing rice showed increased reactive oxygen species-scavenging abilities and transcription levels of many genes functioning in stress response and development that may regulate root development and drought tolerance.OsNCED2^(T)-NILs showed a denser root system and drought resistance,promoting the yield of rice under dryland conditions.OsNCED2^(T)may confer dryland adaptation in upland rice and may find use in breeding dryland-adapted,water-saving rice. 展开更多

Runoff Modeling in Ungauged Catchments Using Machine Learning Algorithm-Based Model Parameters Regionalization Methodology

Model parameters estimation is a pivotal issue for runoff modeling in ungauged catchments.The nonlinear relationship between model parameters and catchment descriptors is a major obstacle for parameter regionalization,which is the most widely used approach.Runoff modeling was studied in 38 catchments located in the Yellow–Huai–Hai River Basin(YHHRB).The values of the Nash–Sutcliffe efficiency coefficient(NSE),coefficient of determination(R2),and percent bias(PBIAS)indicated the acceptable performance of the soil and water assessment tool(SWAT)model in the YHHRB.Nine descriptors belonging to the categories of climate,soil,vegetation,and topography were used to express the catchment characteristics related to the hydrological processes.The quantitative relationships between the parameters of the SWAT model and the catchment descriptors were analyzed by six regression-based models,including linear regression(LR)equations,support vector regression(SVR),random forest(RF),k-nearest neighbor(kNN),decision tree(DT),and radial basis function(RBF).Each of the 38 catchments was assumed to be an ungauged catchment in turn.Then,the parameters in each target catchment were estimated by the constructed regression models based on the remaining 37 donor catchments.Furthermore,the similaritybased regionalization scheme was used for comparison with the regression-based approach.The results indicated that the runoff with the highest accuracy was modeled by the SVR-based scheme in ungauged catchments.Compared with the traditional LR-based approach,the accuracy of the runoff modeling in ungauged catchments was improved by the machine learning algorithms because of the outstanding capability to deal with nonlinear relationships.The performances of different approaches were similar in humid regions,while the advantages of the machine learning techniques were more evident in arid regions.When the study area contained nested catchments,the best result was calculated with the similarity-based parameter regionalization scheme because of the high catchment density and short spatial distance.The new findings could improve flood forecasting and water resources planning in regions that lack observed data. 展开更多

Single nucleotide polymorphism(SNP)genotyping arrays provide an optimal high-throughput platform for genetic research and molecular breeding programs in both animals and plants.In this study,a highquality and custom-designed Rice3K56 SNP array was developed with the resequencing data of 3024 rice accessions worldwide,which was then tested extensively in 192 representative rice samples.Printed on the Gene Titan chips of Affymetrix Axiom each containing 56,606 SNP markers,the Rice3K56 array has a high genotyping reliability(99.6%),high and uniform genome coverage(an average of 6.7-kb between adjacent SNPs),abundant polymorphic information and easy automation,compared with previously developed rice SNP arrays.When applied in rice varietal differentiation,population diversity analysis,gene mapping of 13 complex traits by a genome-wide association study analysis(GWAS),and genome selection experiments in a recombinant inbred line and a multi-parent advanced generation inter-cross populations,these properties of the Rice3K56 array were well demonstrated for its power and great potential to be a highly efficient tool for rice genetic research and genomic breeding. 展开更多

SDN (Software Defined Network) has many security problems, and DDoS attack is undoubtedly the most serious harm to SDN architecture network. How to accurately and effectively detect DDoS attacks has always been a difficult point and focus of SDN security research. Based on the characteristics of SDN, a DDoS attack detection method combining generalized entropy and PSOBP neural network is proposed. The traffic is pre-detected by the generalized entropy method deployed on the switch, and the detection result is divided into normal and abnormal. Locate the switch that issued the abnormal alarm. The controller uses the PSO-BP neural network to detect whether a DDoS attack occurs by further extracting the flow features of the abnormal switch. Experiments show that compared with other methods, the detection accurate rate is guaranteed while the CPU load of the controller is reduced, and the detection capability is better. 展开更多

Identification of metabolites responsible for tolerance to low nitrogen availability(low-N)will aid in the genetic improvement of rice yield under nitrogen deficiency.In this study,a backcross introgression line(G9)and its recurrent parent Shuhui 527(SH527),which show differential responses to low-N stress,were used to identify metabolites associated with lowN tolerance in rice.Differences in metabolite contents in the leaves of G9 and SH527 at three growth stages under low-N stress were assessed by gas chromatography–mass spectrometry.Many metabolites,including amino acids and derivatives,were highly enriched in G9 compared with SH527 under the control condition,suggesting that the two genotypes had basal metabolite differences.Low-N stress induced genotype-specific as well as growth stagedependent metabolite changes.Metabolites induced specifically in G9 that were involved in glycolysis and tricarboxylic acid metabolism were enriched at the tillering and grain filling stages,and metabolites involved in nitrogen and proline metabolism were enriched at the booting stage.Enrichment of pyroglutamate,glutamate,2-oxoglutarate,sorbose,glycerate-2-P,and phosphoenolpyruvic acid in G9 suggests that these metabolites could be involved in low-N stress tolerance.The results presented here provide valuable information for further elucidation of the molecular mechanisms of low-N tolerance in crops. 展开更多

Plant microRNAs(miRNAs)play important roles in biological processes such as development and stress responses.Although the diverse functions of miRNAs in model organisms have been well studied,their function in wild rice is poorly understood.In this study,high-throughput small RNA sequencing was performed to characterize tissue-specific transcriptomes in Oryza longistaminata.A total of 603 miRNAs,380 known rice miRNAs,72 conserved plant miRNAs,and151 predicted novel miRNAs were identified as being expressed in aerial shoots and rhizomes.Additionally,99 and 79 miRNAs were expressed exclusively or differentially,respectively,in the two tissues,and 144 potential targets were predicted for the differentially expressed miRNAs in the rhizomes.Functional annotation of these targets suggested that transcription factors,including squamosa promoter binding proteins and auxin response factors,function in rhizome growth and development.The expression levels of several miRNAs and target genes in the rhizomes were quantified by RT-PCR,and the results indicated the existence of complex regulatory mechanisms between the miRNAs and their targets.Eight target cleavage sites were verified by RNA ligase-mediated rapid 5′end amplification.These results provide valuable information on the composition,expression and function of miRNAs in O.longistaminata,and will aid in understanding the molecular mechanisms of rhizome development. 展开更多

Dominant early heading(DEH)in rice(Oryza sativa L.)is of interest in both breeding and genetics.The genetic mechanisms underlying DEH have remained largely unclear.We have developed a near-isogenic DEH line without yield drag named DEH_229 by sister-line backcross(BC)breeding with MH63,a restorer,as the genetic background.We conducted a pilot genetic investigation under both short-day(SD)and long-day(LD)conditions.The DEH line harbored only 1.06%variation in the genome sequence relative to MH63.The variants were distributed throughout the genome.Using QTL mapping by sequencing(QTL-seq)on an F_(2) population derived from a cross of MH63×DEH_229,57 loci were detected under the SD condition.Joint mapping employing a genome-wide association study with accessions from the 3000 rice genome sequencing project(3K-RG),reduced the number of QTL by 43.9%.Using Rice Functional Genomics&Breeding(RFGB)database,the number of SNP cluster regions within the QTL regions reduced by 27.3%.Further comparison of the genome variation between DEH_229 and MH63 in addition to gene annotation information revealed a new DEH allele of DTH3 with multiple variable sites as a possible major factor underlying the early-heading phenotype of DEH_229.An InDel marker,ZMEH_1,was designed based on the variation between DEH_229 and MH63 within this region.It accounted for 86.0%of heading date variation under both SD and LD conditions in 109 randomly chosen progeny derived from extreme lines of the MH63×DEH_229 population.This study reveals the genetic complexity of DEH in the near-isogenic line and may provide useful material and marker information for plant molecular breeding. 展开更多

A finite element model updating technique for complicated beam-type structures is presented in this study.Firstly, a complicated beam-type structure is reduced to a reduced super beam model with a much smaller degree of freedom by using the reduced super beam method, which is based on the classic plane cross-section assumption and displacement interpolation function of beam theory.Then based on the reduced super beam, the analysis of eigensolutions and eigensensitivities from the reduced eigenequation are processed for model updating, which will greatly reduce the computational effort when compared to the traditional model updating methods performed on the global model.Optimization techniques are adopted for updating the difference of modal dynamic properties, resulting in optimal values of the structural parameters.Finally, a complicated stiffened cylindrical shell model and a practical missile structure, served as the illustrative examples, are employed for model updating application, which demonstrate that the reduced super beam-based method is both effective and highly efficient. 展开更多

Effect of Cardioplegia for Myocardial Protection in Pediatric Cardiac Surgery:A Network Meta-Analysis

Cardioplegia has been widely used to reduce myocardial injury during pediatric cardiac surgery;however,which cardioplegia solution has the best protective effect has not been established.Thus,we compared the myocardial protective effects of different cardioplegia solutions used in pediatric cardiac surgery.Seven databases were searched to identify the relevant randomized controlled trials.A network meta-analysis with a Bayesian framework was conducted.The outcomes included the following biochemical and clinical outcomes:serum concentrations of the creatine kinase-myocardial band at 6 h postoperatively;cardiac troponin I(cTnI)at 4,12,and 24 h postoperatively;spontaneous beating after declamping;postoperative arrhythmias;inotropic support percentage and duration;mechanical ventilation hours;intensive care unit stay in days;hospital stay in days;and mortality.The group treated with cold crystalloid cardioplegia(cCCP)was chosen as the control group.The 22 studies involved 1529 patients.Six types of cardioplegia solutions were described in these studies,including cold blood cardioplegia,cCCP,del Nido,histidine-tryptophan-ketoglutarate(HTK),terminal warm blood cardioplegia,and warm blood cardioplegia(wBCP).The serum concentrations of the 24-h cTnI with wBCP(MD=−2.52,95%CI:−4.74 to−0.27)was significantly lower than cCCP.The serum concentrations of the 24-h cTnI with HTK(MD=4.91,95%CI:2.84–7.24)was significantly higher than cCCP.There was no significant difference in other biochemical and clinical outcomes when compared to cCCP.In conclusion,wBCP may have a superior myocardial protective effect with lower 24-h cTnI levels postoperatively and similar clinical outcomes after pediatric cardiac surgery. 展开更多

Objective To investigate the application of oncoplastic surgery in breast-conserving surgery.Methods We retrospectively analyzed the clinical data of 103 breast cancer patients who underwent breast-conserving surgery in the First Affiliated Hospital of Henan University.All the patients were female whose tumor volume-to-breast volume ratio was greater than 20%.Fifty-two patients were treated with oncoplastic breast-conversing surgery(observation group),and 51 patients were treated with traditional breast-conserving surgery(control group).The volume of resected tissue,subjective satisfaction with breast shape,objective score of breast shape,and follow-up were compared between the two groups.Results In the observation group,the weight of resected breast tissue was 64.2–172.1 g,with a median of 98.7 g.In the control group,the weight of resected breast tissue was 67.5–175.7 g,with a median of 102.3 g.After 12 months of follow-up,the subjective satisfaction rate and objective score of breast shape in the observation group were significantly better than those in the traditional breast-conserving surgery group(P<0.05).There was no recurrence,metastasis,or death in the two groups.There was no significant difference in postoperative complications between the two groups(P>0.05).Conclusion Oncoplastic breast-conserving surgery leads to better cosmetic results and a more satisfactory clinical results. 展开更多

Gorham-Stout disease(GSD)is a sporadic chronic disease characterized by progressive bone dissolution,absorption,and disappearance along with lymphatic vessel infiltration in bone-marrow cavities.Although the osteolytic mechanism of GSD has been widely studied,the cause of lymphatic hyperplasia in GSD is rarely investigated.In this study,by comparing the RNA expression profile of osteoclasts(OCs)with that of OC precursors(OCPs)by RNA sequencing,we identified a new factor,semaphorin 3A(Sema3A),which is an osteoprotective factor involved in the lymphatic expansion of GSD.Compared to OCPs,OCs enhanced the growth,migration,and tube formation of lymphatic endothelial cells(LECs),in which the expression of Sema3A is low compared to that in OCPs.In the presence of recombinant Sema3A,the growth,migration,and tube formation of LECs were inhibited,further confirming the inhibitory effect of Sema3A on LECs in vitro.Using an LEC-induced GSD mouse model,the effect of Sema3A was examined by injecting lentivirus-expressing Sema3A into the tibiae in vivo.We found that the overexpression of Sema3A in tibiae suppressed the expansion of LECs and alleviated bone loss,whereas the injection of lentivirus expressing Sema3A short hairpin RNA(shRNA)into the tibiae caused GSD-like phenotypes.Histological staining further demonstrated that OCs decreased and osteocalcin increased after Sema3A lentiviral treatment,compared with the control.Based on the above results,we propose that reduced Sema3A in OCs is one of the mechanisms contributing to the pathogeneses of GSD and that expressing Sema3A represents a new approach for the treatment of GSD. 展开更多

The plant hormone abscisic acid(ABA)is crucial for plant seed germination and abiotic stress tolerance.However,the association between ABA sensitivity and plant abiotic stress tolerance remains largely unknown.In this study,436 rice accessions were assessed for their sensitivity to ABA during seed germination.The considerable diversity in ABA sensitivity among rice germplasm accessions was primarily reflected by the differentiation between the Xian(indica)and Geng(japonica)subspecies and between the upland-Geng and lowland-Geng ecotypes.The upland-Geng accessions were most sensitive to ABA.Genome-wide association analyses identified four major quantitative trait loci containing21 candidate genes associated with ABA sensitivity of which a basic helix-loop-helix transcription factor gene,OsbHLH38,was the most important for ABA sensitivity.Comprehensive functional analyses using knockout and overexpression transgenic lines revealed that OsbHLH38 expression was responsive to multiple abiotic stresses.Overexpression of OsbHLH38 increased seedling salt tolerance,while knockout of OsbHLH38 increased sensitivity to salt stress.A salt-responsive transcription factor,OsDREB2A,interacted with OsbHLH38 and was directly regulated by OsbHLH38.Moreover,OsbHLH38 affected rice abiotic stress tolerance by mediating the expression of a large set of transporter genes of phytohormones,transcription factor genes,and many downstream genes with diverse functions,including photosynthesis,redox homeostasis,and abiotic stress responsiveness.These results demonstrated that OsbHLH38 is a key regulator in plant abiotic stress tolerance. 展开更多

DNA methylation,one of the most important epigenetic phenomena,plays a vital role in tuning gene expression during plant development as well as in response to environmental stimuli.In the present study,a methylation-sensitive amplified polymorphism(MSAP) analysis was performed to profile DNA methylation changes in two contrasting rice genotypes under salt stress.Consistent with visibly different phenotypes in response to salt stress,epigenetic markers classified as stable inter-cultivar DNA methylation differences were determined between salt-tolerant FL478 and salt-sensitive IR29.In addition,most tissue-specific DNA methylation loci were conserved,while many of the growth stage-dependent DNA methylation loci were dynamic between the two genotypes.Strikingly,salt stress induced a decrease in DNA methylation specifically in roots at the seedling stage that was more profound in IR29 than in the FL478.This result may indicate that demethylation of genes is an active epigenetic response to salt stress in roots at the seedling stage,and helps to further elucidate the implications of DNA methylation in crop growth and development. 展开更多

Flavonoids constitute a major group of plant phenolic compounds.While extensively studied in Arabidopsis,profiling and naturally occurring variation of these compounds in rice(Oryza sativa),the monocot model plant,are less reported.Using a collection of rice germplasm,comprehensive profiling and natural variation of flavonoids were presented in this report.Application of a widely targeted metabolomics method facilitated the simultaneous identification and quantification of more than 90 flavonoids using liquid chromatography tandem mass spectrometry(LC-MS/MS).Comparing flavonoid contents in various tissues during different developmental stages revealed tissue-specific accumulation of most flavonoids.Further investigation indicated that flavone mono-C-glycosides,malonylated flavonoid O-hexosides,and some flavonoid O-glycosides accumulated at significantly higher levels in indica than in japonica,while the opposite was observed for aromatic acylated flavone C-hexosyl-O-hexosides.In contrast to the highly differential accumulation between the two subspecies,relatively small variations within subspecies were detected for most flavonoids.Besides,an association analysis between flavonoid accumulation and its biosynthetic gene sequence polymorphisms disclosed that natural variation of flavonoids was probably caused by sequence polymorphisms in the coding region of flavonoid biosynthetic genes.Our work paves the way for future dissection of biosynthesis and regulation of flavonoid pathway in rice. 展开更多
