Interleukin 17(IL-17)and its main producer,T cell receptorγδcells,have neurotoxic effects in the pathogenesis of intracerebral hemorrhage(ICH),aggravating brain injuries.To investigate the correlation between IL-17 ...Interleukin 17(IL-17)and its main producer,T cell receptorγδcells,have neurotoxic effects in the pathogenesis of intracerebral hemorrhage(ICH),aggravating brain injuries.To investigate the correlation between IL-17 and ICH,we dynamically screened serum IL-17 concentrations using enzyme-linked immunosorbent assay and explored the clinical values of IL-17 in ICH patients.There was a significant negative correlation between serum IL-17 level and neurological recovery status in ICH patients(r=–0.498,P<0.01).To study the neurotoxic role of IL-17,C57 BL/6 mice were used to establish an ICH model by injecting autologous blood into the caudate nucleus.Subsequently,the mice were treated with mouse neural stem cells(NSCs)and/or IL-17 neutralizing antibody for 72 hours.Flow cytometry,brain water content detection,Nissl staining,and terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling results indicated that NSC transplantation significantly reduced IL-17 expression in peri-hematoma tissue,but there was no difference in T cell receptorγδcells.Compared with the ICH group,there were fewer apoptotic bodies and more Nissl bodies in the ICH+NSC group and the ICH+NSC+IL-17 group.To investigate the potential effect of IL-17 on directional differentiation of NSCs,we cultured mouse NSCs(NE-4 C)alone or co-cultured them with T cell receptorγδcells,which were isolated from mouse peripheral blood mononuclear cells,for 7 days.The results of western blot assays revealed that IL-17 secreted by T cell receptorγδcells reduced the differentiation of NSCs into astrocytes and neurons,while IL-17 neutralization relieved the inhibition of directional differentiation into astrocytes rather than neurons.In conclusion,serum IL-17 levels were elevated in the early stage of ICH and were negatively correlated with outcome in ICH patients.Animal experiments and cytological investigations therefore demonstrated that IL-17 probably has neurotoxic roles in ICH because of its inhibitory effects on the directional differentiation of NSCs.The application of IL-17 neutralizing antibody may promote the directional differentiation of NSCs into astrocytes.This study was approved by the Clinical Research Ethics Committee of Anhui Medical University of China(For human study:Approval No.20170135)in December 2016.All animal handling and experimentation were reviewed and approved by the Institutional Animal Care and Use Committee of Anhui Medical University(approval No.20180248)in December 2017.展开更多
Dexamethasone has been widely used after various neurosurgical procedures due to its anti-inflammatory property and the abilities to restore vascular permeability,inhibit free radicals,and reduce cerebrospinal fluid p...Dexamethasone has been widely used after various neurosurgical procedures due to its anti-inflammatory property and the abilities to restore vascular permeability,inhibit free radicals,and reduce cerebrospinal fluid production.According to the latest guidelines for the treatment of traumatic brain injury in the United States,high-dose glucocorticoids cause neurological damage.To investigate the reason why high-dose glucocorticoids after traumatic brain injury exhibit harmful effect,rat controlled cortical impact models of traumatic brain injury were established.At 1 hour and 2 days after surgery,rat models were intraperitoneally administered dexamethasone 10 mg/kg.The results revealed that 31 proteins were significantly upregulated and 12 proteins were significantly downregulated in rat models of traumatic brain injury after dexamethasone treatment.The Ingenuity Pathway Analysis results showed that differentially expressed proteins were enriched in the mitochondrial dysfunction pathway and synaptogenesis signaling pathway.Western blot analysis and immunohistochemistry results showed that Ndufv2,Maob and Gria3 expression and positive cell count in the dexamethasone-treated group were significantly greater than those in the model group.These findings suggest that dexamethasone may promote a compensatory increase in complex I subunits(Ndufs2 and Ndufv2),increase the expression of mitochondrial enzyme Maob,and upregulate synaptic-transmission-related protein Gria3.These changes may be caused by nerve injury after traumatic brain injury treatment by dexamethasone.The study was approved by Institutional Ethics Committee of Beijing Neurosurgical Institute(approval No.201802001)on June 6,2018.展开更多

Zero Poisson’s ratio(ZPR)mechanical metamaterials can yield no transverse displacements when unidi-rectionally compressed,and cylindrical sandwich meta-structures composed of semi re-entrant(SRE)ZPR metamaterials are...Zero Poisson’s ratio(ZPR)mechanical metamaterials can yield no transverse displacements when unidi-rectionally compressed,and cylindrical sandwich meta-structures composed of semi re-entrant(SRE)ZPR metamaterials are thus explored for applications on cylindrical shells of underwater equipment or sub-mersible structures.A group of ZPR unit cells with specified pre-strained wave propagation characteristics and adequate load-bearing capabilities is optimally designed based on the periodic boundary condition(PBC)and Bloch’s Theorem.The sound transmission and pressure-resistant performance of cylindrical sandwich meta-structures comprising the homogeneous and graded SRE ZPR unit cells are then investi-gated.The results show that the designed meta-structures can perfectly yield better vibroacoustic atten-uation behavior within the specified frequency regions corresponding to the pre-strained band gaps and safely bear the hydrostatic pressure equivalent to 1000 m depth with low weight-bulk ratios.In addition,the functionally graded metamaterial core can boost vibroacoustic performance within broader frequency ranges.展开更多
ObjectiveTo explore the clinical effectiveness of xingnao kaiqiao (regaining consciousness and opening orifice) acupuncture therapy for patients with acute ischemic stroke, and establish a prediction model for prognos...ObjectiveTo explore the clinical effectiveness of xingnao kaiqiao (regaining consciousness and opening orifice) acupuncture therapy for patients with acute ischemic stroke, and establish a prediction model for prognosis for the target population through Logistic regression analysis.MethodsNinety-six patients with acute ischemic stroke were randomly assigned to either an ozone therapy group or an ozone therapy plus xingnao kaiqiao acupuncture group (OA group) with a 1:1 ratio. The patients in the ozone group was treated with ozone therapy, and those in the OA group were treated with acupuncture at Nèiguān (内关PC6), Shuĭgōu (水沟GV26), Sānyīnjiāo (三阴交SP6), lower Jíquán (下极泉Extra, lower HT1), Wĕizhōng (委中BL40) and Chĭzé (尺泽LU5) on the basis of the treatment as the ozone therapy group. The treatment was given once daily for 7 days as a session and 3 consecutive sessions were required. Before treatment and 48 h after treatment, the serological indicators (interleukin-6 [IL-6], tumor necrosis factor-α [TNF-α] and hypersensitive C-reactive protein [hs-CRP]) were tested. Using the National Institute of Health stroke scale (NIHSS) score and Montreal cognitive assessment (MoCA) score, the neurological and cognitive functions of patients were assessed. The clinical efficacy was evaluated with the modified Rankin scale, and the incidence of adverse effects was analyzed statistically in two groups. The multivariate Logistic regression analysis was adopted to detect the risk factors of the prognosis in patients and the nomographic prediction model was established.Results(1) clinical therapeutic effect: after treatment, the levels of IL-6, TNF-α and Hs-CRP in the OA group were significantly lower than that in the ozone group (all P < 0.05). After treatment, compared with the ozone therapy group, NIHSS score was lower and the scores of BI and MoCA were significantly higher in the OA group (all P < 0.05). The total effective rate of the OA group was 91.67%, higher than that of the ozone therapy group (62.50%, P < 0.05). The incidence of adverse effects in the OA group was lower when compared with that in the ozone group (P < 0.05);(2) establishment of prediction model for prognosis: age, history of diabetes, cardiogenic cerebral embolism, high density lipoprotein-cholesterol (HDL-C), white blood cell (WBC) and treatment methods were independent risk factors of the unfavorable prognosis for the patients with acute stroke. The established model had sound accuracy for prediction of prognosis.ConclusionXingnao kaiqiao acupuncture was effective on acute ischemic stroke, and the prognosis of this therapy was increased significantly when combined with the ozone therapy. The preventive measures should be set up for patients with risk factors, e.g. advanced age, diabetes, cardiogenic cerebral embolism, low HDL-C and abnormal WBC, in order to further reduce the risk of unfavorable prognosis in patients with acute stroke.展开更多
Background Surgical site infection(SSI)is a common complication following craniotomy that increases morbidity,mortality,and medical expenses.The objectives of this study were to determine the relevant risk factors ass...Background Surgical site infection(SSI)is a common complication following craniotomy that increases morbidity,mortality,and medical expenses.The objectives of this study were to determine the relevant risk factors associated with SSI after elective craniotomy for brain tumor and analyse the treatments for SSI.Methods A retrospective nested case-control study was conducted using data from patients who underwent craniotomy for brain tumor resection at the Neurosurgical Oncology Department No.6 of Beijing Tiantan Hospital,Capital Medical University,between January 2019 and December 2021.Risk factors for SSI were determined using multivariate logistic regression analysis.We analyzed microbiological and related treatment data for different SSI types.Results Among 2061 patients who underwent craniotomy for brain tumor,31 had SSI(1.50%).In the multivariate logistic regression analysis,body mass index(BMI)and operative duration were identified as independent risk factors for SSI.The most common microorganism isolated from SSIs was Staphylococcus epidermidis(22.9%),and drug sensitivity results showed that gram-positive bacteria were sensitive to linezolid,vancomycin and tigecycline,whereas gram-negative bacteria were sensitive to meropenem,cefepime and ceftazidime.Six of the seven patients who underwent bone flap removal due to osteomyelitis were infected with gram-negative bacteria.Conclusions BMI and operative duration were identified as independent risk factors for SSI.Diabetes mellitus,previous ratio therapy,type of incision,recurrence tumor and other risk factors were not found to be associated with the occurrence of SSI in this study.展开更多

Here we present SHRIMP zircon data for the rhyolite in Well Baocan 1, one of the deepest wells in the basin. The 283.9±2.7 Ma reported in our study provides the best precise age determination for the Yierxitu Formation, the oldest Permian layer of Hami Depression, one of the three substructural units of the Turpan-Hami Basin, and a potential hydrocarbon reservoir in this basin. Our data refines earlier imprecise 39^Ar-40^Ar ages and shows that the volcanic rocks both inside the Turpan-Hami Basin and along its margin are almost coeval. We delineate a collisional orogenesis, and the new age of 283.9±2.7 Ma may limit the latest time of the collision orogenesis.展开更多

The Pb^2+-induced aggregation in Au NP solution is monitored by both colorimetric response and UV-vis spectroscopy. A rather broad linear range (from 0.1 to 30 pmol/L) and low detection limit (0.1 pmol/L) are explored for Au NP sensors used for detection of Pb^2+ ions. Furthermore, the response of GSH-stabilized Au NPs toward Pb^2+ ions is specific compared with other possible interferants (Hg^2+, Mg^2+, Zn^2+, Ni^2+, Cu^2+, Co^2+, Ca^2+, Mn^2+, Cd^2+, and Ba^2+).展开更多
Local drug delivery has received increasing attention in recent years.However,the therapeutic efficacy of local delivery of drugs is still limited under certain scenarios,such as in the oral cavity or in wound beds af...Local drug delivery has received increasing attention in recent years.However,the therapeutic efficacy of local delivery of drugs is still limited under certain scenarios,such as in the oral cavity or in wound beds after resection of tumors.In this study,we introduce a bioinspired adhesive hydrogel derived from the skin secretions of Andrias davidianus(SSAD)as a wound dressing for localized drug elution.The hydrogel was loaded with aminoguanidine or doxorubicin,and its controlled drug release and healing-promoting properties were verified in a diabetic rat palatal mucosal defect model and a C57BL/6 mouse melanoma-bearing model,respectively.The results showed that SSAD hydrogels with different pore sizes could release drugs in a controllable manner and accelerate wound healing.Transcriptome analyses of the palatal mucosa suggested that SSAD could significantly upregulate pathways linked to cell adhesion and extracellular matrix deposition and had the ability to recruit keratinocyte stem cells to defect sites.Taken together,these findings indicate that property-controllable SSAD hydrogels could be a promising biofunctional wound dressing for local drug delivery and promotion of wound healing.展开更多

The preventive measures should be set up for patients with risk factors, e.g. advanced age, diabetes, cardiogenic cerebral embolism, low HDL-C and abnormal WBC, in order to further reduce the risk of unfavorable prognosis in patients with acute stroke.
文摘Background Surgical site infection(SSI)is a common complication following craniotomy that increases morbidity,mortality,and medical expenses.The objectives of this study were to determine the relevant risk factors associated with SSI after elective craniotomy for brain tumor and analyse the treatments for SSI.Methods A retrospective nested case-control study was conducted using data from patients who underwent craniotomy for brain tumor resection at the Neurosurgical Oncology Department No.6 of Beijing Tiantan Hospital,Capital Medical University,between January 2019 and December 2021.Risk factors for SSI were determined using multivariate logistic regression analysis.We analyzed microbiological and related treatment data for different SSI types.Results Among 2061 patients who underwent craniotomy for brain tumor,31 had SSI(1.50%).In the multivariate logistic regression analysis,body mass index(BMI)and operative duration were identified as independent risk factors for SSI.The most common microorganism isolated from SSIs was Staphylococcus epidermidis(22.9%),and drug sensitivity results showed that gram-positive bacteria were sensitive to linezolid,vancomycin and tigecycline,whereas gram-negative bacteria were sensitive to meropenem,cefepime and ceftazidime.Six of the seven patients who underwent bone flap removal due to osteomyelitis were infected with gram-negative bacteria.Conclusions BMI and operative duration were identified as independent risk factors for SSI.Diabetes mellitus,previous ratio therapy,type of incision,recurrence tumor and other risk factors were not found to be associated with the occurrence of SSI in this study.

文摘Permian volcanic rocks are widely distributed in the Turpan-Hami Basin, which is part of the Central Asian orogenic belt. Here we present SHRIMP zircon data for the rhyolite in Well Baocan 1, one of the deepest wells in the basin. The 283.9±2.7 Ma reported in our study provides the best precise age determination for the Yierxitu Formation, the oldest Permian layer of Hami Depression, one of the three substructural units of the Turpan-Hami Basin, and a potential hydrocarbon reservoir in this basin. Our data refines earlier imprecise 39^Ar-40^Ar ages and shows that the volcanic rocks both inside the Turpan-Hami Basin and along its margin are almost coeval. We delineate a collisional orogenesis, and the new age of 283.9±2.7 Ma may limit the latest time of the collision orogenesis.
文摘Direct mixture of Au^3+ with glutathione (GSH), which act as both reduction agents and stabilizers, in aqueous solution gave rise to production of gold nanoparticles (Au NPs) with uniform sizes of around 21 nm. The GSH stabilizer Au NPs in solution show immediate aggregation after addition of 1 mol/L NaCI aqueous solution containing Pb^2+ ions. The Pb^2+-induced aggregation in Au NP solution is monitored by both colorimetric response and UV-vis spectroscopy. A rather broad linear range (from 0.1 to 30 pmol/L) and low detection limit (0.1 pmol/L) are explored for Au NP sensors used for detection of Pb^2+ ions. Furthermore, the response of GSH-stabilized Au NPs toward Pb^2+ ions is specific compared with other possible interferants (Hg^2+, Mg^2+, Zn^2+, Ni^2+, Cu^2+, Co^2+, Ca^2+, Mn^2+, Cd^2+, and Ba^2+).

文摘Local drug delivery has received increasing attention in recent years.However,the therapeutic efficacy of local delivery of drugs is still limited under certain scenarios,such as in the oral cavity or in wound beds after resection of tumors.In this study,we introduce a bioinspired adhesive hydrogel derived from the skin secretions of Andrias davidianus(SSAD)as a wound dressing for localized drug elution.The hydrogel was loaded with aminoguanidine or doxorubicin,and its controlled drug release and healing-promoting properties were verified in a diabetic rat palatal mucosal defect model and a C57BL/6 mouse melanoma-bearing model,respectively.The results showed that SSAD hydrogels with different pore sizes could release drugs in a controllable manner and accelerate wound healing.Transcriptome analyses of the palatal mucosa suggested that SSAD could significantly upregulate pathways linked to cell adhesion and extracellular matrix deposition and had the ability to recruit keratinocyte stem cells to defect sites.Taken together,these findings indicate that property-controllable SSAD hydrogels could be a promising biofunctional wound dressing for local drug delivery and promotion of wound healing.
