The proliferation of Internet of Things(IoT)systems has resulted in the generation of substantial data,presenting new challenges in reliable storage and trustworthy sharing.Conventional distributed storage systems are hindered by centralized management and lack traceability,while blockchain systems are limited by low capacity and high latency.To address these challenges,the present study investigates the reliable storage and trustworthy sharing of IoT data,and presents a novel system architecture that integrates on-chain and off-chain data manage systems.This architecture,integrating blockchain and distributed storage technologies,provides high-capacity,high-performance,traceable,and verifiable data storage and access.The on-chain system,built on Hyperledger Fabric,manages metadata,verification data,and permission information of the raw data.The off-chain system,implemented using IPFS Cluster,ensures the reliable storage and efficient access to massive files.A collaborative storage server is designed to integrate on-chain and off-chain operation interfaces,facilitating comprehensive data operations.We provide a unified access interface for user-friendly system interaction.Extensive testing validates the system’s reliability and stable performance.The proposed approach significantly enhances storage capacity compared to standalone blockchain systems.Rigorous reliability tests consistently yield positive outcomes.With average upload and download throughputs of roughly 20 and 30 MB/s,respectively,the system’s throughput surpasses the blockchain system by a factor of 4 to 18. 展开更多

Advances in noble metal-modified g-C_(3)N_(4) heterostructures toward enhanced photocatalytic redox ability

The photocatalytic activity of catalysts depends on the energy-harvesting ability and the separation or transport of photogenerated carriers.The light absorption capacity of graphitic carbon nitride(g-C_(3)N_(4))-based composites can be enhanced by adjusting the surface plasmon resonance(SPR)of noble metal nanoparticles(e.g.,Cu,Au,and Pd)in the entire visible region.Adjustments can be carried out by varying the nanocomponents of the materials.The SPR of noble metals can enhance the local electromagnetic field and improve interband transition,and resonant energy transfer occurs from plasmonic dipoles to electron-hole pairs via near-field electromagnetic interactions.Thus,noble metals have emerged as relevant nanocomponents for g-C_(3)N_(4) used in CO_(2) photoreduction and water splitting.Herein,recent key advances in noble metals(either in single atom,cluster,or nanoparticle forms)and composite photocatalysts based on inorganic or organic nanocomponent-incorporated g-C_(3)N_(4) nanosheets are systematically discussed,including the applications of these photocatalysts,which exhibit improved photoinduced charge mobility in CO_(2) photoconversion and H2 production.Issues related to the different types of multi-nanocomponent heterostructures(involving Schottky junctions,Z-/S-scheme heterostructures,noble metals,and additional semiconductor nanocomponents)and the adjustment of dimensionality of heterostructures(by incorporating noble metal nanoplates on g-C_(3)N_(4) forming 2D/2D heterostructures)are explored.The current prospects and possible challenges of g-C_(3)N_(4) composite photocatalysts incorporated with noble metals(e.g.,Au,Pt,Pd,and Cu),particularly in water splitting,CO_(2) reduction,pollution degradation,and chemical conversion applications,are summarized. 展开更多

Background:Osteoporosis is a chronic bone disease characterized by bone loss and decreased bone strength.However,current anti-resorptive drugs carry a risk of various complications.The deep learning-based efficacy prediction system(DLEPS)is a forecasting tool that can effectively compete in drug screening and prediction based on gene expression changes.This study aimed to explore the protective effect and potential mechanisms of cinobufotalin(CB),a traditional Chinese medicine(TCM),on bone loss.Methods:DLEPS was employed for screening anti-osteoporotic agents according to gene profile changes in primary osteoporosis.Micro-CT,histological and morphological analysis were applied for the bone protective detection of CB,and the osteogenic differentiation/function in human bone marrow mesenchymal stem cells(hBMMSCs)were also investigated.The underlying mechanism was verified using qRT-PCR,Western blot(WB),immunofluorescence(IF),etc.Results:A safe concentration(0.25mg/kg in vivo,0.05μM in vitro)of CB could effectively preserve bone mass in estrogen deficiency-induced bone loss and promote osteogenic differentiation/function of hBMMSCs.Both BMPs/SMAD and Wnt/β-catenin signaling pathways participated in CB-induced osteogenic differentiation,further regulating the expression of osteogenesis-associated factors,and ultimately promoting osteogenesis.Conclusion:Our study demonstrated that CB could significantly reverse estrogen deficiency-induced bone loss,further promoting osteogenic differentiation/function of hBMMSCs,with BMPs/SMAD and Wnt/β-catenin signaling pathways involved. 展开更多

Nitrogen(N)is a key factor in the positive response of cereal crops that follow leguminous crops when compared to gramineous crops in rotations,with the nonrecyclable rhizosphere-derived N playing an important role.However,quantitative assessments of differences in the N derived from rhizodeposition(NdfR)between legumes and gramineous crops are lacking,and comparative studies on their contributions to the subsequent cereals are scarce.In this study,we conducted a meta-analysis of NdfR from leguminous and gramineous crops based on 34 observations published worldwide.In addition,pot experiments were conducted to study the differences in the NdfR amounts,distributions and subsequent effects of two major wheat(Triticum aestivum L.)-preceding crops,corn(Zea mays L.)and soybean(Glycine max L.),by the cotton wick-labelling method in the main wheat-producing areas of China.The meta-analysis results showed that the NdfR of legumes was significantly greater by 138.93%compared to gramineous crops.In our pot experiment,the NdfR values from corn and soybean were 502.32 and 944.12 mg/pot,respectively,and soybean was also significantly higher than corn,accounting for 76.91 and 84.15%of the total belowground nitrogen of the plants,respectively.Moreover,in different soil particle sizes,NdfR was mainly enriched in the large macro-aggregates(>2 mm),followed by the small macro-aggregates(2–0.25 mm).The amount and proportion of NdfR in the macro-aggregates(>0.25 mm)of soybean were 3.48 and 1.66 times higher than those of corn,respectively,indicating the high utilization potential of soybean NdfR.Regarding the N accumulation of subsequent wheat,the contribution of soybean NdfR to wheat was approximately 3 times that of corn,accounting for 8.37 and 4.04%of the total N uptake of wheat,respectively.In conclusion,soybean NdfR is superior to corn in terms of the quantity and distribution ratio of soil macro-aggregates.In future field production,legume NdfR should be included in the nitrogen pool that can be absorbed and utilized by subsequent crops,and the role and potential of leguminous plants as nitrogen source providers in crop rotation systems should be fully utilized. 展开更多

Multifunctional Film Assembled from N‑Doped Carbon Nanofiber with Co–N_(4)–O Single Atoms for Highly Efficient Electromagnetic Energy Attenuation

Single-atom materials have demonstrated attractive physicochemical characteristics.However,understanding the relationships between the coordination environment of single atoms and their properties at the atomic level remains a considerable challenge.Herein,a facile waterassisted carbonization approach is developed to fabricate well-defined asymmetrically coordinated Co–N_(4)–O sites on biomass-derived carbon nanofiber(Co–N_(4)–O/NCF)for electromagnetic wave(EMW)absorption.In such nanofiber,one atomically dispersed Co site is coordinated with four N atoms in the graphene basal plane and one oxygen atom in the axial direction.In-depth experimental and theoretical studies reveal that the axial Co–O coordination breaks the charge distribution symmetry in the planar porphyrin-like Co–N_(4) structure,leading to significantly enhanced dielectric polarization loss relevant to the planar Co–N_(4) sites.Importantly,the film based on Co–N_(4)–O/NCF exhibits light weight,flexibility,excellent mechanical properties,great thermal insulating feature,and excellent EMW absorption with a reflection loss of−45.82 dB along with an effective absorption bandwidth of 4.8 GHz.The findings of this work offer insight into the relationships between the single-atom coordination environment and the dielectric performance,and the proposed strategy can be extended toward the engineering of asymmetrically coordinated single atoms for various applications. 展开更多

The path planning of Unmanned Aerial Vehicle(UAV)is a critical issue in emergency communication and rescue operations,especially in adversarial urban environments.Due to the continuity of the flying space,complex building obstacles,and the aircraft's high dynamics,traditional algorithms cannot find the optimal collision-free flying path between the UAV station and the destination.Accordingly,in this paper,we study the fast UAV path planning problem in a 3D urban environment from a source point to a target point and propose a Three-Step Experience Buffer Deep Deterministic Policy Gradient(TSEB-DDPG)algorithm.We first build the 3D model of a complex urban environment with buildings and project the 3D building surface into many 2D geometric shapes.After transformation,we propose the Hierarchical Learning Particle Swarm Optimization(HL-PSO)to obtain the empirical path.Then,to ensure the accuracy of the obtained paths,the empirical path,the collision information and fast transition information are stored in the three experience buffers of the TSEB-DDPG algorithm as dynamic guidance information.The sampling ratio of each buffer is dynamically adapted to the training stages.Moreover,we designed a reward mechanism to improve the convergence speed of the DDPG algorithm for UAV path planning.The proposed TSEB-DDPG algorithm has also been compared to three widely used competitors experimentally,and the results show that the TSEB-DDPG algorithm can archive the fastest convergence speed and the highest accuracy.We also conduct experiments in real scenarios and compare the real path planning obtained by the HL-PSO algorithm,DDPG algorithm,and TSEB-DDPG algorithm.The results show that the TSEBDDPG algorithm can archive almost the best in terms of accuracy,the average time of actual path planning,and the success rate. 展开更多

China is a hotspot of relict plant species that were once widespread throughout the Northern Hemisphere.Recent research has demonstrated that the occurrence of long-term stable refugia in the mountainous regions of central and south-western China allowed their persistence through the late Neogene climate fluctuations.One of these relict lineages is Dipteronia,an oligotypic tree genus with a fossil record extending to the Paleocene.Here,we investigated the genetic variability,demographic dynamics and diversification patterns of the two currently recognized Dipteronia species(Dipteronia sinensis and D.dyeriana).Molecular data were obtained from 45 populations of Dipteronia by genotyping three cpDNA regions,two single copy nuclear genes and 15 simple sequence repeat loci.The genetic study was combined with niche comparison analyses on the environmental space,ecological niche modeling,and landscape connectivity analysis.We found that the two Dipteronia species have highly diverged both in genetic and ecological terms.Despite the incipient speciation processes that can be observed in D.sinensis,the occurrence of long-term stable refugia and,particularly,a dispersal corridor along Daba Shan-west Qinling,likely ensured its genetic and ecological integrity to date.Our study will not only help us to understand how populations of Dipteronia species responded to the tectonic and climatic changes of the Cenozoic,but also provide insight into how Arcto-Tertiary relict plants in East Asia survived,evolved,and diversified. 展开更多

Background Diabetic cardiomyopathy (DCM) causes the myocardium to rely on fatty acid β-oxidation for energy. The accumulation of intracellular lipids and fatty acids in the myocardium usually results in lipotoxicity, which impairs myocardial function. Adipsin may play an important protective role in the pathogenesis of DCM. The aim of this study is to investigate the regulatory effect of Adipsin on DCM lipotoxicity and its molecular mechanism.MethodsA high-fat diet (HFD)-induced type 2 diabetes mellitus model was constructed in mice with adipose tissue-specific overexpression of Adipsin (Adipsin-Tg). Liquid chromatography-tandem mass spectrometry (LC–MS/MS), glutathione-S-transferase (GST) pull-down technique, Co-immunoprecipitation (Co-IP) and immunofluorescence colocalization analyses were used to investigate the molecules which can directly interact with Adipsin. The immunocolloidal gold method was also used to detect the interaction between Adipsin and its downstream modulator.ResultsThe expression of Adipsin was significantly downregulated in the HFD-induced DCM model (P < 0.05). Adipose tissue-specific overexpression of Adipsin significantly improved cardiac function and alleviated cardiac remodeling in DCM (P < 0.05). Adipsin overexpression also alleviated mitochondrial oxidative phosphorylation function in diabetic stress (P < 0.05). LC–MS/MS analysis, GST pull-down technique and Co-IP studies revealed that interleukin-1 receptor-associated kinase-like 2 (Irak2) was a downstream regulator of Adipsin. Immunofluorescence analysis also revealed that Adipsin was co-localized with Irak2 in cardiomyocytes. Immunocolloidal gold electron microscopy and Western blotting analysis indicated that Adipsin inhibited the mitochondrial translocation of Irak2 in DCM, thus dampening the interaction between Irak2 and prohibitin (Phb)-optic atrophy protein 1 (Opa1) on mitochondria and improving the structural integrity and function of mitochondria (P < 0.05). Interestingly, in the presence of Irak2 knockdown, Adipsin overexpression did not further alleviate myocardial mitochondrial destruction and cardiac dysfunction, suggesting a downstream role of Irak2 in Adipsin-induced responses (P < 0.05). Consistent with these findings, overexpression of Adipsin after Irak2 knockdown did not further reduce the accumulation of lipids and their metabolites in the cardiac myocardium, nor did it enhance the oxidation capacity of cardiomyocytes expose to palmitate (PA) (P < 0.05). These results indicated that Irak2 may be a downstream regulator of Adipsin.ConclusionsAdipsin improves fatty acid β-oxidation and alleviates mitochondrial injury in DCM. The mechanism is related to Irak2 interaction and inhibition of Irak2 mitochondrial translocation. 展开更多

To better understand the community patterns mediated by connectivity in artificial reefs of coastal areas, it is necessary to understand the distribution and coexistence of organisms with artificial reefs area and adjacent waters. This study was conducted to examine main catches assemblages collected by trawls in Haizhou Bay,which included five habitats: the artificial reef area(AR), aquaculture area(AA), natural area(NA), estuary area(EA) and comprehensive effect area(CEA). The result shows that the total abundances of species in the five habitats were highly different(univariate PERMANOVA: P = 0.001, n = 24), but some species were also unique in their habitat(e.g. Scapharca subcrenata and Glossaulax didyma in AA). The body size distribution of specific species between habitats are different. For Collichthys lucidus, their body size in AR(14.63 cm ± 1.64 cm) and EA(14.3 cm ± 0.85 cm) is higher than that in NA(10.65 cm ± 1.64 cm), CEA(11.28 cm ± 1.85 cm) and AA(12.1 cm ±0.43 cm), which indicates the potential connection from AR to EA mediated by their adult population. We concluded that artificial reefs in AR can be considered key components that have the ability to support species assemblages in adjacent habitats. This study has implications for the conservation and monitoring of species assemblages in coastal areas in terms of that artificial reefs can be applied in different stages of habitat protection implementation and in different combinations of scenarios. 展开更多

Genomic alterations in oral multiple primary cancers

Oral squamous cell carcinoma (OSCC) is the predominant type of oral cancer, while some patients may develop oral multiple primary cancers (MPCs) with unclear etiology. This study aimed to investigate the clinicopathological characteristics and genomic alterations of oral MPCs. Clinicopathological data from patients with oral single primary carcinoma (SPC, n=202) and oral MPCs (n=34) were collected and compared. Copy number alteration (CNA) analysis was conducted to identify chromosomal-instability differences among oral MPCs, recurrent OSCC cases, and OSCC patients with lymph node metastasis. Whole-exome sequencing was employed to identify potential unique gene mutations in oral MPCs patients. Additionally, CNA and phylogenetic tree analyses were used to gain preliminary insights into the molecular characteristics of different primary tumors within individual patients. Our findings revealed that, in contrast to oral SPC, females predominated the oral MPCs (70.59%), while smoking and alcohol use were not frequent in MPCs.Moreover, long-term survival outcomes were poorer in oral MPCs. From a CNA perspective, no significant differences were observed between oral MPCs patients and those with recurrence and lymph node metastasis. In addition to commonly mutated genes such as CASP8, TP53 and MUC16, in oral MPCs we also detected relatively rare mutations, such as HS3ST6 and RFPL4A. Furthermore, this study also demonstrated that most MPCs patients exhibited similarities in certain genomic regions within individuals, and distinct differences of the similarity degree were observed between synchronous and metachronous oral MPCs. 展开更多

BACKGROUND Intestinal flora disorder(IFD)poses a significant challenge after laparoscopic colonic surgery,and no standard criteria exists for its diagnosis and treatment.AIM To analyze the clinical features and risk factors of IFD.METHODS Patients with colon cancer receiving laparoscopic surgery were included using propensity-score-matching(PSM)methods.Based on the occurrence of IFD,patients were categorized into IFD and non-IFD groups.The clinical characteristics and treatment approaches for patients with IFD were analyzed.Multivariate regression analysis was performed to identify the risk factors of IFD.RESULTS The IFD incidence after laparoscopic surgery was 9.0%(97 of 1073 patients).After PSM,97 and 194 patients were identified in the IFD and non-IFD groups,respectively.The most common symptoms of IFD were diarrhea and abdominal,typically occurring on post-operative days 3 and 4.All patients were managed conservatively,including modulation of the intestinal flora(90.7%),oral/intravenous application of vancomycin(74.2%),and insertion of a gastric/ileus tube for decompression(23.7%).Multivariate regression analysis identified that preoperative intestinal obstruction[odds ratio(OR)=2.79,95%CI:1.04–7.47,P=0.041]and post-operative antibiotics(OR=8.57,95%CI:3.31–23.49,P<0.001)were independent risk factors for IFD,whereas pre-operative parenteral nutrition(OR=0.12,95%CI:0.06–0.26,P<0.001)emerged as a protective factor.CONCLUSION A stepwise approach of probiotics,vancomycin,and decompression could be an alternative treatment for IFD.Special attention is warranted post-operatively for patients with pre-operative obstruction or early use of antibiotics. 展开更多

BACKGROUND Chemoresistance is the primary contributor to distant metastasis in the context of neoadjuvant chemoradiotherapy(nCRT)for rectal cancer.However,the underlying mechanisms remain elusive.AIM To detect the differential expression profiles of plasma exosomal microRNAs(miRNAs)in poor and good responders and explore the potential mechanisms of chemoresistance.METHODS In this study,the profiles of plasma exosomal miRNAs were compared in two dimensions according to treatment responses(poor/good responders)and treatment courses(pre/post-nCRT)using RNA sequencing.RESULTS Exosome hsa-miR-483-5p was up-regulated in good responders post-nCRT.Bioinformatics analysis revealed that the target genes of hsa-miR-483-5p were mainly enriched in tumor-specific pathways,such as the MAPK signaling pathway,EGFR tyrosine kinase inhibitor resistance,Toll-like receptor signaling pathway,VEGF signaling pathway,and mTOR signaling pathway.Further analysis indicated that MAPK3,RAX2,and RNF165 were associated with inferior recurrence-free survival in patients with rectal cancer,and the profiles of MAPK3,TSPYL5,and ZNF-417 were correlated with tumor stage.In addition,the expression profiles of MAPK3,RNF165,and ZNF417 were negatively correlated with inhibitory concentration 50 values.Accordingly,an hsa-miR-483-5p/MAPK3/RNF 165/ZNF417 network was constructed.CONCLUSION This study provides insights into the mechanism of chemoresistance in terms of exosomal miRNAs.However,further research is required within the framework of our established miRNA-mRNA network. 展开更多

Background: Uveal melanoma (UVM) is the most common primary intraocular tumor in adults. However, identification of the effective biomarker for the diagnosis and treatment of UVM remains to be explored. Calcium and integrin-binding protein 1 (CIB1) is emerging as an important factor in tumor progression. Purpose: To determine the contribution of CIB1 in the diagnosis of UVM. Method: Immunohistochemical staining is used to detect the CIB1 expression level, while Gene Expression Profiling Interactive Analysis 2 (GEPIA2) and UALCAN online tools were used to analyze patient survival and CIB1 correlation genes in UVM. Integrative analysis using STRING and GeneMANIA predicted the correlated genes with CIB1 in UVM. Results: CIB1 expression level in UVM was significantly enhanced when compared with that in paracancerous tissues. A higher CIB1 expression level resulted in a significantly worse disease-free survival as well as overall survival. Moreover, the survival probability of patients was associated with body weight and gender of the patients with UVM. The correlated genes with CIB1 in UVM, and the similarity of the genes in UVM expression and survival heatmap were verified. Furthermore, Gene ontology enrichment analysis revealed that CIB1 and its correlated genes are significantly enriched in ITGA2B-ITGB3-CIB1 complex, regulation of intracellular protein transport and regulation of ion transport. Conclusions: Our novel findings suggested that CIB1 might be a potential diagnostic predictor for UVM, and might contribute to the potential strategy for UVM treatment by targeting CIB1. 展开更多

为解决复杂环境下油茶果的检测精度不高的问题,提出一种YOLOv5-CP的油茶果检测方法。首先利用RealSense D435i深度相机在自然场景下采集各种环境下的油茶果图像,使用LabelImg软件进行油茶果的标注;然后引入Cutout数据增强方法和坐标注... 为解决复杂环境下油茶果的检测精度不高的问题,提出一种YOLOv5-CP的油茶果检测方法。首先利用RealSense D435i深度相机在自然场景下采集各种环境下的油茶果图像,使用LabelImg软件进行油茶果的标注;然后引入Cutout数据增强方法和坐标注意力模块(Coordinate Attention),以及提出一种改进的PANet特征提取层对YOLOv5模型进行优化,构建一种新的油茶果检测模型YOLOv5-CP;最后将YOLOv5-CP与现有模型在复杂环境下进行油茶果检测对比试验。试验表明:YOLOv5-CP模型的检测准确率、召回率以及平均精度分别为98%、94.6%以及98.4%,遮挡和重叠环境下对比原YOLOv5模型检测准确率分别提升11.3%和10.8%。本文方法有效提升油茶果检测过程中遮挡、重叠等复杂环境下果实的检测准确率,为后续开发油茶采摘机器人提供理论基础。 展开更多
