Dysfunction of neuronal nitric oxide synthase contributes to neurotoxicity,which triggers cell death in various neuropathological diseases,including epilepsy.Studies have shown that inhibition of neuronal nitric oxide...Dysfunction of neuronal nitric oxide synthase contributes to neurotoxicity,which triggers cell death in various neuropathological diseases,including epilepsy.Studies have shown that inhibition of neuronal nitric oxide synthase activity increases the epilepsy threshold,that is,has an anticonvulsant effect.However,the exact role and potential mechanism of neuronal nitric oxide synthase in seizures are still unclear.In this study,we performed RNA sequencing,functional enrichment analysis,and weighted gene coexpression network analysis of the hippocampus of tremor rats,a rat model of genetic epilepsy.We found damaged hippocampal mitochondria and abnormal succinate dehydrogenase level and Na+-K+-ATPase activity.In addition,we used a pilocarpine-induced N2a cell model to mimic epileptic injury.After application of neuronal nitric oxide synthase inhibitor 7-nitroindazole,changes in malondialdehyde,lactate dehydrogenase and superoxide dismutase,which are associated with oxidative stress,were reversed,and the increase in reactive oxygen species level was reversed by 7-nitroindazole or reactive oxygen species inhibitor N-acetylcysteine.Application of 7-nitroindazole or N-acetylcysteine downregulated the expression of caspase-3 and cytochrome c and reversed the apoptosis of epileptic cells.Furthermore,7-nitroindazole or N-acetylcysteine downregulated the abnormally high expression of NLRP3,gasdermin-D,interleukin-1βand interleukin-18.This indicated that 7-nitroindazole and N-acetylcysteine each reversed epileptic cell death.Taken together,our findings suggest that the neuronal nitric oxide synthase/reactive oxygen species pathway is involved in pyroptosis of epileptic cells,and inhibiting neuronal nitric oxide synthase activity or its induced oxidative stress may play a neuroprotective role in epilepsy.展开更多

METHODS: A total of 53 patients with IEP diagnosed by clinical features and laboratory findings were und...AIM: To study the appearances of acute interstitial edematous pancreatitis (IEP) on non-enhanced MR imaging. METHODS: A total of 53 patients with IEP diagnosed by clinical features and laboratory findings were underwent MR imaging. MR imaging sequences included fast spoiled gradient echo (FSPGR) fat saturation axial T1-weighted imaging, gradient echo T1-weighted (in phase), single shot fast spin echo (SSFSE) T2-weighted, respiratory triggered (R-T) T2-weighted with fat saturation, and MR cholangiopancreatography. Using the MR severity score index, pancreatitis was graded as mild (0-2 points), moderate (3-6 points) and severe (7-10 points). RESULTS: Among the 53 patients, IEP was graded as mild in 37 patients and as moderate in 16 patients. Forty-seven of 53 (89%) patients had at least one abnormality on MR images. Pancreas was hypointense relative to liver on FSPGR T1-weighted images in 18.9% of patients, and hyperintense in 25% and 30% on SSFSE T2-weighted and R-T T2-weighted images, respectively. The prevalences of the findings of IEP on R-T T2-weighted images were, respectively, 85% for pancreatic fascial plane, 77% for left renal fascial plane, 55% for peripancreatic fat stranding, 42% for right renal fascial plane, 45% for perivascular fluid, 40% for thickened pancreatic Iobular septum and 25% for peripancreatic fluid, which were markedly higher than those on in-phase or SSFSE T2-weighted images (P 〈 0.001). CONCLUSION: IEP primarily manifests on non- enhanced MR images as thickened pancreatic fascial plane, left renal fascial plane, peripancreatic fat stranding, and peripancreatic fluid. AIM To retrospectively compare the outcomes of catheter drainage,urokinase and ozone in management of empyema.METHODS Retrospective study included 209 patients(111 males and 98 females; age range 19 to 72 years) who w...AIM To retrospectively compare the outcomes of catheter drainage,urokinase and ozone in management of empyema.METHODS Retrospective study included 209 patients(111 males and 98 females; age range 19 to 72 years) who were diagnosed with empyema.The patients were divided into 3 groups based on the therapy instituted: catheter drainage only(group I); catheter drainage and urokinase(group II); catheter drainage,urokinase and ozone(group III).Drainage was considered successful if empyema was resolved with closure of cavity,clinical symptoms were resolved,and need for any further surgical procedure was avoided.Success rate,length of stay(LOS),need for further surgery and hospital costs were compared between the three groups using the Kruskall-Wallis nonparametric test,with P < 0.05 considered significant.RESULTS Of the 209 patients with empyema,all catheters were placed successfully under CT guidance.Sixty-three patients were treated with catheters alone(group I),64 with catheters and urokinase(group II),and 82 with catheters,urokinase and ozone(group III).Group I,group II and group III had success rates of 62%,83% and 95% respectively(P < 0.05).Group I and groupII had statistically longer LOS(P < 0.05) and higher hospital costs(P < 0.05) compared to group III.There were statistically significant differences between the three groups when comparing patients who converted into further surgery.CONCLUSION The combination of chest tube drainage,urokinase and ozone is a safe and effective therapeutic modality in thoracic empyema.展开更多

Data were calculated with Meta-Disc version 1.4 and Rev Man version 5.2 software. From the pooled data, we calculated sensitivity (Sen), specificity (Spe), positive likelihood ratio (+LR), negative likelihood ratio (-LR), and diagnostic odds ratio (DOR). Summary receiver operating characteristic (SROC) curves were constructed and incidence of complications was recorded.RESULTS: Four documents included in this present meta-analysis met the criteria for analysis. The pooled Sen, Spe, +LR, -LR and DOR with 95%CI were 0.91 (0.86-0.95), 1.0 (0.91-1.0), 18.64 (4.83-71.93), 0.11 (0.05-0.26) and 153.17 (30.78-762.33), respectively. The area under the SROC curve was 0.98. The incidence of pneumothorax and hemoptysis was 17.86%-51.80% and 10.50%-19.40%, respectively.CONCLUSION: CT fuoroscopy-guided PTNB, which has an acceptable incidence of complications, can be used as a primary examination method for lung GGO, with moderate sensitivity and specifcity.展开更多
Formaldehyde(FA),as the simplest endogenous carbonyl molecule,participates in many biosynthesis and metabolism in living organisms,such as nucleotides and adenosine triphosphate(ATP).FA concentrations are sub-millimol...Formaldehyde(FA),as the simplest endogenous carbonyl molecule,participates in many biosynthesis and metabolism in living organisms,such as nucleotides and adenosine triphosphate(ATP).FA concentrations are sub-millimolar in the normal healthy body,but can rise significantly in a number of disease pathologies.As a result,detecting endogenous FA is critical for illness diagnosis and rehabilitation therapy monitoring.Recent studies have focused on the FA-responsive turn-on fluorescence probe,which has huge promise in the detection and visualization of FA in living cells and organisms,as well as exceptional use in disease diagnosis and therapeutic monitoring.This review summarizes the fluorescence luminescence mechanism and design concepts of FA fluorescent probes,as well as their recent applications in bioimaging and illness diagnostics.Additionally,this article indicates the present dilemma of FA-responsive fluorescent probe,including selectivity,specificity,and detection mode,which may provide references for the development of FA-responsive fluorescent probes.展开更多
